Description
本発明は、複合体(conjugates)の生成に関し、ならびにある方法およびその方法を実施するキットに関する。
複合体は、生物科学的な研究、診断薬および医薬において広く使用されている。最も単純な場合においては、複合体は、第2の化学物質(B)、例えば標識分子などに連結される、第1の化学物質(A)、典型的には、例えば生体分子などの分子、ABハイブリッドを形成する。式AjBk(式中、jおよびkは整数である)によって表わされるオリゴマーの形態も可能である。複合体は、通常、特定の目的で設計され、しばしば、自然には生じない材料の新規な組み合わせを含む。典型的には、複合体の1つの構成要素は、他の分子(例えば抗原)と相互作用する能力を有し、例えば抗体であり、第2の構成要素は、幾つかの他の有用な特性(例えば可測性、ガン細胞を殺す能力)を付与することができ、例えば標識である。
リングの方法論を用いて標識化複合体を、コスト効率のよい生産を可能にするような量で購入することができない。
オン酸ヒドラジド(BMPH)などが存在する。
1つの態様においては、本発明は、第1の化学物質および第2の化学物質を反応させて、第1および第2の化学物質が互いに共有結合により結合されている複合体を形成するための方法であって、第1の化学物質、第2の化学物質およびチオール発生剤を同時に接触させることを含み、チオール発生剤がチオール化反応において第1の化学物質と反応して、第1の化学物質上に遊離スルフヒドリル基の形成をもたらし、遊離スルフヒドリル基が第2の化学物質と反応して複合体を形成し、第2の化学物質が、スルフヒドリル基との反応性に関して多価である(即ち、第2の化学物質の1つの分子は、2つ以上のスルフヒドリル基と反応することができる)方法を提供する。
以上のチオール基を含む第1の化学物質をA−SHで表わすことができる。
AとTGとの間の反応を起源としないチオールがB上の限られた数のY官能基に対してA−SH分子と競合し得るため、使用できる複合条件の範囲に関しては特定の制約が存在する。種々の試薬の初期純度、特にTGの初期純度は、これが望ましくない遊離チオール(例えば、TGの加水分解物)の潜在的な供給源であるため、1つの重要な制約である。複合反応のpHを増大させると、A上のアミンを脱プロトン化し易くなり、結果としてアミンとTGとの反応を速くすることになるが、TGの加水分解速度も増大する可能性がある。従って、複合の効率は、あらゆる化学反応の場合と同様に、反応物の濃度に依存するであろうが、チオール汚染の初期レベル、他のチオールの場合と比較した(AとTGとの反応からの)A−SHの生成速度、B−Yとの種々の異なるチオール含有分子の相対的な反応性、および複合に利用できるY官能基の総量にも依存するであろう。
、その特定のB−Y分子があらゆる他のカップリング反応に関与するのを阻止する。従って、「n」が小さい場合、確実に各A−SH分子がB−Yと反応できるようにするためには、大量に過剰のB−Yが必要になり得る。過剰量のB−Yを使用することは、特にBが大きな生体分子である場合、現実的でなく、非経済的である。その上、最終的な複合体中における高レベルの遊離Bの存在は、特定の用途においては厄介であり得る。その代りに、本発明者は、望ましくないチオールとの不所望の競合反応に直面した状況下においてさえ複合体を調製する改善された方法は、高値の「n」を有するB−Y試薬を用いることにより、A−SHとB−Ynとの間での複合効率を高めることであることが分かった。

)(Sigma、P6782)(B)を、スルホ−SMCC(4mM)(Pierce、22322)を用いて25℃で1時間活性化した。マレイミド活性化HRP(「mal−HRP」)(B−Y)を、10mMのリン酸ナトリウム、150mMのNaCl、1mMのエチレンジアミンテトラ酢酸(EDTA)(pH7.03)で平衡化されたPD10カラム(Amersham Biosciences)で脱塩した。活性化されたタンパク質(2.5mg/ml)は、直ちに使用され、または凍結乾燥された後に使用された。
0.5mlの100mMのリン酸ナトリウム(pH7.2)中におけるグルコースオキシダーゼ(5mg)(Biozyme GO3B3)を、スルホ−SMCC(2mM)を用いて25℃で30分間活性化した。マレイミド活性化Gox(mal−Gox)を実施例1で説明されているように、脱塩および処理した。
ウサギIgG(Sigma I5006)を、1mg/mlにおいて、Tris緩衝生理食塩水(TBS)(50mMのTris/150mMのNaCl、pH8.0)中に溶解し、小さなアリコートにおいて−70℃で保存した。被覆されたELISAプレートを調製するため、IgGを解凍し、TBS中において1ml当たり20μgにまで希釈した。Nunc maxisorbプレート(透明、96−ウェル)(コード071832)を、1つのウェル当たり50μl(1μg)のIgGと共にインキュベートした。プレートは、室温で>1時間被覆され、その後、フォイルでくるまれ、貯蔵用に4℃に移された。被覆されたプレートは10日以内に使用された。使用する直前に、プレートはTBS/0.1%のBSA(pH8.0)を用いて>30分間ブロッキングされた。ELISAにより複合体を試験するため、2重または3重に複製されたウェルをTBS/0.1%のBSA中において適切に希釈された50μlの複合体と共にインキュベートした。25℃で60分後、プレートをTBSで5回洗った。適切な基質(以下参照)を加え、適切な波長(使用した標識に依存する)において2分または10分後、Victor3モデル1420マルチラベルカウンタ(Perkin Elmer)を用いて吸光度を測定した。HRP活性は、1mlの試薬当たり1μlのH2O2を含有する、50mMの酢酸ナトリウム(pH5.0)中における1mMの2,2’−アジノ−ビス−(3−エチルベンゾチアゾリン−6−スルホン酸)(ABTS)基質を用いて測定された。Gox活性は、HRP(50μg/ml)および2mMのABTSを含有する、100mMの酢酸ナトリウム、100mMのグルコース(pH5.0)を用いるカップリングアッセイシステムにおいて測定された。アルカリ性ホスファターゼ活性は、1mMのMgCl2および0.5mMのZnSO4を含有する、50mMのグリシン緩衝液(pH9.6)中における5mMのリン酸p−ニトロフェニル(PNPP)を用いて測定された。
凍結乾燥された抗−ウサギIgG抗体(1mg)(Sigma R2004)(「Ab1」)を1mlの150mMのNaCl中に再懸濁させ、4℃で保存した。1.2mMのHCl中における2−イミノチオラン(2IT)の以下のストックを調製した:1000mM、100mM、10mM、1mM、0.1mMおよび0.01mM。Ab1(A)、mal−HRP(B−Y)(実施例1で説明されているとおりにして調製されたもの)および緩衝液(100mMのリン酸ナトリウム、1mMのEDTA、pH7.4)を1:1:2の比で混ぜ合わせ、20μlに小分けした部分を1.5ml用のEppendorfチューブに分注した。それらのチューブは、200mM(C7)、20mM(C8)、2mM(C9)、0.2mM(C10)、0.02mM(C11)および0.002mM(C12)の最終的な2IT濃度を与えるべく、5μlの2IT(TG)を受け入れた。対照チューブ(neg)は、2ITの代わりに1.2mMのHClを受け入れた。「Pos」は、1/1000に希釈された陽性対照抗体である(Sigma A6667)。90

pHを様々に変えることにより複合効率に及ぼされる影響を、様々な割合で0.2MのNa2HPO4および0.2MのNaH2PO4を混合することにより調製された一連のリン酸塩緩衝液を用いて調べた:10:0(緩衝液P1、pH9.29);9:1(緩衝液P2、pH7.72);4:1(緩衝液P3、pH7.38);7:3(緩衝液P4、pH7.14);3:2(緩衝液P5、pH6.94);1:1(緩衝液P6、pH6.75);2:3(緩衝液P7、pH6.58);3:7(緩衝液P8、pH6.39);1:4(緩衝液P9、pH6.12);1:9(緩衝液P10、pH5.81);0:10(緩衝液P11、pH4.29)。Ab1(実施例4で説明されているように、調製されたもの)およびmal−HRP(2.5mg/ml)(実施例1から;水で再構成された凍結乾燥物(lyophilisate))を1:1で混合し、20μlのアリコートをEppendorfチューブに分注した。その後、各チューブは、緩衝液(P1からP11のうちの1つ)(20μl)を受け入れ、続いて10μlの5mMのTG(即ち、1mMの最終濃度)を受け入れた。60分後、950μlのTBS/0.1%のBSA(pH8.0)を加え、それらのサンプルをウサギIgG−被覆プレート(実施例3参照)を用いてELISAにより分析した。その結果が図2に示されている。
pHを様々に変えることによりmal−GOXを用いる複合効率に及ぼされる影響を、サンプルの量が半分であったことと、反応が975μlのTBS/0.1%のBSAを加えることにより停止させられた点を除き、実施例5で説明されているように、調べた。サンプルは、ウサギIgG−被覆プレート(実施例3)を用いてELISAにより分析され
た。その結果が図3に示されている。
pH値を7.4に固定した状態で緩衝液の種類を様々に変えることによる影響について調べた。緩衝液(200mM)およびAb1を1:1で混合し、10μlのアリコートをEppendorfチューブに分注し、続いて、5μlの2.5mg/mlのmal−HRP(実施例1)および5μlの5mMの2ITを分注した。対照の反応は、2ITの代わりに1.2mMのHClを用いて開始された。各緩衝液(Tris、HEPESまたはリン酸ナトリウム)の最終濃度は50mMであった。その結果が図4に示されている。
2ITからのチオールの放出に及ぼすpHの影響を、一連のリン酸塩緩衝液またはTris緩衝液を用いて調査した。90μlの各緩衝液のサンプルを10μlの100mMの2ITと混合し、複製したアリコート(20μl)を透明なマイクロプレート内において25℃で45分間インキュベートした。200μlのDTNB(200mMのリン酸ナトリウム、1mMのEDTA(pH8.0)中における80μg/mlのストックから)を加え、1分後、プレートをA405で読み取った。結果が図5に示されている。
リン酸塩緩衝液中における複合体形成の速度を3つの異なるpH値で調べた。50μlの反応物は、10μlのAb1(実施例4)、20μlの緩衝液、10μlのmal−HRP(実施例1)(2.5mg/ml)および10μlの1mMの2ITを含んだ。特定の時点(5分、20分、60分および2時間)で5μlのサンプルを回収し、TBS/0.1%のBSA中において1/200に希釈した後、ウサギIgG被覆プレート(実施例3)を用いるELISAにより試験した。結果が図6に示されている。
HRP(2.5mg/ml)と緩衝液(P4;実施例5)(サンプル1から4)またはAb1(実施例4)とP4緩衝液(サンプル9から6)を混合(1:1)し、10μlの小分けした部分を、時間0分(この時点で、いずれかの抵抗性(outstanding)材料(Ab1またはmal−HRPのどちらか)が加えられた)に関して相対的に−30分、−15分、−5分および−1分の時点における2ITの互い違い(staggered)付加による5μlの2IT(1mM)と混合した。これは、この実験の半分において、mal−HRPの付加に先立つチオール化Ab1の形成を可能にし、この実験のもう一方の半分において、Ab1の導入に先立つmal−HRPの潜在的な重合を可能にした。参照サンプル(チューブ5)は、時間0分において、Ab1およびHRPを2ITに同時に加えることにより作成された。更に60分のインキュベーションの後、975μlのTBS/0.1%のBSAを加え、サンプルを、ウサギIgG被覆プレート(実施例3)を用いるELISAにより試験した。結果が図7に示されている。
160μlの過ヨウ素酸ナトリウム(0.1M)を2mlのHRP(0.1Mのリン酸Na(pH7.2)中において12.5mg/ml)に加え、暗所において、25℃で25分間インキュベートした。結果として生じたアルデヒド−HRPをSephadex G−25で脱塩して過剰な過ヨウ素酸塩を除去し、0.5Mの重炭酸ナトリウム(pH9.2)中における2,2’(エチレンジオキシ)ビス−エチルアミン(1vol%の「EDBA」最終濃度)と反応させた。室温で1時間後、シッフ塩基を安定化させるため、(5Mストックからの)50mMにシアノ水素化ホウ素ナトリウムを加えた。更に1時間後、そのEDBA修飾HRPサンプルを0.1Mのリン酸ナトリウム(pH7.2)中に脱塩し、5mg/mlに調整した。

mal−EDBA−HRP(EDBA−HRPの2つの異なるバッチから調製)およびmal−OLA−HRP(実施例11から)(10μl;25μg)をそれぞれ2μlの5mg/mlのヤギ抗−ウサギIgG(200mMのHepes(pH7.5)中における)および1.3μlの8mMのTG1と複合させた。25℃で4時間後、サンプルをTBS/0.1%のBSAで1mlに希釈し、そこから段階希釈液を調製し、ウサギIgG被覆プレートまたは対照プレート(ウサギIgGなし)のいずれかを用いてELISAで試験した。結果が図8に示されている。
上で説明されているように、調製されたmal−EDBA−HRP(10μl)を、5μlの2mg/mlのヤギ抗−ウサギIgG(20mMのリン酸Na/150mMのNaCl)、2μlのTG1(8mM)および3μlの以下の1M緩衝液のうちの1つ:MOPS(pH6.5)、MOPS(pH7)、Hepes(pH7.5)またはEPPS(pH8)と混合した。25℃で一晩インキュベーション後、段階希釈液を調製し、ウサギIgG被覆プレートまたは対照プレート(ウサギIgGなし)のいずれかを用いてELISAで試験した。結果が図9に示されている。
複合体の性能を増強することにおけるEDBA処理の適切性が、生来、HRPよりももっと利用可能なアミンであるグルコースオキシダーゼを用いて更に例証される。とは言うものの、EDBA処理と組み合わせた過ヨウ素酸塩酸化は、アミンではなくヒドロキシルが付け加えられている対照複合体(エタノールアミンで処理されたもの)よりも実質的に良好な複合体をもたらす。
ヨードアセチル−EDBA−HRP(実施例11で説明されているように、調製)(10μl;25μg)を2μlの5mg/mlのヤギ抗−ウサギIgG(200mMのHepes(pH7.5)中における)および1.3μlの8mMのTG1と複合させた。暗所において25℃で一晩(〜16時間)インキュベーションした後、複合体をTBS/0.1%のBSAで1mlに希釈し、そこから段階希釈液を調製し、ウサギIgG被覆プレートまたは対照プレート(ウサギIgGなし)のいずれかを用いてELISAで試験した。結果が図11に示されている。
mal−EDBA−HRPまたはmal−OLA−HRP(10μl;25μg)のいずれかを、10μlの様々な濃度(200mMのHepes(pH7.5)中における)のヤギおよびウサギIgG、ならびに2μlのTG1(8mM)と混合した。25℃で4時間のインキュベーションの後、サンプルを1ml当たり250ngの抗体濃度に希釈し、ウサギIgG被覆プレートまたは対照プレート(ウサギIgGなし)のいずれかを用いてELISAで試験した。結果が図12に表わされている。
ン反応物の濃度は〜6μM(即ち、1mg/mlの抗体に対して)であり、効果的には60μMのアミン(約10個のリジンがTGと反応することができるものと仮定して)である。mal−OLA−HRPの場合においては、マレイミド官能基の濃度はSMCC処理前の初期アミン含有量よりも何ら大きくなり得ず、従って、〜50から100μMより大きくない。アミンは、生理学的pH値において、水分子よりも求核性は高いが、典型的な複合反応においては、反応物および溶媒(即ち、水)の濃度が好適ではない。
必要な場合に複合反応を休止させるために使用され得る潜在的なクエンチ剤として種々のアミンを調査するため、本発明者らは、標準的なDTNBアッセイを用いて、2ITからのチオール放出を調べた。グリシン、エタノールアミンまたは1,3−ジアミノプロパン(DAP)の5mMの溶液を100mMのリン酸ナトリウム緩衝液(pH7.4)中において調製した。各緩衝液の980μlのアリコートを1.2mMのHCl中における20μlの100mMの2ITと混合した。新たに調製されたDTNB試薬(100mMのリン酸ナトリウム、1mMのEDTA(pH8.0)中における200μlの80μg/mlの溶液)を、あるタイムコースにわたって、各反応混合物の20μlのアリコートに加えた。DTNBを付加してから1分以内にサンプルを読み取った。結果が図13に示されている。
潜在的な失活剤を用いた場合の初期チオール放出速度についてのより良い認識を得るため、実施例17の実験が、放出された任意のチオールをDTNBとの反応により即座に「捕捉」することができるように変更された。pH8でDTNB反応を測定する必要があるため、実施例17と同じpHを使用することは不可能であったが、同じ3種類のアミンについて調べた。DTNB(80μg/ml)を0.2Mのリン酸ナトリウム、1mMのEDTA(pH8.0)中において新たに調製し、透明な96−ウェルプレート内に分注した(1つのウェル当たり200μl)。20μlの100mMの2ITを加え、自動プレート循環機能を用い、2分毎にA405で吸光度を読み取った。結果が図14に示されている。
凍結乾燥は、タンパク質をベースとした製品を安定化させ、かつ、貯蔵寿命を延ばすため、広く用いられている。活性成分の安定化を助けるためにしばしば使用される賦形剤は、それの主要な機能が凍結ステップ中の保護をもたらすことである凍結保護剤、およびそれの機能が凍結乾燥中および/または保管中の劣質化を防止することである溶解保護剤を含む。任意の新しいサンプルに対する凍結乾燥のための最良の調合物は経験によってのみ決定され得る。本発明者らの知る限りにおいては、TGの凍結乾燥またはB−Y分子の存在下におけるTGの凍結乾燥に関する先例は存在しない。TGおよびB−Yの混合物の凍結乾燥は、Y官能基(例えばマレイミド)、TG(例えば2−IT)およびBの生物学的活性(例えばHRP、アルカリ性ホスファターゼ)を安定化させるための単一調合物の開発を必要とする。明らかなことに、1つの活性成分に有意な損傷が存在する場合には、たとえそれ以外の成分が満足に保存されていたとしても、あらゆるそれ以降の複合反応は劣質化することになるであろう。
5mMのTris/5mMのMgCl2/0.1mMのZnCl2(pH7.0)中におけるアルカリ性ホスファターゼ(16.2mg/ml:BiozymeコードALP112G)を0.1Mのリン酸ナトリウム(pH7.2)を用いて5mg/mlに希釈し、4mMのスルホ−SMCCを用いて25℃で1時間活性化した。この後、サンプルを10mMのリン酸ナトリウム(pH5.8)中に脱塩することにより、3.125mg/mlの濃度を得た。2.必要に応じて賦形剤を加え、最後に、典型的には400μMまたは800μMの濃度で2−ITを加えた。凍結乾燥する前の酵素の最終的な濃度は2.5mg/mlであった。サンプルを、ポリプロピレン製チューブまたはガラス製バイアルに入った液体窒素中において急速に凍らせた後、24時間サイクルを用いて、Advantage ES凍結乾燥器内において凍結乾燥させた。ステップ1:−40℃の棚温度で1320分間;ステップ2:−10℃の棚温度で60分間;およびステップ3:+20℃の棚温度で60分間。凍結乾燥後、サンプルは、−20℃または37℃のいずれかで数日間貯蔵された。
マレイミド活性化EDBA修飾HRPを(実施例19において上で説明されている如く)pH5.8の緩衝液中に脱塩し、TG1を加えて最終濃度を800μMにした後、40μl(100μgのHRP)の小分けした部分を凍結乾燥した。20mMのリン酸ナトリウム/150mMのNaCl(pH7.2)中における320μlの1mg/mlのヤギ抗−マウスIgG抗体に32μlの2MのHepes/10mMのEDTA(pH7.25)を補給した。100μlのこの材料を用いて各バイアルの凍結乾燥混合物を再構成した。25℃で、一晩、複合が進められ、結果として生じた複合体が、実施例3で説明されているように、調製されたマウスIgG被覆プレートでのELISAにより試験された。
糖類は、しばしば、凍結乾燥および/または貯蔵中のタンパク質を安定化させるのに役立ち、37℃でマレイミド−EDBA−HRP/TGおよびマレイミド−アルカリ性ホスファターゼ/TGの凍結乾燥混合物へのトレハロースの包含は、(混合物から調製された複合体のELISAにおける性能によって測定したときに)糖を含んでいないサンプルに比べて安定性を有意に高めた(以下の表および実施例20を参照のこと)。アルカリ性ホスファターゼは、一般的に、Mg2+およびZn2+イオンの存在下において分析されるため、本発明者らは、金属イオンが、凍結乾燥または貯蔵中のこの酵素を安定化させる上で更に役立っていたのかもしれないという可能性について考察した。しかし、金属イオンは、凍結乾燥中に他の成分(例えば、2−IT)に及ぼす損傷効果を有している可能性があり、またはその後の複合反応を妨害する心配があるため、本発明者らは、アルカリ性ホスファターゼよりも安上がりのHRPを用いて幾つかの初期試験を実施した。
Claims (15)
- タンパク質である第1の化学物質および酵素または蛍光物質である第2の化学物質を反応させて、第1および第2の化学物質が互いに共有結合により結合されている複合体を形成する方法であって、
前記方法は、第1の化学物質、第2の化学物質およびチオール発生剤を水溶液条件中で同時に接触させることを含み、
チオール発生剤が、チオール化反応において第1の化学物質と反応して、第1の化学物質上に遊離スルフヒドリル基の形成をもたらし、
遊離スルフヒドリル基が、第2の化学物質と反応して、複合体を形成し、
第2の化学物質が、スルフヒドリル基との反応性に関して多価であり、
さらに、第2の化学物質:チオール発生剤のモル比は1:1以下である、形成方法。 - チオール発生剤が2−イミノチオランを含む、請求項1に記載の方法。
- 前記第2の化学物質とチオール発生剤とのモル比が、1:1から1:20の範囲である、請求項1または2に記載の方法。
- 前記モル比が、1:10から1:15の範囲である、請求項3に記載の方法。
- 前記第1の化学物質、前記第2の化学物質および前記チオール発生剤が1段階手順において同時に混ぜ合わされる、請求項1から4のいずれか1項に記載の方法。
- 前記チオール発生剤および前記第1の化学物質が混ぜ合わされる第1ステップ、ならびに前記第2の化学物質が前記チオール発生剤および前記第1の化学物質と混ぜ合わされる第2ステップを含む、請求項1から4のいずれか1項に記載の方法。
- 前記第1および第2の化学物質が混ぜ合わされる第1ステップ、ならびに前記チオール発生剤が第1および第2の化学物質と混ぜ合わされる第2ステップを含む、請求項1から4のいずれか1項に記載の方法。
- 前記チオール発生剤および前記第2の化学物質が混ぜ合わされる第1ステップ、ならびに前記第1の化学物質が前記チオール発生剤および前記第2の化学物質と混ぜ合わされる第2ステップを含む、請求項1から4のいずれか1項に記載の方法。
- 前記第1の化学物質が、前記第2の化学物質および前記チオール発生剤を含む乾燥混合物に液状の形態で加えられる、請求項8に記載の方法。
- 更に、複合反応を終結させるために求核試薬を加えることを含む、請求項1から9のいずれか1項に記載の方法。
- 前記第2の化学物質が酵素を含む、請求項1から10のいずれか1項に記載の方法。
- 請求項1から11のいずれか1項に記載の方法において使用するための複合キットであって、
前記第1の化学物質、前記第2の化学物質および前記チオール発生剤から選択される少なくとも1つの試薬のサンプル、ならびに請求項1に記載の方法を実施するための取り扱い説明書を含む、複合キット。 - 前記第2の化学物質のサンプルおよび前記チオール発生剤のサンプルを含む、請求項12に記載のキット。
- チオラクトン、イミノチオラクトン、エピスルフィドおよびチアゾリジンから選択されるチオール発生剤の少なくとも1つのサンプルを含む、請求項12または13に記載のキット。
- 前記少なくとも1つの試薬のサンプルが付加的にポリオールを含む、請求項12から14のいずれか1項に記載のキット。
