以下の実施例で使用したビニルアルコキシシロキサンポリマーは、特に指定のない限り、Gelestによって供給されたそのままで使用した。図1は、ビニルメトキシシロキサンホモポリマー1:(Gelest、カタログ番号VMM−005)及びビニルエトキシシロキサンホモポリマー2:(Gelest、カタログ番号VEE−005)を例示説明する。図2は、ビニルエトキシシロキサン−プロピルエトキシシロキサンコポリマー3:(Gelest、カタログ番号VPE−005)を例示説明する。
相10:20gの乾燥した多孔質球状シリカ粒子(dp3μm、表面積225m2/g、孔径175Å)を、250mLの丸底フラスコに移し、その後、トルエン(60mL)中の5gのビニルエトキシシロキサンホモポリマー2と0.5gのテトラメチルエチレンジアミン(例えば、Aldrich)との混合物を添加した。スラリーの上側で慎重に分散させた後、反応混合物を安定的に還流させ、72時間撹拌した。シリカ粒子を濾過し、トルエン及びアセトンで十分に洗浄した。次に、結合したシリカを、5%の酢酸溶液(CH3CN:H2O=1:1、v/v)の100mLの混合物中に分散させ、2時間超音波処理した。濾過及びアセトンでの洗浄後、得られたシリカを105℃で一晩、真空下で乾燥させた。乾燥したシリカを、60mLのトルエンに再び溶解し、その後、5gのビニルジメチルエトキシシラン(例えば、Gelest)及び0.5gのテトラメチルエチレンジアミン(例えば、Aldrich)を添加した。得られた混合物を、24時間還流した。官能化シリカ粒子を濾過し、トルエン及びアセトンで十分に洗浄して、相10を得た。

相41:15mLの溶媒(例えば、ジクロロメタン)を、5gのビニル官能化シリカ(相19)、3gの1−オクタデセン6(例えば、Aldrich)、及び0.8gのジクミルペルオキシド(例えば、Aldrich)に添加した。得られた混合物を、均一になるまで超音波処理し、次いで、全ての揮発物を、回転蒸発装置を用いて減圧で除去した。次に、得られた混合物を、100mLのガラス瓶に移して、ガス入口及びガス出口の両方を有するねじ蓋を取り付ける。瓶を不活性ガス(例えば、窒素またはアルゴン)で15分間パージした後、(1大気圧で)瓶を封止し、所望の温度(50〜300℃)まで加熱した。同じ温度で16時間維持した後、反応を冷まし、反応混合物を、トルエン中に分散させ、30分間超音波処理した。濾過後、ケーキをトルエン及びアセトンで十分に洗浄して、相41を得た。
相47:15mLの溶媒(例えば、ジクロロメタン)に、5gのビニル官能化シリカ(相22)、0.53gの1−オクタンチオール9(例えば、Aldrich)及び0.35gのジクミルペルオキシド(例えば、Aldrich)を添加した。得られた混合物を、均一になるまで超音波処理し、次いで、全ての揮発物を、回転蒸発装置を用いて減圧で除去した。次に、得られた混合物を、100mLのガラス瓶に移して、ガス入口及びガス出口の両方を有するねじ蓋を取り付ける。瓶を不活性ガス(例えば、窒素またはアルゴン)で15分間パージした後、瓶を封止し、所望の温度(50〜300℃)まで加熱した。同じ温度で16時間維持した後、反応を冷まし、反応混合物を、トルエン中に分散させ、30分間超音波処理した。濾過後、ケーキをトルエン及びアセトンで十分に洗浄して、相47を得た。
試験は、酸ストレス条件とカラム試験とのサイクルの反復からなった。
性能試験条件:固定相、相42、及び市販のC18;カラムの寸法は3×150mm;移動相は10%のアセトニトリル/90%の10mMの酢酸アンモニウム(pH=5.2);流量は0.425mL/分;注入体積は2μL;温度は30℃;検出は220nmのUV;試験プローブはウラシル(0.15mg/mL)、アセトアニリド(2mg/mL)。
酸ストレス条件:移動相、0.1MのTFA;流量、0.425mL/分;温度、80℃;持続時間、3時間。
試験は、塩基ストレス条件とカラム試験とのサイクルの反復からなった。性能試験条件は、以下の通りであった。カラムは相43で充填、寸法3×150mm;移動相は10%のアセトニトリル/90%の10mMの酢酸アンモニウム(pH=5.2);流量は0.425mL/分;注入体積は2μL;温度は30℃;検出は220nmのUV;試験プローブはウラシル(0.15mg/mL)、プロカインアミド(0.15mg/mL)、トシル酸ナトリウム(0.15mg/mL)、アセトアニリド(2mg/mL)。
塩基ストレス条件は以下の通りであった。移動相は10%のメタノール中に0.1MのNaOH;温度は30℃;持続時間は1時間。
相42で充填されたカラムの性能を評価するために、ウラシル、フタル酸ジメチル、及びフェナントレンを含有する混合物を使用して、クロマトグラフィー分離を行った。
試験条件は、次の通りであった。カラムは相42で充填、粒径は5μm、カラム寸法は3×150mm;移動相はアセトニトリル/脱イオン水(70/30、v/v);流量は0.425mL/分;注入体積は2μL;温度は30℃;検出は220nmのUV;ならびに試験プローブはウラシル(ピーク1、0.15mg/mL)、フタル酸ジメチル(ピーク2、0.75mg/mL)、及びフェナントレン(ピーク3、0.15mg/mL)。
一連のアミノグリコシド抗生物質(例えば、ゲンタマイシン、スペクチノマイシン、カナマイシン、リボスタマイシン、ストレプトマイシン、アプラマイシン、パロモマイシン、ジヒドロストレプトマイシン、ネオマイシン、硫酸ネチルマイシン、トブラマイシン、アミカシン、アルベカシン)は、極めて親水性であり、かつ従来の逆相カラム上に保持するのは困難であるため、これらは、イオン対逆相液体クロマトグラフィー(RPLC)を使用して相42で充填されたカラムで分析した。トリフルオロ酢酸(TFA)及びヘプタフルオロ酪酸(HFBA)を、アミノグリコシドを保持するのを助けるためのイオン対試薬として使用した。約pH1の脱イオン中の100mMのTFAを、移動相として主に使用した。少量のHFBAを、保持を上昇させ、選択性を調整するために、移動相に添加した。
アミノグリコシド抗生物質に必要なクロマトグラフィー条件に対する耐久性を評価するために、相42で充填されたカラムを、100mMのTFA(約pH1)を移動相として使用して、50℃でゲンタマイシン分離の500回を超える連続運転に供した。図14は、本プロセスの間の分離のオーバーレイを例示説明する。全体的に見て、相42は、研究の間、並外れた化学安定性及びクロマトグラフィー安定性を呈し、従来のシラン化学構造から作製された市販のC18相の50%を超える保持損失(retention loss)と比較して、わずか4%未満の保持損失が認められた。

約1以下のpHを有する移動相が、20時間を超えて該クロマトグラフィー材料を通して流される間、疎水性中性化合物のクロマトグラフ分析の保持時間が、+/−10%を超えて変化しない、前記クロマトグラフィー材料。 - 前記プロセスは、
前記第1の段階の間であるが、前記第2の段階の前に、少なくとも約100℃及び500mbar未満の条件下で、前記シリカを前記少なくとも1つの第1の官能基化化合物と反応させる工程を反復することと、
前記第2の段階の間に、少なくとも約100℃及び500mbar未満の条件下で、前記表面結合された第1の官能基化化合物の前記1つ以上の第1の反応性基を、前記少なくとも1つの第2の官能基化化合物と反応させる工程を反復することと、を更に含む、請求項37に記載のクロマトグラフィー材料。 - 前記第1の段階での、シリカと少なくとも1つの第1の官能基化化合物との前記反応は、溶媒の非存在下で行われる、請求項37に記載のクロマトグラフィー材料。
- 前記第2の段階での、前記表面結合された第1の官能基化化合物の前記1つ以上の第1の反応性基と、前記少なくとも1つの第2の官能基化化合物との反応は、溶媒の非存在下で行われる、請求項39に記載のクロマトグラフィー材料。
- 前記第1の段階での、シリカと少なくとも1つの第1の官能基化化合物との反応は、触媒の存在下で行われる、請求項37に記載のクロマトグラフィー材料。
- 前記第1の官能基化化合物は、ビニルシロキサンポリマーを含む、請求項37に記載のクロマトグラフィー材料。
- 前記ビニルシロキサンポリマーは、式I:
- 前記第1の反応性基は、ビニル基及びアリル基からなる群から選択されるメンバーを含む、請求項37に記載のクロマトグラフィー材料。
