細胞培養物
0.1%コラゲナーゼ消化(Worthington Biochemical、Bedford、MA)を用いてヒト臍帯静脈内皮細胞(HUVEC)を単離した後、ゼラチンを被覆(0.1%)しておいた組織培養板に付着させ、熱で不活性にしておいたウシ胎児血清(BioWhittaker、Walkersville、MD)を20%と内皮細胞分裂促進因子(Biomedical Technologies、Stoughton、MA)を50μg/mlとヘパリン(APP、ロサンジェルス、CA)を8Ul/mlとペニシリンを50Ul/mlとストレプトマイシンを15μg/ml補充しておいた培地199(Gibco BRL、Grand Island、NY)の中で増殖させた。報告した実験で用いた継代は2および3のみであった。
ゼラチンを0.1%被覆しておいた96個ウエル板(96−well plates)にHUVEC(5x103)を入れて室温で1時間培養した。24時間後、培地を除去して、ウシ胎児血清を5%と組換え型ヒトVEGF165(R&D Systems、ミネアポリス、MN)、LTD4またはLTB4をいろいろな濃度で補充しておいた新鮮な培地199に交換した。72時間後にMTT(商標)アッセイ(Sigma、セントルイス、MO)(15)を用いて内皮細胞の数を数えた。ATL−1の調製をClish他(13)と同様にして実施した。抑制パーセントを同様な様式で評価し、この評価に、作動薬を添加する前に15−エピ−リポキシンA4、15−エピ−16−(パラ−フルオロ)−フェノキシ−リポキシンA4のメチルエステル(ATL−1)、LXA4、15−エピ−LXA4、または15−R/S−メチル,LXA4と一緒に15分間インキュベート(37℃)することを含めた。あらゆるインキュベーションを3回重複して実施した。各実験を行う前に物理的方法(LC/MS/MSおよびUVを包含)(13)を用いてATL−1、LXA4、15−エピ−LXA4または15−R/S−メチル,LXA4の一体性および濃度を評価した。
48個ウエルの化学走性用チャンバ(NeuroProbe、Cabin John、MD)の下方のウエルにVEGF、ATL−1または媒体を加えた。これらのウエルの上をゼラチンが0.1%被覆されている孔サイズが10μmのポリカーボネート製フィルターで覆った。HUVEC(1x106)を上方のウエルの中に入れて、前記チャンバをインキュベート(37℃、5%CO2、12時間)した。インキュベーション後、フィルターを取り出し、上表面から細胞をかき取り、固定した後、Diff−Quik(Dade
Behring、Newark、DE)で染色した。前記フィルターを横切って下方の表面に移動した細胞の数を光顕微鏡で数えたが、ここでは、4つの場(fields)を高倍率(100X)で数えた。インキュベーションを3回重複して実施した。抑制の評価では、媒体またはATL−1を入れた培地の中に内皮細胞を入れて15分間懸濁させた後、前記チャンバの中に入れた。
光度計による酵素免疫検定(細胞消滅検定キット:R&D Systems)を用いて
個々の細胞消滅した細胞におけるDNA断片化を量化した。96個ウエルのミクロタイタープレートの中で増殖させたHUVEC(5X103細胞/ウエル)を3日間インキュベートし、3.7%のホルムアルデヒドで固定した後、標識を付ける前にプロテイナーゼKを用いて透過性を上げた。そのDNA断片にビオチニル化(biotinylated)ヌクレオチドを混合した後、ステプタビジン−西洋わさびペルオキシダーゼ接合体に続いて基質であるTACS−Sapphireを用いてそれを検出した。
麻酔をかけたマウス(BALB/c、6−8週零のオス)の背側皮膚の下に無菌空気(3ml)を皮下注射することで拡大させたマウス空気嚢(air pouches)を用いて脈管形成の評価を行った。24時間後、ATL−1(10μg/嚢)または媒体を局所的に搬送した直後にVEGF(1μg/嚢)を注入した。血管円柱(vascular
casts)の形成を用いて血管含有量を評価した(16に示されているように)。簡単に述べると、マウスに麻酔をかけ(144時間の時)そしてその動物を加熱されているジャケットの中に入れる(40℃、10分間)ことで抹消血管拡張を起こさせた。カルミンレッド(carmine red)(Sigma)を5%ゼラチン溶液に5%入れて40℃に温めた溶液を1ml i.v.注射することで血管円柱を生じさせた。空気嚢内層(linings)を切開して重量を測定した。次に、その組織を2mlの3N NaOH溶液に入れて21℃で0.5時間かけて溶解させた後、熱水(56℃)の中で10分かけて完全に消化させた。その消化させたサンプルを遠心分離(2500rpm、15分間)にかけた後、0.45μmのフィルターに通して濾過した。96個ウエル板の分光光度計(較正曲線を使用)を530nmで用いて染料の含有量を定量した。その結果を血管指数(VI)として組織1ミリグラム重量当たりのカルミン染料のミクログラムで表した(n=4動物/群)。脈管構造を可視化する目的で前記嚢の背側表面を切り取った後、ホルムアルデヒドに入れて48時間固定した。その組織に脱水を100%エタノール(5日間、4℃)を用いて受けさせた後、杉脂に2週間浸けることで洗浄した。別の組の実験では、マウスに麻酔をかけ、フルオレセインイソチオシアネート−デキストラン(Sigma)をPBSに0.05g/ml入れたもの(200μl)を屠殺直前144時間の時にi.v.注射した。内層を切開し、固定し、ガラススライドの上に乗せた後、蛍光を検査した(Nikon EclipseモデルE600)。両方のプロトコルとも、その観察者は処理を判断することができた。
空気嚢膜組織を10%緩衝ホルマリンに入れて一晩固定した後、処理してパラフィンの中に埋め込んだ。オンフェイス(on face)で切り取った5ミクロメートルのパラフィン断面(膜組織が入っている)をCD31発現に関する免疫組織化学で用いた。簡単に述べると、スライドからパラフィンを除去し、0.25%のトリプシン(Sigma Chemical)の中に37℃で20分間入れることで予め処理しておいた後、蒸留水に入れて洗浄した。さらなる段階の全部を室温の含水チャンバ(hydrated chamber)の中で実施した。スライドをPeroxidase Block(DAKO、Carpinteria、CA)で5分間予備処理することで内因性ペルオキシダーゼ活性を抑制した後、ヤギ血清を50MmのTris−Cl(pH7.4)で1:5に希釈したもので20分間予備処理することで、非特異的結合部位を遮断しておいた。一次ラット抗マウスCD31抗体(BD PharMingen、サンディエゴ、CA)を50mMのTris−Cl(pH7.4)で1:100に希釈(3%のヤギ血清と一緒にして)加えて1時間置いた。50MmのTris−Cl(pH7.4)で洗浄した後、二次ラビット抗ラット抗体(DAKO)を50MmのTris−Cl(pH7.4)で1:200に希釈(3%のヤギ血清と一緒に)して加えて30分間置いた。スライドを再び50mMのTris−Cl(pH7.4)で洗浄した後、ヤギ抗ラビット−西洋ワサビペルオキシダーゼ接合抗体(Envision検出キット:DAKO)を加えて30分間置いた。更
に洗浄した後、DABクロモゲンキット(DAKO)を製造業者に従って用いてイムノペルオキシダーゼ染色を生じさせた後、ヘマトキシリンを用いて対比染色を行った。
結果を平均±S.E.M.として表す。分散分析を用いて結果の統計学的評価を実施しそしてP<0.05の値を群平均間の差が統計学的に有意であることを示すとして採用した。
安定なアスピリン引き金リポキシンA4類似物(ATL−1と表示)はVEGFで刺激されたHUVEC増殖の効力のある抑制剤であることを確認した(IC50は〜3nM)(図1)。抑制は濃度に依存し、最大の高原部が10nMの所に存在し、そしてその細胞をゲニステイン(genistein)、即ちチロシンキナーゼ活性の抑制剤と一緒にインキュベート(50μM、5分間、37℃、n=3)するとある程度ではあるが可逆的であった(VEGF刺激増殖が38.0±2.5から78.0±6.21%に、P<0.05)。ATL−1類似物単独では濃度を高く(100nM)してもHUVEC増殖に対する作用は全くみられなかった(図1の挿入図)。極めて対照的に、安定なLXB4類似物は増殖を増加させた。それらは関連した構造物であることから、ATLとLXB4類似物が前記細胞に対して個別の作用を示したことは、ATL−1の反応が非常に立体選択的であることを示している。また、代表的な実験でATL−1を未変性LXA4および15−エピ−LXA4と等モル濃度(10nM)で直接比較した結果、活性等級の順位はATL−1>15−エピ−LXA4>LXA4であることが分かった(例えば抑制度合はそれぞれ63.3±3.3、59.8±1.8および38.1±2.0%)。前記細胞をATL−1に接触させた後、トリパンブルーエクスクルージョンアッセイ(trypan blue exclusion assay)で測定した時、HUVECの〜98%が生存できる状態のままであり、このことは、その化合物は細胞毒性を持たないことを示している。VEGFは、内皮細胞に特異的な分裂促進因子であることに加えて、また、内皮細胞生存因子でもあり、従って、細胞増殖を刺激するばかりでなくまた内皮細胞消滅(17)を抑制することでも脈管形成を助長する。
(代表例としてATL−1を示す)は濃度に応じてVEGF刺激HUVEC移動を抑制し、10nMのATLの時に最大の抑制度合(〜45%)が得られたことを示している。増殖検定で観察したように、ATL−1もLXA4も15−エピ−LXA4も15−R/S−メチル,LXA4も単独では濃度を高く(100nM)しても内皮細胞移動を誘発せず(図2A)、この確認は、ATL−1、LXA4、15−エピ−LXA4または15−R/S−メチル,LXA4は細胞が新血管新生部位に移動する初期段階の遮断で役割を果たしていることを示唆している。
同じ用量(1嚢当たり10μgのLTB4)でLTB4刺激新血管新生を与えた(n=2)。
の所で完成する(complete)ことも確認した。加うるに、そのような新規なアスピリン引き金媒介物(aspirin−triggered mediators)はサイトカインの放出を抑制しかつ遺伝子転写レベルで局所的サイトカイン−ケモカイン軸(cytokine−chemokineaxis)(30)を向け直す働きをする可能性があり(29)、両方の作用とも脈管形成過程で興味が持たれる(3)。今日までに試験された他のエイコサノイドは全部でないにしても大部分が脈管形成前駆体(pro−angiogenic)(ロイコトリエンを包含)であることを注目すべきである(例えばLTD4およびLTB4、下記の表および図3を参照)(5、6、28)。このことを鑑み、ATL−1、LXA4、15−エピ−LXA4および15−R/S−メチル,LXA4が抗脈管形成活性を示したことは驚くべきことであった。
