一観点において、本発明は丸葉留紅草抽出物を含有する糖尿病及びその合併症の予防または治療用医薬組成物及び健康機能食品に関する。
本発明で用いられた丸葉留紅草(Quamoclit angulata)は、ナス目ヒルガオ科ツル性一年草である。熱帯アメリカ原産であり、主に観賞用で栽培されており、ツルはアサガオのように生長しながら左に巻き上がる特徴がある。全草の長さは3m前後である。葉は長い葉柄を持ち交互についており、心臓型円形をなす。葉先が尖って、下の部分の両側端が尖った角となる。花は8〜9月に咲き黄色を帯びた紅色であり、長い花茎の端に3〜5個ずつ垂れ下がる。花はアサガオを縮小したような形を取っており、萼・雄蕊は、各々5個ずつであり、雌蕊は1個である。実はさく果で丸く、9月に熟して萼が残る。留紅草と似ているが、葉が分かれない。
また、本発明の丸葉留紅草抽出物は、糖尿病及びその合併症の予防または改善するための目的で食品の形態で製造することができる。従って、本発明は、丸葉留紅草抽出物を有効性分として含有する食品組成物である健康機能食品に関する。
糖尿病と糖尿病合併症に係る研究から糖尿病が酸化的ストレスと密接な関連があることが報告されている。糖尿病において現れる慢性的な高血糖は、ブドウ糖の自動酸化、蛋白質糖化等多様な経路を介して遊離基の生成を増加させて、反応性の高いこれらの物質によって酸化的ストレスが高まる。本発明の丸葉留紅草抽出物は、最終糖化産物(advanced glycation end products,AGEs)の生成を抑制して、異物による酸化的ストレス誘発を減少させて、老化防止及び老化を遅らせる効果が期待される。
従って、本発明は、前記丸葉留紅草抽出物を対象に投与する工程を含む、癌の予防または治療方法に関する。
丸葉留紅草は、済州(チェジュ)南郡安徳面(アンドクミョン)、ソグァンリで採集し、丸葉留紅草51gをアルコール100%で室温で超音波抽出機で2時間毎に15分ずつ2〜3日の間、抽出した。抽出した後、抽出物を真空回転濃縮機で、常温で減圧濃縮し、抽出された残渣を真空凍結乾燥機で乾燥し、乾燥された丸葉留紅草抽出物7gを水に0.075mg/mLになるよう溶解した。
韓国済州(チェジュ)南郡安徳面(アンドクミョン)、ソグァンリで採集した丸葉留紅草50gを細切し、前記細切した丸葉留紅草から水を溶媒として、室温で、超音波抽出機で2時間毎に15分ずつ2〜3日抽出した。前記抽出された抽出液をろ過した後、減圧濃縮し、濃縮液を取得した。前記濃縮液を水に懸濁して、活性炭が充填されたカラムクロマトグラフィーによりエタノール50%を移動相で溶出させて、分離精製し、前記分離精製された丸葉留紅草抽出物を減圧濃縮させた後、前記濃縮された丸葉留紅草抽出物5gに水を可溶して丸葉留紅草抽出物の濃度が20mg/mLになるよう溶解させた。
実施例1と実施例2で製造された丸葉留紅草抽出物で処理されたヒト網膜色素上皮細胞株であるARPE19細胞株で糖尿病性網膜病症等の糖尿病合併症を誘発する血管内皮細胞成長因子(VEGF)の抑制能を確認した。
前記方法で培養されたARPE19細胞からトータルRNAを分離するために、培地を取り除いた後、TRIzol reagentを用いて分離し、配列番号1〜2のプライマーを利用して、RT−PCR(reverse transcriptase-polymerase chain reaction)を行って、VEGF mRNAの発現を確認した。
その結果、図1に示したように、実施例1で製造された丸葉留紅草抽出物を処理した実験群でVEGF mRNA発現が抑制されることを確認した。

前記方法で培養されたARPE19細胞の培養液を取得して、VEGF ELISA kit(R&D,UK)を利用して分泌されるVEGFの量を測定した。
その結果、図2に示したように、実施例1と実施例2で製造された丸葉留紅草抽出物を処理した実験群でVEGF発現が減少することを確認した。
実施例1と実施例2で製造された丸葉留紅草抽出物で処理されたマウスすい臓β細胞株のMin6細胞株でインシュリン分泌促進の有無を確認した。
正常マウスに実施例1で製造された丸葉留紅草抽出物を処理することによって、糖尿病の指標として、体重、血糖、及び尿蛋白の変化を調べた。
正常マウスと実際に糖尿病にかかったマウスに実施例2で製造された丸葉留紅草抽出物を処理することによって、糖尿病の指標の血糖、糖化ヘモグロビン及び尿蛋白の変化を確認した。
糖尿病治療を受ける患者に発生する合併症を減らすためには、血糖値を適切なレベルに維持することが大事である。ある時点において測定する血糖値は、多くの要因によって変動が生じることがあるため、長期間の血糖調節推移を把握する目的で最も広く用いられる検査が糖化ヘモグロビン(HbA1c)である。糖化ヘモグロビンは、赤血球に正常に存在する血色素に糖が結合された形態で、血糖が高く維持された場合に糖化ヘモグロビン数値も高まる。糖化ヘモグロビンは、2〜4ヶ月の間の平均血糖値を反映するため、長期間の血糖調節程度を把握するのに役立つ。
6週齢のマウスと6週齢の糖尿病マウスを購入して、一定の温度(25℃)と湿度(50%)下で1週間適応させた後、肝毒性検査に用いた。

糖尿患者の場合には、白内障が早期に始まり、また急速に悪化して視力が急速に落ちる。糖尿病にかかると、水晶体に不透明現象が促される。そこで、レンズ培養実験を介して、糖尿病性白内障発生抑制効能を調べた。
Claims (5)
- 室温において、水又はアルコールで抽出された丸葉留紅草(Quamoclit angulata)抽出物を含有する糖尿病またはその合併症の予防または治療用医薬組成物。
- 前記合併症は、慢性高血糖症、アテローム性動脈硬化症、微小血管症、腎臓疾患、心臓疾患、糖尿病性網膜症、及びその他一つ以上の眼科疾患からなる群より選択されることを特徴とする請求項1に記載の糖尿病またはその合併症の予防または治療用医薬組成物。
- 室温において、水又はアルコールで抽出された丸葉留紅草抽出物を含有する糖尿病またはその合併症の予防用健康機能食品。
- 前記合併症は、慢性高血糖症、アテローム性動脈硬化症、微小血管症、腎臓疾患、心臓疾患、糖尿病性網膜症、及びその他一つ以上の眼科疾患からなる群より選択されることを特徴とする請求項3に記載の糖尿病及びその合併症の予防用健康機能食品。
