Description
米国特許第4775525号明細書(Pera)は、口腔衛生を促進するため、特に歯垢を除去することによって虫歯を減少させるためにアルギン酸ナトリウムを含む製剤および方法に関する。
Hcモデルは、Pashleyら(Greenhill JD,Pashley DH.The effects of desensitising agents on the hydraulic conductance of human dentin in vitro.J.Dent Res 1981;60:686−698)によるオリジナルのデザインに基づく。エッチングした象牙質ディスクをヒトの臼歯から準備し、スプリットチャンバ装置内に歯髄を下にして置く。チャンバの底部は、模倣象牙質流体を含有する流体リザーバーに接続されている単一の入口部を除いて閉じられており、チャンバの上部は空気に対して開口している。模倣象牙質流体を含有する圧力容器の内側の圧力を1p.s.iまで増加させることによって底部の入口部を介して静水圧を象牙質ディスクに適用する。安定化時間の後、象牙質ディスクを通過する流体の流速を測定する。
象牙質透過率を、ベースラインにて、および各剤(水中に分散した3%w/wアルギン酸塩)での処理後、各試料について水力学的コンダクタンスの関数として測定した。各ディスクはその独自のコントロールとしての機能を果たした。処理する前に、各試料の水力学的コンダクタンスを唾液薄膜の適用後に測定した。この値を100%の透過率を表すように設定し、ディスクのベースライン透過率と呼ぶ。シーラントの適用後、水力学的コンダクタンスを評価した(5分にわたって)。この値を使用して、特定のシーラントについての透過率減少の割合を算出した。処理の後、ディスクを模倣した経口チャレンジに供した。順に、これらは(i)10%クエン酸での5秒のリンス、(ii)0.17%KClでリンスしながら30秒のブラシ、および(iii)10秒のパージ圧力の適用(ディスクの歯髄側上に12p.s.i)。各処理群において3個の象牙質ディスクを使用した。
Hc実験から得た結果を表1に詳細に示す。Hcモデルにおける象牙質透過率の減少の初期の割合を図1のグラフに示す。象牙質透過率(水力学的コンダクタンス)の80%以上の減少の達成は望ましい(「良」と記載している)細管閉塞システムとみなす。
Claims (4)
- 象牙質知覚過敏症の治療の有効成分として、1.5:1.0より高いα−L−グルロン酸対β−D−マンヌロン酸の比を有し、2×10 4 〜1.35×10 5 ダルトンの分子量範囲を有し、かつBrookfield粘度計で37℃にて1%w/w水溶液として測定した場合に1〜100cPの範囲の粘度を有するアルギン酸塩を含む、象牙質知覚過敏症の治療に使用するための口腔ケア組成物。
- 前記アルギン酸塩が、前記組成物の0.5%wt〜6%wtの量で存在する、請求項1に記載の口腔ケア組成物。
- 口内洗浄液の形態である、請求項1または2に記載の口腔ケア組成物。
- 請求項1〜3のいずれか一項に記載の口腔ケア組成物を調製する方法であって、都合の良い任意の順序で必要な割合で前記口腔ケア組成物の成分を混合することを含む方法。
