ヒト初代培養細胞系、動脈内皮細胞(HAECやHPAEC等)、静脈内皮細胞HUVEC(Cambrex BioScience Rockland Inc.から入手)、及び、肺線維芽細胞LL47(American Type Culture Collectionから入手)を使用説明書に従って培養し、それらを使用して、CellTiter−Glo Lunminescent Cell viability Assayにより、ヒト内皮細胞及び線維芽細胞それぞれに対する各目的化合物の特異的な活性を、当該各細胞を比較しつつ評価する。発光シグナルの生成を利用する当該アッセイは、細胞培養中のATP濃度の定量化に基づいている。細胞培養中のATP生成量は生存細胞の数を反映している。故に、当該アッセイは、試験化合物の細胞増殖効果及び細胞毒性効果を評価するのに用いられることが多い。96ウェルプレートに増殖培地を加え、細胞を約5×103細胞/ウェルになるように播種する。24時間後、各種投与量の目的化合物を培地に添加し、各投与量につき同じものを4サンプル作製する。72時間処理した後、CellTiter−Glo試薬をメーカーの使用説明書に従って培地に添加し、発光を測定する。対照群にはDMSOビヒクルのみを与える。増殖培地中の内皮細胞及び線維芽細胞の増殖に対する目的化合物のIC50を用量反応曲線に基づいて求め、濃度に対してプロットする。
内皮細胞遊走は血管新生過程の鍵となるステップであり、新たにその場で血管形成を行う上で極めて重要である。Biocoat Endothelial Cell Migration Angiogenesis System(BD Biosciences)のトランス−ウェル・フィルター/インサーツ・チャンバー(trans−well filter/inserts chamber)を内皮細胞遊走アッセイに使用する。当該チャンバーは、ヒト・フィブロネクチンでコーティングした孔径3μmのインサートを有する24−トランスウェル・チャンバー・プレートである。内皮細胞基礎培地を含ませた0.1%ウシ血清アルブミンをインサートに加え、37℃で1時間培養する。内皮細胞(HUVEC)を内皮細胞基礎培地中の0.1%ウシ血清アルブミンと共に4〜5時間培養して飢餓状態とし、細胞を採取した後、内皮細胞基礎培地中の0.1%ウシ血清アルブミン100μlに含ませた各種処理剤を添加したトランスウェルプレートの上部チャンバーに播種(1×105/ウェル)する。各種の化学誘引物質を含む完全増殖培地を下部チャンバーに添加する。細胞を22±1時間37℃で遊走させる。インサート内部の未遊走細胞をQチップで慎重に取り除く。次に、トランスウェルインサートの下部に遊走してきた細胞を4%パラホルムアルデヒドで固定し、ヘキスト33342で染色し、蛍光顕微鏡撮影を行う。各フィルター/インサートの3つの顕微鏡視野について、遊走した細胞の数を分析する。各試験濃度群及び対照群においてはフィルター/インサート・チャンバー培養を3連で行う。10倍率の顕微鏡視野当たりの遊走した細胞の平均数をデータとして
示し、さらに、用量曲線を基にIC50を計算する。
ヒト内皮細胞を細胞外マトリックスでインビトロ培養すると、当該細胞外マトリックスによって内皮細胞の付着及び分化が促進され、細管が形成する。内皮管形成アッセイはこの現象に基づく。細胞外マトリックスでコーティングした96ウェルプレートに内皮細胞(HUVEC)を播種(1.5×104/ウェル)し、目的化合物を完全増殖培地を用いて各種濃度とし、それぞれにつき同じものを3サンプル作製して、各目的化合物による処理を行う。細胞を37℃で約18時間処理して、内皮管を形成させ、倒立光学顕微鏡撮影を行う。画像解析ソフトウェアのImage−Pro Plus(メリーランド州、Siler Spring、Media Cybernetics,Inc.)を用いて細管長を測定する。各ウェル、及び、各処理条件の3連ウェルについて3つの視野での平均管長をデータとして示す。用量曲線を基にIC50値を計算する。
鶏胚漿尿膜により、血管新生の生理学的過程の理想的なインビボモデルが得られる。鶏胚漿尿膜の上に配置したメチルセルロースディスクに血管新生調節剤を塗布することで、新生血管系の発現を変化させることが可能である。文献:Staton et al,Current Methods for Assaying Angiogenesis in vitro and in vivo,INT.J.EXP.PATHOL.,85:233−48(2004)を参照されたい。当該文献の全内容を本明細書に引用して援用する。血管新生漿尿膜アッセイを用いて、各合成化合物の血管新生阻害能をエキソビボで評価する。強制的に空気を循環させる加湿機能付き孵卵器で受精鶏卵を37.5℃で培養する。胚発生3日目に、卵を割り、100mm3のペトリプレートに胚を移し、細胞培養インキュベーター中、37.5℃で卵の発生を継続する。予め作製した約2mm径のメチルセルロースディスクを、胚発生5日目の胚漿尿膜の上に静かに植え付けた後、試験化合物の溶液又は対照ビヒクルをメチルセルロースディスクの上に塗布する。胚を細胞培養インキュベーター中でさらに2日間培養する。胚発生7日目に、この漿尿膜を検査し、新生血管形成を定量分析する。生存鶏胚の各CAMの血管密度指数(VDI)を求めて、血管新生に対する処理効果を評価する。VDIは、メチルセルロースディスクで覆われた部分において3つの等距離同心円中に見られる血管の交差数であり、Image Pro
Plusソフトウェアを使用して求めたものである。各処理群(N≧5)の定量分析に基づく平均VDIを標準偏差と共にデータとして示す。
マトリゲルプラグアッセイは、インビボの新生血管形成アッセイであり、合成化合物及び組み換えタンパク質の血管新生阻害活性の評価に広く用いられている。約8〜10週齢の雌のC57BL/6マウスと高濃度マトリゲルマトリックスとをマトリゲルプラグ実験に使用する。各処理群に4匹又は5匹のマウスを割り当て、各マウスに対しマトリゲルプラグを2つとする。50ng/mlのVEGF、50ng/mlのFGFb、及び、3ng/mlのヘパリンを血管新生促進剤としてマトリゲルに混合する。各種投与量の目的化合物をマトリゲルに混合するか、又は、腹腔内投与、静脈内投与、若しくは経口投与する。4℃のマトリゲルを、1プラグ当たり500μlのマトリゲルを使用して、マウスの両側の剃毛部に皮下注射する。注射したマトリゲルはすぐに単一の固形ゲルプラグを形成する。マトリゲルを接種してから約2週間後に各群のプラグを採取する。CO2を吸入させて各マウスを安楽死させ、マウスの皮膚を剥いで、プラグを露出させる。無傷のプラグを取り出して、10%ホルマリンで固定し、組織学的に分析する。パラフィン包埋したプラグ
の各切片(厚さ:5μm)をCD31に特異的な抗体で免疫染色し、H&Eで対比染色する。各マトリゲルプラグの全断面内に見られるCD31陽性微小血管をカウントする。各マウス群において、約6つのマトリゲルプラグを定量分析し、微小血管密度に関して、対照群と化合物処理群との間に何らかの統計学的有意差がみられるかどうかを評価する。
腫瘍増殖は血管新生に依存する。腫瘍血管新生の阻害は、癌に対する有効な治療法となっている。標準的な異種移植ヌードマウスモデルを使用して、考えられる血管新生阻害薬の抗腫瘍活性を評価する。ヒト腫瘍細胞又はその断片を5〜7週齢ヌードマウスに移植する。目的化合物を各マウス群に腹腔内投与、静脈内投与、又は経口投与する。腫瘍サイズ及びマウス体重を週に2回確認する。標準偏差と共に各時点での平均腫瘍体積を、異種移植片接種後の経過日数である腫瘍増殖時間に対して示す。
本開示の化合物は、医薬品と共に使用することが可能な任意の従来手段によって投与することができ、単一の治療剤としてもよく、又は、治療剤を組み合わせてもよい。本開示の化合物は単独で投与してもよいが、一般的には医薬基剤と共に投与する。当該医薬基剤は、選択された投与経路と医薬分野の標準的な慣行とに基づいて選択される。上記化合物はまた、インターフェロン(IFN)、インターフェロンα−2a、インターフェロンα−2b、コンセンサス・インターフェロン(CIFN)、リバビリン、アマンタジン、リマンタジン、インターロイキン−12、ウルソデオキシコール酸(UDCA)及びグリシリジン等の他の治療剤と共に投与してもよい。
はない。薬学的に許容される基剤としては、ポリマー及びポリマーマトリックスが挙げられる。
者の血液と等張にする溶質を含んでいてもよい、水性及び非水性の等張無菌注射液;並びに、懸濁化剤、溶解剤、増粘剤、安定剤、及び保存料を含んでいてもよい、水性及び非水性無菌懸濁液が挙げられる。化合物は、医薬基剤中の生理学的に許容される希釈剤に含ませて投与してもよい。この生理学的に許容される希釈剤は、例えば無菌の液体又は液体混合物であり、水;塩水;デキストロース水溶液及び関連する糖液;エタノール、イソプロパノール又はヘキサデシルアルコール等のアルコール;プロピレングリコール、又は、例えばポリ(エチレングリコール)400等のポリエチレングリコールなどといったグリコール類;2,2−ジメチル−1,3−ジオキソラン−4−メタノール等のグリセロールケタル類;エーテル;オイル;脂肪酸;脂肪酸エステル又はグリセリド;あるいは、アセチル化脂肪酸グリセリドが挙げられる。これらには、薬学的に許容される界面活性剤(石鹸又は洗浄剤など)、懸濁化剤(ペクチン、カルボマー、メチルセルロース、ヒドロキシプロピルメチルセルロース又はカルボキシメチルセルロース等)、又は乳化剤、及び、他の医薬アジュバントが添加されていても、いなくてもよい。

標準的な2ピース(two−piece)ハードゼラチンカプセル毎に、粉末状の有効成分100mg、ラクトース150mg、セルロース50mg、及び、ステアリン酸マグネシウム6mgを充填して、多数のカプセル単位を調製する。
大豆油、綿実油又はオリーブ油等の消化可能なオイル中に有効成分を含ませて混合物を調製し、それを容積移送式ポンプで、溶かしたゼラチンに注入し、有効成分100mgを含有するソフトゼラチンカプセルを形成する。カプセルを洗浄し、乾燥させる。ポリエチレングリコールとグリセリンとソルビトールとの混合物に有効成分を溶解させ、水混和性の
混合薬を調製してもよい。
有効成分100mg、コロイド二酸化ケイ素0.2mg、ステアリン酸マグネシウム5mg、微結晶セルロース275mg、デンプン11mg、及び、ラクトース98.8mgを含む投与単位となるように、従来の手順で多数の錠剤を調製する。嗜好性を高めるため、外観及び安定性を向上させるため、又は、吸収を遅らせるために適当な水性及び非水性コーティングを施してもよい。
従来のプロセスや新規のプロセスで調製される、経口投与用の固体投与形態である。これらの投与単位は、すぐに溶解して薬物を送達するので、水無しで経口的に摂取される。有効成分を、糖、ゼラチン、ペクチン及び甘味料などの成分を含有する液体に混合する。これらの液体を、フリーズドライ技術や固体状態で抽出する技術によって固体の錠剤又はカプレットへと固体化する。薬剤化合物を、粘弾性及び熱弾性の糖やポリマーあるいは起泡性成分と共に圧縮して、水無しで即時放出されるような多孔質マトリックスを調製してもよい。
Claims (5)
- ビチオノール、又は、その薬学的に許容される塩若しくは溶媒和物を含有する、充実性腫瘍の増殖又は転移を阻害する医薬組成物。
- 充実性腫瘍の増殖を阻害する、請求項1に記載の医薬組成物。
- 充実性腫瘍の転移を阻害する、請求項1に記載の医薬組成物。
- 充実性腫瘍が乳がんである、請求項1〜3のいずれかに記載の医薬組成物。
- 充実性腫瘍が卵巣がんである、請求項1〜3のいずれかに記載の医薬組成物。
