又は酸化亜鉛などが用いられる。
乾燥された大山蓮華の花(キョンドン市場、国内産)100gに水及びエタノールの比率を7:3に調整した70%エタノール1.0Lを加え、実温で3日間浸出した。これを濾過し、濾液を真空濃縮して乾燥抽出物を得て、抽出収率は7.8g/100gであった。前記乾燥抽出物を1%(w/v)濃度となるように30%1,3−ブチレングリコール溶液で溶解し、濾過した後、本発明の実験に適用した。
乾燥された大山蓮華の全草100gに水及びエタノールの比率を7:3に調整した70%エタノール1.0Lを加え、実温で3日間浸出した。これを濾過し、濾液を真空濃縮して乾燥抽出物を得て、抽出収率は7.0g/100gであった。前記乾燥抽出物を1%(w/v)濃度となるように30%1,3−ブチレングリコール溶液で溶解し、濾過した後、本発明の実験に適用した。
ケラチノサイトに紫外線が照射されると、細胞内で活性酸素が分泌される。前記現象を人為的に誘導するために、人体の正常角質細胞であるHacat細胞(韓国細胞主銀行)を48−ウェルマイクロプレート(48−well microplate)の各ウェルに1×104細胞となるように接種し、DMEM培地(シグマ社、米合衆国)で24時間37℃で培養した。ここに紫外線(UVB)を照射した後、各々のウェルに製造例1の大山蓮華抽出物を培養液に0.1、0.5、1.0、2.0%(v/v)添加した後、24時間経過後、細胞活性をMIT分析法で確認した。
繊維芽細胞に紫外線が照射されると、細胞内で活性酸素が分泌され、細胞は損傷を受ける。前記現象を人為的に誘導するために、人体の繊維芽細胞(韓国細胞主銀行)を48−ウェルマイクロプレート(48−well microplate)の各ウェルに1×104細胞となるように接種し、DMEM培地(シグマ社、米合衆国)で24時間37℃で培養した。ここに紫外線(UVA)を照射した後、各々のウェルに製造例1の大山蓮華抽出物を0.1、0.5、1.0、2.0%(v/v)に調整して培養液に添加した後、24時間経過後、細胞活性をMIT分析法で確認した。
繊維芽細胞は紫外線照射によってDNA損傷が誘発されるが、この防止は皮膚癌の予防及び老化の遅延に重要な要素となる。従って、本発明の大山蓮華抽出物に前記DNA損傷誘発を抑制する効果があるのか確認するために、次の実験を実施した。
前記実施例1で得た試料を用いて本実験を実施したが、実験は次のように進行された。まず、人体の正常繊維芽細胞を48−ウェルマイクロプレート(ナンク社、デンマーク)の各ウェルに1×106細胞となるように接種し、DMEM培地(シグマ社、米合衆国)で24時間37℃で培養した。次いで、製造例1及び製造例2の大山蓮華抽出物を0.01、0.1、1.0、2.0及び5.0%(v/v)にし、血清のないDMEM倍地に交替した後、48時間、追加培養した。この際に、対象群として大山蓮華抽出物が含まれていない、血清のないDMEM倍地に交替して実験した(対照例2)。培養後、各ウェルの上層液を集めてコラーゲン分解酵素分析キット(Amersham、米合衆国)を用いてコラーゲン分解酵素生成の程度を測定し、その結果を下記表4に示した。
人体の正常繊維芽細胞を48−ウェルマイクロプレートの各ウェルに1×106細胞となるように接種し、DMEM培地で24時間37℃で培養した。次いで、製造例1及び製造例2の大山蓮華抽出物を0.01、0.1、1.0、2.0及び5.0%(v/v)にし、血清のないDMEM倍地に交替した実験群(実施例13〜22)と、大山蓮華抽出物が含まれていない、血清のないDMEM倍地に交替した対象群(対照例3)を24時間、追加培養した。培養後、各ウェルの上層液を集めてプロコラーゲン(procollagen)タイプIC−ペプチド(PICP) 量をキット(Takara、日本)を用いて新しく合成されたコラーゲン量を測定し、下記表5に示した。
上記で収得した大山蓮華抽出物を含む化粧料のうち、柔軟化粧水(スキン)の剤形例は次のようである。

本発明の大山蓮華抽出物を含む化粧料のうち、マッサージクリームの剤形例は次のようである。
上記で収得した大山蓮華抽出物を含む化粧料のうち、 パックの剤形例は次のようである。
本発明の大山蓮華抽出物が含有された化粧料の紫外線による皮膚保護を確認するために、次のように実験を実施した。まず、人為的に人の皮膚に紫外線を照射した後(1.5MED)、剤形例3で製造した栄養クリームを6時間間隔で塗布した。比較例としては、剤形例3から大山蓮華抽出物を抜いたものを使用した。実験は20名を対象にし、紫外線を照射してから48時間後、紅斑生成程度の目視評価及び機器評価を実施した。機器評価は皮膚色度計(Skin colorimeter, Minolta社、日本)を用いて測定し、結果は下記表11に示した。
本発明の化粧料の皺改善効果を実際使用テストを通して評価した。製造例1の大山蓮華抽出物を1%(v/v)含有している剤形例3の栄養クリーム、及び剤形例3の栄養クリームにおいて大山蓮華抽出物を精製水に代替したクリームを比較例として使用した。30名の女性を対象に、顔の一方面には剤形例3の栄養クリームを、他方面には比較例のクリームを使用して、6週後の皺改善効果を専門家の目視評価を通して皺の改善程度を評価した。この評価に基づいた皺改善効果の結果は、下記の表12に示した通りである。
Claims (3)
- 大山蓮華の花のアルコール抽出物を有効成分として含むコラーゲン合成増進用化粧料組成物であって、前記大山蓮華の花抽出物の含量は前記化粧料組成物の総重量に対し0.01乃至10.0重量%であるコラーゲン合成増進用化粧料組成物。
- 前記化粧料組成物は、溶液、懸濁液、乳濁液、ペースト、ゲル、クリーム、ローション、パウダー、石鹸、界面活性剤‐含有クレンジング、オイル、粉末ファンデーション、乳濁液ファンデーション、ワックスファンデーション、及びスプレーから構成された群から選択される剤形を有することを特徴とする請求項1に記載の化粧料組成物。
- 第1項又は第2項の化粧料組成物を人間の皮膚に塗布することを特徴とする化粧方法。
