Description

フィブロネクチン

選択的スプライシングとは、異なるmRNAを産生するDNAの一次RNA転写物のスプライシングの異なるパターンの出現を指す。イントロンの切り出し後、選択は、どのエクソンが一緒にスプライシングされてmRNAを形成するかを決定することができる。選択的スプライシングは、異なるエクソンを含む異なるアイソフォームおよび/または異なる数のエクソンの産生を誘導する。例えば、1個のアイソフォームは、1個以上のドメインを含んでもよい、1個以上のエクソンに対応する追加のアミノ酸配列を含んでもよい。
これは、互いに結合する一対の分子の一方のメンバーを記載するものである。結合対のメンバーは、天然由来であっても、または全体的もしくは部分的に合成的に作製されたものであってもよい。前記対の分子の一方のメンバーは、その表面上、または空洞上に、前記対の分子の他方のメンバーの特定の空間構造および極性構造に結合し、従ってそれと相補的である領域を有する。結合対の型の例は、抗原-抗体、ビオチン-アビジン、ホルモン-ホルモン受容体、受容体-リガンド、酵素-基質である。本発明は、抗原-抗体型反応に関する。
これは、天然に産生された、または部分的もしくは全体的に合成的に産生された免疫グロブリンを記載する。この用語はまた、抗体抗原結合部位を含む任意のポリペプチドまたはタンパク質をも包含する。本発明は天然形態の抗体に関するものではない、すなわち、それらはその天然の環境にはないが、それらを天然の起源からの精製により単離もしくは取得するか、または他に遺伝子組換え、もしくは化学的合成により取得することができたものであること、ならびにその後、それらが、後に記載するように非天然アミノ酸を含んでもよいことを理解しなければならない。抗体抗原結合部位を含む抗体フラグメントとしては、限定されるものではないが、Fab、Fab'、Fab'-SH、scFv、Fv、dAb、Fdなどの抗体分子;ならびにダイアボディが挙げられる。
これは標的抗原の全部または一部に結合し、かつこれと相補的である分子の部分を記載する。抗体分子中では、これを抗体抗原結合部位と呼び、標的抗原の全部または一部に結合し、かつこれと相補的である抗体の部分を含む。抗原が大きい場合、抗体は該抗原の特定の部分にのみ結合することができ、その部分をエピトープと呼ぶ。抗体抗原結合部位を、1個以上の抗体可変ドメインにより提供することができる。抗体抗原結合部位は、抗体軽鎖可変領域(VL)と抗体重鎖可変領域(VH)を含んでもよい。
これは、本発明の結合メンバーまたはそのような結合メンバーをコードする核酸が、一般的には本発明に従うであろう状態を指す。かくして、本発明の結合メンバー、VHおよび/またはVLドメインを、例えば、その天然の環境から、実質的に純粋な、もしくは均質な形態で、または核酸の場合、必要な機能を有するポリペプチドをコードする配列以外の起源の核酸もしくは遺伝子を含まないか、もしくは実質的に含まない状態で、単離および/もしくは精製して提供することができる。単離されたメンバーおよび単離された核酸は、それらがその天然の環境中に、または調製がin vitroもしくはin vivoで実行される組換えDNA技術による場合、それらが調製される環境(例えば、細胞培養物)中に認められる他のポリペプチドもしくは核酸などの、それらが天然に結合する材料を含まないか、または実質的に含まないであろう。メンバーおよび核酸を、希釈剤もしくはアジュバントと共に製剤化し、さらなる実用的な目的のために、単離することができる。例えば、前記メンバーを、イムノアッセイにおける使用のためにマイクロタイタープレートをコーティングするのに用いる場合、通常はゼラチンもしくは他の担体と混合するか、または診断もしくは治療において用いる場合、製薬上許容し得る担体もしくは希釈剤と混合することができる。結合メンバーを、天然に、もしくは異種真核細胞の系(例えば、CHOもしくはNS0(ECACC 85110503)細胞)によりグリコシル化するか、またはそれらを非グリコシル化(例えば、原核細胞中での発現により製造する場合)することができる。

を含み、ヒトまたは動物中での原発腫瘍により現在もしくは以前に占有された部位から遠くの部位への該結合メンバーの局在化が、腫瘍転移の存在を示す、前記方法も提供する。前記結合メンバーを検出可能な標識で標識する場合、検出可能な標識の存在または非存在を、該標識を検出することにより決定することができる。
実験
材料および方法
動物モデル
動物実験は、Swiss Federal Veterinary Officeにより認可されたものであり、Swiss Animal Protection Ordinanceに従って実施した。マウスを規則的にモニターした。疼痛もしくは苦痛の徴候を示す場合、または体重減少が15%を超えた場合、動物を安楽死させた。雄のSv129マウス(RCC, Fullingsdorf, Switzerland)は、Dario Rusciano (SIFI, Catania, Italy)の厚意により提供された約106個の突然変異F9マウス奇形癌細胞(Terranaら、1987)の静脈内注入を受けた。マウスを、腫瘍細胞注入の3週間後に、in vivoでのビオチン化、標的化実験または免疫組織化学のための器官切除のために用いた。
in vivoでのビオチン化実験を、以前に記載のように実施した(Roesliら、2006, Rybakら、2005)。簡単に述べると、麻酔したマウスの胸部を、胸骨正中切開術を介して切開した。左心室をかん流針を用いて穿刺し、小切片を右心房中で作製して、かん流溶液の流出を可能にした。その直後、全身循環のかん流を、1.5 ml/分の流速、100 mm Hgの圧力で実施した。第1の工程においては、血漿増量剤として10%(w/v)デキストラン-40(Amersham Biosciences, Uppsala, Sweden)を補給した、PBS、pH 7.4中の1 mg/mlのスルホ-NHS-LC-ビオチン(Pierce, Rockford, IL, USA)を含有する15 mlのビオチン化溶液(38℃に予め温める)を用いて、かん流を実行した。その後、ビオチン化反応と競合することができる血液成分は、かん流の最初の数分以内に循環から排除され、様々な組織中の使用し得る一次アミン含有タンパク質(ならびに特定の糖脂質およびリン脂質)を、ビオチンを用いて共有的に改変することができる。未反応のビオチン化試薬を中和するために、in vivoでのビオチン化、次いで、38℃に予め温めた、PBS, pH 7.4中の50 mM Tris、10%(w/v)デキストラン-40を用いる10分間の洗浄工程を行った。ビオチン化試薬を用いるかん流の間(およびクエンチング溶液を用いる以後のかん流の最初の3分間)に、心臓の周囲の領域を、PBS, pH 7.4(38℃)中の50 mM Trisを用いて洗浄して、流出する未反応のビオチン化試薬をクエンチし、器官表面の分子の望ましくない標識を回避した。かん流後、器官および腫瘍を切除し、標本を、器官ホモジェネートの調製のために新鮮に簡易凍結するか、または組織化学的分析のための凍結切片の調製のために、凍結埋込み化合物(Microm, Walldorf, Germany)中に埋込み、液体窒素中のイソペンタン中で凍結した。かん流していないマウスを、プロテオミクス分析のための陰性対照として用いた。
標本を、組織1 mgあたり40μlの溶解バッファー(PBS, pH 7.4中の2%SDS、50 mM Tris、10 mM EDTA、完全Eプロテイナーゼ阻害剤カクテル(Roche Diagnostics, Mnnheim, Germany))に再懸濁し、Ultra-Turrax T8ディスペンサー(IKA-Werke, Staufen, Germany)を用いて均質化した。ホモジェネートを、Vibra-cell (Sonics, New Town, CT, USA)を用いて超音波処理した後、99℃で15分間インキュベートし、15000 x gで20分間遠心分離した。上清を総タンパク質抽出物として用いた。タンパク質濃度を、BCAタンパク質アッセイ試薬キット(Pierce)を用いて決定した。
各サンプルにつき、960μlのストレプトアビジン-セファロース(Amersham Biosciences, Uppsala, Sweden)スラリーをバッファーA(PBS中のNP40 1%、SDS 0.1%)中で3回洗浄し、ペレット化し、15ミリグラムの総タンパク質抽出物と混合した。ビオチン化タンパク質の捕捉を、回転式ミキサー中、RTで2時間進行させた。上清を除去し、樹脂をバッファーAで3回、バッファーB(PBS中のNP40 0.1%、NaCl 1M)で2回、および50 mM重炭酸アンモニウムで1回洗浄した。最後に、樹脂を重炭酸アンモニウムの50 mM溶液400μlに再懸濁し、20μlの配列決定等級改変ブタトリプシン(50 mM重炭酸アンモニウム中の40 ng/μlのストック溶液)を添加した。プロテアーゼ消化を、一定の攪拌下、37℃で一晩実行した。ペプチドを脱塩し、精製し、C18マイクロカラム(ZipTip C18, Millipore, Billerica, MA, USA)を用いて濃縮した。凍結乾燥後、ペプチドを-20℃で保存した。
トリプシンペプチドを、UltiMateナノスケールLC系およびChromeleonソフトウェア(Dionex, Sunnyvale, CA, USA)により制御されるFAMOSマイクロ自動サンプラー(LC Packings, Amsterdam, The Netherlands)を用いる逆相高速液体クロマトグラフィー(RP-HPLC)により分離した。移動相Aは、水中の2%アセトニトリルおよび0.1%トリフルオロ酢酸(TFA)からなり、移動相Bは水中の80%アセトニトリルおよび0.1% TFAであった。流速は300 nl/分であった。1.5 mgの総タンパク質から精製されたビオチン化タンパク質親和性の消化から誘導された凍結乾燥ペプチドを、5μlのバッファーAに溶解し、カラム(内径:75μm、長さ15 cm、C18 PepMap 100を充填、3μm、100Åビーズ;LC Packings)上に載せた。ペプチドを、0〜30%のBの勾配で7分間、30〜80%のBで67分間、80〜100%のBで3分間、および100%のBで5分間溶出させた;カラムを100%のAで20分間平衡化した後、次のサンプルを分析した。溶出する画分を、水中の3 mg/mlのα-シアノ-4-ヒドロキシ桂皮酸、277 pmol/mlのニューロテンシン(内部標準)、0.1%TFA、および70%アセトニトリルの溶液と混合し、オンラインProbot系(Dionex)を用いて192穴MALDI標的プレート上に示させた(depose)。MALDI-マトリックス溶液の流速を1.083μl/分に設定した。かくして、20秒間に回収された各画分は、361 nlのMALDI-マトリックス溶液および100 nlのサンプルを含んでいた。ニューロテンシンの最終濃度は、ウェルあたり100 fmolであった。
MALDI-TOF/TOF質量スペクトル分析を、4700 Proteomics Analyzer (Applied Biosystems, Framingham, MA, USA)を用いて行った。前駆体イオン選択のために、全画分をMS様式で測定した後、MS/MSを実施した。サンプルスポットあたり最大15個の前駆体を、衝突誘起解離によるその後の断片化のために選択した。コンピューター内で消化されたタンパク質のデータベースに対してMSおよびMS/MSデータを一致させるための内部MASCOT (Matrix Science, London, UK)ソフトウェアを用いる、Global Protein Server Workstation (Applied Biosystems)により、スペクトルを処理および分析した。得られたデータを、NCBIホームページ(http://www.ncbi.nlm.nih.gov/)からダウンロードしたマウスデータベースに対してスクリーニングした。MASCOTソフトウェアにより実施されたタンパク質同定を、最良のペプチドイオンに関して95%の信頼区間内で正確なコールであると考えた。
抗ED-B抗体フラグメントscFv(L19)の単離は、以前に記載されている(Piniら、1998)。親抗ED-A抗体を、公開された手順(Giovannoni, Nucleic. Acid Research, 2001, 29(5):E27)を用いてETH-2ライブラリーから単離した。高親和性抗ED-A抗体をもたらす、親抗ED-A抗体の親和性成熟は、以下の節に記載されている。
親抗ED-A抗体(ETH-2由来抗体)を、親和性成熟ライブラリーの構築のための鋳型として用いた。ライブラリーのVH CDR1 (DP47生殖系列)およびVL CDR1 (DPK22生殖系列)中の配列可変性を、VH CDR1の位置31、32および33ならびにVL CDR1の位置31、31aおよび32で無作為突然変異を作製するプロセスにおいて、部分的縮重プライマー5'-CTGGAGCCTGGCGGACCCAGCTCATMNNMNNMNNGCTAAAGGTGAATCCAGA-3'(配列番号17)(VH用)および5'-CCAGGTTTCTGCTGGTACCAGGCTAAMNNMNNMNNGCTAACACTCTGACTGGCCCTGC-3'(配列番号18)(VL用) (全てのオリゴヌクレオチドを、Operon Biotechnologies, Cologne, Germanyから購入した)を用いるPCRにより導入した。VHVL組合せを、鋳型としてゲル精製されたVHおよびVL断片を用いる、プライマーLMB3long (5'-CAGGAAACAGCTATGACCATGATTAC-3')(配列番号19)およびfdseqlong (5'-GACGTTAGTAAATGAATTTTCTGTATGAGG-3') (配列番号20)を用いるPCR集合によりscFv形式で集合させた。集合させたVH-VL断片を、NcoI/NotIで二重消化し、NcoI/NotI消化されたpHEN1ファージミドベクター(Hoogenboomら、1991)中にクローニングした。得られる連結産物を、Vitiら、2000に従ってエレクトロコンピテント大腸菌TG-1細胞中にエレクトロポレーションし、1.5 x 107個の抗体クローンを含有するライブラリーを生じさせ、改善された親和性でED-Aに結合する抗体についてスクリーニングした。
上記の抗体ライブラリーを、BIAcore分析を用いて、親抗ED-A抗体よりも高い親和性でED-Aに結合した抗体についてスクリーニングした。BIAcore分析において用いた抗原(11A12)は、ヒトフィブロネクチンのED-Aドメインを含み、以下のアミノ酸配列(配列番号120)を有する:
抗ED-A抗体を以下のように発現させ、精製した:10 mlの2TY、Amp、1%グルコース中のTG1エレクトロコンピテント前培養物を、抗ED-A抗体の1つのDNAミニプレップ1μlの存在下でエレクトロポレーションした。次いで、前培養物を1:100に希釈し(800 mlの2TY、Amp、0.1%グルコース中の8 ml)、0.4〜0.6のOD600まで増殖させた後、IPTGで一晩誘導した。次の日、細胞を遠心分離し、上清を濾過した(Millipore 0.22μm)。scFvをAタンパク質-セファロースカラム上で精製し、トリエチレンアミンを用いて、カラムからscFvを溶出させた。溶出したscFvを含む画分を、PBS中、4℃で一晩透析した。次いで、scFv画分を、PBSと共にSuperdex 75カラム上に入れ、0.5 ml/分で流出させ、0.25 mlの画分を回収した。モノマー画分をBIAcore分析に用いた。
BIAcore Chipを、HBS-EPバッファーBIACORE、0.01 M Hepes pH 7.4、0.15 M NaCl、3 mM EDTA、0.005%界面活性剤P20(アッセイに用いたのと同じバッファー)を用いて、5μl/分の流速で一晩フラッシュした。抗原(11A12)を、酢酸バッファー(pH 4.0)中に50μg/mlの濃度に希釈し、チップ上のCOOH基を、N-ヒドロキシスクシンイミド(NHS)およびエチル-N-(ジメチルアミノプロピル)-カルボジイミド(EDC)の混合物50μlの注入により活性化した。40μlの11A12抗原をチップ上に注入し、残留した遊離COOH基を30μlのエタノールアミンで阻害した。0.22μmの濾過後、20μlの各細菌上清をチップ上に注入し、抗原との相互作用をリアルタイムでモニターした。
親抗ED-A抗体および抗ED-A抗体B2、C5、D5、C8、F8、B7およびG9のkon、koffおよびKDを、表面プラスモン共鳴を用いて評価した。チップを、5μl/分のバッファー流速で、アッセイの間に用いたのと同じバッファーを用いて一晩平衡化した。コーティング手順全体をこの流速で実施した。抗原11A12を、酢酸バッファーpH 4.00(BIACOREにより提供)で1:25に希釈して、20μg/mlの最終濃度を得た。次いで、NHSおよびEDCを混合し、50μlを注入して、CM5チップ上のCOOH基を活性化した。次いで、40μlの抗原を注入した(これは約40分続く)。次いで、30μlのエタノールアミンを注入して、最終的な遊離COOHの反応性を阻害した。
in vivoでのビオチン化の成功を検証するために、ビオチン化構造の染色をRybakら、2005に記載のように実施した。切片(10μm)を新鮮に凍結された標本から切断し、アセトンで固定し、ストレプトアビジン:ビオチン化アルカリホスファターゼ複合体(Biospa, Milano, Italy)およびFast-Red TR (Sigma)[内因性アルカリホスファターゼを阻害するために1 mMのLevamisoleの存在下で]と共に連続的にインキュベートし、ヘマトキシリン溶液(Sigma)で対抗染色した。
親抗ED-A抗体scFvを、提供者のプロトコルに従って、市販の赤外フルオロフォア誘導体Alexa Fluor 750カルボン酸スクシニジルエステル(Invitrogen)で標識した。標識された抗体を、PD-10カラム(GE Healthcare)を用いるゲル濾過により未反応の色素から分離した。Invitrogenの標識プロトコルに従って見積もられた標識の程度は、抗体分子あたり5色素分子であった。Alexa 750標識された親抗ED-A scFv抗体(0.3 mg/mlの最終濃度)を、F9DR腫瘍細胞の注入の3週間後にSv190マウスの尾静脈中に注入した(200μl/マウス、すなわち、60μgの抗体/マウス)。マウスの器官を、標識された抗体の注入の6時間後に切除し、タングステンハロゲンランプ、Alexa 750に特異的な励起および放射フィルター、ならびにモノクロCCDカメラを備えた自作の赤外蛍光画像化装置(Birchlerら、1999)を用いて画像化した。
F8ダイアボディは、例えば、scFv形式で用いられるように、抗ED-A抗体F8と同じVHおよびVLドメインを含む。F8ダイアボディおよび抗ED-A scFv F8は、VHおよびVLドメインの間に異なるリンカー配列を有する。F8ダイアボディリンカーのアミノ酸配列は、GSSGG(配列番号28)(ヌクレオチド配列:gggtccagtggcggt(配列番号29))である。従って、F8ダイアボディリンカー配列は、5アミノ酸長であるが、抗ED-A scFv F8においては、リンカーは20アミノ酸長である(配列番号11を参照)。VLおよびVHドメインの間のリンカーの長さの減少は、VLおよびVHドメインの分子内対形成よりも分子間対形成の方が好ましいことを意味している。結果として、1個のF8ポリペプチドのVLドメインは、それが同じF8ポリペプチドのVHドメインと対形成するよりも、別のF8ポリペプチドのVHドメインと対形成する可能性が高い。
示差的に発現されたタンパク質およびスプライス変異体の同定
肝臓およびF9肝臓転移におけるアクセス可能なタンパク質の比較分析に用いたかん流に基づく化学的プロテオミクス方法(Terranaら、1987)を、図1Aに示す。これらの腫瘍は、マウス肝臓の表面および内側に大きい転移巣を形成する(図1B)。末端麻酔下で、腫瘍担持マウスを、血漿増量剤として10%デキストラン-40を補給した、PBS, pH 7.4中のスルホスクシンイミジル-6-[ビオチン-アミド]ヘキサノン酸(1 mg/ml)の1.8 mM溶液15 mlを用いてかん流した。典型的には、この手順を10分間継続し、血管の内腔および外側面の両方上での、主要な循環器の全てからの血管の除去およびアクセス可能なタンパク質の選択的ビオチン化が可能になった。実際には、ストレプトアビジン-アルカリホスファターゼコンジュゲートを用いる組織化学的染色により確認されるように、F9肝臓転移の全ての血管が、この手順で効率的かつ選択的に標識された(図1C)。正常な肝臓においては、血管は強く染色されたが、肝臓の生理学的濾過機能と適合して、いくつかの類洞の標識も検出された(図1C)。in vivoでのビオチン化を、一次アミンを含む溶液を用いるかん流によりクエンチした。続いて、肝臓転移の標本を肝臓から切除し、ホモジェナイズし、ストレプトアビジン樹脂上での親和性クロマトグラフィーによる強い界面活性剤SDSの存在下でのビオチン化タンパク質の回収に用いた(図1A)。宿主の肝臓中での転移性タンパク質の拡散の危険性を最小化するために、in vivoでビオチン化された健康なマウスに由来する肝臓を、正常な肝臓の血管の研究に用いた。また、F9腫瘍マウスに由来する宿主の肝臓の使用は、残存する健康な組織が少ないため、および微小転移巣の不在を巨視的に排除するのが困難であるため、問題が多かった。総合すれば、7匹のin vivoでビオチン化された健康なマウスおよび9匹のin vivoでビオチン化されたF9腫瘍担持マウスに由来するサンプルを、プロテオミクス分析に用いた。さらに、2匹の健康なマウスおよび3匹の転移を担持する非ビオチン化マウスに由来する標本を、陰性対照として用いた。ストリンジェントな洗浄手順ならびにF9転移および正常肝臓に由来するストレプトアビジン捕捉タンパク質の樹脂上でのトリプシン消化(平行して処理)によりペプチドの集合が得られ、ナノ-HPLCおよびMALDI-TOF/TOF質量スペクトル分析手順を用いて、これを分離、同定および比較することができる(Roesliら、2006)。
肝臓構造と転移性血管新生との間の最も顕著な区別が、フィブロネクチンのED-AおよびED-Bドメインについて観察された。両方の場合において、転移血管の強く特異的な染色が観察されたが、正常肝臓および実質的に全ての正常器官(増殖期の子宮内膜および卵巣のいくらかの血管については例外)はこの免疫組織化学分析において陰性のスコアであった(図3A)。重要なことに、ED-Aはヒト肺転移および肝臓転移の血管新生において強く発現されることもわかった(図4)。
リガンドに基づく転移の血管標的化のための標的としてのED-Aの有用性を試験するために、近赤外線蛍光画像化を用いるin vivo標的化実験を実施した。親抗ED-A scFv抗体を、Alexa Fluor 750で標識し、F9転移を担持するマウスに静脈内注入した。切除された器官の近赤外線蛍光画像化により、転移病変における標的化剤の顕著な蓄積が示された(図3B)。
BIAcore分析1
BIAcore分析は、以下のように抗原に対する抗体の親和性を推定するために分析されたそれぞれの抗ED-A抗体に関するグラフをもたらした:それぞれのグラフのx軸は時間に対応し、y軸は共鳴単位(BIAcoreチップ上にコーティングされた抗原に対する試験された抗体の結合親和性を示す尺度)に対応する。それぞれのグラフは、3個のピークおよびバッファーの交換に対応し、従って、結果の解釈について無関係である1個の落ち込みを示す。
各抗ED-A抗体のkon、koffおよびKD値を、BIAevaluationソフトウェアを用いて評価した。抗原11A12に対する親抗ED-A抗体および抗ED-A抗体B2、C5、D5、C8、F8、B7およびG9のkon、koffおよびKD値を、表3に詳細に記載する。抗ED-A抗体B2、C5、D5、C8、F8、B7およびG9は全て、それらが誘導された親抗ED-A抗体よりも良好な抗原11A12のKD値を有し、これは、それらが親抗ED-A抗体よりも高い親和性でED-Aに結合し、従ってA-FNにも結合することを示している。結果として、抗ED-A抗体B2、C5、D5、C8、F8、B7およびG9は、本明細書の他の場所に記載された親抗ED-A抗体を用いて行われた同じin vivoおよび免疫組織化学試験において用いられた場合、同じか、またはより良好な結果を生じる可能性が非常に高い。従って、親抗ED-A抗体を用いて得られたin vivoおよび免疫組織化学データは、抗ED-A抗体B2、C5、D5、C8、F8、B7およびG9を腫瘍転移の治療に用いることができるという証拠を提供する。
マウス組織1グラム(g)あたり検出されたI125標識(ヨウ素化)F8ダイアボディの注入用量(ID)の割合(%)は、肝臓、肺、脾臓、心臓、腎臓、腸、尾および血管について非常に類似しており、また、腎臓を除いて、全て2% ID/g未満を示した(図8)。対照的に、F9腫瘍は、分析した他のマウス組織のいずれかよりも、平均で約4倍以上のIDを含んでいた(図8)。これは、F8ダイアボディがF9マウス腫瘍に選択的に標的化されることを証明している。他の組織において検出されるIDの割合は、マウスに存在するF8ダイアボディのバックグラウンド負荷または他のマウス組織の非特異的標識を示す可能性が最も高い。本明細書の他の場所に記載のように、4匹のマウスを用いて生体内分布実験を行ったが、F9腫瘍において検出されるF8ダイアボディの割合を変化させたマウス組織あたり検出されるIDの割合(図8の誤差バーを参照されたい)は、いずれの試験した他のマウス組織におけるよりも一貫して高かった。
抗ED-A抗体H1、B2、C5、D5、E5、C8、F8、F1、B7、E8およびG9は全てscFv抗体であり、従来の方法を用いてこれらを配列決定した。抗ED-A抗体H1のヌクレオチド配列を図6に示す。抗ED-A抗体H1のアミノ酸配列を図7に示す。

Claims (21)
- フィブロネクチンのエキストラドメイン-A (ED-A)アイソフォームに結合する抗体分子またはその抗原結合部位であって、
(i) 配列番号81に示されるアミノ酸配列を含むVHドメイン、ここで配列番号81に示されるアミノ酸配列の第5位のアミノ酸がバリン残基(V)ではなくロイシン残基(L)である、および
(ii) 配列番号82に示されるアミノ酸配列の第1-108位のアミノ酸を含むVLドメイン、ここで配列番号82に示されるアミノ酸配列の第18位のアミノ酸がリシン残基(K)の代わりにアルギニン残基(R)である、
を含む前記抗体分子またはその抗原結合部位。 - 抗体分子またはその抗原結合部位が一本鎖Fvを含むか又はダイアボディである、請求項1に記載の抗体分子またはその抗原結合部位。
- VHドメインがペプチドリンカーを介してVLドメインにコンジュゲートされている、請求項1または2に記載の抗体分子またはその抗原結合部位。
- 前記ペプチドリンカーが5〜25アミノ酸を含む、請求項3に記載の抗体分子またはその抗原結合部位。
- 前記ペプチドリンカーが5アミノ酸を含む、請求項4に記載の抗体分子またはその抗原結合部位。
- 抗体分子またはその抗原結合部位が検出可能な標識にコンジュゲートされている、請求項1〜5のいずれか1項に記載の抗体分子またはその抗原結合部位。
- 抗体分子またはその抗原結合部位が、殺菌活性若しくは細胞傷害活性を有する分子、又は放射性アイソトープにコンジュゲートしている、請求項1〜5のいずれか1項に記載の抗体分子またはその抗原結合部位。
- ペプチドリンカーを介して殺菌活性若しくは細胞傷害活性を有する分子にコンジュゲートされている、請求項7に記載の抗体分子またはその抗原結合部位。
- 抗体分子またはその抗原結合部位が、殺菌活性若しくは細胞傷害活性を有する分子および該抗体分子またはその抗原結合部位を含んでなる融合タンパク質の形態である、請求項7または8に記載の抗体分子またはその抗原結合部位。
- 殺菌活性若しくは細胞傷害活性を有する分子が、サイトカインである、請求項7に記載の抗体分子またはその抗原結合部位。
- 抗体分子またはその抗原結合部位が、ペプチドリンカーを介してサイトカインにコンジュゲートしている、請求項10に記載の抗体分子またはその抗原結合部位。
- 抗体分子またはその抗原結合部位が、サイトカイン及び抗体分子またはその抗原結合部位を含む融合タンパク質の形態で存在する、請求項10または11に記載の抗体分子またはその抗原結合部位。
- 抗体分子またはその抗原結合部位が毒素、標的化部位又は酵素にコンジュゲートしている、請求項1〜5のいずれか1項に記載の抗体分子またはその抗原結合部位。
- 毒素、標的化部位又は酵素がペプチドリンカーを介して抗体分子またはその抗原結合部位にコンジュゲートしている、請求項13に記載の抗体分子またはその抗原結合部位。
- 以下の(i)および(ii)を含む抗体コンジュゲート、
(i) VHドメイン及びVLドメインを含み、かつ、フィブロネクチンのエキストラドメイン-A (ED-A)アイソフォームに結合する抗体またはその抗原結合部位、
(a)ここで前記VHドメインは配列番号81に示されるアミノ酸配列を含み、ただし配列番号81に示されるアミノ酸配列の第5位のアミノ酸がバリン残基(V)の代わりにロイシン残基(L)である、かつ
(b)ここで前記VLドメインは配列番号82に示されるアミノ酸配列の第1-108位のアミノ酸を含み、ただし配列番号82に示されるアミノ酸配列の第18位のアミノ酸がリシン残基(K)の代わりにアルギニン残基(R)である、
ここで前記VHドメインは5アミノ酸のペプチドリンカーを介して前記VLドメインにコンジュゲートされている、ならびに
(ii)殺菌活性若しくは細胞傷害活性を有するサイトカイン。 - 抗体がダイアボディーである、請求項15に記載の抗体コンジュゲート。
- 請求項7〜12のいずれか1項に記載の抗体分子もしくはその抗原結合部位、または請求項15もしくは16に記載の抗体コンジュゲートを、製薬上許容しうる担体中に含む、医薬組成物。
- 請求項1〜5および10〜12のいずれか1項に記載の抗体分子もしくはその抗原結合部位または請求項15もしくは16に記載の抗体コンジュゲートをコードする、単離された核酸。
- 請求項18に記載の核酸を含む宿主細胞。
- 請求項19に記載の宿主細胞を、抗体分子もしくはその抗原結合部位または抗体コンジュゲートを産生する条件下で培養することを含む、請求項1〜5および10〜12のいずれか1項に記載の抗体分子もしくはその抗原結合部位、または請求項15もしくは16に記載の抗体コンジュゲートを製造する方法。
- 前記抗体分子もしくはその抗原結合部位または抗体コンジュゲートを単離する工程をさらに含む、請求項20に記載の方法。
