Description
より具体的には、本発明は、
少なくとも1つの膜親和性(membraneophilic)領域を含む少なくとも1つの支持固体表面;
その膜親和性領域に少なくとも部分的に固定されている被覆層であって、(i)界面活性剤膜、(ii)脂質模倣ポリマー、(iii)界面活性剤又は乳剤系、(iv)液晶、又はその組み合わせ、から成る被覆層;及び
その被覆層に含まれている、結合している、連結している、又は会合している物質
を含んでなる装置に関する。
各々の支持固体表面は、1、2又は数個の膜親和性領域を含むことができる。各々の膜親和性領域は、同様の又は異なる被覆層により被覆することができる。
本装置は、前記被覆層を覆う水溶液も含むこともある。
更に、本装置は、ゲル又はポリマー構造から成る層を含むことがある。この層を用いて、例えば、pH勾配を導入すること、又は例えば抗体のような分子又は官能基を取り付けるための第二の固定マトリックスを提供することができる。この層は、前記被覆層の上及び/又は下に配置することができる。
本発明に従って使用することができる膜親和性領域は、任意の形状又は幾何学的配置の任意の膜親和性表面又は化学的及び/又は物理的な調節若しくは操作により膜親和性にした表面で構成又は形成され得る。
前記物理的な調節若しくは操作は、例えば、プラズマ処理又はその他の適する方法により行うことができる。
更に、前記被覆層は、脂質模倣ポリマーによって構成することができる。このようなポリマーの一例は、ジブロックポリマーである。
本発明の装置は、その被覆層に磁場をかけるための少なくとも1つの磁石を含むこともできる。
更に、本発明の装置は、その被覆層周囲の追加の膜、タンパク質−脂質混合物、洗浄溶液、染色溶液、消化溶液及びその他の溶液又は懸濁液の交換のための、少なくとも1つの入口及び少なくとも1つの出口連結部を含むこともできる。
本装置は、その装置との材料の出し入れ輸送のための少なくとも1つの流体チャネルを含むこともできる。

高分子の集束又は濃縮のために、又は高分子凝集体の形成のために、
適している。
・できるだけ多くのアミノ酸配列に対応するタンパク質配列中のできるだけ多くのペプチドを検出及び同定する最適化プロトコルを含む、いわゆる配列カバー率の検討をはじめとする、タンパク質の同定及び特性付け。翻訳後修飾の検討もこの文脈にあてはまる。
・様々な細胞サンプルからの膜タンパク質のマッピング、つまり、膜プロテオームプロファイリング。広範な存在度レベルを有する非常に複雑なサンプルを消化し、分析する。MS分析前に、生成ペプチドを1つの次元(例えば、逆相HPLC)又は2つの次元(例えば、イオン交換HPLC(SCX−強カチオン交換)、その後、逆相HPLC)で分離する。これにより、低存在度タンパク質の検討及び新薬ターゲットの発見が可能となる。
・膜タンパク質の機能検討、例として、ターゲット検索及び受容体の脱オーファン化、例えばG蛋白結合受容体(GPCR)の脱オーファン化である。ターゲット検索の場合、その目的は、リガンド結合検討を用いて、どのリガンドがどのタンパク質に結合するかを特定することである。受容体の脱オーファン化の場合、その目的は、受容体及びそれらの推定リガンドの機能を解明することである。機能が不明であり、それ故、オーファン受容体という名前の受容体が多く存在する。
・疾病状態及び/又は薬剤療法状態時のタンパク質発現レベルのアップ・アンド・ダウンレギュレーションの調査、いわゆる発現プロファイリング。このような検討は、様々なサンプルの比較及びそれらサンプル間の違いの評価を含む。
・多数の化学反応及び/又は洗浄段階を実行して、このチップでの最適化プロトコルを実現することができる。このような段階は、サンプルを希釈せずに実行することができる。
・同じサンプルを使用して、膜タンパク質の膜外及び膜貫通部分をターゲットにし、様々な段階で回収することができる。
・単一サンプルを種々の酵素に付して、逐次的に及び/又は同時に、同サンプルの様々な部分を消化することができる。
・逐次的消化などと共にこのチップにおける容積に対する高い表面積比率は、最少のサンプルロスで高濃度の生成ペプチドを導き、それが、より高い感度を可能にする。これは、サンプル中の配列カバー率の検討及び低存在度タンパク質の検出を促進する。
・細胞からの種々の細胞内画分を1つのチップで同時に取り扱うことができる。
・リガンド結合、架橋及び/又は消化により、ターゲットのペプチド結合部位を得ることができる。リガンドと結合しているタンパク質と結合していなタンパク質とでは、リガンドが酵素の分解を立体的に妨害する又は結合により構造変化をもたらすことで、酵素的消化後に、異なるペプチドマップを生成する。
・サンプルが表面に拘束される。つまり、分析の間、サンプルが固定マトリックス内に保持され、これにより、標識、化学的な調節、消化などの多数の段階が可能となる。
・MS、蛍光、電気化学、SPR、QCMなどをはじめとする、チップ実装及びチップ非実装の検出技術の蓄積と併用することができる。
・既存のチップベースのプラットフォーム及び従来の分析ツールに本チップを組み込むことが可能。
・サンプル処理の自動化が容易である。
・膜タンパク質のような膜結合成分が、それらの天然脂質二重層環境内に保持される。
・膜結合成分の構造及び機能が、維持される。
・本発明は、膜タンパク質をそれらの天然二分子膜環境内に保つことができるので、タンパク質のような膜結合成分を含有する膜小胞の調製、輸送、堆積及び処理に、界面活性剤を使用する必要がない。界面活性剤は不都合な効果、例えばタンパク質の変性効果を有することがあるので、これは有利である。本発明に関連してほんの少数の又はすべての段階で界面活性剤を使用して、例えば、分析前により純粋な膜タンパク質調製物を得ることも可能である。
1つの次元での電場、その後、例えばpH勾配と併用での第二次元での分離など、2つの次元でのタンパク質の分離に基づくことが多い。タンパク質が、その等電点(pI)を生じる中性電荷に到達すると、そのタンパク質は、電場での移動を停止する。膜タンパク質に伴う問題は、この等電点での沈殿である。しかし、脂質二重層の保護された疎水性内部にあるとき、これは、同程度には起こらない。チップ構造は、pH勾配と組み合わせて、2−Dゲル分離に通常適用されるものと同じ原理を用いて、膜タンパク質の分離を助長することができる。これらのpH勾配は、脂質マトリックス上にpH勾配を作ることができるゲルとチップ構造を組み合わせて生成することができる。また、マイクロ流体工学をチップ構造と組み合わせて、様々なpHの種々の溶液でチップ構造の一部をフラッシュすることができる(図13)。このように、タンパク質は、そのタンパク質の電荷が中性になる(これは、上記に説明したように、その等電点において生じる)まで電場内で移動する。
上述のように、配列カバー率の検討のために多数の異なるプロトコルを実行することができる。図21は、異なるプロテアーゼを使用する逐次的消化の概念を示す。
(実施例)
(材料)
130〜170μmの厚さを有する直径28mmのカバーガラスは、Menzel−Glaser(Braunschweig, Germany)から購入した。電子ビームレジスト、フォトレジスト及び現像液は、AZ Electronic Materials(Somerville, NJ, USA)から入手し、プライマーは、Shipley(Marlborough, MA, USA)から入手した。この製造に使用した他のすべての化学物質は、VLSIグレードのものであり、別様に述べていない限りMerckから入手した。マスク製造に使用した100nmのクロムで被覆された100mm、低反射ブランク石英マスクは、Nanofilm(Westlake Village, CA)から入手した。蒸着に使用した材料は、Nordic High Vacuum(Kullavik, Sweden)から購入した。
カバーガラスをアセトン中で20分間、超音波処理し、80℃のRCA1溶液(H2O2(25%)、NH3(30%)及びMilli−Qの1:1:5溶液)中で10分間洗浄し、緩衝酸化物エッチング溶液(NH4F(30%)とHF(49%)の6:1溶液)に2分間、浸漬し、QDRでMilli−Q水中ですすぎ、N2下でブロー乾燥させた。
基板をAVAC HVC−600薄膜形成装置に挿入し、1.5nmのチタン、13.6nmの金、1.5nmのチタン、そして最後に60nmの二酸化ケイ素からなる薄膜を、電子ビーム蒸着法により基板上に蒸着した。それらの基板に先ず接着促進剤(HMDS)を、次にAZ5214−Eイメージ反転物質を6000rpmで45秒間スピンコートし、露光前にホットプレート上で90℃にて2分間焼成し、Carl Suss MJB−3マスクアライナーを用いて、パターン化された暗視野クロムマスクを通して波長365nmで120mJ/cm2の線量に露光した。露光後にそれらの基板をホットプレート上で125℃にて30秒間焼成し、その後、720mJ/cm2の線量にフラッド露光した。そのパターンを、AZ351現像液とDI水の1:5混合物中で60秒間現像し、QDRですすぎ、N2下でブロー乾燥させた。明視野パターンが得られた。露光された二酸化ケイ素をPlasmatherm RIE m−95 反応性イオンエッチング装置でエッチングした(180秒、100W、25mTorr、CF4:32cm3/秒、H2:8cm3/秒、O2:1cm3/秒)。二酸化ケイ素が完全にエッチングされて除去されたことを確認するために、プロフィロメーター測定、二点抵抗測定及び金湿式エッチングテストを行った。Shiphley1165リムーバーを使用してそれらの基板から残存レジストをストリッピングし、QDRで、アセトン、2−プロパノール及びDI水ですすぎ、N2下でブロー乾燥させた。TePla300PC Microwave Plasma systemを使用してそれらの基板を最終的にデサムして(desummed:60秒、250W、O2の250cm3/秒)、Micropositプライマーから生じた二酸化ケイ素を用いて残存シロキサンを除去した。
エッチングは、幾つかのレジストを用い、湿式エッチング系、例えば緩衝酸化物エッチング溶液(49%フッ化水素酸及び30%フッ化アルミニウムの1:6混合物)を使用して、行うこともできる。それらの表面を化学的及び/又は物理的な調節若しくは操作によって膜親和性にすることができる。
基板をAVAC HVC−600薄膜形成装置に挿入し、薄膜(50nmのAl2O3か又は1.5nmのTi+13.5nmのAuの何れか)を電気ビーム蒸着法によってそれらの基板に蒸着した。それらの基板に4000rpmで30秒間、MicoChem SU−8 2002 ネガ型フォトレジストをスピンコートし、露光前にホットプレート上で95℃にて2分間焼成し、Carl Suss MJB−3 マスクアライナーを用いて、パターン化された明視野クロムマスクを通して波長365nmで120mJ/cm2の線量に露光した。露光後にそれらの基板をホットプレート上で95℃にて60秒間焼成した。そのパターンを、MicroChemのSU−8現像液中で60秒間現像し、2−プロパノール中で簡単にすすぎ、N2下でブロー乾燥させた。これにより、Au又はAl2O3上のSu−8 2002の暗視野パターンが生じた。TePla 300PC Microwave Plasma systemを使用してそれらの基板を最終的にデサム(60秒、250W、O2の250cm3/秒)した。それらの表面を化学的及び/又は物理的な調節若しくは操作によって膜親和性にすることができる。
(材料及び方法)
(小胞調製)
小胞は、原液としてのクロロホルムに溶解した大豆レシチン(SBL)(100mg/mL)から調製し、DiO(一般的には、脂質濃度に対して1mol%)で染色した。Criado及びKeller(Criado, M., Keller, B.U. FEBS Lett. 224, 172-176(1987))によって説明されたものに修正を加えた脱水/再水和法(Karlssonn, M et al. Anal. Chem. 72, 5857-5862(2000))を用いて、単層リポソームを調製した。一言で言えば、5μLの脂質分散物の液滴(1mg/mL)をカバーガラス上に置き、減圧デシケーターの中で25分間、脱水した。その脂質薄膜が乾いたら、それを緩衝溶液(Trizma塩基の5mM、K3PO4の30mM、KH2PO4の30mM、MgSO4の1mM、EDTAの0.5mM、pH7.8)で再水和した。数分後、リポソームが形成された。ガラスピペットを使用することにより、脂質懸濁液の小サンプルを緩衝溶液の液滴に写し、それを試験する表面の上に置いた。
Leica PL Fluotar 40x対物レンズを使用する倒立顕微鏡(Leica DM IRB, Wetzlar, Germany)の台に、カバーガラスを直接置いた。落射蛍光照明には、488nmのAr+レーザーライン(2025−05、Spectra−Physics)を使用した。密着性を壊し、レーザー光を散乱させるために、その光線路に透明回転盤を置いた。多色ミラー(Leica)及び対物レンズによって光を送って、フルオロフォア(fluorophores)を励起させた。蛍光を3チップ方式カラーCCDカメラ(Hamamatsu, Kista, Sweden)により集め、デジタルビデオ(DVCAM, DSR-11, Sony, Japan)により記録した。Adobe Premiereグラフィックソフトウェア及びMatlabを使用してデジタル画像を編集した。
Trizma塩基、グリセロール及びリン酸カリウムは、Sigma−Aldric Sweden ABから入手した。大豆レシチン(SBL、極性脂質抽出物)は、Avanti Polar Lipids,Inc.(700 Industrial Park Drive, Alabaster, AL 35007)から入手した。クロロホルム、EDTA(Titriplex III)、硫酸マグネシウム、リン酸二水素カリウム、塩化カリウム、塩化ナトリウム及び塩化マグネシウムは、Merck(Darmstadt, Germany)からのものであった。脱イオン水(Millipore Corp., Bedford, MA)を用いて緩衝液を調製した。DiO(3,39−ジオクタデシルオキサカルボシアニン過塩素酸塩)は、Molecular Probes(Leiden, The Netherlands)から入手した。極性脂質抽出物は、ホスファチジルコリン(45.7%)、ホスファチジルエタノールアミン(22.1%)、ホスファチジルイノシトール(18.4%)、ホスファチジン酸(6.9%)及びその他(6.9%)の混合物からなるものであった。
パターン化された脂質二重層中での拡散移動の一例を、図10に示す。先ず、脂質を小さな単層小胞(SVU)の融合によって膜親和性表面に堆積させた。このSUVは、蛍光可視化のために1mol%DiOをドープした大豆脂質(総脂質濃度:240μL、大豆脂質の原液の100mg/(mL・クロロホルム)を約5時間、蒸発によって乾燥させ、2mLの緩衝溶液(20mMのNaCl、10mMのTrizma、pH9.5を含む)中で再び水和させた)からなるものであった。膜親和性表面へSUVを接着及び融合した後(待ち時間は、通常、30〜60分)、構成された表面の上の緩衝溶液を数回交換することにより、周囲の流体中の余剰脂質材料を除去した。その緩衝溶液を、浸透圧不均衡を生じさせるカルシウムイオン及びより高いイオン強度を含有する緩衝液(100mMのNaCl、5mMのTrizma、2mMのCaCl2、pH8.6)に換えること、そして短い間隔で完全に装置のフローセルから緩衝液を排出させることによって、小胞の融合を促進した。
本脂質装置技術と質量分析の一体化により、装置中の脂質マトリックスと会合しているタンパク質を同定することができる。これを例証するために、赤血球膜(RBCM)タンパク質を使用して次の実験を行った。先ず、それらの細胞を、pH7.4のリン酸緩衝食塩水(PBS)で洗浄した。コレステロールを、PBS中のβ−シクロデキストリン(6mM)で抽出し、除去した。2mMのEDTA、pH8.3、1mMのDTT及びプロテアーゼ阻害剤で細胞を溶解し、広範囲に洗浄した。その後、そのRBCMを高塩分の溶液(1MのKCl、1mMのDTT、pH7.8)及び高pH溶液(0.1MのNa2CO3、1mMのDTT、pH11.3)で処理して、非内在性膜タンパク質をできる限り除去した。RBCMを、20mMのNaCl、Trizmaの10mM、pH8.0中に、0.8mg/mLに希釈し、合計15分の継続時間(5秒のパルス、その後の5秒の休止の間隔で)で超音波処理して、膜を小胞に縮小した。その小胞溶液を200nm孔径のPVDF膜フィルターに通して、超音波処理装置のtipから形成されるチタン粒子を除去した。脂質装置は、化学的及び/又は物理的な調節若しくは操作によって膜親和性にした底面に、Al2O3被覆チップ(薄層形成装置(AVAC HVC−600)において蒸着により堆積させた、50nm厚の層)を取り付けた。更に、細胞の望ましい高さ(100〜500μm)を提供するためのテフロン(登録商標)スペーサーと、化学的及び/又は物理的な調節若しくは操作によって膜親和性にした、Al2O3で被覆された上蓋を取り付けた。
赤血球膜調製物を用いて、プロファイリング検討に関して評価し、結果を得た。要するに、RBCMと示した、上記と同様の調製物をこの実験で使用した。小胞懸濁液を濾過する最終段階は省いた。LC−MS/MS分析はフィルターからのポリマー汚染物に対して高感度であることが証明されたからである(この場合、ナノスプレー源を装備した7−Tesla(フーリエ変換リニアトラップ四重極型(Linear Trap Quadrupole-Fourier Transform))LTQ−FT質量分析装置(Thermo Electron)を使用した)。また、異なる緩衝液を用いた。高いイオン強度は、表面への膜材料の吸着を増加させるように思われたので、この場合は、10mMのTrizma、300mMのNaCl、pH8を用いた。
要するに、チップ(容量約500μL)に小胞懸濁液(チップ超音波処理した(tipsonicated)RBC)を充填し、約1時間、放置した。この小胞調製物は、濾過しなかった。チップ超音波処理(tipsonication)プロセス中にサンプルに放出された可能性のあるチタン及びガラス粒子を除去するために、その小胞懸濁液を2mLアリコートに分けてエッペンドルフ管に入れ、10000rpmで10分間遠心分離した。
トリプシン(同じ緩衝液中の0.005mg/mL)をチップに再び添加し、そのチップを37℃にて2時間インキュベートした。

配列カバー率の検討の目的は、特定のタンパク質中のアミノ酸をできるだけ多く検出することである。高い配列カバー率は、ペプチドがそのタンパク質の大部分から作られていることを意味する。高い配列カバー率は、様々なプロテアーゼを同時に又は逐次的に使用して多数の部位でターゲットタンパク質を分解し、LC−MS/MS分析に適する多数のペプチドを生じさせることによって、得ることができる。
Claims (18)
- 被覆層を付着する少なくとも1つの膜親和性(membraneophilic)領域をそれぞれに含む少なくとも2つの対向する支持固体表面;
その膜親和性領域に少なくとも部分的に固定されている被覆層であって、(i)脂質の膜、(ii)脂質模倣ポリマーの膜、(iii)脂質又は脂質模倣ポリマーで構成される乳剤系、(iv)脂質又は脂質模倣ポリマーで構成される液晶から成る被覆層;及び
その被覆層に含まれている、結合している、連結している、又は会合している物質;
を含んでなる装置を用いた方法であって、
方法が、(i)前記装置を、前記物質の一部の制御された分解を行う少なくとも1つの消化剤又は分解剤と接触させ、その後、その物質が、実質的に可動な画分と、依然としてその被覆層に含まれている、結合している、連結している、又は会合している固定画分とになる第一段階;及び(ii)その実質的に可動な画分を検出又は分析する第二段階を含んでなる、被覆層に含まれている、結合している、連結している、又は会合している物質を同定する方法。 - 前記消化剤が、酵素である、請求項1に記載の方法。
- 異なる消化剤又は分解剤の混合物が使用される、請求項1又は2に記載の方法。
- 異なる消化剤又は分解剤が逐次的に使用される、請求項1又は2に記載の方法。
- 異なる消化剤又は分解剤が同時に使用される、請求項1又は2に記載の方法。
- (iii)被覆層に含まれている、結合している、連結している、又は会合している物質の残存固定画分を保持する前記被覆層の構造を崩壊させる少なくとも1つの崩壊剤と前記装置を接触させ、その後、その物質が、実質的に可動性であるその基本的要素から成る第三段階;及び(iv)前記被覆層から放出される、その被覆層に含まれていた、結合していた、連結していた、又は会合していた物質の残存画分を検出又は分析する第四段階を更に含んでなる、請求項1〜5の何れか一項に記載の方法。
- 前記崩壊剤が、界面活性剤又は有機溶媒である、請求項6に記載の方法。
- 検出又は分析段階(工程(ii)及び/又は工程(iv))の一方又は両方が、分離段階に先行して実施される、請求項1〜7の何れか一項に記載の方法。
- 前記検出又は分析が、質量分析法、表面プラズモン共鳴法(SPR)、水晶発振子マイクロバランス法(QCM)、及び蛍光ベースの技術から選択される方法により実施される、請求項1〜8の何れか一項に記載の方法。
- 前記質量分析法が、MALDI MS法及びエレクトロスプレーイオン化法(ESI MS−MS)から選択される、請求項9に記載の方法。
- 用いられるMALDI MS法が、MALDI−TOF法及びMALDI−TOF−TOF法から選択される、請求項10に記載の方法。
- 前記装置が、支持固体表面と被覆層の間に捕捉された水層を更に含んでなる、請求項1〜11の何れか一項に記載の方法。
- 前記装置が、前記被覆層を覆う液体又は水溶液を更に含んでなる、請求項1〜12の何れか一項に記載の方法。
- 前記装置が、前記支持固体表面上に金属薄膜を更に含んでなる、請求項1〜13の何れか一項に記載の方法。
- 前記装置の膜親和性領域が、二酸化ケイ素、ガラス、マイカ若しくはポリマー若しくはそれらの組み合わせ、金属酸化物、金属若しくはそれらの組み合わせ、又は、酸化アルミニウム、金若しくはそれらの組み合わせの何れかでできた表面である、請求項1〜14の何れか一項に記載の方法。
- 前記装置の被覆層が、脂質二重膜、細胞膜、無傷細胞、脂質小胞、脂質ナノチューブ、脂質単層、細胞小器官及びそれらの組み合わせから成る群より選択されるいずれかで構成される、請求項1〜15の何れか一項に記載の方法。
- 前記装置の被覆層に含まれている、結合している、連結している、又は会合している物質が、表在性及び内在性膜タンパク質、グリコシルホスファチジルイノシトール(GPI)アンカー型タンパク質、リン脂質、スフィンゴ脂質、薬剤、両親媒性物質、親油性物質、ステロール、糖、オリゴヌクレオチド、ポリマー及びDNAから成る群より選択される、請求項1〜16の何れか一項に記載の方法。
- 前記装置が、被覆層の周囲の追加の膜、タンパク質−脂質混合物、洗浄溶液、染色溶液、消化溶液及びその他の溶液又は懸濁液の交換のために、前記被覆層を覆う液体又は水溶液の流れを作るための少なくとも1つの入口及び少なくとも1つの出口連結部を更に含んでなる、請求項1〜17の何れか一項に記載の方法。
