Description
本発明は、変化した空間配置を有するコンセンサスインターフェロン(rSIFN‐co)(「組換え高効率複合インターフェロン」とも言う)またはその等価物を提供する。本発明は、同じ一次配列を持つタンパク質が異なる生物学的活性を有する可能性を明らかにする。本出願で説明するように、同一のアミノ酸配列を持つタンパク質は異なった活性を有する。これらのタンパク質の有効性は時に高まることもあり、そして、変化した空間配置を有するタンパク質の新しい機能が明らかになることもある。Wei(2005年;公開番号WO2005/02177 A2; 国際出願番号 PCT/US2004/028068)は、タンパク質の空間配置を調整する一連の方法を提供している。
前記組換え高効率複合インターフェロン(rSIFN‐co)は、他のインターフェロンと比べて副作用が少ない。そのためインターフェロン治療が必要な患者に、より大量に投与できる。またこれらの副作用が少ないrSIFN‐coを他の病気の予防及び/または治療に用いることもできる。故に、本発明は患者に投与する時に、より副作用が少ない前記組換え高効率複合インターフェロン(rSIFN‐co)を提供する。
(人工遺伝子)
本発明はまた、高効率複合インターフェロンまたはその等価物をコードする人工遺伝子を提供する。人工遺伝子を設計することは、通常の技術の範囲内である。ヌクレオチド配列を生成する様々な方法や他の分子生物学技術は、従来、述べられている。以下の例、Joseph Sambrook and David W. Russell, 分子クローニング:研究室マニュアル、2000年12月、Cold Spring Harbor Laboratory Press出版を参照。

(a)前記インターフェロンをコードする核酸分子を、発現用の優先コドンにより適切な宿主に導入する。
(b)導入宿主を前記インターフェロンの発現が可能な状態にする。
本発明は、組換え高効率複合インターフェロン(rSIFN‐co)を生産する方法を提供し、さらに発現したインターフェロンの再生を含む。
上記に述べた組換え高効率複合インターフェロン(rSIFN‐co)は、特別なプロモーター、エンハンサー、または他の調節要素を用いる高効率の発現系により生産されてもよい。一実施形態では前記プロモーターは誘導性である。前記誘導プロモーターはPBAD、熱ショックプロモーターまたは重金属誘導性プロモーターを含むが、それに限定されるものではない。熱ショックプロモーターは物理的手段により活性化され、一方で他のプロモーターは、例えばIPTGやテトラサイクリンといった化学的手段により活性化される。IPTGは下流の遺伝子を活性させるため細胞に加えられるか、遺伝子を不活性化させるため除去される。テトラサイクリンはプロモーターを誘発するため、またはプロモーターの強度を制御するために利用される。

結果的に、本発明及び医学的治療のための、多量の高精製であるとともに空間配置が変化したrSIFN‐coタンパク質は、封入体の変性と再生、そして一連の精製工程から得られた。前記精製は精製タンパク質の生物活性に影響は及ぼさない。
本発明は、患者のウイルス性疾患あるいは腫瘍を、治療もしくは予防する方法を提供する。前記方法は、患者に対し、組換え高効率複合インターフェロン(r-SIFN-co)またはその等価物を有効量投与する段階からなる。Wei (2005; 国際公報番号 WO 2005/034853A2号)は、患者の重症急性呼吸器症候群を、予防もしくは治療する方法を提供する。
組換え高効率複合インターフェロン(rSIFN−co)はDNA複製とB型肝炎のB型肝炎表面抗原及びB型肝炎e抗原の分泌を抑制する。
本発明は、患者の重症急性呼吸器症候群または、ウイルス誘起の上部呼吸器感染疾患を予防もしくは治療する方法を提供する。前記方法は、患者に対し、組換え高効率複合インターフェロンの有効量または組換え高効率複合インターフェロンの機能的等価物の有効量を患者に投与する段階を備える。上記方法の一実施形態として、インターフェロンは、α、β、γ、εまたは、これらの組み合わせである。
ウイルス性上部呼吸器感染疾患は、別の病名として、感冒、風邪と呼ばれる。これは上気道の伝染性のウイルス感染症であり、この伝染性の感染症は粘膜の炎症、くしゃみ、そして咽頭炎をもたらすといった特徴を有している。通常、この感染症は、200以上の異なるウイルスによって引き起こされる。これらの200以上の異なるウイルスは、ライノウイルスであることが知られている。風邪は、インフルエンザを引き起こすウイルスと同一のウイルスによって、引き起こされるのではない。風邪を患う人の咳の飛沫あるいはくしゃみの飛沫、もしくは風邪を患う人によって汚染された物に接触することで、風邪は広まる。風邪をひくのは、子供が最も多く、年齢を経るに従って、減少する。これは、風邪を引き起こすウイルスに対する免疫が、病気になった後、獲得されるからである。成人になると、次第に、風邪を引き起こす幅広いウイルスに対する免疫が発達していく。子供は1年に10回風邪をひくが、大人は1年に3回風邪をひく。

約70〜80%のURIはRSウイルス(respiratory Syncytial virus)、アデノウイルス、ライノウイルス、コクサッキーウイルス(cox-sackie virus)、コロナウイルスやその変異体、A型インフルエンザウイルスやその変異体、B型インフルエンザウイルスやその変異体、パラインフルエンザウイルスやその変異体、あるいはエンテロウイルスやその変異体などのウイルスによって引き起こされる。大人の発症するURIの主なる原因は、ライノウイルスによるものである。子供の発症するURIの主要な2つの原因は、RSウイルスとパラインフルエンザウイルスによるものである。
感受性細胞表面に付着させ、抗ウイルスタンパクの産出を誘導し、細胞内でウイルスの複製及び増殖を遮断する。
組換え高効率複合インターフェロン(rSIFN−co)は免疫応答を調整する。この応答には、T細胞の免疫応答、NK細胞の活性、モノカリオンの食作用、さらに生体内での抗体産出が含まれる。
四川省(中華人民共和国における省)におけるSARSの予防及び制御に取り組む作業グループの同意を得て、組換え高効率複合インターフェロン(rSIFN−co)の配付が2003年5月に始まった。組換え高効率複合インターフェロンのスプレーは、SARSに感染する危険性が高い人口集中地域における病院の医者や看護士に割り当てられた。また、前記スプレーは、SARSの予防及び制御に取り組む国立研究グループに割り当てられた。2003年12月19日現在、3000人のスプレー使用者の中で、スプレー使用に関連した副作用の報告はなかった。さらに、この組換え高効率複合インターフェロンのスプレーを使用した医者や看護士、四川省の人、あるいは他の組織の人は、誰もSARSには感染しなかった。
この組換え高効率複合インターフェロン(rSIFN−co)は次に示す癌もしくは腫瘍の抑制、予防、または治療に対し有用である。
本発明はまた、上記過程を用いることで、組換え高効率複合インターフェロンの生成を提供する。
以下に示すのは、いくつかのrSIFN−coの製剤である。錠剤、カプセル、経口摂取用の液体、ペースト剤、注射、スプレー、座薬、そして液剤があげられる。薬を皮下注射、あるいは静脈注射することが一般的である。この薬担体は、いかなる許容可能な薬担体であってもよい。例えば、炭水化物、セルロース、接着剤、崩壊剤、皮膚軟化剤、充填剤、付加溶解剤保存料、増粘剤、整合剤などがあげられる。
(ペグ化)
ペグ化は、ポリエチレングリコールの分子鎖が、タンパク薬物とペプチド薬に結合することにより薬物動態を向上させる過程である。この向上は、前記タンパク薬物やペプチド薬をタンパク分解酵素から遮断することにより達成される。Harris and Chess, Effect of pegylation on pharmaceuticals. Nat Rev Drug Discov. 2003 Mar; 2 (3):214-21を参照のこと。
持続放出の送達マトリクス及び持続放出の送達リポソームは、rSIFN−coとともに用いられ、持続放出製剤及び制御放出製剤を生成する。Robinson and Talmadge, Sustained Release of Growth Factors. In Vivo 2002 Nov-Dec; 16(6):535-40を参照。著者らは、ペグ化と持続放出の送達マトリクス及び持続放出の送達リポソームの両方が、組み換え分子の薬物速度論的特性と薬理学的特性を改善すると述べている。このようにしてこれらの臨床上の有用性を向上させることで、上記研究が患者の薬剤服用順守を増大させると述べている。
IFN−conは、新しいインターフェロン分子であって、遺伝子工学法を用いて、ヒトIFN−αのサブタイプの保存アミノ酸に準じて構築された。IFN−conは、広域スペクトラムIFN活性を持つとして立証されている。前記IFN活性とは、特にC型肝炎の治療に効果的である高い抗ウイルス性、腫瘍抑制活性などを示す。
純粋なrSIFN−coのタンパク質を得るために、rSIFN−coのcDNAを、大腸菌の高発現ベクターにクローニングした。そして、ベクターの強力なPBADプロモーターを活性化できるL−アラビノースを用いて、rSIFN−co遺伝子の高発現を誘発した。
(エボラウイルスによる組換え高効率複合インターフェロン(rSIFN−co)の効果)
背景:エボラウイルスは、恐ろしい症状を引き起こす致死性のウイルスとして有名であり、最も顕著な症状は、高熱と大量の内出血である。エボラウイルスは、感染者の90%をも死に至らしめる。エボラウイルスは、出血(血液)熱を引き起こす能力を有するウイルスの一つである。この病気には具体的な治療法はない。現在、患者は支持療法を受けている。この療法は、患者の体液と電解質の平衡を保ち、酸素濃度と血圧を維持し、合併症を起こさないように治療する方法からなる。患者は症状の発現から10日以内に死に至る。
1.1 薬剤:四川バイオテクノロジーリサーチセンター(Sichuan Biotechnology Research Center)提供のrSIFN−co
1.2 ウイルス:軍事医学科学院微生物流行病研究所(Military Medical Science,Institute of Microbiology Epidemiology)提供のエボラウイルス
1.3 実験の安全性レベル:ウイルス実験は、生物安全レベル3実験室で実施された。
1.4 動物:60匹のBALB/Cマウス
2.1 60匹のマウスは、無作為に6群に分類され、各群は、10匹のマウスからなる。群1には、エボラウイルスの接種当日に、rSIFN−coを1μg与えた。群2には、エボラウイルスの接種から1日後に、rSIFN−coを1μg与えた。群に3は、エボラウイルスの接種から2日後に、rSIFN−coを1μg与えた。群に4は、エボラウイルスの接種から3日後に、rSIFN−coを1μg与えた。群5には、エボラウイルスの接種から4日後に、rSIFN−coを1μg与えた。群6には、rSIFN−coを与えず、これを対照群とした。
2.2 薬剤の投与:1μgのrSIFN−coを、6日間連続して、1日に1回投与した。
群6(対照群)の10匹のマウス全てが死亡した。群1、群2、群3の全てのマウスが、毒作用は観察されずに生存した。群4、群5においては、いくらかの毒作用が見られた。
結論
これらの結果から、エボラウイルスに対するrSIFN−coの有効性が明確に示された。
(組換え高効率複合インターフェロン(rSIFN−co)の抗HIV効果)
材料
1.1 野生型HIV

1.7 96ウェルプレート、NUNC製
1.8 二酸化炭素インキュベーター
1.9 層流フード
1.10 蛍光光度計
1.11 UV吸光光度計
1.12 その他
2.1 指数増殖期(Log)の293−CD4−CCR5細胞を得て、0.25%のパンクレアチンで消化し、トリパンブルー染色液で染色することにより細胞数を決定し、DMEM培地で、1ミリリットル当たり2.0×105個の細胞濃度(cell/ml)まで希釈した。
2.2 96ウェルプレートのそれぞれのウェルに、100μl(マイクロリットル)の293−CD4−CCR5DMEM培地の懸濁液を注入した。該プレートを5%二酸化炭素インキュベーターで37℃下に放置した。次の日、ウェルの基底面積の70%が再生していることが観察された。
2.3 浮遊物を取り除いた後、様々な濃度のrSIFN−coを、100μl(マイクロリットル)ずつそれぞれのウェルに注いだ。リン酸緩衝生理食塩水(PBS)用と培養基用として、対照群を2群用いた。
2.4 前記プレートを二酸化炭素インキュベーターで37℃下に、約18〜20時間放置した。
2.5 試験ウェル:異なる濃度の野生型HIVと薬剤耐性HIVウイルスをそれぞれのウェルに100μl(マイクロリットル)ずつ注入した。
対照ウェル:ウイルスは添加せず、それぞれのウェルに100μl(マイクロリットル)ずつDMEM培地のみを注いだ。
2.6 前記プレートを二酸化炭素インキュベーターで37℃下に、約24時間放置した。
2.7 ルシフェラーゼルーチン分析が行われ、浮遊物のタンパク質濃度が測定された。ルシフェラーゼはRLU/mg単位により測定された。
rSIFN−coは、1ミリリットル当たり4ナノグラム(ng/ml)以上の量でHIVを抑制できる。表4と図14−15を参照。エクセル(Excel)を用いて、ルシフェラーゼをY軸、rSIFN−co濃度をX軸として用いた場合、1ミリリットル当たり4ナノグラム(ng/ml)以上の量である時、ルシフェラーゼ活性がPBSと培地の対照群よりも明らかに低いことが明確に示されている。明らかな用量依存性の逆応答が示されている。

1.3 軍事医学科学院微生物流行病研究所 分子生物学部門(Molecular Biology Department of Institute of microbiology and epidemiology,Academy of Military Medical Science)提供のインフルエンザウイルス
1.4 DMEM培地 Gibco製
1.5 新生ウシ胎仔血清
1.6 96ウェルプレート、NUNC製
1.7 二酸化炭素インキュベーター
1.8 層流フード
1.9 倒立顕微鏡
1.10 その他
2.1 指数増殖期(Log)中の10日齢のニワトリ胚膜細胞を得て、0.25%のパンクレアチンで消化し、トリパンブルー染色液で染色することにより細胞数を決定し、DMEM培地で、1ミリリットル当たり2.0×105個の細胞濃度(cell/ml)まで希釈した。
2.2 96ウェルプレートのそれぞれのウェルに、100μl(マイクロリットル)の293−CD4−CCR5DMEM培地の懸濁液を注入した。該プレートを二酸化炭素インキュベーターで37℃下に放置した。次の日、細胞は単層細胞まで成長した。
2.3 浮遊物を取り除いた後、様々な濃度のrSIFN−coを100μl(マイクロリットル)ずつ、それぞれのウェルに注いだ。rSIFNを添加しない対照群を2群用いた。
2.4 前記プレートを二酸化炭素インキュベーターで37℃下に、約18〜20時間放置した。
2.5 試験ウェル:異なる濃度のインフルエンザウイルスをそれぞれのウェルに100μl(マイクロリットル)ずつ注入した。
対照ウェル:インフルエンザウイルスは添加せず、それぞれのウェルに100μl(マイクロリットル)ずつDMEM培地のみを注いだ。
2.6 前記プレートを二酸化炭素インキュベーターで37℃下に、約24時間放置した。
2.7 倒立顕微鏡により細胞を観察した。
3.1 倒立顕微鏡によると、インフルエンザウイルスを添加してインターフェロンを加えていない対照ウェルにおける細胞は、明らかな細胞変性効果(CPE)を有していた。前記CPEは、細胞の円形化、細胞壊死、反射光の減少及び細胞の剥離などを示す。
3.2 試験ウェルにおける細胞は、1ミリリットル当たり10ナノグラム以上の濃度であるrSIFN−coを含んでおり、CPEを有しておらず、正常細胞に匹敵する形態であった。図16を参照。
3.3 インフルエンザウイルスとインターフェロンが入っていない対照ウェルは、全くCPEを有していなかった。
rSIFN−coは、1ミリリットルあたり10ナノグラム(ng/ml)以上の濃度でインフルエンザウイルスに対して効果的である。
(肺癌患者の臨床的観察)
患者には、2005年10月7日に非小細胞肺癌が認められた。化学療法による治療を1ヶ月行ったが、2005年10月22日と2005年10月25日の造影検査では、改善は見られなかった。2006年1月4日の検査においても向上は見られなかった。患者は高効率複合インターフェロンにより1ヶ月治療した。投与計画は次のとおりである。患者は1日おきに注射を受けた。1回目は9マイクログラムの用量が与えられた。その後の注射の用量は全て2倍の18マイクログラムとした。2006年2月23日の検査では急速な改善が示された。
Claims (3)
- エボラウイルスによって引き起こされるウイルス性疾患を処置するための、ナノグラム量からマイクログラム量の有効量の組換えインターフェロンを含有する組成物であって、前記組換えインターフェロンは、下記で表わされるアミノ酸配列を有し、および下記で表わされる配列を有するポリヌクレオチドによってコードされることを特徴とする組成物。
- 前記組換えインターフェロンが、経口投与、静脈注射、筋肉注射、腹腔注射、皮下注射、鼻腔もしくは粘膜への投与、または呼吸機で吸入することにより投与されることを特徴とする請求項1に記載の組成物。
- 前記組成物は、さらに薬剤を含有し、または薬剤によりカプセル化され、および前記薬剤が、ポリエチレングリコール(PEG)であることを特徴とする請求項1に記載の組成物。
