Description
下記表1に記載の各加工食品をミキサーにかけて均質化し、均質化したもの1gに、下記表2に示す抽出液19mlを加えて約100rpmのサイクルで12〜16時間振とうした。振とうしたものを3000×gで20分間遠心分離して、上清を濾紙(アドバンテックNo.5A)で濾過し、食品抽出液を調製した。
下記表3に記載の各加工食品を用いて、試験例1と同じ操作により、それら加工食品からの食品抽出液を調製した。抽出液は表2の組成のものを使用した。
下記に示す加工食品の原材料に牛乳を凍結乾燥したものを1g当り10μg配合して、牛乳カゼインを含有するモデル加工食品を調製した。各加工食品の調製は常法に従い行なった。
・イチゴジャム:いちご58部、糖類41部、ペクチン0.8部、クエン酸0.2部
・ジュース:オレンジ濃縮果汁9部、砂糖9部、クエン酸0.16部、アスコルビン酸0.02部、水81.8部
・ビスケット:小麦粉67部、ショートニング6.7部、砂糖13.4部、食塩0.5部、膨剤0.9部、酸剤0.1部、乳化剤0.07部、プロテアーゼ0.01部
Claims (8)
- 抗体を利用したELISA法により食品中の特定原材料の存在の有無及び/又はその量を検査する食品検査方法のための食品成分抽出方法であって、食品と、亜硫酸塩を含有する抽出液とを混合し、前記食品中の成分を抽出することを特徴とする食品成分抽出方法。
- 前記抽出液に界面活性剤を含有していることを特徴とする請求項1記載の食品成分抽出方法。
- 前記界面活性剤がドデシル硫酸ナトリウムである請求項2記載の食品成分抽出方法。
- 食品中の特定原材料の存在の有無及び/又はその量を検査する食品検査方法であって、食品に亜硫酸塩を含有する抽出液を接触させて該食品中の成分を抽出した食品抽出液を調製した後、前記食品抽出液を検査目的とされている特定原材料に含まれている物質を特異的に認識する抗体に接触させ、ELISA法により食品中の特定原材料の存在の有無及び/又はその量を検査することを特徴とする食品検査方法。
- 前記抽出液に界面活性剤を含有している請求項4に記載の食品検査方法。
- 前記界面活性剤がドデシル硫酸ナトリウムである請求項5に記載の食品検査方法。
- 抗体を利用したELISA法により食品中の特定原材料の存在の有無及び/又はその量を検査するための食品検査キットであって、(1)抽出液及び該抽出液に添加するための亜硫酸塩、又は、亜硫酸塩が添加された抽出液、及び(2)検査目的とされる特定原材料に含まれる物質を特異的に認識する抗体とを含むことを特徴とする食品検査キット。
- 亜硫酸塩として亜硫酸ナトリウム、亜硫酸水素ナトリウム、亜硫酸カリウム、亜硫酸アンモニウム及び亜硫酸鉄から選ばれた少なくとも1種を含有する請求項7記載の食品検査キット。
