Description
本発明は、新規かつ有用なヒト抗体κ型軽鎖を提供する。本明細書において、「ヒト抗体κ型軽鎖」は、ヒト由来の免疫グロブリンのκ型の軽鎖(Light chain)を指す。
1つの局面において、本発明は、本発明に係るヒト抗体κ型軽鎖または本発明に係るポリペプチドをコードする遺伝子を提供する。本明細書中で使用される場合、「ヒト抗体κ型軽鎖をコードする遺伝子」は、ヒト抗体κ型軽鎖またはそのフラグメントであるポリペプチドをコードするポリヌクレオチドが意図される。
・配列番号配列番号2、4、6、8、21、25、29、34、39、49もしくは53の何れかの塩基配列において、1または数個の塩基が置換、欠失または付加されている塩基配列によって示されるポリヌクレオチド;
・配列番号2、4、6、8、21、25、29、34、39、49もしくは53の何れかの塩基配列によって示される相補鎖と、ストリンジェントな条件下でハイブリダイズし得るポリヌクレオチド;または
・配列番号2、4、6、8、21、25、29、34、39、49もしくは53の何れかの塩基配列において、システインをコードする末尾のTGTが、他のアミノ酸をコードする塩基配列に置換されたポリヌクレオチド
であり得る。
本発明はまた、抗ウイルス剤を提供する。本明細書において、抗ウイルス活性とは、ウイルスの感染性、増殖能または免疫回避能を低下させる活性を意味する。ウイルスの感染性とは、ウイルスが宿主細胞に吸着または侵入する性質を意味する。このときの抗ウイルス活性は、例えば、ヒト型抗体酵素の活性によってウイルス粒子の表面タンパク質を、少なくとも部分的に切断または分解して、宿主細胞に対するウイルスの吸着または侵入を抑制する活性を示す。つまり、当該抗ウイルス活性は、ウイルスの中和活性と言い換えることができる。
本発明者らは、酵素活性を有するマウスのモノクローナル抗体(抗体酵素)の配列および立体構造を解析することによって、抗体酵素はκ型の軽鎖を有し、当該軽鎖に触媒三つ組残基様構造を有している場合が多いことを見出した。なお、触媒三つ組残基様構造とは、例えば、セリン残基、ヒスチジン残基およびアスパラギン残基によって形成された、触媒活性を有していると考えられる構造である。
本発明は、ポリヌクレオチドの製造方法を提供する。一実施形態において、本発明に係るポリヌクレオチドの製造方法は、上述したプライマーセットを用いた2段階のPCR反応により、ヒトcDNAからヒト抗体のκ型軽鎖の可変領域を少なくともコードするポリヌクレオチドを増幅する工程を包含している。用いるプライマーの設計、PCR反応の条件、鋳型cDNA等は、上述したとおりである。
本発明にかかるポリヌクレオチドの製造方法によって製造したポリヌクレオチドは、サブグループIIに属するVκ遺伝子を有する抗体軽鎖遺伝子または可変領域を少なくともコードするそのフラグメントであるので、適当な宿主(例えば細菌、酵母)に導入して、サブグループIIに属するVκ遺伝子を有する抗体軽鎖または可変領域を少なくとも含むそのフラグメントを発現させることができる。
すなわち、本発明に係る抗ウイルス剤は、可変領域が配列番号26、14、22、30、50、54または35のアミノ酸配列によって示されるポリペプチドからなるヒト抗体κ型軽鎖を含有することを特徴としている。
(1−1.プライマーの設計)
図1に示すように、1段階目のPCR反応のためのフォワードプライマーおよびリバースプライマー、2段階目のPCR反応のためのフォワードプライマーおよびリバースプライマーを設計した。
狂犬病ウイルスのワクチンを用いて、複数回にわたって過剰免疫されたボランティアから得られた末梢血から、Ficoll−paqueを用いてリンパ球を分離した。RNA extraction kit(Stratagene)を用いて、分離した約3.0×107個のリンパ球からトータルRNAを得た。TheromoScript RT−PCR System(Invitrogen)を用い、oligo(dT)をプライマーとして、トータルRNAを逆転写することにより、目的とするcDNA(cDNAライブラリ−)を調製した。
1段階目のPCR反応では、1−2.で調製したヒト末梢血cDNAを鋳型として使用した。フォワードプライマーとしては、塩基配列が、agttCCATGGAGCTTCTGGGGCTGCTAATG(配列番号9)であるオリゴヌクレオチドを用いた。なお、5〜10番目(CCATGG)は、制限酵素サイトである。リバースプライマーとしては、塩基配列が、ccgtCTCGAGACACTCTCCCCTGTTGAAG(配列番号10)であるオリゴヌクレオチドを用いた。なお、5〜10番目(CTCGAG)は、制限酵素サイトである。プライマーの詳細を表1にまとめた。
2段階目のPCR反応では、1−3.で得た一段階目のPCR産物を鋳型として使用した。フォワードプライマーとしては、塩基配列が、agttCCATGGATRTTGTGATGACYCAG(配列番号11)であるオリゴヌクレオチドを用いた。なお、5〜10番目(CCATGG)は、制限酵素サイトである。リバースプライマーとしては、塩基配列が、ccgtCTCGAGACACTCTCCCCTGTTGAAG(配列番号10)であるオリゴヌクレオチドを用いた。なお、5〜10番目(CTCGAG)は、制限酵素サイトである。プライマーの詳細を表2にまとめた。
1−4.における二段階のPCR反応により得られた約750bpのPCR産物(サブグループIIに属するκ型軽鎖遺伝子)をpCR Blunt II−TOPOベクターへ組み込んだ。上記PCR産物(約750bp):0.5μl、食塩水:0.5μl、ミリQ水:1.5μl、およびTOPOベクター:0.5μlから、総量2.5μlの反応液を調製し、23℃で5分間反応させた。
33.3μlのコンピテントセル(大腸菌DH5α)に、サブグループIIに属するκ型軽鎖遺伝子をクローニングしたTOPOベクターを2μl加え、氷中に10分間静置した。次に、42℃のウォーターバスで45秒間ヒートショックを行い、直ちに氷中に戻して2分間静置した。その後、クリーンベンチ内において、300mlのSOC培地を加え、37℃で1時間、震とう培養した(復活培養)。
20個のコロニー(コロニー#1〜#20)について、インサートを確認した。まず、定法を用いて菌体からプラスミドを回収した。回収したプラスミド:1.0μl、10×Hバッファー:1.5μl、Eco RI:0.3μl、および滅菌ミリQ水:12.2μlからなる総量15.0μlの反応液を調製して、37℃で一晩制限酵素反応を行った。その後、2%アガロースゲル(NuSieve GTGアガロース)を用いて電気泳動を行った。図4に示すように、コロニー#1〜#16、#18、#19について、目的遺伝子の挿入を確認した。以降、これらをクローン#1〜#16、#18、#19と称する。
クローン#1〜16、#18および#19についてシーケンス解析を行い、相同性検索により、それぞれの生殖細胞系列遺伝子におけるVκ遺伝子を判定した。結果を表3に示す。得られた18クローンすべてがサブグループIIに属していた。
クローン#1、クローン#16、クローン#7、およびクローン#11をそれぞれ、Hisタグ配列サイトを有するプラスミドに導入した。濃度5ng/μLに調製した上記プラスミドDNA1μlを、氷上にて融解させた50μLのBL21(DE3)pLysSに加え、5分間にわたって氷上に静置した。その後、42℃のウォーターバスにて30秒間インキュベーションした後、2分以上氷上に静置した。クリーンベンチ内において、さらに、37℃に温めておいた250μLのSOC培地を加え、ラウンドチューブに移し替えて、37℃、200rpmにて1時間振とう培養(復活培養)した。復活培養の後、プレートに50μLまたは10μLの培養液を播いて、37℃で一晩インキュベーションした後、プレート上に形成されたコロニーの個数をカウントした。
1−9.におけるヒートショックによる形質転換とは別に、エレクトロポレーションによる形質転換についても実施した。50μLのコンピテントセルに5μLの上記プラスミドを加えた後に、これをキュベットに素早く移して、1分間氷上に放置した。エレクトロポレーターを2.5kVに設定し、キュベットをエレクトロポレーターに配置してパルスを印加した。直後に450μLのSOC培地を加え、転倒混和し、2mLのチューブに移して、37℃で1時間振とう培養した。
3mLのLB培地および6μLのアンピシリン(最終濃度100μg/mL)を試験管に入れ、形質転換した菌のグリセロールストックから竹串を用いて菌の一部を試験管に移し、37℃にて一晩前培養した。前培養の後、5mLのLB培地および5μLのアンピシリン(最終濃度5ng/mL)を試験管に入れ、前培養した培養液をLB培地の1/100量(50μL)だけ当該試験管に移し、25℃にて本培養を開始した。本培養は、O.D.660が約0.6〜0.8になるまで続けた。O.D.660が約0.6〜0.8に達した後、0.1MのIPTG50μLを試験管内に加え、37℃にて6時間培養した。6時間後に5mLチューブに培養液を入れ、4℃、18000×gにて10分間遠心した。培地(上清)をデカンテーションによってファルコンチューブに移した。また、菌体(ペレット)に1×PBSを加え、菌体をピペットによって懸濁し、別のファルコンチューブに移した。培地および菌体の各々を凍結して保存した。
また、1−11.において得られた菌体(ペレット)の懸濁液を、粘性がなくなるまで液体窒素を用いて繰り返し凍結融解させた後、14000rpm、4℃にて25分間遠心した。得られたペレットを不溶性画分とし、上清を可溶性画分としてそれぞれ回収した。不溶性画分には、サンプバッファーを加えてペレットを溶解させた。
タンパク質の発現を確認することができたため、続いて、発現されたタンパク質が抗体軽鎖であるか否かを同定するために、抗ヒト型(Fab’)2抗体を用いてウエスタンブロッティングを行った。まず、2−4.において得られたIPTGによる誘導あり(+)およびIPTGによる誘導なし(−)の各々の培地を、トリクロロ酢酸(TCA)沈殿により濃縮し、電気泳動用ゲルの各レーンに流して、SDS−PAGEを行った。続いて、電極を用いてメンブレンへの転写を行い、当該メンブレンをブロッキングした後、抗ヒト型(Fab’)2抗体と免疫反応させた。その後、発色基質液を加え、メンブレンを観察した。結果を図11に示す。図11は、目的タンパク質のバンドを可視化したメンブレンを示す写真である。示すように、図7、図9および図10においてバンドが確認された31kDa付近に、強いバンドが検出された。これにより、大腸菌において発現したタンパク質がヒト型抗体軽鎖であることを同定することができた。
発現タンパク質(ヒト型抗体軽鎖)を、一次精製および二次精製した。
1−14.において精製したクローン#1、#7、#11および#16由来のヒト型抗体軽鎖のそれぞれについて、酵素活性を測定した。酵素活性の測定には、市販されている配列の異なるペプチドにMCA(Methyl−Coumaryl−Amide)を結合させた基質(MCA標識ペプチド)を使用した。MCA標識ペプチドを使用するプロテアーゼ活性試験では、標識ペプチドが切断されて遊離する部分が、切断前の標識ペプチドと異なる波長の蛍光を発することを利用してプロテアーゼ活性を測定する。より詳細には、MCA標識ペプチドでは、蛍光物質であるMCAに隣接するリジン残基のεアミノ基がアセチル化されている。MCA標識ペプチドが、上記リジン残基のカルボキシル末端側において切断されると、ペプチド部分とAMC(Amino−Methyl−Coumarin)とに分かれる。AMCは、切断前のMCAに由来する。遊離したAMCが発する蛍光の波長は、ペプチジル−MCAとは異なるので、AMCによって発せられる蛍光の強度の変化を利用して、どれだけの基質が切断を受けたのかを測定することができる。
サンプルとして、1−14.においてクローン#1、#7、#11および#16から生産し、精製したヒト型抗体軽鎖を用いた。ネガティブコントロールとして、ヒト型抗体軽鎖遺伝子が挿入されていないpET20b(+)からの発現生成物を使用した。試薬などの調製に際しては、滅菌したミリQ水を使用し、使用するマイクロチューブおよびチップは、オートクレーブ処理によって滅菌されたものを用いた。無菌操作は、全てクリーンベンチ内で行った。測定時の励起波長は360nmとし、測定する蛍光波長は465nmとした。
精製した各ヒト型抗体軽鎖の濃縮サンプル(10μM、5μMおよび1μM)を各MCA標識ペプチドに加えた反応液を調製して、25℃の気相インキュベータおよび37℃の気相インキュベータの中で反応させた。
クローン#1、#7、#11および#16由来のヒト型抗体軽鎖のMCA分解試験の結果を図16〜19に示す。
クローン#6およびクローン#18についても、1−9.から1−18.までと同様に、形質転換、発現タンパク質の精製および酵素活性の測定を行った。結果、クローン#6は酵素活性を有しておらず、クローン#18は酵素活性を有していた。
以下のような手順により、LCAライブラリおよびLC2ライブラリの二つのライブラリを構築した。
狂犬病ウイルスのワクチンを用いて、複数回にわたって被験者を過剰免疫し、血清の中和活性を測定した。血清の中和活性が最も高かった(7.2IU)被験者をドナーとして末梢血を採取し、Ficoll−paqueを用いて当該末梢血からリンパ球を分離した。RNA extraction kit(Stratagene)を用いて、分離した約3.0×107個のリンパ球からトータルRNAを得た。TheromoScript RT−PCR System(Invitrogen)を用い、oligo(dT)をプライマーとして、トータルRNAを逆転写することにより、後述するPCR反応において鋳型とするcDNAを調製した。
まず、図28に示すようなプライマーを設計した。詳しくは、NCBIのIgBLAST(http:/www.ncbi.nlm.nih.gov/igblast/)に登録されているヒト抗体軽鎖遺伝子の配列情報に基づき、これらのヒト抗体軽鎖遺伝子を網羅的に増幅するためのプライマーセットとして、5’(フォワード)プライマー20種類と、3’(リバース)プライマー1種類とからなる計20ペアのプライマーセットを設計した。5’プライマーは、ヒト抗体軽鎖のV領域のN末端領域に対応する塩基配列を有するオリゴヌクレオチドとし、3’プライマーは、ヒト抗体軽鎖の定常(C)領域のC末端領域に対応する塩基配列に対する相補配列を有するオリゴヌクレオチドとした。なお、5’プライマーには、大腸菌発現ベクターpET101/D−TOPOベクター(登録商標、Invitrogen)に挿入するための4塩基(CACC)を付加した。
サブグループIIに属するVκ遺伝子を有するヒト抗体軽鎖遺伝子のみを増幅するために、5’プライマーとして、サブグループIIに対応するプライマー(表4に示すVk2aTOPO、Vk2bATOPO、Vk2bGTOPOおよびVk2cTOPOの4種類)を用い、3’プライマーとして、VCR2862を用いて、2−2.と同様の手順でPCR反応を実施した。
構築した2つのライブラリ(LCAライブラリ、LC2ライブラリ)のそれぞれについて、大腸菌TOP10を形質転換した。TOP10内で、プラスミドを十分に増幅させた後、菌体を破壊してプラスミドを回収した。回収したプラスミドを精製した後、抗体軽鎖の効率のよい発現系である大腸菌BL21の形質転換に供した。得られた形質転換体から、ライブラリ毎に384クローンをランダムに選び(計768クローン)、以下のスクリーニングに供した。
以下のように第1スクリーニングを実施した。具体的には、各大腸菌クローンを150μlのLB培地において培養し、その上清について、ELISA法により、抗体軽鎖の発現および狂犬病ウイルス抗原2種類に対する結合活性を測定した。測定結果に基づき、LCAライブラリから20クローン、LCライブラリから23クローン、計43クローンを、狂犬病ウイルス抗原2種類に対する結合活性がみられるクローンとして選択した。
上記7クローンについて、100mlの培養系で培養を行った。抗体軽鎖を発現させた菌体を、凍結融解を繰り返して破砕し、遠心分離して上清を回収した。回収した上清を、抗体軽鎖を含むサンプルとして、発現ベクター由来のHisタグを利用して抗体軽鎖を粗精製した。精製は、オープンカラムに担体(Ni Sepharose TM6 fast Flow)を充填し、緩衝液を自然落下させることにより実施した。カラムの平衡化、バインディング、およびサンプルアプライ後のウォッシュのための緩衝液の組成としては、20mMリン酸ナトリウム、0.5M塩化ナトリウム、および20mMイミダゾール(pH7.4)を用いた。溶出のための緩衝液の組成としては、20mMリン酸ナトリウム、0.5M演歌ナトリウム、および500mMイミダゾール(pH7.4)を用いた。溶出後、タンパク質溶出画分を、PBS(−)によって一晩透析した後、分画分子量10000の限外ろ過膜(MILLIPORE)を用いて遠心操作により濃縮して粗精製品とした。粗製製品について、SDS電気泳動およびクーマシーブリリアントブルー染色によって確認した(図31)。図31中、四角枠で示したバンドは、低分子量側が抗体軽鎖のモノマーを、高分子量側が抗体軽鎖のダイマーを示すと考えられる。なお、抗体軽鎖モノマーの分子量は、タグ等が付加されているため、約27kDaとなった。
得られた粗精製品のうち、精製の際に沈殿を形成しなかったものについて、ELISAプレートに固定した狂犬病ウイルス抗原(狂犬病ウイルス標品(αCVS)および精製ニワトリ胚細胞狂犬病ワクチン(αPECE))との親和性(kd)を測定した。
(3−1.クローン#1由来のヒト型抗体軽鎖の精製)
評価に用いるクローン#1由来のヒト型抗体軽鎖を以下の様に一次精製および二次精製した。図20の(a)は、一次精製におけるNi−NTAカラムクロマトグラムおよびSDS−PAGE分析の結果を示す図である。図20の(b)は、二次精製における陽イオン交換クロマトグラムおよびSDS−PAGE分析の結果を示す図である。
続いて、ヒト型抗体酵素およびウイルスを用いたウイルスの中和試験によって、ヒト型抗体軽鎖の抗ウイルス活性を調べた。試験に用いたウイルスは、狂犬病ウイルス(以下、RABVと記載)のCVS−11株(以下、CVSと記載)、ERA株(以下、ERAと記載)およびHEP−Flury株(以下、HEPと記載)、水疱口内炎ウイルス(以下、VSVと記載)ならびにレオウイルス(以下、ReoVと記載)である。ウイルスを感染させる細胞として、RABV用にNA細胞、他のウイルス用にL929細胞を用いた。
固定した細胞を洗浄し、クリスタルバイオレットを各ウェルにいれて細胞を染色した。細胞の染色後にクリスタルバイオレットを捨て、ウェルを水洗した。各ウェルを肉眼または実態顕微鏡によって観察し、ウイルス感染によって細胞がウェルから剥離して、染色されていない「抜け」をプラークとして計測した。PBSおよびウイルスの混合液をかけたウェルに形成されたプラーク数を100%として、ヒト型抗体酵素およびウイルスの混合液をかけたウェルに形成されたプラーク数のパーセンテージを算出した。結果を図21に示す。図21の(a)は、ヒト型抗体酵素が高活性を示す温度について評価した図である。図21の(b)は、ヒト型抗体酵素が高活性を示す濃度について評価した図である。
ヒト型抗体酵素とウイルスとのインキュベーションの手順は、3−2.において確認した通りであり、ウイルス感染の手順は、3−3.に記載の通りである。ヒト型抗体酵素は、単量体のみを含むもの、および二量体のみを含むものに分けて、その抗ウイルス活性を調べた。ここでは、3種の株の狂犬病ウイルス株、VSVおよびReoVに対するヒト型抗体酵素の抗ウイルス活性について試験した。3−3.と同様に、プラークアッセイを用いて、PBSおよび各ウイルスの混合液をかけたウェルに形成されたプラーク数を100%として、ヒト型抗体酵素および各ウイルスの混合液をかけたウェルに形成されたプラーク数のパーセンテージを算出した。この結果をまとめたものをグラフ化して図22に示す。
図22において確認したように、本発明に係るヒト型抗体酵素は、ラブドウイルス科のウイルスに高い抗ウイルス活性を示し、ReoVにはほぼ活性を示さなかった。ここで、ラブドウイルス科のウイルスは、宿主細胞の膜に由来するエンベロープを有している。エンベロープウイルスは、エンベロープが壊されると感染性を失ってしまう。もし、本発明に係るヒト型抗体酵素がエンベロープを破壊する活性を有する場合、宿主細胞にも傷害性であり得る。そこで、本発明に係るヒト型抗体酵素の安全性を確認するために、トリ赤血球の凝集作用の有無について調べた。本発明に係るヒト型抗体酵素またはPBSと1%のトリ赤血球浮遊液とを混合して、25℃にて48時間反応させた。その結果を図24に示す。図24に示すように、本発明に係るヒト型抗体酵素によって、赤血球の凝集は起こっていない。よって、本発明に係るヒト型抗体酵素は、膜融合活性を有していないため、宿主細胞を傷害する活性を有していないと考えられる。つまり、本発明に係るヒト型抗体酵素は、ウイルスに特異的なタンパク質に作用して抗ウイルス活性を示す、安全性の高い抗ウイルス剤として使用可能と考えら得る。
上述のように、#1クローン(germline:A18b)のヒト型抗体酵素は、単量体の形態において特に高い抗ウイルス活性を示した。このため、S−S結合を介して二量体の形成に必須と考えられる220番目のシステインに変異を導入して、単量体のヒト型抗体酵素のみが形成されるように、cDNAを設計した。この設計の詳細を図25に示す。図25の(a)に示すように、全長のヒト型抗体酵素の遺伝子における220番目のシステインをコードするTGTを、GCTに置換した。これによって、図25の(b)に示すように、元のアミノ酸配列では220番目がシステインであるために、単量体および二量体が混在していたが、置換後のアミノ酸では220番目がアラニンであるために、S−S結合が形成されず、単量体のみが得られた。アミノ酸置換によるコンフォメーションの変化などの影響を受けて、抗ウイルス活性が変化するおそれがあったが、3−4.において確認された抗ウイルス活性と同様に、この220番目をアラニン置換したヒト型抗体酵素は、ラブドウイルス科に属するウイルスに対する抗ウイルス活性を示した。以下、その試験手順および結果を説明する。
クローン#7由来のヒト型抗体酵素についても、クローン#1由来のヒト型抗体酵素と同様にウイルス抑制効果を試験した。ヒト型抗体酵素とウイルスとのインキュベーションの手順は、3−2.において確認した通りであり、ウイルス感染の手順は、3−3.に記載の通りである。ヒト型抗体酵素としては、単量体の抗ウイルス活性を調べた。ウイルスとしては、CVSウイルスに対するヒト型抗体酵素の抗ウイルス活性について試験した。3−3.と同様に、プラークアッセイを用いて、PBSおよび各ウイルスの混合液をかけたウェルに形成されたプラーク数を100%として、ヒト型抗体酵素および各ウイルスの混合液をかけたウェルに形成されたプラーク数のパーセンテージを算出した。この結果をまとめたものをグラフ化して図27に示す。
クローン23D4由来のヒト型抗体酵素についても、クローン#1由来のヒト型抗体酵素と同様にウイルス抑制効果を試験した。ヒト型抗体酵素とウイルスとのインキュベーションの手順は、3−2.において確認した通りであり、ウイルス感染の手順は、3−3.に記載の通りである。ヒト型抗体酵素は、単量体のみを含むもの、および二量体のみを含むものに分けて、その抗ウイルス活性を調べた。ここでは、3種の株の狂犬病ウイルス株、VSVおよびReoVに対するヒト型抗体酵素の抗ウイルス活性について試験した。3−3.と同様に、プラークアッセイを用いて、PBSおよび各ウイルスの混合液をかけたウェルに形成されたプラーク数を100%として、ヒト型抗体酵素および各ウイルスの混合液をかけたウェルに形成されたプラーク数のパーセンテージを算出した。この結果をまとめたものをグラフ化して図33に示す。
3−5.と同様に、クローン23D4についても膜融合活性を検討した。その結果、図34に示すように、クローン23D4に係るヒト型抗体酵素によって、赤血球の凝集は起こっていない。よって、本発明に係るヒト型抗体酵素は、膜融合活性を有していないため、宿主細胞を傷害する活性を有していないと考えられる。つまり、本発明に係るヒト型抗体酵素は、ウイルスに特異的なタンパク質に作用して抗ウイルス活性を示す、安全性の高い抗ウイルス剤として使用可能と考えら得る。
上述のように、本発明に係るヒト型抗体軽鎖は、単量体の形態において特に高い抗ウイルス活性を示した。このため、各クローンに係るヒト型抗体軽鎖について、3−6.と同様に、S−S結合を介して二量体の形成に必須と考えられるC末端のシステインに変異を導入して、単量体のヒト型抗体軽鎖のみが形成されるように、cDNAを設計した。すなわち、全長のヒト型抗体酵素の遺伝子における上記システインをコードするTGTを、ALEHHHHHH(配列番号12)(+終止コドン)をコードするGCTCTCGAGCACCACCACCACCACCACTGA(配列番号13)に置換した。例えば、クローン23D4については、配列番号32の塩基配列とし、クローン22F6については、配列番号37の塩基配列とし、クローン23F1については、配列番号42の塩基配列とした。
続いて、本発明に係るヒト抗体軽鎖クローンが、インフルエンザウイルスに対して感染抑制能を有するか否かを試験した。インフルエンザウイルスとしては、A/広島/71/2001(H3N2)株を用いた。ウイルスは、孵化後11日目のニワトリ卵の尿膜腔において培養され、感染性の尿膜腔液として、使用時まで−80℃にて保管した。感染させる細胞としては、10%の牛血清が加えられたEagle’s最小培地(MEM)中で培養されたMDCK細胞を用いた。
本発明に係るヒト抗体軽鎖クローンの抗ウイルス活性について、国際標準法を用いてインビボにおける中和試験を行った。すなわち、狂犬病ウイルスCVS(Challenge Virus Strain)と、抗体とをインビトロにて一定時間反応させた後、マウス脳内に反応液を接種し、マウスの生存率から、抗体のウイルス中和能を評価した。
本発明に係るヒト抗体軽鎖クローンの核酸分解活性を試験した。各サンプルは、His−Tag精製の後、陽イオンカラムクロマトグラフィーにより精製したものを用いた。各サンプルの濃度は、後述の表9に示す。基質となる核酸としては、プラスミドDNA(pBR322)を用いた。ネガティブコントロールとしては、反応させていない基質(MasterMix)を用いた。ポジティブコントロールとしては、DNase1を反応させたものを用いた。各サンプルについては、37℃のサーマルサイクラーにおいて、24時間または48時間反応させた。ポジティブコントロールについては、DNase1を30分間反応させた。ネガティブコントロールについては、サーマルサイクラーにおいて、0時間、24時間または48時間インキュベートした。そして、反応後のサンプルに、10×ローディングバッファおよびミリQ水を加え、−30℃で凍結した。その後、アガロースゲル電気泳動を行った。結果の一部を図42(a)に示す。
本発明に係るヒト抗体軽鎖クローンのがん細胞に対する細胞障害性について試験した。まず、前述したような手法を用いて以下のヒト型抗体型鎖を作製した:#1_C220A(#1クローンのモノマー)、#1_dimer(#1クローンのダイマー)、23D4_C220A(23D4クローンのモノマー)、23D4_dimer(23D4クローンのダイマー)、#4_C220A(#4クローンのモノマー)、#9a_C220A(#9クローンのモノマー)、および、#13_C220A(#13クローンのモノマー)。なお、本明細書において、「C220A」は、ジスルフィド結合がされないように220番目のシステインをアラニンに改変して得たモノマーを示し、「dimer」は、ワイルタイプから調製したダイマーを示す。
Claims (7)
- 狂犬病ウイルスに対するヒト抗体κ型軽鎖であって、抗ラブドウイルス活性、および抗インフルエンザウイルス活性を有しており、その可変領域が、配列番号26のアミノ酸配列によって示されるポリペプチドからなることを特徴とするヒト型の抗体酵素。
- 狂犬病ウイルスに対するヒト抗体κ型軽鎖であって、抗ラブドウイルス活性、抗インフルエンザウイルス活性、およびがん細胞傷害性を有しており、その可変領域が、配列番号14のアミノ酸配列によって示されるポリペプチドからなることを特徴とするヒト型の抗体酵素。
- 狂犬病ウイルスに対するヒト抗体κ型軽鎖であって、抗ラブドウイルス活性およびがん細胞傷害性を有しており、その可変領域が、配列番号30のアミノ酸配列によって示されるポリペプチドからなるヒト型の抗体酵素を含有していることを特徴とする抗ラブドウイルス剤。
- 狂犬病ウイルスに対するヒト抗体κ型軽鎖であって、抗インフルエンザウイルス活性、および核酸分解活性を有しており、その可変領域が、配列番号50のアミノ酸配列によって示されるポリペプチドからなることを特徴とするヒト型の抗体酵素。
- 狂犬病ウイルスに対するヒト抗体κ型軽鎖であって、抗インフルエンザウイルス活性を有しており、その可変領域が、配列番号35のアミノ酸配列によって示されるポリペプチドからなることを特徴とするヒト型の抗体酵素。
- 狂犬病ウイルスに対するヒト抗体κ型軽鎖であって、抗インフルエンザウイルス活性を有しており、その可変領域が、配列番号54のアミノ酸配列によって示されるポリペプチドからなることを特徴とするヒト型の抗体酵素。
- 狂犬病ウイルスに対するヒト抗体κ型軽鎖であって、抗ラブドウイルス活性を有しており、その可変領域が、配列番号22のアミノ酸配列によって示されるポリペプチドからなることを特徴とするヒト型の抗体酵素。
