我々は、正常な脳と比較して、多形性神経膠芽腫(GBM)における発現を増加させることが分かった1つのタンパク質は、正常な神経幹細胞プールのレギュレーターである、核受容体コリプレッサー(N−CoR)であることを発見した。GBMにおけるN−CoRの表現は、免疫組織化学およびウェスタンブロットによって確認された。
エンドタール無水物(1.68g;10mmol)およびN−エチルピペラジン(3.42g;30mmol)を10mLのトルエンに添加し、還流しながら18時間加熱した。その後、溶媒を減圧下で蒸発させ、残留物をメチルt−ブチルエーテルから結晶化し粗製の生成物(1.8g)を得た。同じ溶媒から再結晶することで、2つのまとまりで、計1.2g(収率42.5%)を得た(m.p.215−218℃(分解を伴う))。D2O中のナトリウム塩の1H NMRおよびMS陰イオンスペクトルから、それぞれ構造および分子量(m/z282.2ams)が確認される。ナトリウム塩.1H NMR:(D2O):0.95 (t, 3H), 1.42−1.65 (m, 4H), 2.20−2.42 (m, 4H), 2.43−2.55 (m, 2H), 2.93 (d, 1H), 3.08 (d, 1H), 3.20−3.33 (m, 2H), 3.42−3.58 (m, 2H), 4.45 (q, 1H), 4.75 (q, 1H)。MSスペクトルは、m/z563.3amsで会合する二量体の存在を示し、これは、双性イオンと予想できる。さらに、エンドタール(MSm/z185ams)の痕跡が存在するが、これはNMRスペクトルからは明らかではない。陰イオン質量スペクトルから、282.2m/z(理論値282.2)で親イオンが、分子量が282.2質量単位であることが確認された。
エンドタール無水物(1)(4g、24mモル)を、乾燥させたメタノール(20mL)にて、還流しながら3時間加熱した。反応混合物を室温まで冷却し、分離した固体(6)をろ過し、メタノールから結晶化した。収量4.6g(96%).Mp114−146℃。
塩化メチレン(40mL)に酸誘導体(6)(2.6g、13mモル)を含む混合物に、EDC.HCL(2.75g、15mモル)を加え、その後HOBt(150mg)を加えた。混合物は室温で10分間撹拌し、その後N−ベンジルピペラジン(7)(1.76g)、続いてDIPEA(3.5mL、20mモル)を添加した。反応混合物を室温で一晩撹拌した。水を反応混合物に添加し、塩化メチレン層を分離し、水性NaHCO3で一度洗浄し、NaSO4で乾燥し、ろ過して濃縮した。粗製の残留物は、溶離剤として塩化メチレンに1−2%のメタノールを含むものを使用したカラムクロマトグラフィにて精製し、必要な純粋な物質(8)を得た。収量2.7g(58%)。
メタノール(20mL)にエステル(8)(2.50g、7mモル)を含む溶液に、水性NaOH(5mLの水に360mgを溶解)を添加し、室温で一晩撹拌した。その後、溶媒を蒸発させ、水(20mL)を添加し、溶液のpHを、6NのNClを使用してpH5に調整した。その後、それを蒸発させ、塩化メチレン(30mL)を添加した。残留する固体NaClをろ過によって除去し、ろ液を濃縮した。生じた固体をジイソプロピルエーテルで粉末にし、純粋な酸(9)(化合物103)を得た。収量1.8g(75%)mp>190℃(dec)。質量スペクトルおよび1HNMRデータから、化合物103の同一性が確認された。1H NMR:(CDCl3):1.45−1.84 (m, 4H), 2.45−2.68 (m, 2H), 2.75−2.95 (m, 2H), 3.12−3.35 (m, 2H), 3.38−3.55 (m, 2H), 3.60−3.80 (m, 2H), 3.95−4.20 (m, 2H), 4.75−4.85 (m, 2H), 7.40 (s, 5H)。化合物103の質量電荷比を測定する質量分析データから、177.2m/z、345m/zおよび711m/zにおけるピークが示された。
エンドタール無水物(1.68g;10mモル)およびunsym−N,N−ジメチルエチレンジアミン(2.64g;30mモル)を、トルエン(10mL)に添加し、還流しながら2時間加熱した。その後、溶媒を、減圧下で蒸発して除去し、残留物は少量のジイソプロピルエーテルで粉末にし、再結晶させた。その後、生成物は、−20℃まで冷却して、ヘキサンから2度再結晶した。純粋な生成物(1.9g;収率74%)は48−50℃溶解した。1HNMRスペクトルは双性イオンの構造に完全に一致し、D2Oで処理した場合に、2つの陽子の交換を生じた。さらに、質量スペクトルで化合物106の同一性を確認した。1H NMR:(CDCl3):1.62−1.75 (m, 2H), 1.85−1.95 (m, 2H), 2.2 (s, 6H), 2.45 (t, 2H), 2.92 (s, 2H), 3.60 (t, 2H), 4.85−4.95 (m, 2H)。化合物106の質量電荷比を測定する質量分析データから、239.2m/z、257m/zおよび513m/zのピークが示された。
多形性神経膠芽腫(GBM)の治療のための新しい治療のターゲットを同定するために、カンタリジンアナログを、多形性神経膠芽腫細胞の増殖を阻害する能力について評価した。特に、GBM細胞株U373を評価に使用した。
図1に示されるように、それぞれのノルカンタリジンアナログは、インビボで、用量依存的な様式でGBMの増殖を阻害した。
核複合体に作用するPP2A抗ホスファターゼおよびレチノイドの組み合わせの効果を決定するために、我々は、ヒトGBMに対してインビトロで活性があることが示された水可溶性のカンタリジン誘導体、エンドタールおよび化合物100に着目した。
図2は、化合物100並びにエンドタールは、それぞれATRAとの組み合わせで、相乗的に、GBM細胞株U373の増殖を阻害したことを示す。2つの薬剤がある状態でのパーセント生存が、組み合わせたときと同じ用量で2つの薬剤を単独で使用した場合のパーセント生存の結果より小さい場合に、組み合わせにおける2つの薬剤の阻害活性の相乗作用(強化)があるとされる。ATRAとの組み合わせにおける、化合物100およびエンドタール(end)の相乗作用の程度は、以下の表3示される。
化合物100のアナログ、レチノイン酸およびトリコスタチンAの阻害特性の腫瘍タイプ特異性があるかどうかを決定するために、我々は、GBM株U373、乳癌株、MCF−7(ATCCから得た)、腎臓癌細胞株UMRC(UMRCはDr.Zhuangから得た、NINDS, NIH from the Intramural Research Support Program, SAIC, National Cancer Institute, Frederick Cancer Research and Development Center)に対する単一の薬剤として阻害効果を測定した。
腎臓癌細胞株UMRC(図3)は、脳腫瘍株U373(図4)よりも感受性が低く、一方、乳癌株MCF−7(図5)は、オールトランスレチノイン酸、エンドタールチオ無水物、ノルカンタリジン、エンドタールおよびトリコスタチンAに対して、U373と同程度であった。GBMに対して、これらの薬剤は、一定の細胞タイプ特異性を有する。MCF−7細胞に対する薬剤の活性は、脳腫瘍治療のために開発されるレジメンが、乳癌ならびにN−CoRを過剰発現するその他の腫瘍にたいしても有用である可能性を示唆する。
化合物100の低いミクロモル濃度の阻害活性に加えて、化合物105もまた、化合物100のように、双性イオンであり、図9に示されるように高い活性がある。さらに、脂質可溶性の化合物102は、神経膠腫細胞株U373(図10)を高度に阻害することができる。ノルカンタリジンのその他の誘導体、化合物101、103、104および106は、活性がより小さい(図11−14)。
材料および方法
MDA−MB−231、HT−29、NCI−H460、NCI−H522、NCI−H69、GXF−209、HepG2、OVAR−3、PANC−1、DU−145、LNCAP、HL−60、K−562およびMOLT−4細胞株はいずれも、アメリカンタイプカルチャーコレクション(ATCC;Manassas,VA)から、または国立癌研究所(NCI;Frederick,MD)から入手した。RPMI−1640培地、L−グルタミンジペプチド(HyQ SG−200)およびHEPESは、Hyclone(Logan,UT)から入手した。
アッセイは、説明書に従って行った。簡潔には、プレートをインキュベーターから取り出し、30分間室温のベンチに置いた。プレートは積み重ねなかった。室温で30分のインキュベーションした後、100uLのCellTiter−Glo試薬、プレートの各々のウェルに添加し、2分間混合し、その後、さらに10分間室温でインキュベーションした。その後、PerkinElmer Microbetaシンチレーションおよびルミネセンスカウンター(Trilux)を使用して、発光を記録した。
これらの研究は、材料および方法において記述されるように行い、生データおよびプレートの計画は付属物にて概説される。それぞれの細胞株におけるそれぞれの薬剤について得られたIC50値は、表5に概説される。それぞれの細胞株に対する化合物の効果のグラフ表示は、関連する曲線あてはめと共に、図19A−Nに示される。
移植、HIV/AIDSおよび癌、主に白血病による、免疫無防備状態の患者の人口の増大は、重篤な真菌感染の増加をもたらした。これらの患者における感染症から最もよく採取された菌類は、アスペルギルス種およびカンジダ種である。有効な治療は、カンジダ種の治療に利用可能であるが、アスペルギルス種によって引き起こされる感染症の治療に関する懸念が残っており、これは、免疫無防備状態の宿主における高い死亡率に関与する。そのような感染症は、この群の患者にて取り除くことは困難なため、これらの菌類に対して良好な活性を示す薬剤の必要性が増大している。加えて、足および手の爪および皮膚のそれほど重大ではないが慢性の面倒な真菌感染である皮膚糸状菌症が、世界的に何百万もの人々において発症している。真菌感染の変化する情勢およびこれらの感染症に対する全体的な治療法の不足の結果、化合物100および102は、潜在的な将来の発生のために試験が行われている。
抗菌試験を、化合物100および102の共通のロットで完了した。
3つのカンジダ・アルビカンス、3つのカンジダ・グラブラタ(glabrata)、3つのクリプトコッカス・ネオフォルマンス(neoformans)、3つのアスペルギルス・フミガーツス、3つのリゾパス・オリザエ(Rhizopus oryzae)、3つのフザリウム・ソラニ、3つのシュードアレシェリア・ボイジイ(Pseudallescheria boydii)、および2つのトリコスポロン・ルブラム(Trichosporon rubrum)を含む計23の単離株を試験した。全ての単離株は、評価用の菌類試験室に供された臨床分離株であった。抗真菌感受性試験は、National Committee for Clinical Laboratory Standards, M−27A2, Reference Method for Broth Dilution 抗真菌感受性試験of Yeasts;Approved StandardおよびM38−A ”Reference Method for Broth Dilution 抗真菌感受性試験of Conidium−Forming Filamentous Fungi; Approved Standardに概説された方法によって行った。これは、グルタミンを含みおよび重炭酸ソーダを含まないRPMI−1640、酵母は0.5−2.5x103または糸状菌は1−5×104の接種サイズ、35℃で24および48時間のインキュベーションにおける試験を含む。最小発育阻止濃度(MIC)は、酵母では薬剤フリーのコントロールと比較して50%の濁度の減少および糸状菌では80%の阻害がもたらされる最小濃度として定義した。
ヒト対象、安全性プロフィールおよびその他の適切な因子におけるこの化合物の達成できる濃度に依存して、この化合物は、トリコスポロン・ルブラムによって引き起こされる皮膚糸状菌の感染症のための実行可能な競合物である可能性がある。

- 以下の構造を有する化合物、または前記化合物の塩、エナンチオマーもしくは双性イオン:
結合αは、存在しまたは存在せず;
R1およびR2は、一緒に=Oであり;
R3は、OH、O−、OR9、SH、S−、SR9であり、
ここにおいて、R9は、H、アルキル、アルケニル、アルキニルまたはアリールであり、
R4は、
ここにおいて、Xは、NR10またはN+R10R10であり、
ここにおいて、それぞれのR10は、独立に、アルキル、置換C2−C12アルキル、
アルケニル、置換C4−C12アルケニル、アルキニル、置換アルキニル、アリール、
置換アリール(ここにおいて、R1およびR2が=Oの場合、置換基はクロロ以外で
ある)、
R5およびR6は一緒に=Oであり;および
R7およびR8は、それぞれ、Hである。 - 以下の構造を有する請求項1記載の化合物、または前記化合物の塩、エナンチオマーもしくは双性イオン:

または、以下の構造を有する請求項8に記載の化合物、または前記化合物の塩、双性イオンもしくはエナンチオマー:
結合αは存在せず;Xは、NR10であり、
ここにおいて、R10は、C1−C12アルキル、置換C1−C12アルキル、
C2−C12アルケニル、置換C4−C12アルケニル、C2−C12アルキニル
または置換C2−C12アルキニルである。 - R10は、メチルである、請求項8から10の何れか1項に記載の化合物:
- 請求項1から17の何れか1項に記載の化合物および医薬的に許容可能な担体を含む医薬組成物。
- 医薬的に許容可能な担体が、リポソームを含むか、または化合物がリポソームもしくはマイクロカプセル中に含まれる請求項18に記載の医薬組成物。
- レチノイドレセプターリガンドがオールトランスレチノイン酸(ATRA)から選択され、ヒストン脱アセチル化酵素リガンドは、2−アミノ−8−オキソ−9,10−エポキシ−デカノイル、3−(4−アロイル−1H−ピロール−2−イル)−N−ヒドロキシ−2−プロペンアミド、3−(1−メチル−4−フェニルアセチル−1H−2−ピロリル)−N−ヒドロキシ−2−プロペンアミド、アピシジン(apicidin)、アルギニン酪酸塩、酪酸、デプシペプチド、デプデシン(depudecin)、m−カルボキシケイ皮酸ビス−ヒドロキサミド(m−carboxycinnamic acid bis−hydroxamide)、N−(2−アミノフェニル)−4−[N−(ピリジン−3−イルメトキシカルボニル)アミノメチル]ベンズアミド、エンチノスタット(entinostat)、オキサムフラチン(oxamfiatin)、フェニルブチレート、ピロキサミド(pyroxamide)、スクリプタイド(scriptaid)、サーチノール(sirtinol)、ナトリウム酪酸塩、スベリックビスヒドロキサン酸(suberic bishydroxamic acid)、スベロイルアニリドヒドロキサン酸(suberoylanilide hydroxamic acid)、トリコスタチンA、トラポキシンA、トラポキシンBおよびバルプロ酸から成る群から選択される、請求項21または22記載の使用。
- 癌が、乳癌、大腸癌、大細胞肺癌、肺の腺癌、小細胞肺癌、胃癌、肝臓癌、卵巣腺癌、膵臓癌、前立腺癌、前骨髄球性白血病、慢性骨髄球性白血病または急性リンパ球性白血病、多形性神経膠芽腫、結腸直腸癌、卵巣癌を治療するための薬剤を製造するための請求項21から23の何れか1項に記載の使用。
