本発明の化合物の全ての部分およびこれらの組合せに関して、適切な原子価が維持される。
PARPの阻害
ニコチンアミド[2,5’,8−3H]アデニンジヌクレオチドおよびストレプトアビジンSPAビーズをAmersham Biosiences(UK)から購入した。大腸菌(E.coli)から精製された組換えヒトポリ(ADP−リボース)ポリメラーゼ(Recombinant Human Poly(ADP−Ribose)Polymerase)(PARP)および6−ビオチン−17−NAD+をTrevigen,Gaithersburg,MDから購入した。NAD+、ヒストン、アミノベンズアミド、3−アミノベンズアミドおよびウシ胸腺DNA(dcDNA)をSigma,St.Louis,MOから購入した。MCAT配列を含有するステムループオリゴヌクレオチドをQiagenから購入した。該オリゴを、10mM Tris HCl(pH7.5)、1mM EDTAおよび50mM NaClを含有するアニーリングバッファー中に1mMまで溶解し、95℃で5分間温置し、ついで45℃で45分間アニーリングさせた。ヒストンH1(電気泳動的に95%の純度)をRoche,Indianapolis,INから購入した。ビオチン化ヒストンH1を、該タンパク質をスルホ−NHS−LC−ビオチン(Pierce Rockford,IL)で処理することにより調製した。該ビオチン化反応は、穏やかに渦巻撹拌しながら3当量の10mM スルホ−NHS−LC−ビオチンをリン酸緩衝食塩水(pH7.5)中の100μM ヒストンH1に4℃で1分間にわたってゆっくり且つ断続的に加え、ついで4℃で1時間の温置を行うことにより行った。ストレプトアビジン被覆(FlashPlate Plus)マイクロプレートをPerkin Elmer,Boston,MAから購入した。
この研究は、脚部にHCT−116腫瘍を担持するヌードマウスにおいて行った。放射線療法の開始の3日前(−3)に、A−620223を0、6.25、12.5または25mg/kg/日で14日間にわたって運搬するOMPをマウスの腹腔内に移植した。開始日である第0日に、10線量の放射線治療(2Gy/日)を単独で、または3つの異なる用量の2−(N−プロピルピペリジン−4−イル)ベンゾイミダゾール−4−カルボキサミドと組合せてマウスに適用した。
この研究は、B16F10マウスメラノーマを有するマウスにおいて行った。マウスをマウス8〜10匹/群の6つの治療群に分けた。治療群に関して、図2を参照されたい。第0日にB16F10細胞をC57BL/6マウスに皮下注射した。第1日に投与を開始した。第1〜14日にA−861695を1日2回経口投与した。第3〜7日にテモゾロミド(TMZ)を毎日経口投与した(TMZおよびA−861695の両方を投与した群では、A−861695の投与の2時間後にTMZを投与した)。
この研究はFisher344ラットで行った。9Lは、Fisher344ラットにおいて正位神経膠肉腫を生成する移植可能なラット神経膠腫細胞系である。9Lは正位に移植されるため、このモデルは、薬物が血液脳関門を通過しなければならない環境において化合物が有効となる可能性を評価するために使用されうる。血液脳関門を通過する、TMZのような物質は、このモデルにおいて、そうでない物質より有効である。
scidマウスにおけるMX−1乳癌異種移植モデルを使用して、白金に基づく物質の効力をA−861695が増強する能力を試験した。この細胞系は、十分には分化していない乳癌を有する29歳の女性から誘導された。MX−1は細胞毒性物質に感受性である。
この研究においては、シスプラチンと組合されたA−861695の効力をヌードマウスにおけるMX−1乳癌異種移植モデルにおいて評価した。第16日に腫瘍のサイズを100mm3にマッチングさせ、PARPインヒビター療法(経口、1日2回、×8)を同じ日に開始させた。第18日に6.0mg/kg/日のシスプラチンの1用量を腹腔内投与した。図5に示すデータは9匹/治療群のマウスの平均±S.E.M.である。
本出願人は、2−[(2R)−2−メチルピロリジン−2−イル]−1H−ベンゾイミダゾール−4−カルボキサミドのような式(I)の化合物のヒトにおける投与を研究した。本出願人は、ABT−888およびテモゾロミドを投与する場合に、表2に示す以下の計画を用いた。この投与プロトコールは12サイクルまでにわたって継続されうる。
TMZおよびABT−888で治療した後の小脳内神経髄芽細胞腫異種移植を有するマウスの生存率を図7および8に示す。時間の単位は日である。
Claims (4)
- 哺乳動物における白血病を治療するためのキットであって、
前記キットは、(i)PARPインヒビターを含む医薬組成物および(ii)細胞毒性物質を含む医薬組成物を含有し、
前記PARPインヒビターは、2−[(2R)−2−メチルピロリジン−2−イル]−lH−ベンゾイミダゾール−4−カルボキサミドまたはその治療上許容される塩であり、
前記細胞毒性物質は、テモゾロミド、シスプラチンおよびカルボプラチンよりなる群から選ばれる、前記キット。 - 哺乳動物における中枢神経系の腫瘍を治療するためのキットであって、
前記キットは、(i)PARPインヒビターを含む医薬組成物および(ii)細胞毒性物質を含む医薬組成物を含有し、
前記PARPインヒビターは、2−[(2R)−2−メチルピロリジン−2−イル]−lH−ベンゾイミダゾール−4−カルボキサミドまたはその治療上許容される塩であり、
前記細胞毒性物質は、テモゾロミド、シスプラチンおよびカルボプラチンよりなる群から選ばれる、前記キット。 - PARPインヒビターを含む、哺乳動物における白血病を治療するための医薬組成物であって、
