本出願は、2004年10月7日出願の米国仮出願第60/522,512号について優先権を主張するものであり、前記明細書の内容は、その開示の一部として本明細書に引用される。
本発明は、成熟樹状細胞の産生および細胞治療におけるその使用、および免疫エフェクター細胞の感作に関する。成熟樹状細胞は、未成熟樹状細胞から産生させることができる。
細胞治療は、修飾抗原提示細胞(APC)または免疫エフェクター細胞を利用し、患者における免疫応答を引き起こす。抗原提示細胞が免疫応答を引き起こすため、それらは細胞治療にとって中枢を成す。実際に、それらは、Tリンパ球から一次免疫応答を誘導することができる唯一の細胞である。

選択可能マーカー遺伝子、例えば、哺乳類細胞中の安定または一過性トランスフェクタントを選択するためのネオマイシン遺伝子、高レベルの転写を行うためのヒトCMVの極初期遺伝子由来のエンハンサー/プロモーター配列、mRNA安定性のためのSV40由来の転写終止およびRNAプロセシングシグナル、複製のSV40ポリオーマ源および適正なエピソーム複製のためのColE1、万能(versatile)多クローニング部位、およびセンスRNAおよびアンチセンスRNAのイン・ビトロ転写のためのT7およびSP6 RNAプロモーター。他の手段は、当該技術分野で知られており、利用可能である。
ポリペプチドおよびタンパク質は、本発明の様々な方法の必要成分である。タンパク質およびポリペプチドは、Perkin Elmer/Applied Biosystems, Inc., Model 430Aまたは431A(フォスター・シティー, カリフォルニア, 米国)製などの市販の自動ペプチド合成装置を用いる化学合成によって得ることができる。合成したタンパク質またはポリペプチドを沈澱させ、例えば高性能液体クロマトグラフィー(HPLC)によってさらに精製することができる。あるいは、タンパク質およびポリペプチドは、本明細書に記載の既知の組換え法によって下記のような宿主細胞およびベクター系を用いて得ることができる。
イン・ビトロでの樹状細胞への増殖および分化を目的とするCD34+幹細胞の単離および増殖については、多くの方法が当該技術分野で知られている。例えば、米国特許第5,199,942号を参照されたい。前記明細書の内容は、その開示の一部として本明細書に引用される。下記の記載は単なる例示のためのものであり、本発明の範囲を限定するものではない。
CD34+幹細胞は、この細胞を適当なサイトカインとインキュベートすることによって樹状細胞に分化させることができる。Inabaら(1994)(前記引用)の文献には、ネズミ幹細胞をネズミGM-CSFと共にインキュベートすることによる幹細胞の樹状細胞へのイン・ビトロでの分化が記載されている。簡単に説明すれば、単離した幹細胞を1 - 200ng/mLのネズミGM-CSF、好ましくは約20ng/mL GM-CSFと共に標準的RPMI成長培地でインキュベートする。培地は、一日置きに約一回ずつ新鮮な培地に置換する。約5-7日間培養した後、表面マーカーの発現および形態学によって評価したところ、大部分の細胞は樹状である。樹状細胞を、蛍光活性化セルソーター(FACS)または他の標準的方法によって単離する。
DCは、末梢血中の普通の非増殖性CD14+前駆体(単球)からGM-CSFおよびIL-4、またはGM-CSFおよびIL-13を含む培地で培養することによって生成させることができる(例えば、WO 97/29182号参照)。この方法は、Sallusto and Lanzavecchia (1994) J. Exp. Med. 179:1109およびRomani et al. (1994) J. Exp. Med. 180:83に記載されている。簡単に説明すれば、CD14+前駆体は多量に存在するので、(末梢血中のCD34+細胞およびさらに関与する前駆体を増加させるのに用いられる)G-CSFのようなサイトカインによる患者の予備治療はほとんどの場合に不必要であることが報告されている(Romani et al. (1996) J. Immunol. Methods 196:137)。他の研究者らは、この方法によって生成したDCはむしろ相同であると思われ、未成熟状態で、または完全に分化した、または成熟した状態で産生させることができることを報告している。FCS(ウシ胎児血清)のような非ヒトタンパク質を回避し、完全かつ不可逆的に成熟した安定なDCを成熟刺激として自己由来単球ならし培地を用いることによって得ることができることが示された(Romani et al. (1996) Immunol. Methods 196:137、Bender et al. (1996) J. Immunol. Methods 196:121)。しかしながら、本発明とは対照的に、これらの研究では、IL-12のレベルが増加し、および/またはIL-10のレベルが減少した成熟DCは得られなかった。
樹状細胞に抗原を負荷する方法は、当業者に知られている。一つの態様では、樹状細胞は、抗原を含む培地で培養される。次いで、DCはMHC分子と会合している細胞表面上の抗原を吸収して処理する。好ましくは、DCは、抗原をコードする核酸によるトランスフェクションによって抗原が負荷される。DCのトランスフェクションの方法は、当業者に知られている。
本発明の幾つかの方法では、T細胞は哺乳類から単離され、成熟した修飾DCによってイン・ビトロで感作(または活性化)することができる。一つの方法では、フィコール-ハイパーク密度勾配遠心分離を用いて、確立された手続きに従って赤血球および好中球からPBMCを分離する。細胞を、1%ウシ胎児血清(FBS)を加えた改質AIM-V(AIM-V(GIBCO)に2mMグルタミン、10μg/mL硫酸ゲンタマイシン、50μg/mLストレプトマイシンを加えたものからなる)で洗浄する。T細胞を標準的手法に準じてカラムまたは磁性ビーズにカップリングした適当なモノクローナル抗体によるネガティブ選択またはポジティブ選択によって増殖させる。細胞の一定分量を、CD4、CD8、CD3、およびCD14などの細胞表面表現型について分析する。例示の目的で、細胞を洗浄し、前記のように改質し、5%FBSと100U/mLの組換えIL-2 (rIL-2)を含むAIM-V(補足したAIM-V)1mL当たり約5 x 105個の細胞の濃度で再懸濁する。細胞がHIV+患者から単離される場合には、25nMのCD4-PE40 (緑膿菌外毒素Aの転座およびADP-リボシル化ドメインに結合したHIV結合CD4ドメインからなる組換えタンパク質)、またはHIVに選択的にハイブリダイゼーションする他の同様な組換え細胞傷害性分子を細胞培養物に加え、細胞増殖の残りについて、培養物からHIVに感染した細胞を選択的に除去する。CD4-PE40は、HIVに感染した細胞培養物でのp24産生を阻害し、HIV-1に感染した細胞を選択的に殺すことが示されている。
もう一つの態様では、細胞表面マーカーを用いて、本発明の方法を実施するのに必要な細胞を単離することができる。例えば、ヒト幹細胞は典型的にはCD34抗原を発現し、一方、DCはMHC分子および補助的刺激分子(例えば、B7-1およびB7-2)である、顆粒球、NK細胞、B細胞、およびT細胞に特異的なマーカーを欠くものを発現する。表面マーカーの発現により、これらの細胞の同定および精製が促進される。これらの同定および単離の方法としては、FACS、カラムクロマトグラフィー、磁性ビーズによるパンニング、ウェスタンブロット法、X線撮影法、電気泳動、高性能液体クロマトグラフィー(HPLC)、薄層クロマトグラフィー(TLC)、超核酸クロマトグラフィーなど、および流体またはゲル沈降反応、免疫拡散(単純または二重)、免疫電気泳動、ラジオイムノアッセイ(RIA)、酵素結合免疫吸着定量法(ELISA)、免疫蛍光分析法などのような様々な免疫学的方法が挙げられる。免疫学的および免疫分析法の手順の一般的な総説については、Stites and Terr (監修)1991 「基礎および臨床免疫学(Basic and Clinical Immunology)」(第7版)、およびPaul(前記引用)を参照されたい。抗原を選択するための抗体の作製方法の論文については、Harlow and Lane (1989)(前記引用)を参照されたい。
本発明の方法の別の態様では、抗体を用いる必要がある。このような抗体は、モノクローナルまたはポリクローナルであってもよい。それらは、抗体誘導体または抗体変異体であってもよい。それらは、キメラ、ヒト化、またはヒト自体であってもよい。タンパク質またはポリペプチドを用いると、当業者であれば、この受容体に特異的に結合する抗体をさらに生成させることができる。Fab、Fab'、Fab2、Fab'2、および一本鎖可変領域などの抗体の機能性断片または誘導体を用いることもできる。抗体は、細胞培養、ファージ、またはウシ、ウサギ、ヤギ、マウス、ラット、ハムスター、モルモット、ヒツジ、イヌ、ネコ、サル、チンパンジー、ゴリラなどの様々な動物で産生させることができるがこれらに限定されない。これらの断片または誘導体は、タンパク質またはその断片についての結合の特異性を保持している限り、用いることができる。抗体は、結合の特異性について適切な抗原に対する結合を不適切な抗原または抗原混合物に対する結合と所定の条件の組で比較することによって試験することができる。抗体が不適切な抗原の少なくとも2、5、7、および好ましくは10倍の適切な抗原に結合している場合には、この抗体は特異的であると考えられる。
目的の遺伝子(例えば、CD40Lおよび/またはIL-12p35)の発現を定量するための様々な方法が知られており、ハイブリダイゼーション分析法(ノーザンブロット分析)およびPCRに基づくハイブリダイゼーション分析法が挙げられるが、これらに限定されない。IL-12 p35 mRNAまたはCD40L mRNAのようなmRNAレベルの変更についての分析では、試料に含まれる核酸を最初に抽出することができる。例えば、mRNAは、Sambrook et al. (1989)(前記引用)に記載の手順に従って様々な細胞溶解酵素または化学的溶液を用いて単離し、または市販の核酸結合樹脂により製造元によって提供された添付説明書に従って抽出することができる。抽出した核酸試料に含まれるmRNAは、次に標準的手順に準じてそれぞれ核酸プローブおよび/またはプライマーを用いてハイブリダイゼーション(例えば、ノーザンブロット分析)および/または増幅法によって検出することができる。
タンパク質分析については様々な技術が当該技術分野で利用可能であり、放射免疫測定(radioimmunoassay)、ELISA(酵素結合免疫吸着定量法)、「サンドイッチ」免疫測定法、免役放射定量測定法、イン・サイチュー(in situ)免疫測定法(例えば、金コロイド、酵素、または放射性同位体標識を用いる)、ウェスタンブロット分析、免疫沈降分析法、免疫蛍光分析法、およびPAGE-SDSが挙げられるが、これらに限定されない。
前記のように、本発明は、本発明の方法によって産生された樹状細胞または教育されたT細胞を用いるエクス・ビボ療法も提供する。例えば、免疫原で形質転換した樹状細胞を用いて、細胞傷害性およびヘルパーT細胞をイン・ビトロで活性化することができる。あるいは、形質転換した樹状細胞を哺乳類に導入して、T細胞をイン・ビボで活性化する。さらにまた、イン・ビトロで感作されたT細胞を、T細胞を活性化してクラスIのMHC分子を認識するものに対応する抗原性ペプチドを有する標的細胞に対してT細胞が細胞傷害性である哺乳類に導入することができる。これらの標的細胞は、典型的には癌細胞、またはMHCクラスI表面で特有の抗原性ペプチドを発現する感染細胞である。
本発明の方法によって産生されるT細胞または樹状細胞は、直接対象者に投与して、選択された免疫原に対して活性なT細胞を産生することができる。投与は、細胞を良好に送達して、最終的に対象者の血液または組織細胞と接触させるための当該技術分野で知られている方法によることができる。
本発明は、本発明の成熟化法を実施するための商業上の重要なキットを提供する。一つの態様では、キットは、IFNγポリペプチドまたはIFNγ mRNAをイン・ビボまたはイン・ビトロで発現する発現カセット、およびCD40Lポリペプチド、またはCD40LのCD40L mRNAイン・ビボまたはイン・ビトロ発現を発現する発現カセットを含んでなる。もう一つの態様では、このキットは、TNFαポリペプチドまたはTNFαmRNAをイン・ビボまたはイン・ビトロで発現する発現カセット、およびCD40Lポリペプチド、またはCD40LのCD40L mRNAイン・ビボまたはイン・ビトロ発現を発現する発現カセットを含んでなる。これらのキットは、イン・ビトロ転写のためのRNAポリメラーゼをさらに含んでもよい。
本発明の方法によって産生される抗原提示細胞または教育されたT細胞の免疫原性は、下記のものを包含するがそれらに限定されない周知の方法によって決定することができる。
ペプチドをパルスし、51Crを標識した標的の抗原特異的T細胞による溶解を比較することができる。「一層活性の高い」組成物は、経時的な標的のより大きな溶解を示す。溶解の速度論ならびに固定時点(例えば、四時間)の全般的標的溶解を用いて、性能を評価することができる。Ware et al. (1983) 3. Immunol. 131:1312。
修飾APCに接触したT細胞によって分泌されるサイトカインの種類および量の分析は、機能活性の尺度とすることができる。サイトカインはELISAまたはELISPOT分析法によって測定して、サイトカイン産生の速度および総量を決定することができる。Fujihashi et al. (1993) J. Immunol. Meth. 160:181、Tanquay and Killion (1994) Lymphokine Cytokine Res. 13:259。
本発明の組成物を、正常な提供者または患者由来のPBMCからの反応性T細胞個体群を誘発する能力について分析することができる。この系では、誘導したT細胞を溶解活性、サイトカイン放出、ポリクローン性、および抗原性エピトープに対する交差反応性について試験することができる。Parkhurst et al. (1996) Immunol. 157:2539。
免疫原性は、HLAトランスジェニックマウスを本発明の組成物でワクチン接種して、誘導された免疫応答の性質および大きさを測定することによってイン・ビボで評価することができる。あるいは、hu-PBL-SCIDマウスモデルは、ヒトPBLの養子免疫伝達によってマウスにヒト免疫系を再構築することができる。これらの動物に組成物をワクチン接種し、Shirai et al. (1995) J. Immunol. 154:273、Mosier et al. (1993) Proc. Natl. Acad. Sci. USA 90:2443に以前に記載されているように免疫応答について分析することができる。
T細胞は、反応性組成物に応答して増殖する。増殖は、例えば3H-チミジン接種を測定することによって定量的に観察することができる。Caruso et al. (1997) Cytometry 27:71。
非ヒト霊長類(チンパンジー)モデル系を用いて、HLA制限リガンドのイン・ビボ免疫原性を観察することができる。チンパンジーは、ヒトMHC分子と重複しているMHC-リガンド特異性を共有しており、従って相対的イン・ビボ免疫原性についてHLA制限リガンドを試験することができる。Bertoni et al. (1998) Immunol. 161:4447。
幾つかの細胞内シグナル形質導入結果(例えば、リン酸化)は、MHC-リガンド複合体によるTCR関与の成功と関連している。これらの事象の定性的および定量的分析は、TCR関与によりエフェクター細胞を活性化する組成物の相対的能力と相関している。Salazar et al. (2000) Tnt. J. Cancer 85:829、Isakov et al. (1995) J. Exp. Med. 181:375。
Histopaque 1077およびTween 20は、Sigma (セントルイス, ミズーリー)から購入した。PBSおよびX-VIVO 15は、Cambrex (イースト・ラザフォード, Ni)から購入した。AIM-V培地、イスコブの改良ダルベッコ培地、およびRPMI 1640培地ならびにトリパン・ブルーおよびウシ胎児血清(FBS)は、Invitrogen (カールスバード(Carlsbad), カリフォルニア)から購入した。Viaspanは、Dupont Pharma Labs (ウィルミントン, デラウェア)から購入した。GM-CSF、IL-4、TNF-α、IL-1β、IL-6、およびIFN-γは、全てR&D Sytems (ミネアポリス, ミネソタ)から購入した。PGE2は、Cayman Chemicals (アナーバー, カリフォルニア)から購入した。可溶性CD40Lは、Alexis Biochemicals (サンディエゴ, カリフォルニア)から購入した。ヒトAB血清は、Valley Biochemical (ウインチェスター, バージニア)から購入した。
ヒトPBMCは、Life Blood (メンフィス, テネシー)によって提供された健康なボランティアからの白血球分離採血法によるコレクションから単離した。PBMCは、フィコール-ヒストペイク(Histopaque)密度遠心分離によって調製し、PBSで四回、室温にて洗浄した。2 x 108個のPBMCを30mLのAIM-V培地に再懸濁し、150cm3のプラスチックフラスコに37℃で二時間接着させた。非接着細胞を除き、残りの細胞をGM-CSF (1000U/mL)およびIL-4 (1000U/mL)を補ったX-VIVO 15培地で5%CO2にて37℃で5-6日間培養した。
T細胞を、RPMI中でPMAで一時間刺激した。細胞を回収し、PBSで一回洗浄した。総RNAは、QIAGEN RNイージー法(QIAGEN RNeasy procedure)を用いて抽出した。活性化T細胞からの総RNA 1μgを、高適合性Advantage Polymerase (Clontech)を用いるGene Amp Goldキット(Applied Bioscience)を用いて一つのチューブRT-PCR(one tube RT-PCR)反応に入れた。CD40L配列の遺伝子特異的プライマーは、CD40L配列 CD40L 5'プライマー: 5'-GCATCATCGAAACATACAACC-3'(配列番号:11)およびCD40 3'プライマー: 5'-GTATTATGAAGACTCCCAGCG-3' (配列番号:12)の塩基47および塩基859に対応している。PCR断片を精製して、T4 DNAリガーゼ(Invitrogen)を用いてpCR2.1ベクターにサブクローニングした。ジェンバンク(GenBank)コンセンサス配列を有するCD40Lオープンリーディングフレームおよび配列の配列分析により、二つの突然変異の存在が明らかになった。一つの突然変異は保存的であり、アミノ酸変化を生じなかった。もう一つの置換は、機能性アミノ酸変化Asn-Serを生じた。位置指定突然変異誘発は、非保存的アミノ酸変化を修正して、アスパラギンに戻すために行った。簡単に説明すれば、10-40ngのCD40L PCR2.1プラスミドDNAを、特注の5'リン酸化し、HPLC精製したプライマー(QIAGEN)、PFU Ultra酵素、ならびに10 x PCR緩衝剤(Stratagene)およびdNTPS(Clontech)を用いる位置指定突然変異誘発で用いた。PCR反応の後、Dpn I制限酵素(Promega)を加え、37℃で一時間インキュベートし、親鋳型を消化した。次に、この反応物5μLをOneshot MACH T1Rコンピテント細胞(Invitrogen)に形質転換し、新たに作製したアンピシリン含有LBプレートで培養した。六個のコロニーを選択し、アンピシリンを含むLBで一晩培養し、3mL培養物として生育した。DNAは、プラスミドミニプレップ(miniprep)(QIAGEN)を用いて単離した。それぞれのクローンについて精製したDNAの一部を、M13FプライマーおよびM13Rプライマー(Invitrogen)を用いるCD40Lオープンリーディングフレームの配列分析のために、ノースカロライナ大学(UNC)のシークエンシング施設に提出した。次に、全てのクローンを、DNASTAR Seqman分析ソフトウエアを用いてCD40Lについてコンセンサスジェンバンク(GenBank)配列に対して整列させた。正確な突然変異誘発塩基を含むことについて、クローン番号2(CD40L WT PCR 2.1と改名)を選択した。
CD40L WT PCR2.1プラスミドをSpel制限酵素を用いて線形化し、フェノール/クロロホルム抽出に続いてエタノール沈澱によって精製した。線形鋳型を水で再構成し、mMessage mMachine T7 Ultraキット(Ambion)を用いて製造元の指示に従ってイン・ビトロで転写した。ポリアデニル化反応に進む前に、RNAの一部を最終分析のために取っておいた。ポリアデニル化RNAは、RNイージーカラム(QIAGEN)を用いてRNAクリーンアップのプロトコールに従って精製した。RNAを水で溶出し、個別に-150℃以下で保管した。ポリA尾部の長さは、RNA Bioanalyzer 2100を用いる非ポリアデニル化RNAと最終生成物の比較分析によって決定した。
エレクトロポレーションの前に、DCを採取してPBSで洗浄した後、冷却したViaspan(Barr Laboratories)に0. 5mL中に4 x 107/mLでまたは0.2mL中に2.5 x 107/mLで再懸濁し、氷上に置いた。DCをmRNA (mRNAコード抗原について1または2μg/106およびCD40L mRNAについて4μg/106)と混合し、4mmギャップのエレクトロポレーションキュベットに入れ、Biorad装置を用いてエレクトロポレートした。エレクトロポレーションの直後に、DCをX-VIVO 15培地で洗浄し、最後にGM-CSF (800U/mL)およびIL-4 (500U/mL)を1 x 106/mLで補ったX-VIVO 15に再懸濁し、低粘着性の6ウェルプレート(BD Biosciences, フランクリンレイクス, ニュージャージー)で37℃にて四時間または二十四時間培養した。下記の追加の成熟刺激も、この時点で添加した。
エレクトロポレーションの後、CD40L mRNAでトランスフェクトしたDCを、IFN-γ(1000U/mL)、TNF-α(10ng/mL)、またはIFN-γとPGE2 (1μg/mL)の組合せで処理した。比較すると、未成熟DCは様々な抗原コードmRNAでトランスフェクトした後、TNF-α (10ng/mL)、IL-1β (10ng/mL)、IL-6 (100ng/mL)、およびPGE2 (1μg/mL)、または可溶性CD40L (200ng/mL) + エンハンサー (1μg/mL)を含んでなり、同時または連続的に1000U/mLのIFN-γを加えた「サイトカインカクテル」で処理した。
未成熟DCを、TNF-α (10ng/mL)、IFN-γ (1000U/mL)、およびPGE2 (1μg/mL)で培養五日目で表現型に成熟した。六日目に、DCを採取し、前記のように抗原とCD40L mRNAを用いてエレクトロポレーションを行い、800U/mL GM-CSFと500U/mL IL-4を含むX-VIVO 15培地で四時間培養した後、採取し、またはワクチン生産のために処方した。
100ng/mLのKRN7000を、500U/mLのIFN-γと1μg/mLのPGE2による二十四時間の培養と組み合わせた、エレクトロポレーションの直後にCD40Lベース工程のDCに適用した。
106個のDCを採取し、冷却したPBS/1%FCSに再懸濁した。MHC分子(HLA-ABC、HLA-DR)、補助刺激分子(CD80、CD86)、成熟マーカー(CD83)、および単球マーカー(CD14)に特異的なフィコエリトリン(PE)またはFITC接合抗体を、96ウェルプレート(BD Biosciences)中でウェル当たり1 x l05個のDCと混合し、4℃で少なくとも十五分間インキュベートした。同位体と調和した抗体を、コントロールとして用いた。十分に洗浄した後、CellQuestソフトウェア(BD Biosciences)を用いるFACScaliburフローサイトメーター(BD Biosciences)で蛍光分析を行った。
HeLa細胞を10% FBS/DMEMで成長させた後、採取して、4mmキュベット中でGFPおよびCD40L RNA (それぞれ20 μg/5 x l06細胞)でエレクトロポレートした。トランスフェクション後の回収率は約70%であり、細胞を6ウェル皿で培養し、一晩培養した。一晩培養した後、トランスフェクトしたHELA細胞を剥離により採取し、マウスIgG1-PEまたは抗ヒトCD40L-PE (両方とも、BD Biosciences,サンディエゴ, カリフォルニア)で染色し、CD40Lの細胞表面発現を探した。2 x 105細胞/試験管を、1% FBS/PBS中10μg/mLの抗体で4℃にて三十分間染色した。細胞を、FACScaliberフローサイトメーターとCellquestソフトウェア(BD Biosciences)を用いて分析した。HeLaで発現したCD40Lの機能を分析するため、1 x 106個の未成熟樹状細胞を6ウェル皿(2mL総量)中で、1000U/mLのIFN-γ(R&D Systems, ミネアポリス, ミネソタ)を加えた5% huAB血清/RPM1中で1 x 106個のHeLa細胞と一晩共培養した。ブロッキングCD40Lモノクローナル抗体 (eBioscienceからの24-31)を10μg/mLで適合したウェルに加え、タンパク質の細胞表面発現が樹状細胞の刺激に必要なことを確かめた。培養上清を18-24時間後に採取し、サイトカインIL-10およびIL-12の発現をELISA(BD Biosciences)によって分析した。
DCの走化性を、24ウエルのトランスウェルチャンバー(Corning Costar, アクトン, マサチューセッツ)中で孔の大きさが8μmのポリカーボネートフィルターを介する遊走により測定した。イスコブス(Iscoves)改良ダルベッコ培地または3-300ng/mLのCCL19、5-250ng/mLのCCL21両者の組合せを含むAIM-V培地、または培地単独中の5%ヒトAB血清を、下のチャンバーに加えた。0.1mL中1-5 x 105個のDCを上のチャンバーに加え、37℃で2-3時間インキュベートした。下のチャンバーを5mL試験管(BD Biosciences)に採取し、0.1mL PBSに再懸濁し、生細胞数をトリパンブルーを用いて計数した。
PVDF膜ELISポット(ELISpot)プレート(Millipore, バレリカ, マサチューセッツ)に5μg/mLモノクローナル抗IFN-γまたは抗IL-2捕捉抗体(BD Pharmingen, サンディエゴ, カリフォルニア)をコートして、4℃で二十四時間インキュベートした。インキュベーションの後、プレートをPBS/0.05% Tween 20で洗浄し、5%ヒトAB血清/RPMI 1640培地で一時間ブロックした。PBMC、T細胞、またはCD8増加T細胞を105個の細胞/ウェル、および1 x 104個の細胞/ウェルのmRNAをトランスフェクトし、抗原を負荷したDC標的を10 : 1のエフェクター:標的比で培養し、37℃、5% CO2で少なくとも十六時間インキュベートした。
IL-12およびIL-10 ELISAセット(BD Pharmingen)については、BD Pharmingenによって公表され、BD Opt EIA試薬セットB pH9.5を用いる方法である。簡単に説明すれば、ELISAプレート(BD Biosciences)に抗IL-12p70または抗IL-10 ELISA捕捉抗体のコーティング緩衝液を4℃で二十四時間コートした。プレートを200μL/ウェルの10% FCS/PBSで一時間ブロックした後、標準物(BD Pharmingen)および上清試料を100μL/ウェルずつ二回加え、室温で二時間インキュベートした。プレートを洗浄し、抗サイトカイン検出抗体を加え、一時間インキュベートし、プレートを洗浄して、溶液を100μLのストレプトアビジン-HRPに代えて、室温でさらに一時間インキュベートした。プレートを再度洗浄し、発色基質を10-20分間加えた後、停止溶液を用いて発色を停止した。プレート分析は、KCジュニアソフトウェア(Winooski, バーモント)を備えたBio-Tek装置EL x 800プレートリーダーを用いて行った。結果は、ピコグラム数/mL/106 DCを示す。この分析法は1mLが106個のDCに対応するように設定されているので、これらの結果はピコグラム数/mL/106DCとして表すこともできる。例えば、3000pg/mL/106DCは、3000pg/106DCと同等である。
mRNAでトランスフェクトした成熟樹状細胞を、CD8精製T細胞と共培養した。全ての共培養は、R-10培地(10mM HEPES pH7.4、1mMピルビン酸ナトリウム、0.1mM非必須アミノ酸、2mMグルタミン酸ナトリウム、55μMβ-メルカプトエタノールを加えた10%FBS、RPMI-1640)で行った。全ての細胞培養試薬は、Invitrogen(カールスバード, カリフォルニア)製であった。CD8+細胞は、単球粘着工程から採取した非接着細胞由来のCD8+T細胞単離キットII(Miltenyi Biotec, オーバーン, カリフォルニア)を用いて精製した。CD8+細胞を、前記の方法で調製した樹状細胞と10 : 1のCD8+:DCで混合した。最初の七日間、細胞を0.2U/mLのIL-2(R&D Systems, ミネアポリス, ミネソタ)を加えた培地で培養した後、24ウエルの組織培養皿に1mL(1 x 106個のCD8+細胞)/ウェルずつ加えた。この最初の七日間の培養後、CD8+細胞を採取し、計数して、5U/mLのIL-2を加えた培地で新鮮なDC刺激因子と共に10:1で再培養した。再度細胞を一週間培養した後、新鮮なDCと20U/mLのIL-2で再刺激した。CTL分析法は、三回目の刺激の後三日間または七日間行った。
T2細胞を、10μg/mLのHLA-A201制限MART-APLペプチド(LAGIGILTV:配列番号:24)または天然ペプチド(AAGIGILTV:配列番号:25)またはPSA-1ペプチド(FLTPKKLQCV:配列番号:26)を用いてFBS/RPMI培地中で一晩インキュベートすることによってパルスを加え、洗浄した後、CTLとして使用した。樹状細胞標的を前記の方法でGFP mRNA、MART-1 APL mRNA、Flu-M1 mRNAでトランスフェクトし、未成熟で一晩インキュベートした。パルスを加えたT2細胞は、100μCiのNa51Cr(Perkin-Elmer Life and Analytical Sciences, Inc., ボストン, マサチューセッツ)と共に37℃で九十分間インキュベートした。過剰の51Crを洗浄によって除き、5000の標識した標的を様々なE:T比のCD8+細胞とともに四時間インキュベートした。非特異的溶解は、パルスを加えないT2細胞を25,000細胞/ウェル加えることによって減少した。放出された51Crを、上清中でシンチレーション計測によって測定した。総放出量は1% Triton X-100を標的に加えることによって計算され、自然放出は培地のみを加えることによって計算された。溶解率は、式(放出された試料のcpm - 自然cpm)/(総放出 - 自然放出cpm)を用いて計算した。
DCは、前記のように「CD40Lベース工程」を用いて生成させ、MART-1をコードするmRNAを負荷した。エレクトロポレーションの後、DCをKRN7000、IFN-γ、およびPGE2とインキュベートした。DCとPBMCを1 : 10の比で20U/mLのIL-2の存在下にて共培養した。PBMCを同一条件下で三回再刺激し、CTL誘導の頻度をMART-1/A2四量体で染色することによって測定し、NKT細胞の増殖をKRN7000/CD1d四量体を用いてFACSによって数えた。
インターフェロン-γおよびCD40Lを用いる連続成熟は、IL-12p70の分泌を最適にする。
未成熟DCを、GM-CSFおよびIL-4を含むX-VIVO 15培地で接着細胞PBMCを六日間培養することによって調製した。DCを六日目に回収し、106個のDC当たり2μgのeGFPをコードするmRNAでエレクトロポレートし、「サイトカインカクテル」で三十六時間成熟させた。あるいは、同時または連続的に加えられるIFN-γおよび可溶性CD40Lの存在下にて、DCを培養することによって成熟させた。補助刺激分子の発現の増加についてDCを観察したが、最も重要なことには、IL-12p70対IL-10の分泌について観察した。図1は、サイトカインカクテルで成熟したDCは、三十六時間の培養期間にわたってIL-12p70と比較して過剰のIL-10を培養上清中に分泌することを示している。対照的に、可溶性CD40LとIFN-γで同時に成熟させたDCは、過剰のIL-12p70を分泌する。しかしながら、十八時間のIFN-γの連続添加の後に可溶性CD40Lを直接培養物に加え、さらに十八時間培養したところ、IL-12p70の分泌レベルは有意に高まった。意外なことには、可溶性CD40Lに続いてIFN-γを加えたところ、IL-12p70の有意な分泌は防止された。結論として、「プライム(prime)」DC成熟に対する本質的刺激(IFN-γ)に続く可溶性CD40Lによって送達される代理Tヘルパー細胞シグナルの連続的送達により、IL-12p70分泌に対するDC成熟が最適になる。
図2は、CD40LをコードするmRNAでトランスフェクトしたHeLa細胞が、抗CD40L抗体とフローサイトメトリーによって明らかされるように、二十四時間の培養の後にCD40Lタンパク質の有意な細胞表面発現を生じることを示している。CD40L mRNAでトランスフェクトしたHeLa細胞を、1000U/mLのIFN-γの存在下にて未成熟DCと共培養した。表IIは、伸長したポリ-A尾部(>400「A」)でトランスフェクトしたHeLa細胞は、未成熟DCと十八時間の培養期間にわたって培養する場合、有意なIL-12p70分泌を誘導することができることを示している。重要なことは、ブロッキング抗CD40L抗体を含むと、IL-12p70の分泌が妨げられることであり、トランスフェクトしたmRNA配列によってコードされるタンパク質の同一性および機能上の重要性が確かめられる。
未成熟DCをGM-CSFとIL-4との培養において六日後に採取し、CD40L mRNA (400-ポリA)を滴定することによってトランスフェクトし、直ちに1000U/mLのIFN-γの存在下で培養した。図3は、十八時間の培養後に回収した上清がIL-10と比較して過剰のIL-12p70を含み、最適サイトカイン分泌には少なくとも4μgのCD40LmRNA/106DCが必要とされることを示している。CD40L mRNAペイロードを4μg/106DC以上に増加すると、成熟後のDC収率が相当減少する(データは示さない)。並行実験において、未成熟DCを4μgのCD40L mRNA/106細胞でトランスフェクトし、IFN-γを直ちに培養物に滴加した。図4は、IL-12p70の最適誘導を支持するには、少なくとも100U/mLのIFN-γが必要であることを示す。図5aは、IL-12p70がトランスフェクション後およびIFN-γとの共培養の6-8時間後に検出可能なレベルで現れ、培養上清における最適蓄積は20-24時間、記録されることを示す。対照的に、10ng/mLの TNF-αをIFN-γの代わりに用いても、IL-12産生が維持されるが、レベルは減少する(図5b)。さらに、IFN-γは、TNF-αよりも低いレベルのIL-10産生を同時に生じる(図5c)。
図2は、mRNAから翻訳されたCD40Lタンパク質はトランスフェクト細胞の細胞表面で発現することができ、タンパク質は、DC上の対応物、すなわちCD40との相互作用の結果IL-12p70分泌のために適切にシグナル化する能力を保持することを示す。トランスフェクトDCでのCD40Lの細胞分布を決定するため、DCをトランスフェクション後の様々な時点で採取し、細胞表面または細胞内コンパートメントのCD40Lの存在を測定した。図6aおよび図6bは、CD40Lの大半が細胞内コンパートメント内に局在しており、有意なタンパク質発現(27% DC CD40Lポジティブ)はトランスフェクションの60分後まで現れないことを示す。従って、IFN-γはトランスフェクションの直後に加えられるが、成熟事象の送達は、CD40Lに先行するIFN-γシグナルにより連続的(sequential)である。図1に示されるように、IFN-γおよびCD40LによるDCの連続的(sequential)成熟がIL-12p70分泌を最適にする。さらに、図7は、CD40Lでトランスフェクトされ、かつIFN-γで処理されたDCをトランスフェクション後の十八時間過剰のブロッキング抗CD40L抗体の存在下にて培養すると、有意なレベルのIL-12p70をなお分泌することを示す。このデータは、この系でIL-12p70の産生に必要なCD40L/CD40相互作用が細胞内コンパートメント内で起こり得ることを示す。
未成熟DCを、4μgのCD40L mRNA/106DCでトランスフェクトし、1000U/mLのIFN-γで補助成熟した。あるいは、CD40L染色のネガティブコントロールとして、未成熟DCを「サイトカインカクテル」で成熟させた。最大頻度の発現はCD40L RNAによるトランスフェクションの約3-4時間後に達成され(図6b参照)、細胞を固定して浸透すると、DCの80%はCD40Lを発現するが、細胞表面染色ではDCの約15%しか検出されない(図6c)。このデータは、CD40Lタンパク質の大半がDC内に保持され、細胞表面では発現しないことを示す。CD40Lタンパク質は一時的に発現し、DCの大半はトランスフェクションの二十六時間後にはCD40Lネガティブになる。CD40Lの同起源の受容体分子であるCD40の発現は、「サイトカインカクテル」のみを受け取るDCと比較すると、CD40LをコードするmRNAによるDCのトランスフェクションによって変更されない。
IL-12p70を分泌し、かつCD80、CD83、およびCD86(表III参照)のような補助刺激分子を高レベルで発現する細胞として定義された成熟表現型を示す能力に加えて、DCがイン・ビボでリンパ節へ回帰することができるようになるには、それらは遊走能力を発揮しなければならない。幾つかの研究は、PGE2が成熟DCの遊走をプライム(prime)することを示している(Luft et al. (2002) Blood 100: 1362, Scandella et al. (2002) Blood 100:1354)。図8は、IFN-γの他に1μg/mLのPGE2が含まれると成熟するDCを遊走させることができ、この遊走能力の獲得はCD40L mRNAペイロードに比例することを示している。従って、CD40Lは成熟するDC表現型および主要なIL-12p70プロファイルに寄与するだけでなく(表II参照)、遊走のプライミングにも寄与する。対照的に、DCは、CD40L mRNAによるトランスフェクションによって成熟し、IFN-γの存在下であるが、PGE2の非存在下で培養したDCは、ケモカイン受容体CCR7の有意な細胞表面発現を示すにも関わらず、遊走することができない(データは示さない)。
CD40L mRNAトランスフェクションおよびIFN-γ/PGE2を介して成熟したDCの「免疫力(immunopotency)」を測定するため、DCを、CD40L mRNAおよびIFN-γ/PGE2培養環境の他にfluマトリックスタンパク質をコードする2μg/106DCのmRNAで同時トランスフェクトした。トランスフェクションの十八時間後に、DCを採取し、洗浄して、自己由来T細胞と共にIFN-γ ELISポット(ELISpot)分析法で共培養した。図9は、CD40L/IFN-γ/PGE2により成熟したDCが、この分析法でflu特異的IFN-γスポットの頻度によって定義されるように、flu mRNAでトランスフェクトして「サイトカインカクテル」で成熟したDCと比較して、免疫力が増加していることを示す。
図9に記載されているようなリコール応答は、最適化補助刺激分子を発現し、かつサイトカイン環境を維持するDCの存在によってあまり変化しない。従って、DCを、多数の健康な提供者が高いナイーブT細胞前駆体出現率を保持している黒色腫関連抗原MART-1に対する一次免疫応答を誘発する能力について試験した。HLA-A201提供者を優先的に用いたので、DCをMART-1をコードするmRNAでトランスフェクトし、A2制限決定基を位置指定突然変異誘発によるmRNA配列の突然変異によって最適化し、27位のアラニンはロイシンによって置換され、本明細書ではMART-APLと呼ばれている (Valmori, D et al (1998) J. Immunol. 160:1750)。2μgのMART-APL mRNAと4μgのCD40L mRNAとで同時トランスフェクトし、直ちにIFN-γ/PGE2で十八時間パルスを加えたDCを、MART-APLだけ負荷し「サイトカインカクテル」で一晩成熟したDCと比較した。抗原を負荷して成熟したDCを精製した自己由来CD8+T細胞に加え、0.2U/mLのヒトIL-2の存在下にて七日間培養した。この期間の後、T細胞を回収して、IL-2 ELISポット(ELISpot)分析法において適当な抗原を負荷したDC刺激因子で二回目の共培養を行った。図10は、CD40LおよびIFN-γ/PGE2によって成熟したCDの存在下で培養したCD8+T細胞は、MART-APL mRNA配列内で最初にコードされた最適化MART-APLエピトープに対する特異的応答においてIL-2を分泌することができるT細胞を極めて有意に増加させることを示している。結論としては、IFN-γ/PGE2およびCD40Lを介する連続的成熟に暴露されたDCは、現在受け入れられている標準的「サイトカインカクテル」で成熟したDCよりも一次免疫応答を生じる点で有意に一層強力である。さらに、図11は、「サイトカインカクテル」で成熟したMART-APLを負荷したDCを用いて生成したCTLは、適切なHLA-A2制限MART-APLペプチドでパルスを加えたT2細胞に対するCD4独立でCD8依存性の細胞傷害性を伝達することができないことを示している(図11b)。対照的に、CD40L/IFN-γ/PGE2で成熟したDC上に生成し
たCTLは完全に活性であり、MART-APLペプチドでパルスしたT2標的を殺す(図11a)。
DCを、本明細書に記載のPME-CD40L工程で五日目に成熟させた。具体的に説明すれば、単球をGM-CSFおよびIL-4を含む培地で五日間培養して、未成熟CD83-DCを産生した。五日目に、未成熟DCにTNFα、IFNγ、およびPGE2(TIP)を供給した。六日目に、TIP表現型を測定した(表IV参照)。表IVに示されるように、細胞の大半はCD80、CD83、CD86、およびCD209に対してポジティブであった。これらのDCは、CCR7ネガティブでもあった(データは示さない)。低割合のCD14+細胞は、樹状細胞に分化しなかった単球を表す。六日目に、CD83+CCR7-DCを、増幅した腎細胞癌RNAから調製した1μgのmRNAと4μgのCD40L mRNA/106細胞で(エレクトロポレーションにより)同時にトランスフェクトした。CD40L発現を、トランスフェクションの四時間後に測定した。細胞をトランスフェクションの四時間後に、液体窒素に低温保存した。融解直後および融解の二十四時間後、融解後回収率および生存率を測定した。融解の二十四時間後には、DCの大半はCCR7+となることが分かる。CCR7+ DCは、CD80、CD83、およびCD86にもポジティブであった。三回の別々の実験結果を表IVに示す。
PME-CD40Lで成熟したDCを、総増幅RCC RNAおよびCD40L RNAを用いる同時トランスフェクションの二十四時間後のケモカインCCL19およびCCL21に応答する遊走について分析した。図12は、四人の独立した提供者を用いると、PME-CD40Lで成熟したDCは遊走性が高く、PME-CD40L工程を用いるエレクトロポレーションの二十四時間後に達成された極めて高いレベルのCCR7発現と一致することを示している(表IV参照)。
「CD40Lベース法」による一次免疫応答が誘導されるにも拘わらず、DCをTNF-α、IFN-γ、およびPGE2で最初に成熟させた後、CD40L + 抗原コードmRNAを用いるエレクトロポレーションを行う「成熟後エレクトロポレーション-CD40L」法では、MART抗原モデル系を用いるCTL活性が有意に向上する(図13)。さらに、PME-CD40L工程を、腎細胞癌患者に由来する完全に自己由来の材料を用いるIFN-γおよびIL-2応答の誘導について試験した。患者DCは、PME-CD40L工程について前記した方法で調製し、自己由来の全増幅RCC腫瘍RNAでエレクトロポレーションした。抗原を負荷したDCを自己由来の患者CD8T細胞と培養し、生成するレスポンダーCTLを、誘導されるDC、および腫瘍関連抗原、hTERT、スルビビン(Survivin)、およびRCC特異性抗原G250でトランスフェクトした個々のDCに応答する細胞内サイトカイン染色によって検討した。eGFPコードmRNAでトランスフェクトしたDCは、ネガティブコントロール刺激因子として用いた。図14は、患者T細胞が全増幅RCC RNAを負荷したDC、および三種類の腫瘍関連抗原にも応答し、eGFP mRNAでトランスフェクトしたネガティブコントロールによって誘導されるより高いIFN-γおよびIL-2の両方の出現頻度を有することを示す(それぞれのRCC関連DC標的に対する全応答から差し引いたeGFPに対する応答)。
MART-1 mRNAを負荷したCD40Lベース法によって成熟し、KRN7000でパルスを加えたDCは、CD1d/KRN7000-四量体染色によって定義されるように、KRN7000の代わりにビヒクルでパルスした同じ成熟したRNAを負荷したDCと比較してPBMC培養物中のNKT細胞の出現頻度を増加する(図15a)。レスポンダーCTL(MART-1/HLA-A2)について四量体分析を用いると、KRN7000でパルスを加え、MART-1 mRNAでトランスフェクトしたDCが存在するすることにより、MART反応性T細胞の出現頻度が有意に増加する(図15b)。従って、PBMC培養物中のNKT細胞の増殖により、NKT細胞由来の「補助」によって達成されると思われる増幅ループが提供され、これにより一次CD8 CTL成長を支持することができる。
成熟の好ましい道筋を示す最初のDC実験で用いたCD40L RNAを、プラスミド鋳型pCR2.1 CD40L WTから転写した。好ましいCD40L RNAは、ARCAキャップ類似体とポリA尾部を含む。プラスミドpCR2.1 CD40L WTを、イニシエーターATGコドンの5'に位置したXbal - EcoRV断片を除去することによって修飾した。この断片は、ベクター配列の32ヌクレオチドを含み、三個の曖昧な潜在的イニシエーターATGコドンを含んでいる。この修飾の理論的根拠は、これらの追加のATGが、正確なCD40L翻訳開始部位と競合することによってCD40L翻訳の効率的開始を妨げることであった。CD40Lのコード配列は、これらの修飾によって影響を受けないままであった。修飾したプラスミド鋳型から転写したCD40L RNAは、IL-12発現の誘導によって測定されるように、二つの独立したDCトランスフェクション実験における現行のCD40L参照標準物より良好に機能した。修飾プラスミドは、pCR2.1 CD40L WT δX-Eと表される。
Claims (33)
- 成熟樹状細胞(DC)をインビトロで調製する方法であって、
(a)単離した未成熟樹状細胞(iDC)を、TNF-αであるTNF-αRアゴニストの存在下で、IFN-γであるインターフェロンγ受容体(IFN-γR)アゴニストを含んでなる第一のシグナルによりインビトロでシグナル化して、CD83+成熟樹状細胞を産生し、
(b)前記CD83+成熟樹状細胞を、有効量のCD40アゴニストを含んでなる第二の一過性シグナルによりインビトロでシグナル化して、CD83+CCR7+成熟樹状細胞を産生させること(前記TNF-αRアゴニストおよびIFN-γRアゴニストの第一のシグナルによるシグナル化の開始後の12〜30時間後に、前記第二の一過性シグナルによるシグナル化が開始され、前記第二シグナルがCD40アゴニストをコードするmRNAの翻訳時に生じ、さらに前記CD83 + 成熟樹状細胞が前記mRNAによりトランスフェクトされたものである)
を含んでなる方法。 - 前記iDCまたは前記CD83 + 成熟DCを、PGE2と接触させることをさらに含んでなる、請求項1に記載の方法。
