Abstract
【解決手段】植物の葉用ポリフェノール増加剤が、プロアントシアニジンとトレハロースとを含む。植物の葉用ポリフェノール増加剤は、プロアントシアニジンとトレハロースとが1:15乃至1:60の重量比で含まれていることが好ましい。植物の葉保存用シートが、植物の葉用ポリフェノール増加剤を含んでいる。

Description
植物の葉保存用シートで茶葉を包み、チャック付きの袋に入れて、4℃で15日間冷蔵保存した。また、比較試験として、茶葉を包まずそのままの状態でチャック付きの袋に入れたもの(以下、「初発」という)、茶葉を不織布で包んでチャック付きの袋に入れたもの(以下、「不織布コントロール」という)、標準濃度本発明シートのトレハロースの代わりにソルビトールを配合した不織布のシートで茶葉を包み、チャック付きの袋に入れたもの(以下、「ソルビトールシート」という)を、同じ条件で保存した。
保存試験を行った各試料について、ニンヒドリン反応法にてアミノ酸測定を行った。ニンヒドリン試薬は、L−8900用ニンヒドリン発色溶液セット(和光純薬工業株式会社製)を用いた。測定試料の調製は、96Wellマルチプレートに、試料50μl、発色溶液A 50μl、発色溶液B 50μlをそれぞれ加えて撹拌後、80℃のオーブンにて5分間加熱した。これをマルチプレートリーダー(製品名「MTP−450」、コロナ電気株式会社製)を用い、550nmにて測定した。検量線は、アミノ酸混合標準液AN−II型とB型とを混合した物(40種類のアミノ酸各2nmol含有)を用いて作成した。
植物の葉保存用シートで桑葉を包み、気温が4℃の場所で40日間保存した(以下、「シート被覆試料」という)。また、比較試験として、桑葉を包まずそのままの状態のもの(以下、「コントロール試料」という)を、気温が4℃の場所で40日間保存した。
シート被覆試料およびコントロール試料のそれぞれについて、桑葉の湿重量に対して4倍量のエタノールを加えて磨砕、攪拌し、これを遠心分離して得られた上清を、桑葉ポリフェノール抽出液とした。この桑葉ポリフェノール抽出液を用いて、比色検定法であるFolin Ciocalteu’s法によりポリフェノール含有量の測定を行った。
ポリフェノールの抗酸化力を調べるために、DPPH法にて、DPPHラジカル消去活性の測定を行った。DPPHラジカル消去活性は、80%エタノール溶液を100%として算出した。測定は、測定容器として96wellマルチプレートを、測定機器としてマルチプレートリーダー(製品名「DTX−800」、Beckman Coulter,Inc.製)を用い、測定波長を520nmとして行った。
シート被覆試料についてアミノ酸分析を行った。比較のため、市販の桑茶(商品名「更木桑茶」)についてもアミノ酸分析を行った。また、初期状態を確認するため、冷凍保存した生桑葉についてもアミノ酸分析を行った。
アミノ酸分析用の試料は、実施例2と同様にして、以下のように作成した。まず、桑葉湿重量約7.5gに対して4倍量のエタノール(約30ml)と、80%エタノールを30mlとを加えて磨砕、攪拌し、これを遠心分離して得られた上清を、抽出液とする。これを、各試料について3検体ずつ調製した。

- プロアントシアニジンとトレハロースとを含むことを特徴とする収穫された茶葉用ポリフェノール増加剤。
- プロアントシアニジンとトレハロースとを含むことを特徴とする収穫された茶葉用ポリフェノールおよびアミノ酸増加剤。
- 前記プロアントシアニジンと前記トレハロースとが1:15乃至1:60の重量比で含まれていることを特徴とする請求項1記載の収穫された茶葉用ポリフェノール増加剤または請求項2記載の収穫された茶葉用ポリフェノールおよびアミノ酸増加剤。
- 請求項1記載の収穫された茶葉用ポリフェノール増加剤または請求項2記載の収穫された茶葉用ポリフェノールおよびアミノ酸増加剤を含む樹脂ペレットをシート状に加工して成ることを特徴とする収穫された茶葉保存用シート。
- 請求項1記載の収穫された茶葉用ポリフェノール増加剤または請求項2記載の収穫された茶葉用ポリフェノールおよびアミノ酸増加剤を含んでいることを特徴とする、請求項4記載の収穫された茶葉保存用シートを製造するための樹脂ペレット。
- 請求項5記載の樹脂ペレットを150乃至200℃で融解した後、厚さ20乃至50μmのシート状に加工することを特徴とする収穫された茶葉保存用シートの製造方法。
