Description
α-受容体アゴニストであるドブタミン(dobutamine)の左旋性異性体は、β−受容体に若干影響を及ぼす。同様に、β受容体アゴニストであるドブタミンの右旋性異性体はα受容体に若干影響を及ぼす。ラセミ化合物を投与すれば、心拍数や血圧を上げることなく、心筋収縮能を増加させる効果が得られる。
プロメタジンは、一つのキラル分子を有する抗ヒスタミンである。この受容体は医薬の鏡像体に対し選択性を有しないため、2つの鏡像体は同じ薬理作用と、同じ強度の薬理活性を有する。
β‐受容体としてプロプラノロールは、その左旋性異性体に依存してアンタゴニスト活性を示す。それは、左旋性異性体がβ‐受容体と同じ配置(configuration)を有し、β‐受容体に選択的に結合することができるが、その右旋性異性体はそうでないからである。
ニフェジピン構造類似体であるBay k8644に関し、その右旋性異性体は、カルシウムアンタゴニストであるが、その左旋性異性体はカルシウムアゴニストである。これらの鏡像体は、正反対の活性を有する。
プロポキシフェンのD‐鏡像体はL‐鏡像体の活性の6倍の強力な鎮痛効果を有するが、鎮咳活性は有しない。逆に、プロポキシフェンのL−鏡像体は、強力な鎮咳効果を有する。文献(Medicinal Chemistry, edited by Wensheng Ji, Anliang Li, Higher Education Press: 25-26)参照。

(8)ピノセンブリンのラセミ化合物は、急性虚血性脳卒中によって引き起こされた損傷から神経細胞を保護することができる。
1.テスト薬
注射用ピノセンブリンラセミ化合物、注射用(R)−ピノセンブリン、および、注射用(S)−ピノセンブリンは、New Drug Development Laboratory of Drug Research Institute of the Chinese Academy of Medical Sciencesから入手し(バッチ番号:20050601、含量:2.36%)、CN200810084682.3に記載した方法、即ち、ピノセンブリンラセミ化合物又はピノセンブリン鏡像体とシクロデキストリン又はその誘導体との包接錯体を形成し、使用の際にその包接錯体を生理食塩水に溶かすことで製造した。バイエル社(ドイツ)から陽性対照群としてのニモジピン(Nimodipine)を購入した。
実験動物:110匹のSHRSPラットおよび10匹の正常なウィスターラット(6週齢)
実験グループ分け:ウィスターラットを正常グループに、SHRSPラットをモデルグループ、ニモジピングループ(3mg/kg)、ピノセンブリンラセミ化合物の低、中、高容量グループ(それぞれ3、10、および、30mg/kg/d)、(R)―ピノセンブリンの低、中、高容量グループ(その容量は、それぞれピノセンブリンラセミ化合物の容量と一致する)、および、(S)―ピノセンブリンの低、中、高容量グループ(その容量は、それぞれピノセンブリンラセミ化合物の容量と一致する)に分けた。
正常グループのマウスを除く、すべてのSHRAPラットには、1%塩化ナトリウム溶液を毎日飲ませた。ニモジピングループのラット(3mg/kg)、ピノセンブリンンラセミ化合物の低、中、高容量のグループ(それぞれ3,10、および30mg/kg/d)、(R)−ピノセンブリンンの低、中、高容量のグループ(それぞれ3,10、および30mg/kg/d)、および、(S)−ピノセンブリンンの低、中、高容量のグループ(それぞれ3,10、および30mg/kg/d)には所定量の薬を投与した。モデルグループには同量の生理食塩水を投与した。投与は、脳卒中の発症後14日間続いた。
(1)一方の肢、両方の前肢、又は、全身におけるけいれんの現れ
(2)片麻痺(hemiplegia)または進行麻痺(general paralysis)の現れ、腹部を地面に当ててからでないと立ち上がれない
(3)より深刻には、全身の微小振戦(fine tremor)、はう(groveling)、弛緩性麻痺(flaccid paralysis)が現れ、ひいては死に至る。上記症状のほかに、一部の動物では過敏(ジャンプしたり、飛び跳ねたりする)が見られた。上記いずれかの症状が現れたら、脳卒中の発症と判断し(決定し)、各動物における発症時刻を記録した。
(1)最後に投与した後、ラットを6時間禁食させ、眼底における血液サンプリングによって血液サンプルを収集した。ラットを致死させて、使用のために製氷皿にラットの全脳を分離した。
(2)0.074%ヘパリン1mlを含んだキュベットに血液3mlを迅速に滴下して、混合した。血液レオロジー検出器を用いて、全血粘度、血漿粘度、赤血球(RBC)凝集指数、および、RBC変形指数を決定した。
(3)0.13mol/Lのクエン酸ナトリウム0.2mLを含んだキュベットに血液2mLを迅速に滴下した。その混合物を1000rpmにて10分間遠心分離にかけて、 血小板凝集メーターを用いて、血小板凝集を決定した。
(4)1mlの血液を、0.109mol/Lのクエン酸ナトリウム0.1mlを含んだキュベットに迅速に滴下して、混合した。その混合物を3000rpmにて15分間遠心分離にかけ、 ELISAによって上澄み中血漿フィブリノゲンを決定した。
(5)7.5%EDTA−2Na30μLおよびアプロチニン(aprotinin)40μLを含んだキュベットに血液2mlを迅速に滴下して、混合した。3000rpmにて15分間遠心分離にかけた後、 放射免疫測定によって上澄み中血漿ET−1を決定した。
(6)全脳組織の重量を一つずつ量り、110±5℃にて48時間真空下で乾燥させ、再度量った。
(1)身体の外観、精神状態、応答性、運動の姿勢、自発活動、水や食料の量、および、体重等を含めてモデリング後の各グループの動物の身体状態(body status)を観察する
(2)血液レオロジー:全血の粘度、血漿粘度、赤血球凝集指数、赤血球変形指数、血小板凝集率などを含む。
(3)血漿フィブリノゲン
(4)血漿ET-1
(5)脳の水分含有量(脳水分含量):真空オーブンに入れ前後の全脳組織の重量に基づいて、そのデータは次の式に当てはめて計算した:脳の水分含有量=(湿重量−乾重量)/湿重量×100%
結果は、平均±標準偏差として表し、データ間の比較は一元配置分散分析(one-way analysis of variance (ANOVA))によって分析・評価された。
(1)ラットの死亡
総24匹のラットは、実験中に死に至った。病理解剖の解析の結果、24匹のうち4匹は出血性脳卒中で死亡し、その他の20匹は虚血性脳卒中で死亡したことが分かった。詳細については、表2を参照されたい。
正常グループのラットは、身体の状態、摂食、及び、運動の観点から正常であった。第4週目の初めのころから、高、中容量のピノセンブリンラセミ化合物及びニフェジピングループを除く他のグループのラットには、過敏(irritation)、発作性痙攣、麻痺、重い頭、食欲不振、尿失禁、毛幹、痩せといった症状を特徴とする脳卒中が絶え間なく現れた。前述した脳卒中発症後の神経学的症状に関するスコアの基準に基づけば、脳卒中を発症した日から、高及び中容量のピノセンブリンラセミ化合物グループのスコアは著しく減少された。発症から2〜14日内に、そのスコアは、それぞれお1.6及び2.3未満と安定していた。これは、高、中容量のピノセンブリンラセミ化合物は神経学的機能に有意な改善効果を有していることを示唆する。低容量のピノセンブリンラセミ化合物グループ及び高容量の(R)−ピノセンブリングループのスコアは、当初数日間は比較的に高かったが、前述した14日間で2.5以上に維持された。これは、低容量のピノセンブリンラセミ化合物、及び、高容量の(R)−ピノセンブリンが改善効果を有しないことを示唆する。脳卒中の発症後、ニモジピングループのスコアは、比較的に低く、6日後2.2を下回る水準で安定していた。
(3)血液レオロジーの分析
表4に示した血小板凝集に対する抵抗性(resistance)の結果から、モデルグループの血小板凝集率が正常グループの血小板凝集率に比べて高いことが分かる(P<0.01)。モデルグループに比べて、高、中容量のピノセンブリンラセミ化合物及び高容量の(R)−ピノセンブリンは血小板凝集率に有意な減少を示したが(P<0.01、P<0.05)、(S)−ピノセンブリンは全く活性を示さなかった。
そこで、ピノセンブリンラセミ化合物は容量依存的に血小板凝集を妨げるが、(R)−ピノセンブリンは高容量で同様の効果を有するといえる。
モデルグループにおける血漿フィブリノゲンの含量は正常グループに比べて高かった(P<0.01)。モデルグループに比べて、高、中、低容量のピノセンブリンラセミ化合物グループ及び高容量の(R)−ピノセンブリングループでは血漿フィブリノゲンの含量に著しい減少が見られた(P<0.01)。それによって、凝固性亢進(hypercoagulability)状態が変わったが、(S)−ピノセンブリンは全く活性を示さなかった。結果の詳細については表4を参照されたい。
表4に示したように、モデルグループでは血漿ET-1が急速に増加し、正常グループと比較すれば有意な差が見られた(P<0.01)。モデルグループに比べて、高、中、低容量のピノセンブリンラセミ化合物グループ及び高容量の(R)−ピノセンブリングループにおける血漿ET-1の含量は有意に減少した(P<0.01、P<0.05)。別の容量のピノセンブリンラセミ化合物や(R)−ピノセンブリン、及び(S)−ピノセンブリンは全く活性を示さなかった。
表4に示したように、モデルグループにおける脳水分量は著しく増加し、正常グループとは有意な差を示した(P<0.05)。モデルグループに比べて、高容量のピノセンブリンラセミ化合物グループにおける脳水分量は有意に減少した(P<0.05)。その他のグループにおいては、注目すべき活性は見られなかった。
モデルグループに比べて、ピノセンブリンラセミ化合物は、容量依存的に脳卒中の発症時期を遅らせることができる(P<0.01)。低容量のピノセンブリンラセミ化合物及び高容量の(R)−ピノセンブリンも、脳卒中の発症時期を遅らせることができる(P<0.05)。ニモジピンは、脳卒中の発症時期を遅延することができる(P<0.01)。ピノセンブリンンが脳卒中の予防効果を有し、かつ、ピノセンブリンラセミ化合物のほうが(R)−ピノセンブリンよりその効果に優れ、そして、(S)−ピノセンブリンは全く効果を示さないといえる。結果の詳細については表2を参照されたい。
1.テスト薬:注射用ピノセンブリンラセミ化合物、注射用(R)−ピノセンブリン、および、注射用(S)−ピノセンブリンは、New Drug Development Laboratory of Drug Research Institute of the Chinese Academy of Medical Sciencesから入手し(バッチ番号:20050601、含量:2.36%)、CN200810084682.3に記載した方法、即ち、ピノセンブリンラセミ化合物又はピノセンブリン鏡像体とシクロデキストリン又はその誘導体との包接錯体を形成し、使用の際にその包接錯体を生理食塩水に溶かすことで製造した。バイエル社(ドイツ)から陽性対照群としてのニモジピンを購入した。
1.神経行動学的検査(ベッダーソンスコア)
神経行動学的検査は、動物を致死させる前に行った。ラットは、2つの前肢の状態を観察するために約1カイ(1χ= 1/3メートル)だけ地面から持ち上げられた。ラットは地面に置かれ、その抵抗の違いを観察するためにラットを押した。ラットは地面に置かれ、その歩行を観察された。4段階スコア法(0〜5スコア)を採択した。スコアが高ければ高いほど、その神経行動の面からより深刻な損傷を受けていることを意味する。
(1)スコア0:正常、異常なし
(2)スコア1:尾を持って動物を持ち上げたとき,前肢(手術対側)が屈曲する
(3)スコア2:地面におかれたラットを手で押圧することで両側の抵抗を調べたとき、手術対側の抵抗が弱い
(4)スコア3:地面におかれたラットの歩行を観察したとき、手術対側を中心に回転行動あり
(5)スコア4:ラットに損傷が大きく、歩けない
神経行動額的なスコアを求めたら、そのラットを致死させ、その脳組織を速やかに回収して、−20℃の冷蔵庫に入れた。10分後、その脳組織を室温雰囲気に移した。嗅球(olfactory bulb)、小脳、および、下位脳幹を切除した後、脳組織を、2mmの間隔で5個の連続的な冠状スライスに切断し、第一に脳前極(brain anterior pole)と視神経交叉(optic decussation)の接続中間部を、第二に視神経交叉を、第三に漏斗ハンドル(funnel handle)を、第四に漏斗ハンドルと尾状核の厚い葉(thick leaves caudate nucleus)の中間部を切った。その後、脳スライスを迅速に4%TTC及び0.1mlの1mo1・L-1K2HPO4を含む5ミリリットル溶液に加えて、37℃の一定温度にて30分間インキュベートした(暗闇の状態)。その間、脳スライスを5分ごとにひっくり返した。TTC染色後、梗塞組織が染色されず白く示されたのに対し、正常組織は、バラのような赤色で示された。脳スライスのすべてのグループを順番に入れて撮影した。写真は、画像解析システムによって処理され、各スライスにおける梗塞領域が計算された。全スライスの梗塞体積は、スライス(2ミリメートル)の厚さによって梗塞面積を乗じて計算し、梗塞体積は、すべてのスライスの梗塞体積の和であった。梗塞体積は、脳浮腫の影響を排除するように半球の割合(%)で示した。
定位装置の上にラットをうつ伏せに固定し、開頭を行った。手術視野のために洗浄した後、原点(起点)としてブレグマ(bregma)を用いて、ブレグマから後方に2mm、右方向に3mmの部位を測定点として選択した。その部位を取り巻く領域2〜3cmをデンタル・ドリルを用いて薄くした。その過程において、軽膜は完全に維持され、かつ、大きい血管は回避された。プローブ・ホルダー(probe holder)を配置し、固定した。
手術後、すぐに(即ち、投与グループへ投与直後)動物の尾静脈にEB/NF (0.5%, dissolved in normal saline)の混合溶液0.25mlを注射した。24時間後、生理食塩水をラットの心臓にかん流して、結合していない色素を除去した。そのラットを致死させ、迅速に脳組織を回収し、虚血性半球と非虚血性半球に分離した。2つの半球の重量をそれぞれ量り、7.5% (w/v) トリクロロ酢酸(TCA)を用いて均質化した。 そのホモジネートを2つの部分に分け、この中の1mlのホモジネーにNaOH(5N)52μlを用いて調整して、中性(pH)にした。その中から200μlを取り出して蛍光強度決定した(励起485nm、放射535nm)。そのNFは、一連の濃度のNF溶液で作成された標準曲線に基づいて決定された。ホモジネーとの他の部分を遠心分離にかけた(12000g、4℃、20分)。上澄み200μlをマイクロプレートのほうに取り出して、620nmにてその吸光度を測定し、かつ、一連の濃度のEB溶液を用いて作成された標準曲線に基づいてそのEBを測定した。その結果をμg EB (又は NF)/g (脳の湿重量) で表した。脳浮腫の割合(パーセント)を次のように表した:(虚血半球の脳重量−非虚血半球の脳重量)/非虚血半球の脳重量×100%
その結果を表7に示す。24時間の脳虚血後、モデルグループにおけるエネルギー代謝指数は手術前の42.6%に減少した(P<0.05)。モデルグループに比べて、ピノセンブリンラセミ化合物グループ(3mg/kg、10mg/kg, 30mg/kg)のエネルギー指数は、モデルグループに基づいてそれぞれ34.6%及び45.8%(P<0.05)までに有意に増加した。それは、容量依存的であった。
6.1 血清中NO及びTNF-αの含量
表9によれば、モデルグループにおける血清中NO及びTNF-αの含量は、有意に増加したが、ピノセンブリンラセミ化合物グループ(3mg/kg、10mg/kg、30mg/kg)では、ラットの血清中NO及びTNF-αの含量(虚血後24時間の時点)が有意に減少した(P<0.05、P<0.01)。
図2によれば、虚血後24時間の時点における組織ではサイトカイン(TNF-α及びIL-1β)の発現が有意に増加され、そして、虚血の初期段階(early stage)からピノセンブリンラセミ化合物を投与すると(3mg/kg、10mg/kg、30mg/kg)、TNF-α、IL-1βの発現がある程度阻害された。
図2によれば、組織におけるICAM-1及びVCAM-1の接着分子の発現が虚血後24時間の時点で有意に減少され、虚血の初期段階からピノセンブリンラセミ化合物を投与すると(3mg/kg、10mg/kg、30mg/kg)TNF-α及びIL-1βの発現がある程度阻害された。
図2によれば、虚血後24時間の時点で組織におけるiNOS及びAQP-4の発現が有意に減少され、虚血の初期段階からピノセンブリンラセミ化合物を投与すると(3mg/kg、10mg/kg、30mg/kg)、iNOS及びAQP-4の発現がある程度阻害された。
24時間の脳虚血の後、ラットに10%抱水クロラールを腹腔内注射することによって麻酔させた。ヘパリン添加生理食塩水(10分)および4%パラホルムアルデヒド(30分)を使って心臓を経由してかん流させた後、脳組織を取り出し、4%パラホルムアルデヒドへ移して、固定し、その後、凍結切片器によって冠状切片(6μm)に切断し、各20切片の中から一つの切片を取り出して、染色した。染色した断片を、ポリリシンで処理したスライドガラス上で膨張させ、−40℃の冷蔵庫に保存した。
(1)ピノセンブリンのラセミ化合物は、虚血性脳卒中によって引き起こされた行動異常を軽減することができる
(2)ピノセンブリンのラセミ化合物は、虚血性脳卒中によって引き起こされた脳血流の減少を軽減することができる
(3)ピノセンブリンのラセミ化合物は、虚血性脳卒中によって引き起こされた脳梗塞の体積(volume)を減少させることができる
(4)ピノセンブリンのラセミ化合物は、虚血性脳卒中によって引き起こされる脳浮腫を軽減することができる。
(5)ピノセンブリンのラセミ化合物は、虚血性脳卒中によって引き起こされるエネルギー代謝異常を軽減することができる。
(6)ピノセンブリンのラセミ化合物は、虚血性脳卒中によって引き起こされた急性炎症を軽減することができる。
(7)ピノセンブリンのラセミ化合物は、急性虚血性脳卒中によって引き起こされた神経細胞の損傷を軽減することができる。
実験結果によれば、SDラットに一回静脈注射すると、ピノセンブリンラセミ化合物のLD50値が490.9 (367.6〜746.7)mg/kgであり、(S)−ピノセンブリンのLD50値は、375.3(271.2〜538.5)mg/kgであり、そして、(R)−ピノセンブリンのLD50値は347.8 (257.4〜466.3)mg/kgであった。上記結果に基づけば、ピノセンブリンラセミ化合物は、ピノセンブリンラセミ化合物のほうが安全に使用できるより広い範囲を有するということになる。これらの薬物すべては動物の重量に影響を及ぼさず、そして、毒性の観点から主に四肢まひが見られ、かつ、肝および肺におけるマイルドな鬱血が見られた。
1.テストにおいて、一過性中大脳動脈閉塞(transient middle cerebral artery occlusion(tMCAO))モデルのラットを採用した。2時間の局所脳虚血後、再かん流(reperfusion)を行って、局所の脳虚血後の治療時間ウィンドウを調べた。そのラットに、1時間、4時間、及び、6時間の再かん流後(即ち、1時間の虚血、3時間の虚血、及び、8時間の虚血)ピノセンブリンラセミ化合物(1mg/kg、5mg/kg、静脈注射)を投与した。1時間及び4時間の再かん流後(即ち、3時間及び6時間の虚血後)、(R)−ピノセンブリン、及び、(S)−ピノセンブリン(1mg/kg、5mg/kg、静脈注射)を投与した。局所性脳虚血におけるこれらの薬剤の治療時間ウィンドウは、tMCAO後24時間に亘っての神経学的症状のスコア、梗塞の大きさ、及び、脳水分含量に対するこれらの影響を研究することで評価した。
2.1 実験動物
100匹の雄SDラット(240-280g)をBeijing Weitong Lihua Experimental Animal Technology Co., Ltd.(Certificate of Conformity: SCXK(JING)2007-0001)から入手した。これらの動物を、手術前後を通して23〜25℃で通常通り育てた(餌及び水は特に制限なく与えた)。
注射用ピノセンブリンラセミ化合物、注射用(R)−ピノセンブリン、および、注射用(S)−ピノセンブリンは、New Drug Development Laboratory of Drug Research Institute of the Chinese Academy of Medical Sciencesから入手し(バッチ番号:20050601、含量:2.36%)、CN200810084682.3に記載した方法、即ち、ピノセンブリンラセミ化合物又はピノセンブリン鏡像体とシクロデキストリン又はその誘導体との包接錯体を形成し、使用の際にその包接錯体を生理食塩水に溶かすことで製造した。バイエル社(ドイツ)から陽性対照群としてのニモジピン(Nimodipine)を購入した。ヒドロキシプロピルβ−シクロデキストリンは、New Drug Development Laboratory in the Drug Research Institute of the Chinese Academy of Medical Sciencesから入手した。TTCは、シグマ社製であった。その他の試薬は、分析等級の市販されている試薬を用いた。
ラットの麻酔:ラットの腹腔内に400mg/kgの10%抱水クロラールを注射したら、立ち直り反射(righting reflex)が消失した。ラットを仰向けにして手術台に固定し、前頸部(anterior neck)を切断し、組織層を分離して、右総頸動脈(common carotid artery、CCA)を露出した。内頸動脈(ICA)及び外頸動脈(ECA)の分岐後、CCAをセグメントに分離した。迷走神経と機関への損傷は回避された。スタンドバイ使用(standby use)に備えて、CCA及びECAの下にライン(line)を配置した。
見かけ手術グループ(3時間の虚血後通常の食塩水を静脈注射する)
モデルグループ(3時間の虚血後50mg/kgのヒドロキシプロピルβ-シクロデキストリンを静脈注射する)
ニモジピングループ(1 mg/kg、3時間の虚血後投与する)
ピノセンブリンラセミ化合物グループ(1mg/kg、5mg/kg、3、6、及び8時間の虚血後に投与する)
(R)−ピノセンブリングループ(1mg/kg、5mg/kg、3及び6時間の虚血後に投与する)
(S)−ピノセンブリングループ(1mg/kg、5mg/kg、3及び6時間の虚血後に投与する)
最初は静脈注射によって投与し、12時間後静脈注射の1.5の容量で腹腔内投与を行った(1回)。
動物が手術後起きている間に(動物を致死させる前に)神経行動学的検査を繰り返し行った。ベッダーソンスコアを採用した(詳細については、実施例2.1の項を参照されたい)
神経行動学的スコアを求めた後、ラットは首をはねることによって致死させ、その脳組織を速やかに取り出して、−20℃の冷蔵庫に入れた。10分後、脳組織を室温の環境に移した。嗅球(olfactory bulb)、小脳、および、下位脳幹を切除した後、脳組織は、2mmの間隔で6個の連続冠状スライスに切断した。その後、脳スライスを素早く2%TTCを含む5mlの溶液中に入れ、37℃の一定温度、及び、暗闇の条件下で30分間インキュベートした。この間、脳スライスは5分ごとにひっくり返した。TTC染色後、正常組織はバラ色(赤色)を示したが、梗塞組織は、染色されず、白色を示した。脳スライスのすべてのグループを順番に入れて撮影した。写真は、画像解析システムにより処理され、各スライスの梗塞領域を算出した。各スライスの梗塞体積は、スライスの厚さ(2mm)×梗塞面積によって算出され、梗塞体積は、すべてのスライスの梗塞体積の和であった。梗塞体積の比率は、[病巣側(sick-side)梗塞の体積/総病巣側脳体積]として表すことができる。
スライスする前に、各動物の脳の湿重量(wet weight)を量った。染色後、脳スライスを105℃にて24時間乾燥させてから、各動物の脳の乾燥重量を量った。各グループの動物の脳の水分含有量を比較した。脳の水分含有量=(脳湿重量 - 脳乾燥重量)/脳湿重量×100%
定量的な測定値のデータは、tテストに対し平均値±標準偏差として表した。カウント数は、X2テストに対し%として表した。
3.1tMCAOラットにおけるベッダーソン(Bederson)値に対するピノセンブリンの影響
見かけ手術グループでは、神経行動学的スコアが0であった。溶媒対照群のラットの平均スコアは、3.4±0.6であった。ここで、ほとんどの動物は、押す力に対して抵抗が弱く、前肢屈曲があり、回転運動があったので、スコア3と記録された。小数の動物は、押す力に対して抵抗が弱く、前肢屈曲あったので、スコア2と記録された。一部の動物では、損傷がひどくて歩けない状態だったので、スコア4と記録された。
ラットにおける24時間のtMCAO後、その結果得られた脳冠状スライスをTTCによって染色した。各グループに属する動物では一定の割合で染色されない梗塞領域(白色)が存在していたが、それは、24時間の再かん流以内に多少自然と回復されたことを意味する。統計的プロセスにおいては、梗塞体積が5%未満であるサンプルは各グループから取り除いた。3時間の脳虚血後に初めて投与されたピノセンブリンラセミ化合物(1mg/kg)は、梗塞体積を減少させ、そして、6時間の脳虚血後に初めて投与されたときその活性は有意ではなかった。
モデルグループにおいて、ラットの脳水分含量は有意に減少した。ピノセンブリンラセミ化合物(1mg/kg、5mg/kg)は、3時間及び6時間の脳虚血後の脳水分量を有意に減少させたが(P<0.05、P<0.01)、その活性は、8時間の脳虚血後に初めて投与されたときには有意なものではなかった。これらの活性は、ある程度容量依存的であった。(R)−ピノセンブリン及び(S)−ピノセンブリン(1mg/kg、5mg/kg)は3時間の脳虚血後の脳水分量を減らしたが(P<0.05)、6時間の脳虚血後に初めて投与されたときにはその効果は有意なものではなかった。また、ニモジピン(1mg/kg)は3時間の脳虚血後に初めて投与されると有意な改善を示さなかった。この結果を表10にまとめた。
以上の結果によれば、3時間及び6時間の虚血後にピノセンブリンラセミ化合物を注射すると(1mg/kg、5mg/kg)、神経行動学的損傷が減少し、梗塞の体積も減少し、かつ、脳浮腫が軽減した。その活性は、有意であり、容量依存的なものであった。(R)−ピノセンブリン及び(S)−ピノセンブリンは、(ラットの)3時間の虚血後に初めて注射された場合に有意な効果を示したが、6時間の虚血後に初めて注射された場合には効果的なものではなかった。ニモジピンは実験条件において有意な効果は示さなかった。ピノセンブリンラセミ化合物(1mg/kg、5mg/kg、iv)は、ラットの急性虚血性脳卒中の治療に有効であり、その治療時間ウィンドウは約6時間であった。

- 高血圧によって引き起こされる脳卒中の予防及び治療剤の製造におけるピノセンブリンのラセミ化合物、ピノセンブリン塩のラセミ化合物、ピノセンブリン前駆体のラセミ化合物、又は、ピノセンブリン水和物のラセミ化合物の使用。
