本出願はジフェニルエチレン化合物およびその誘導体とチアゾリジン又はオキサゾリジン中間物質との化学的カップリングによって形成された新規化合物に関連するものである。これらの化合物は種々の有用な薬理効果を与えるために効果的である。例えば、本化合物はII型糖尿病の動物モデル中の血中グルコース値、血清インシュリンおよびトリグリセリド値を下げるのに有用である。驚くべきことに、これらの化合物はレプチン値を増加させることが見い出され、肝臓毒性がない。

さらに、これらの化合物はインシュリン抵抗性と関連した疾患、例えば、多嚢胞性卵巣症候群、そして高脂質血症、冠動脈疾患および抹消血管疾患の治療に有用である。さらに炎症および免疫疾患、特にTNF−α、IL−1、IL−6及び/又はCOX−2といったサイトカインおよびシクロオキシゲナーゼによって仲介される疾患の治療に有用である。

【従来の技術】
I型およびII型の糖尿病の原因は未だ知られておらず、ただ両方とも遺伝的および環境的なものが主な要因であると考えられている。インシュリン依存I型およびインシュリン非依存II型は既知の型である。I型は自己免疫疾患であり、原因自己抗原が未だ知られていない。I型の患者が生存するためにはインシュリンを非経口でまたは皮下に取り込む必要がある。一方II型糖尿病は、より一般的な症例で、体が十分な量のインシュリンを産生できなくなり、または産生されたインシュリンを適切に使用できなくなることより起こる代謝異常である。インシュリン分泌およびインシュリン抵抗性は主要な欠損症であるにもかかわらず、このメカニズムに関与する正確な遺伝因子は未だ知られていない。

上記表からはっきりと理解できるように、糖尿病治療に有用な各現行薬剤は、特定の欠点を有している。従ってなお、新規薬剤、特に、糖尿病治療に用いる経口投与可能な水溶性薬剤の同定および開発に関心がよせられている。

上記表にリストされたチアゾリジンジオン類は、近年、II型糖尿病の治療のためにより広範な使用がされるようになってきた。インシュリン感作物質として特定の有用性を示し、「インシュリン抵抗性」を治療する。インシュリン抵抗性とは、患者がインシュリン効果に鈍感な状態をいう。しかしながら、現在知られているチアゾリジンジオンでは、患者人口の大半には有効とはいえない。さらに、FDAによって認可されたこの種類の最初の薬剤であるトログリタゾンは、肝臓毒性の問題のために市場から回収されてしまった。こうした事情から、無毒性でより広範な有用性を示すインシュリン感作物質が求められている。

上記に示したように、本発明はまた、免疫疾患または炎症、特にサイトカインまたはシクロオキシゲナーゼによって仲介されるような疾患の治療と関係する。免疫系の主要要素はマクロファージ、抗原提示細胞、T細胞およびB細胞である。他の免疫細胞、例えばNK細胞、好塩基球、マスト細胞および樹上細胞の役割は知られているが、初期免疫疾患におけるその役割は確かではない。マクロファージはメディエータとして、炎症を仲介し、T細胞の刺激および増殖のために必要な「助力」を与える。最も重要なことだが、マクロファージはIL1,IL12およびTNFαを産生する。これらの全ては強力な炎症誘発性分子であり、T細胞のために助力する。さらに、マクロファージ活性は、シクロオキシゲナーゼII(COX−2)、誘導性一酸化窒素シンターゼ(iNOS)といった酵素を誘導し、フリーラジカルの発生が通常細胞に損害を与える。多くの因子がマクロファージを活性化する。このような因子としては細菌性生成物、スーパー抗原およびインターフェロンγ(IFNγ)がある。ホスホチロシンキナーゼ(PTKs)および他の不確定の細胞キナーゼは、活性経路中に含まれると考えられている。

インターロイキン−6(IL−6)は、もう一つの炎症誘発性サイトカインであり、多面作用および過剰作用を示す。IL−6は免疫反応、炎症および造血に関与する。IL−6は肝臓急性期応答の強力な誘導物質であり、糖質コルチコイドによる負の制御の下における視床下部下垂体副腎軸(hypothalamic-pituitary-adrenal axis)の強力な刺激物質である。IL−6は成長ホルモンの分泌を促進するが、甲状腺刺激ホルモンの放出を阻害する。IL−6値の上昇は種々の炎症疾患として表れる。そして、IL−6サイトカインサブファミリーの阻害は、慢性関節リウマチの治療向上の方法として示されてきた(Carroll et al., Inflamm Res, 47:1-7,1998)。さらに、IL−6はアテローム性動脈硬化症の進行および冠状動脈性心疾患の病因として関係付けられてきた(Yudkin et al., Atherosclerosis, 148:209-14, 1999)。IL−6の過剰産生はまた、ステロイド中止後症候群に見られ、その状態は調節解除されたバソプレシン分泌、および骨吸収の増加に付随した骨粗鬆症と関係し、上皮小体機能亢進症および性ステロイド欠乏症の原因となる(Papanicolaou et al., Ann Intern Med, 128:127-37, 1998)。

【発明の実施の形態】
実施例1
(a)3−(3,5−ジメトキシフェニル)−2−(4−ヒドロキシフェニル)アクリル酸(化合物IV)の合成
3,5−ジメトキシベンズアルデヒド(500g,3mol)およびp−ヒドロキシフェニル酢酸(457g,3mol)の混合液に、無水酢酸(1L)およびトリエチルアミン(420mL)を添加した。この非均一混合液を加熱すると、〜70℃で均一になる。130℃‐140℃で6時間撹拌した後に、混合液を室温まで冷却した。濃縮HCl(1L)を反応混合液中にゆっくりと添加し、その間、50分間、温度を20℃−30℃の間に保ち続けた。得られた明黄色の沈殿物を濾過し、ブフナー漏斗中でDI水ですすいだ。固形物は3NNaOH(5L)中に溶解し、1時間撹拌して濾過した。濾過液は酸性化し、温度を25℃−30℃に維持するとともに、農塩酸でpH1にした。沈殿生成物を濾過し、水で洗浄して、粗生成物を与えた。粗生成物はMeOH−H2Oから再結晶化され、40℃で6時間乾燥した。収率:黄白色固形物428g(47%)。1HNMR(DMSO−d6):δ12.48(br,1H),9.42(s,1H),7.59(s,1H),6.95(d,J=8.0Hz,2H),6.76(d,J=8.0Hz,2H),6.35(t,J=2.2Hz,1H),6.27(d,J=2.2Hz,2H),3.56(s,6H).

メタノール(3L)を乾燥させた化合物IV(427.5g,1.42mol)にアルゴン下で十分に添加した。この撹拌懸濁液に濃硫酸(100mL)を添加し、還流時に20時間アルゴン下で加熱した。メタノールは減圧下30℃で蒸発させた。残留物はエチルアセテート(3L)中に溶解し、水(2×1L)、飽和水溶性NaHCO3(pH8,2×1L)、飽和水溶性塩化ナトリウム溶液(2×1L)で洗浄した。有機質層を無水硫酸マグネシウム上で乾燥させて濾過し、溶媒は蒸発させた。得られた粗生成物は高真空下で十分に乾燥させ、次の工程に使用した。収率:白色固形物433.6g(97%)。1HNMR(CDCl3):δ7.72(s,1H),7.06(d,J=7.9Hz,2H),6.77(d,J=7.9Hz,2H),6.33(t,J=2.2Hz,1H),6.26(d,J=2.2Hz,2H),5.74(s,1H),3.81(s,3H),3.60(s,6H).

3−(3,5−ジメトキシフェニル)−2−{4−[4−(2,4−ジオキソチアゾリジン−5−イリデンメチル)フェノキシ]フェニル}プロピオン酸メチルエステル(化合物Vc)の合成
化合物Vcは以下の反応系図によって調製した。

(a)3−(3,5−ジメトキシフェニル)−2−(4−ヒドロキシフェニル)プロピオン酸メチルエステルの合成(化合物Va)
ジオキサン(60mL)中の3−(3,5−ジメトキシフェニル)−2−(4−ヒドロキシフェニル)アクリル酸メチルエステル(2.1g)の懸濁液に、炭素(50%ウェット(wet),2.1g)上の10%パラジウムを添加した。水素付加はパーシェーカー(Parr shaker)中で70psi、6.5時間行われた。触媒はセライト層を通して濾過し、溶剤は蒸発させた。収率:定量的。分析:1HNMR(CDCl3):δ7.15(d,J=8.7Hz,2H),6.74(d,J=8.7Hz,2H),6.29(t,J=2.4Hz,1H),6.25(d,J=2.4Hz,2H),3.78(t,J=8.7Hz,1H),3.72(s,6H),3.62(s,3H),3.31(dd,J=13.5&8.4Hz,1H),2.93(dd,J=13.5&6.9Hz,1H)。

(b)3−(3,5−ジメトキシフェニル)−2−[4−(4−ホルミルフェノキシ)フェニル]プロピオン酸メチルエステルの合成(化合物Vb)
アルゴン下、DMF(2mL)中の水酸化ナトリウム(オイル中60%、0.25g,6.3mmol)の懸濁液に、乾燥DMF(3mL)中のVa(2.0g,6.3mmol)を添加した。生じる黄色溶液に、p−フルオロベンズアルデヒド(0.68mL,6.3mmol)を添加し、80℃で18時間加熱した。反応混合液は室温まで冷却し、水(20mL)を添加し、エチルアセテート(3×50mL)で抽出した。有機層は硫酸ナトリウム上で乾燥させ、濾過され、溶剤は蒸発させた。粗生成物は小さなシリカゲル層を通して濾過した。収率:1.83g(68.8%)。分析:1HNMR(DMSO−d6):δ9.91(s,1H),7.84(d,J=8.7Hz,2H),7.33(d,J=8.7Hz,2H),7.04(d,J=5.4Hz,2H),7.01(d,J=5.4Hz,2H),6.30(t,J=2.1Hz,1H),6.25(d,J=2.1Hz,2H),3.86(t,J=7.8Hz,1Hz),3.76(s,6H),3.66(s,3H),3.36(dd,J=12.6および8.1Hz,1H),2.97(dd,J=13.5&7.5Hz,1H)。

(c)3−(3,5−ジメトキシフェニル)−2−{4−[4−(2,4−ジオキソチアゾリジン−5−イリデンメチル)フェノキシ]フェニル}プロピオン酸メチルエステルの合成(化合物Vc)
無水トルエン(25mL)中のVb(1.81g,4.3mmol)の撹拌懸濁液に、2,4−チアゾリジンジオン(0.56g,4.74mmol)、安息香酸(0.68g,5.60mmol)およびピペリジン(0.60mL,6.03mmol)を逐次添加し、ディーン−スターク(Dean-Stark)装置の助けをかりて2時間の間、水の継続的除去をもって還流温度で加熱した。溶剤は蒸発させ、減圧下で乾燥した。得られた残余物はシリカゲルクロマトグラフィで精製され、ヘキサン−エチルアセテート(1:1)で抽出し、Vcを産出した。収率:1.82g(81.3%)分析:1HNMR(DMSO−d6):δ12.53(br,1H),7.76(s,1H),7.60(d,J=8.7Hz,2H),7.35(d,J=8.7Hz,2H),7.07(d,J=4.8Hz,2H),7.03(d,J=4.8Hz,2H),6.33−6.28(m,3H),4.01(t,J=7.5Hz,1H),3.66(s,6H),3.56(s,3H),3.22(dd,J=13.8&8.4Hz,1H),2.90(dd,J=13.5&7.2Hz,1H)。

Claims (1)
- 化学構造式Iの化合物であって、
n、m、qおよびrは、独立に、n+m≦4およびq+r≦4を満たす0から4までの整数であり;pおよびsは、独立に、p+s≦5を満たす0から5までの整数であり;a,bおよびcは二重結合であって、存在している場合と存在しない場合があるり;二重結合が存在している場合は、二重結合によってEまたはZの立体配置をとることになり、二重結合が存在しない場合は、生じる立体中心はR配置またはS配置をとり、
RおよびR''は、独立して、H、C1-C20直鎖または分岐鎖のアルキル基、C2-C20直鎖または分岐鎖のアルケニル基、-CO2Z'基であり、Z'は、薬学的に許容可能な対イオン、-CO2R'''、-NH2、-NHR'''、-NR2'''、-OH、-OR'''、ハロ、置換C1-C20直鎖または分岐鎖のアルキル基または置換C2-C20直鎖または分岐鎖のアルケニル基であり、R'''はC1-C20の直鎖または分岐鎖のアルキル基または直鎖または分岐鎖のアルケニル基であり;
R'は、CONR''''2(式中のR''''は、独立に、水素、メチル、又はメトキシを表す。)または-CO2Z'(式中のZ'は、薬学的に許容可能な対イオンである。)であり;
A、A'およびA''は、独立して、H、C1-C20アシルアミノ、C1-C20アシルオキシ、C1-C20アルカノイル、C1-C20アルコキシカルボニル、C1-C20アルコキシ、C1-C20アルキルアミノ、C1-C20アルキルカルボキシルアミノ、カルボキシル、シアノ、ハロ、またはヒドロキシであり;

または、AおよびBが共に、又はA'およびB'が共に、メチレンジオキシ又はエチレンジオキシ基を形成するように結合され;
X、X'は、独立して、-NH、-NR'''、OまたはSであり;

3−(3,5−ジメトキシフェニル)−2−{4−[4−(2,4−ジオキソチアゾリジン−5−イルメチル)フェノキシ]フェニル}−N−メトキシ−N−メチルアクリルアミド。
