で表される化合物と反応させて、ヒドロシリル化する第二の工程を有することを特徴とする。
で表される化合物と反応させる第二の工程を有する。このように、無機粒子の表面に、炭素数が4〜50のアルキル基又は炭素数が6〜30のアリール基を導入する前に、ヒドロキシル基を有する無機粒子と、シランカップリング剤を反応させるため、無機粒子の表面近傍のヒドロキシル基の残存量を減少させることができる。その結果、塩基性物質のピークのテーリングが小さい充填剤が得られる。

平均粒子径が5μm、比表面積が450mm2/gのシリカゲル1gをアンプルに入れ、120℃で10時間減圧乾燥した。アンプルを冷却した後、窒素雰囲気下でビニルメチルジメトキシシラン0.55mLを加えて密閉し、350℃で12時間反応させた。さらに、生成物を取り出し、クロロホルム10mL及びメタノール10mLで洗浄した後、120℃で10時間減圧乾燥した。
平均粒子径が5μm、比表面積が450mm2/gのシリカゲル15gを脱水トルエン60mL中に分散させた。次に、ピリジン5.79mL及びビニルトリクロロシラン3.12mLを加え、3時間加熱還流した後、冷却し、ろ過した。得られた残渣を、トルエン100mL、アセトニトリル100mL及び60質量%アセトニトリル水溶液100mLで洗浄した。さらに、60質量%アセトニトリル水溶液100mL中に分散させ、室温で1.5時間撹拌し、ろ過した。得られた残渣を、60質量%アセトニトリル100mL及びメタノール100mLで洗浄した後、120℃で10時間減圧乾燥した。得られた粒子は、炭素の含有量が4.00質量%であった。
シリカゲル10gを120℃で10時間減圧乾燥した後、冷却し、脱水トルエン40mL中に分散させた。次に、ピリジン0.8mL及びオクタデシルジメチルクロロシラン3.3mLを加え、3時間加熱還流した後、冷却し、ろ過した。得られた残渣を、トルエン100mL、クロロホルム100mL及びメタノール100mLで洗浄した後、120℃で10時間減圧乾燥した。
実施例1、実施例2及び比較例1の充填剤をカラムに充填した後、20mMリン酸緩衝液:アセトニトリルが60:40(体積比)の移動相を用いて、強塩基性物質のアミトリプチリンのテーリング係数を測定した。
Claims (5)
- 前記第一の工程は、前記無機粒子を、前記炭素数が2以上8以下のアルケニル基及び/又は炭素数が2以上7以下のアルキニル基を有するシランカップリング剤と反応させる工程と、該シランカップリング剤と反応させた無機粒子を、アルケニル基又はアルキニル基を有さないシランカップリング剤と反応させる工程を含むことを特徴とする請求項1に記載の充填剤の製造方法。
- 請求項1又は2に記載の充填剤の製造方法を用いて製造されていることを特徴とする充填剤。
- 請求項3に記載の充填剤が充填されていることを特徴とするカラム。
- アミトリプチリンのテーリング係数が0.9以上2.0以下であることを特徴とする請求項4に記載のカラム。
