Description
本発明は、1つの態様において、標的または標的フラグメントを細胞培養物または患者から得た標本の中の細胞からインシトゥハイブリッド形成によって直接的に検出することを可能にするものである。好適な態様における細胞は病原体である。本方法は数段階を包含し、これらを必ずしもではないが好適には示す順で実施する。培養物または標本のサンプルをスライドの上に置く。そのサンプルを前記スライドの上に熱または標準的な固定剤で固定する。その固定剤は、例えばメタノール、メタノール酢酸、アセトン、ホルムアルデヒドまたはホルマリンなどであり得る。その固定したサンプルをIDF溶液(以下を参照)で処理し、染色または精査しそして観察する。別法として、当該標本をIDF溶液と混合し、インキュベートした後、ガラススライドの上に塗るか或は他の様式で位置させ、空気で乾燥させた後、固定する。前記IDF溶液を下記の反応体の任意組み合わせで構成させてもよい:カオトロピック塩(例えばグアニジンのチオ硫酸塩もしくは塩酸塩)、イオン性界面活性剤(例えばSDS)および/または非イオン性界面活性剤(例えばIPGEL、デオキシコール酸塩、コール酸塩または胆汁塩)または同様な特性を有する他の反応体、メタノールおよび酢酸。前記IDF溶液に入れる各反応体の濃度は、例えば検出すべき病原体の細胞壁などに依存する。本発明を如何なる理論でも機構でも制限するものでないが、そのようなIDF溶液によって当該病原体の細胞壁および/または膜(細胞および核)の中に「通路」が形成されると考えている。そのような通路によって、プローブが前記細胞壁および細胞膜を通り抜けて当該病原体の細胞質および/または核の中に入り込むことができる。本発明のプローブには、DNA、RNA、PNA、ペプチド、糖ペプチド、リポ蛋白質または糖脂質、または前記いずれかの混合物が含まれ得る。当該サンプルの中の固定した細胞の標的をこの標的に特異的なプローブ複合体(このプローブ複合体は当該標的に特異的な結合因子を含有して成る)とハイブリッド形成または結合に適した条件[例えばShahおよびHarrisの米国特許第6,165,723号(これは引用することによって本明細書に組み入れられる)に記述されている如き]下で接触させる。次に、ハイブリッド形成も結合も起こさなかったプローブを当該サンプルから濯ぎ流してもよい。1つの態様では、その後、その濯いだサンプルに適切な対比染色剤(例えばEvans Blue、DAPI、過マンガン酸カリウムなど)による染色を受けさせてもよい。そのハイブリッド形成または結合したプローブ複合体を例えば顕微鏡などで可視的に検出するが、そのプローブ複合体が存在することは当該細胞標的が存在することの指標である。この方法はいろいろなハイブリッド形成用緩衝液を用いて実施可能であり、それの非限定例をいくつか本明細書に開示するが、それらはShahおよびHarrisの米国特許第6,165,723号(引用することによって本明細書に組み入れられる)にも開示されている。使用するハイブリッド形成用緩衝液は使用するプローブの性質によって決まる。本発明の方法は、細胞、細胞成分、好適には標本の中の病原体を検出しようとする時に有効である。ミコバクテリウム種の病原体を検出しようとする時に用いるに有用な特定の典型的な非限定プローブ複合体を本明細書に開示する。

本発明は、細胞の中の標的である核酸、蛋白質、ペプチド、リポペプチド、糖ペプチド、脂質などの存在を培養物または個体から得た標本(例えば血液塗抹標本、生検、パラフィン包埋組織およびダニなど)からインシトゥハイブリッド形成させて直接検出する目的でプローブが細胞(例えば病原体)の細胞壁および/または細胞膜を透過し得るように改良を受けさせた方法を見いだしたことが基になっている。本発明の方法は、特に、病原体、例えば痰、全血、中枢髄液(CSF)、他の体液または感染した組織などの中に存在する病原体に特異的なヌクレオチド配列を検出するに良好に適する。より具体的には、プローブ(例えばオリゴヌクレオチドプローブ)が感染した宿主細胞の内側または外側のいずれかに存在し得る細胞(例えば病原体、例えば細菌、ウイルス、菌・カビ、酵母菌および原生動物など)の中に入り込むことができるように伝統的な固定/前処理方法に対して行った新規な改良を記述する。加うるに、蛍光標識付きプローブを用いたハイブリッド形成後に対比染色剤(例えばDAPI、Evans Blue、過マンガン酸カリウム)を用いた手順を実施することで、培養物または臨床サンプルの中のハイブリッド形成したプローブを保持している有機体を容易に見ることができるようにもする。
リッド形成に適した条件下で接触させる。
1)ホルムアミドの含有量が約10%から50%で2X.SSC(pH7.4)でNP40の含有量が1%の緩衝液;
2)GuSCN緩衝剤が約1.5Mから4M入っている緩衝液[5MのGuSCNストック緩衝液を5MのGuSCN、100mMのTris−HCl(pH7.8)、40mMのEDTA、1%のNP40で構成させる。このストック緩衝液に1xTE(pH7.8)を添加することによる希釈を実施して示したGuSCNのモル濃度にすることでGuSCN緩衝液のモル濃度をこの上に示した濃度にする];
3)GuHCl緩衝剤が約2から6M入っている緩衝液[8MのGuHClストック緩衝液を8MのGuHCl、200mMのTris−HCl(pH7.8)、40mMのEDTA、1%のNP40で構成させる。このストック緩衝液に1xTE(pH7.8)を添加することによる希釈を実施して示したGuHClのモル濃度にすることでGuHCl緩衝液のモル濃度をこの上に示した濃度にする];
4)ホルムアミド(20−50%)とGuSCN緩衝剤(例えば0.5Mから3M)の混合物で構成されている緩衝液;
5)GuSCN緩衝剤(例えば0.5Mから3M)とGuHCL緩衝剤(例えば1Mから5M)の混合物で構成されている緩衝液。
流体(1)を用いて同じハイブリッド形成を約42℃で実施することでも相当する特異性がもたらされるであろう。
数が少なく、従って短時間に実施可能であるように簡素化したものである。本発明の態様は、また、本発明の組成物を含有して成るキットも包含する。前記組成物をキットの形態で提供することにより、本明細書に示すプロトコルのいろいろな態様を実施することが可能になり、それには、幅広く多様な検出方法に関して簡潔さおよび適合性を最適にしたそれらが含まれる。また、特定的に調製した反応体およびプローブを含有するように調製した前記キットは臨床/診断実験室でも使用可能であると期待し、その場合、特定の核酸が存在するか否かを検出することができることから、特定の標的の存在によって特徴づけられる病気状態が陽性または陰性であるかを識別するに役立つであろう。
この上に示した方法の好適な非限定使用は、ヒト型結核菌を培養物または痰から検出する時の使用である。本分野の技術者は、この新規な方法を興味の持たれる特定の核酸のハイブリッド形成(または標的細胞、組織または病原体の他の細胞成分の検出)の目的で細胞の一体性および形態を保存しながら他の幅広く多様な系、細胞、組織培養物および組織に等しく適用することができることを理解しかつ評価するであろう。
痰を1)NALC/NaOHで処理するか或は2)NALC/NaOHで処理した後にその処理した痰を100℃で15分間沸騰させることでサンプルを不活性にするか或は3)カオトロピック溶液、例えばグアニジンの塩酸塩またはチオ硫酸塩などで処理する(簡単に述べると、痰に2−3倍の体積の5M GuSCNまたは8M GuHCLを混合した)。そのサンプルを37℃で20分間インキュベートした。そのサンプルを遠心分離にかけることで細胞を沈澱させた。その細胞を燐酸塩緩衝食塩水で洗浄した。洗浄した細胞を燐酸塩緩衝食塩水に1%のBSAと一緒に入れて懸濁させるか、或は4)カオトロピック溶液、例えばグアニジンの塩酸またはチオ硫酸塩に入れた後に沸騰させる(細胞を緩衝食塩水に1%のBSAと一緒に入れて懸濁させて100℃で15分間沸騰させてミコバクテリウムを死滅させる以外はこの上に示した段階3と同様に)。次に、その調製した培養物または痰サンプルをガラススライドに塗り付けた後、空気で乾燥させる。
1体積の患者の未処理痰を2体積のIDF溶液(前記)と管の中で混合した後、室温(20−25℃)で15分間インキュベートした。次に、その痰−IDF混合物をガラススライドに塗り付け、空気で乾燥させた後、メタノールで固定した。その固定した塗り付け物をハイブリッド形成前にIDFで処理する段階を省いた。ハイブリッド形成を実施する前に前記スライドをPBSで3回洗浄した。
ガラススライド上にメタノールで固定した痰−IDF混合物にPBS中2%のグルタルアルデヒドを用いた処理を20−25℃(周囲温度)で5分間受けさせた後、PBSによる濯ぎを3回そして空気乾燥を受けさせた。その固定した塗り付け物をハイブリッド形成前にIDFで処理する段階を省いた。
1体積の患者の未処理痰を2体積のIDF溶液(前記)と管の中で混合した後、約20−25℃(周囲温度)で15分間インキュベートした。その痰−IDF混合物を15分間沸騰させることで核酸を溶液の中に放出させると同時に前記サンプルが感染しないようにした。その沸騰させたサンプルから核酸を標準的技術で精製してもよいか、或はShah他[Shah J.S.、King W.、Liu J.、Smith J.、Serpe G.およびPopoff S.、および(1997)Assay Improvements.、米国特許第5,629,156号(これは引用することによって本明細書に組み入れられる)]が記述した如き特定のプローブおよび磁気ビードを用いたサンドイッチハイブリッド形成で興味の持たれる標的核酸を選択してもよい。その精製した標的をPCR(DNA標的の場合)またはRT−PCR(RNA標的の場合)で増幅させてもよい。
細胞の中にヒト型結核菌が存在するか否かを検出する時に、好適には、Shah、NietupskiおよびLiu(米国特許第5,521,300号)が記述した如きヒト型結核菌の23 SリボソームRNAと特異的にハイブリッド形成するDNA配列で構成させたオリゴヌクレオチドプローブを用いる。適切なプローブ複合体の例は下記である:

1つの態様では、このプローブ複合体を固定/前処理しておいたサンプルの核酸と37℃のハイブリッド形成用緩衝液[2.5MのGuSCN、50mMのTris(pH7.8)、20mMのEDTAおよび1%のNP40]中で接触させた。代替態様では、前記プローブ複合体を固定しておいたサンプルの核酸と42℃のハイブリッド形成用緩衝液[50%のホルムアミド、2X SSC(pH7.4)、20mMのEDTA、1%のNP40]中で接触させた。

Claims (12)
- ミコバクテリア属の細胞壁、細胞膜および核膜の透過性を向上させるための組成物であって、2.0から3.3Mの濃度のGuSCN(チオシアン酸グアニジン)、10から100mMの濃度のTris−HCL、5から50mMの濃度のEDTA,0.1から2.0パーセントの濃度のIGEPAL(オクチルフェノキシポリ(エチレンオキシ)エタノール)、1.0から10パーセントの濃度の酢酸、20から50パーセントの濃度のメタノール、0.02から2.5パーセントの濃度のコール酸ナトリウムおよび0.02から2.5パーセントの濃度のデオキシコール酸ナトリウムを含有して成る組成物。
- ミコバクテリア属細胞の中の標的を検出する方法であって、
a)前記細胞を2.0から3.3Mの濃度のGuSCN(チオシアン酸グアニジン)、10から100mMの濃度のTris−HCL、5から50mMの濃度のEDTA,0.1から2.0パーセントの濃度のIGEPAL(オクチルフェノキシポリ(エチレンオキシ)エタノール)、1.0から10パーセントの濃度の酢酸、20から50パーセントの濃度のメタノール、0.02から2.5パーセントの濃度のコール酸ナトリウムおよび0.02から2.5パーセントの濃度のデオキシコール酸ナトリウムを含有して成る組成物と接触させることで透過性細胞を生じさせ、
b)前記段階(a)の透過性細胞を前記標的との結合に特異的な結合因子と接触させ、そして
c)前記段階(b)の結合因子を検出する、
ことを含んで成る方法。 - 前記標的を核酸、ペプチド核酸、ペプチド、糖蛋白質、脂質、リポ蛋白質、ウイルスおよびプリオンから成る群から選択する請求項2記載の方法。
- 前記結合因子を核酸、ペプチド核酸、ペプチド、リポ蛋白質、糖蛋白質および脂質から成る群から選択する請求項2記載の方法。
- 前記結合因子が追加的に検出部分も含有して成る請求項2記載の方法。
- 前記検出部分を蛍光マーカー、放射性マーカー、色素、コロイド状金属、ビオチン/アビジンおよび西洋ワサビペルオキシダーゼから成る群から選択する請求項5記載の方法。
- 前記検出を前記結合因子に親和性を示す標識抗体を用いて実施する請求項2記載の方法。
- 前記標的が微生物であるヒト型結核菌に由来する核酸である請求項2記載の方法。
- 前記結合因子が微生物であるヒト型結核菌に由来する核酸に相補的なオリゴヌクレオチドである請求項2記載の方法。
- 追加的に背景を染色することも含んで成る請求項2記載の方法。
- 前記結合因子がミコバクテリウム属に属する微生物の核酸に相補的なオリゴヌクレオチドである請求項2記載の方法。
- ミコバクテリウム属がヒト型結核菌である請求項1記載の組成物。
