Description
本発明の高血糖改善剤は、上記糖取り込み促進剤の優れた糖取り込み促進作用によって、血糖値を低下させる。したがって、高血糖症、糖尿病、糖尿病関連疾患(例えば、肥満、高脂血症、高コレステロール血症、脂質代謝異常、高血圧症、脂肪肝、メタボリックシンドローム、浮腫、心不全、狭心症、心筋梗塞、動脈硬化症、高尿酸血症、痛風)、または糖尿病合併症(例えば、網膜症、腎症、神経障害、白内障、足壊疽、感染症、ケトーシス)の予防または治療剤として用いることができる。
本発明の高血糖改善食品用組成物は、上記薬理効果を備えた健康食品・健康飲料・特定保健用食品・機能性食品・栄養補助剤、その他ヒト以外の動物に対する薬剤や飼料などに用いることができる。中でも、糖類を多く含んだ飲食物素材に本発明の高血糖改善食品組成物を添加した場合には、筋肉細胞の糖取り込み促進作用による効率的なエネルギー供給効果を奏し、また、それに伴う筋肉組織の活性化・増強・増大や肉体疲労軽減、運動能力の向上などの効果を奏することから、優れたエネルギー補助食品を提供することができる。
L6筋芽細胞(DS Pharma Biomedical社製)を2×104/mLの細胞密度になるようにMEM培地(10%(v/v)牛胎児血清含有)で調製し、96穴組織培養プレートに移し(1穴当たり200μL)、37℃の5%炭酸ガス培養器にて2日間培養した。細胞がコンフルエントになった時点で、培地をMEM培地(2%(v/v)牛胎児血清含有)に置換し、さらに細胞を37℃の5%炭酸ガス培養器にて5日間培養した(L6骨格筋細胞の作製)。次いで、培地をMEM培地(0.2%(w/v)牛血清アルブミン(BSA)含有)に置換し、さらに細胞を37℃の5%炭酸ガス培養器にて18時間培養した。このように培養したL6骨格筋細胞を、糖取り込み試験(実施例1)および細胞毒性試験(実施例2)に用いた。
フィセチン、ルテオリン、ケルセチン、アピゲニン、バイカレイン、ユーパチリンおよびロビネチン(ユーパチリンはPhytoLab社製、他の化合物はExtrasynthese社製)の存在下でのL6骨格筋細胞における糖取り込みを調べた。アピゲニン、バイカレイン、ユーパチリンおよびロビネチンの化学構造式を以下に示す。
上記ポリフェノール類をMEM培地(0.2%(w/v)BSA含有)にて100倍希釈し、ポリフェノール類を30μM含有するMEM培地を調製した。陰性対照として、1%(v/v)DMSOを含有するMEM培地を用いた。調製例1のL6骨格筋細胞の培養プレートに、これらの培地を1穴当たり100μL添加し、プレートを37℃の5%炭酸ガス培養器にて24時間静置した。次いで、WST−1試薬(Roche Diagnostics社製)を1穴当たり10μL添加し、プレートを37℃の5%炭酸ガス培養器にて1時間静置した。プレートの各穴について、生細胞により還元されて生じたホルマザン色素の吸光度(測定波長450nm、対照波長630nm)をプレートリーダーにて測定した。さらに、細胞をクリスタルバイオレット染色し、吸光度(測定波長570nm)をプレートリーダーにて測定し、細胞密度を算出した。結果を図2に示す。
実施例1において、特に顕著な糖取り込み促進作用を示したフィセチンおよびルテオリンについて、GLUT4の細胞膜移行を確認する試験を行った。フィセチンおよびルテオリンをDMSOに溶解し、10μMフィセチンまたは10μMルテオリンを含有するMEM培地(0.2%(w/v)BSA含有)を調製した。陰性対照および陽性対照として、0.1%(v/v)DMSOを含有するMEM培地(0.2%(w/v)BSA含有)、および100nMインスリンを含有するMEM培地(0.2%(w/v)BSA含有)をそれぞれ用いた。調製例1のL6骨格筋細胞の培養ディッシュに、これらの培地を1枚当たり4mL添加し、ディッシュを37℃の5%炭酸ガス培養器にて15分間静置した。KRH緩衝液をディッシュ1枚当たり1mL用いて各ディッシュを2回洗浄した後、非特許文献5に記載の方法にて細胞膜画分を調製した。得られた細胞膜画分のタンパク質量を測定し、その2μgをSDS−PAGEに供してタンパク質を分離した。分離後のタンパク質をポリビニリデンフルオリド(PVDF)膜に転写した。このPVDF膜をブロッキング ワン(ナカライテスク株式会社製)でブロッキングし、TBST(20mM Tris−HCl(pH8.0)、0.15M NaCl、0.05% Tween20)溶液で数回洗浄した後、PVDF膜に1次抗体として抗GLUT4抗体または抗GLUT1抗体(いずれもSanta Cruz Biotechnology社製)を、2次抗体としてホースラディッシュペルオキシダーゼ(HRP)標識した抗ヤギIgG抗体(Santa Cruz Biotechnology社製)を反応させた。膜上の免疫複合体をLumiLight Plusウェスタンブロッティング基質(Roche Diagnostics社製)と反応させた後、PVDF膜を用いてX線フィルムを露光させてGLUT4のバンドのシグナルを検出した。バンド強度は、画像解析ソフト「Scion image」(Scion社製)を用いて解析した。GLUT1を内部標準としてGLUT4のバンド強度を補正した後、DMSO処理細胞のGLUT4レベルを基準として、相対値を算出した。結果を図3に示す。

- 前記化合物が、植物抽出物由来である、請求項1に記載の筋肉の糖取り込み促進剤。
- フィセチン、ルテオリン、ケルセチン、およびそれらの配糖体からなる群より選択される少なくとも1種の化合物を含有する植物抽出物を有効成分として少なくとも1種含有する、in vitroでの筋肉の糖取り込み促進剤。
- フィセチン、ルテオリン、ケルセチン、およびそれらの配糖体からなる群より選択される少なくとも1種の化合物を有効成分として含有する、in vitroでの筋肉のGLUT4活性化剤。
