Description
1.送達すべき薬物粒子は、これら粒子が肺深部に吸入及び浸透され得るように、十分に小さくなければならない。気道内での付着は、粒子の物理的または幾何学的な形状よりもむしろ粒子の空気力学的な大きさによって制御されるため、粒子の空気力学的直径(以下の式1)が主にこの挙動に影響を及ぼす。肺での付着は、空気力学的直径が5ミクロン未満の粒子では実質的に改善され、5ミクロンを超える有効空気力学的直径を有する粒子では実質的に低下する。
χは、粒子の形状ファクターである。(完全に球形状の粒子の場合、χ=1.0、そして不規則な粒子の場合、χ≧1。)
2.薬物粒子はまた、乾燥粉末吸入器(「DPI」)装置によって十分に脱凝塊化される必要もある。複数の薬物粒子の大きなクラスターは、単一のまたは極めて小さな粒子クラスターほど効率良く肺深部に浸透しない。従来より、DPIには、微粒子粉末の脱凝塊化を保証するために複雑な機械システムが利用されてきており、その後でも、このようなシステムは、ほんの一部しか成功していない。粒径を小さくすることに対する要求に対向して、粉末の流動性と脱凝塊し易さは、残念ながら、粒径を増加させるにつれて改善される。有効粒子直径5ミクロン未満では、脱凝塊化効率は顕著な減退を示す。
A−B(II)
式II中、A基はナノ粒子の表面と結合できる基または部分である。ナノ粒子及び/またはバルク粉末材料を溶媒中で処理する場合、B基は、ナノ粒子及びバルク粉末材料を処理するために用いられるどんな溶媒とも相溶化(する)基である。ナノ粒子及び/またはバルク粉末材料を溶媒中で処理しない場合、B基は、ナノ粒子の付加逆的なアグロメレーションを阻止することが可能な基または部分である。A及びB構成成分は、結合基が、所望の表面適合性をも付与できる場合には、同一であることが可能性である。前記相溶化基は、反応性であってよいが、バルク粉末相の構成成分とは一般には非反応性である。結合組成物は、2つ以上の構成成分から構成されても、または2段階以上で作製されてもよく、例えば、A組成物は、表面と反応するA’部分から構成され、その後、Bと反応し得るA”部分が後に続いてもよい。付加順序は重要ではなく、すなわち、A’A”B構成成分の反応は、コアとの結合前に全体としてまたは部分的に行うことが可能である。コーティングにおけるナノ粒子の更なる説明は、M.リンゼンビューラー(Linsenbuhler, M.)らの粉末技術(Powder Technology)、158、2003年、3〜20頁に見出すことができる。
(a)正確な測定及び/または向上した均質性が求められる粉末の秤量、計量及び混合において;
(b)粉末形状でのバルク材料の輸送または保管の補助として挙げられる、粉末の「タップ密度」(単位質量当たりの減少した体積)の増加が望まれる任意の用途において;
(c)固体インクジェットインクまたは印刷インク及びトナーの調製において;
(d)塗料顔料の調製、処理、及び混合において;
(e)電池アノードにおいてグラファイト密度を高めるため;
(f)粉末の混合またはブレンドに要するエネルギー量を軽減するため;
(g)受動的乾燥粉末吸入器を準備するため(ここで、該吸入器は、患者の吸気性気流に依存して、粉末をエアロゾル化及び/または脱凝塊する);
(h)1種以上の製薬上の活性物質を有する乾燥粉末吸入器(DPI)装置を準備するため。DPI装置は、マウスピースと粉末収容システムを備える。バルク固体材料(1種以上の製薬上の活性成分を包含するもの)とナノ粒子とを含む本発明の組成物は、この粉末収容システム内に配置され得る;
(i)カプセル内に充填されるかまたは錠剤に形成される医薬組成物の精度及び/または均質性を高めるため;及び
(j)米国特許第6,051,252号(ライボヴィッツ(Liebowitz)ら)に記載されているような、急速溶解性錠剤に使用するための高いタップ密度の粉末を調製するため(該特許を参照として本明細書に組み込む)。
乾燥粉末(実施例1〜9それぞれにおいて500g)をナノ粒子のトルエン懸濁液と混合して、ナノ粒子の乾燥質量百分率が0.01、0.1、0.5または1.0である混合物を提供した。表面改質ナノ粒子の1質量%トルエン分散液の測定されたアリコートを、追加のトルエンと混合して、各粉末との所望の混合物をそれぞれ調製するのに十分な質量の表面改質ナノ粒子を含有する分散液500gを作製した。各混合物は、シルバーストーン・モデルL4R(Silverson Model L4R)(マサチューセッツ州イースト・ロングメドー(East Longmeadow)のシルバーストーン・マシーンズ社(Silverson Machines, Inc.)製)ホモジナイザーを用いて30分間、十分に混合した。各混合物を次に、強制空気オーブンにおいて150℃で一晩乾燥させた。各混合物の組成を表1に示す。各比較例では、表面改質ナノ粒子を前記粉末と混合せずに、前記粉末をトルエンと混合して、上述と同様にして混合及び乾燥させた。表1中の各比較例において、「−−−」は「無し」、すなわち、表面改質ナノ粒子を前記粉末と混合しなかったことを示す。実施例10では、硫酸アルブテロール粉末をイソプロパノールに加えて、1質量%分散液を作製した。この分散液を、表面改質ナノ粒子の1質量%イソプロパノール分散液と混合した。この混合物をスプレー乾燥させて、乾燥粉末を供給した。
実施例20では、硫酸アルブテロールと実施例10の表面改質ナノ粒子との混合物を、ターブヘイラー(TURBUHALER)乾燥粉末吸入器装置(英国ロンドン(London)のアストラ・ファーマシューティカルズ(Astra Pharmaceuticals)製の「DPI」装置)のリザーバに入れた。比較例7では、別のターブヘイラー(TURBUHALER)乾燥粉末吸入器装置のリザーバに硫酸アルブテロール粉末を充填した。次に、乾燥粉末吸入器装置からの各乾燥粉末の送達特性を、USPスロート(米国薬局方(United States Pharmacopeia)、USP24 <601>エアロゾル、定量吸入器、及び乾燥粉末吸入器(Aerosols, Metered Dose Inhalers, and Dry Powder Inhalers)、図4)を備えたモデル160マープル・ミラー衝突板(Model 160 Marple Miller Impactor)(「MMI」)及び体積流量毎分60リットルを用いて評価した。好適なカプラーをUSP導入ポートに貼付して、DPI装置と導入ポートの間に気密シールを提供した。全ての試験においても、MMIのステージカップは界面活性剤でコーティングして、粒子の飛散と再飛散を予防した。各試験において、DPIからの5回の投与は、15秒おきに1回作動させて、各作動毎にDPIを所定の位置で5秒間保持した。最後の作動後、DPIは、所定の位置で5秒間保持してから取り外した。真空源は、その後、電源を切る前に更に15秒間放置した。
0.005g/mLの濃度を有する表面改質ナノ粒子のストック分散液(5nm寸法、イソオクチル/メチルで表面改質されたもの)は、表面改質ナノ粒子(1.0g)を200mLメスフラスコに加えて、残りの容積をヘプタンを満たすことによって調製された。フラスコ内に攪拌子を入れて、ナノ粒子が視覚的外観に基づいて完全に分散するまで混合物を攪拌プレート上で攪拌した。薬物粉末(アルブテロールベース、フルニソリドのヘミ水和物、酢酸ピルブテロールをそれぞれ5.0g)とストックナノ粒子分散液(10mL)を丸底フラスコ(1.0L)に加えた。次に、このフラスコにヘプタンを加えて、全容積を約200mLとした。フラスコをゴム栓で密封し、混合物を約3〜5分間音波処理と手で揺すり、及びフラスコの側面に付着する凝結した材料が見えなくなるまで、脱凝塊した。その後、フラスコをロータリーエバポレーターに配置して溶媒を除去した。ロータリーエバポレーターは公称温度50℃に設定し、真空下で作動させた。溶媒を全て除去した後、残った粉末はフラスコの側面で固まった。フラスコをその後、45℃の真空オーブンに約1時間入れて、いかなる残留溶媒をも更に除去した。堅い剛毛のブラシを用いて、固まった粉末をフラスコの壁面から取り出した後、粉末を400メッシュ篩から押し出して、凝結した材料をバラバラに砕いた。篩い分けした材料を、次に回収して、後で用いるために容器に入れた。改質された薬物粉末組成物は、公称濃度1.0%の表面改質ナノ粒子を有していた。
表面改質ナノ粒子をガラスバブル(3Mカンパニー(3M Company)から取引表記「S60HSガラスバブル(S60HS Glass Bubbles)」として入手可能なもの)に加えて、表5に挙げる質量%を達成した。比較例CE11では、ガラスバブル(3Mカンパニー(3M Company)から取引表記「S60HSガラスバブル(S60HS Glass Bubbles)」として入手可能なもの)のみを用いた。実施例24〜26及び比較例CE11について得られた物理特性を表6に示す。
表面改質ナノ粒子をセラミック微小球(3Mカンパニー(3M Company)から取引表記「3M(商標)セラミック微小球W−410(3M(Trademark)Ceramic Microspheres W-410)」として入手可能なもの)に加えて、表7に挙げた質量%を達成した。比較例CE12では、セラミック微小球(3Mカンパニー(3M Company)から取引表記「3M(商標)セラミック微小球W−410(3M(Trademark)Ceramic Microspheres W-410)」として入手可能なもの)のみを用いた。実施例27及び28並びに比較例CE12について得られた物理特性を表8に示す。

Claims (4)
- 粉末の形状であって製薬上の活性成分及びセラミックバブルからなる群より選択される少なくとも1種のバルク固体材料と、共有結合的に表面改質された凝結していない表面改質ナノ粒子とを含む粉末組成物であって、該表面改質ナノ粒子が、イソオクチルトリメトキシシラン、オクタデシルトリメトキシシラン、及びイソオクチルトリメトキシシランとメチルトリメトキシシランとの混合物からなる群より選択されるシラン表面改質剤で表面改質されたシリカナノ粒子であって、該粉末組成物の質量に基づき0.001〜10質量%の量で該組成物中に存在して、該バルク固体材料の流動性または噴流性を、表面改質ナノ粒子を含まないバルク固体材料に比べて改善する、粉末組成物。
- 粉末の形状であって製薬上の活性成分及びセラミックバブルからなる群より選択される少なくとも1種のバルク固体材料と、共有結合的に表面改質された凝結していない表面改質ナノ粒子との混合物を含む粉末組成物であって、該表面改質ナノ粒子が、イソオクチルトリメトキシシラン、オクタデシルトリメトキシシラン、及びイソオクチルトリメトキシシランとメチルトリメトキシシランとの混合物からなる群より選択されるシラン表面改質剤で表面改質されたシリカナノ粒子であって、該粉末組成物の質量に基づき0.001〜10質量%の量で該混合物中に存在して、該組成物に自由流動性を付与する、粉末組成物。
- バルク固体材料と、共有結合的に表面改質された凝結していない表面改質ナノ粒子とを液体中で混合した後、該液体を除去することを含む、請求項1又は2のいずれかに記載の粉末組成物を製造する方法。
- マウスピースと、粉末収容システムと、請求項2に記載の粉末組成物とを備える乾燥粉末吸入装置であって、該バルク固体材料が少なくとも1種の製薬上の活性成分を含む、乾燥粉末吸入装置。
