Description
で示される化合物の工業的合成方法であって、式(III):
で示される7−メトキシ−1−ナフトエ酸を反応に使用し、それを還元剤の作用に付して、式(IV):
の化合物を得て、式(IV)化合物のOH官能基を脱離基に変換し、それをシアン化試薬と反応させ、式(I)の化合物を得て、それを固体の形態で単離することを特徴とする方法に関する。
本発明の方法における式(III)の酸の還元は、有利には、式(III)の酸を酸誘導体、より詳細にはエステルに変換した後に行ってもよく、続いてその酸誘導体を、MeOH、LiBH4、DIBAL−H又はビス(2−メトキシエトキシ)−水素化アルミニウムナトリウム(Red−Al)と会合しているか、又は会合していない還元剤、LiAlH4、NaBH4を用いて、式(IV)のアルコールに還元する。
工程A:(7−メトキシ−1−ナフチル)メタノール
THE(3.8ml/g)中の7−メトキシ−1−ナフトエ酸(100g)1当量に、1M BH3−THF(1.235リットル)2.5当量を徐々に加えた。添加中、反応混合物の温度を5℃(+/−2℃)に保持し、次に混合物を周囲温度で3時間放置した。減圧下で蒸発させた後、残渣をジクロロメタン中に取り、水で洗浄した。有機層をMgSO4で乾燥させ、次に溶媒を減圧下で蒸発させ、標記化合物を、薄茶色の固体の形態で、収率91%で得た。
融点:72〜74℃
工程Aで得た化合物(100g)の、ジクロロメタン(5ml/g)中の溶液に、塩化チオニル(126.35g)2当量を加えた。混合物を加熱還流し、還流を2時間続けた。周囲温度に戻った後、溶媒を減圧下で蒸発させた。得られた油分を酢酸エチル中に取り、水で、次いで飽和NaCl溶液で、洗浄した。溶媒を減圧下で蒸発させ、粗生成物をシリカパッド(溶離剤:ヘプタン)を通して精製した。標記化合物を、黄色の固体の形態で、収率84%で得た。
工程Bで得た化合物(100g)の、DMSO30ml/g及び水5ml/g中の溶液に、シアン化カリウム(37.8g)1.2等量を加えた。反応混合物を65℃に加熱し、65℃で3時間保持した。周囲温度に戻った後、MTBE/水(1/1)二成分系を混合物に加えた。水相を除去した。有機相を水で数回、次に飽和NaCl溶液で洗浄した。溶媒を留去し、標記化合物を定量的収率で得た。
融点:83℃
Claims (5)
- 式(III)の酸の還元をBH3−THFを用いて行う、請求項1記載の式(I)の化合物の合成方法。
- 式(IV)の化合物のOH基が、塩化チオニルの作用により、塩素に変換される、請求項1記載の式(I)の化合物の合成方法。
- 使用するシアン化試薬がシアン化カリウムである、請求項1記載の式(I)の化合物の合成方法。
- 請求項1〜4のいずれか1項記載の合成方法により式(I)の化合物を得て、それを還元に付し、次に無水酢酸とカップリングさせることを特徴とする、式(I)の化合物から出発するアゴメラチンの合成方法。
