Description
本発明は、果物又は野菜又は植物からのみ由来する水抽出性の生物活性成分から成る水分散性組成物、好ましくは粉末組成物、その調製方法、並びに多様な食品、栄養補助食品、ペットフード製品、ペット用栄養補助食品、或いは化粧品製剤又は医薬製剤の調製における一次組成物としてのその使用に関する。
果物又は植物原料から抽出される色素及び生物活性化合物は、機能性成分として食品業界で広く使用されている。それら全ての中で、クコ(ナガバクコ(Lycium barbarum))は、果実中に存在する多様な生物活性化合物に関連した、特に視力及び免疫系にとっての有益性及びアンチエイジング特性のため、中国で最も価値のある機能性成分の1つである。伝統的に湯で抽出することにより摂取されている。
第一の目的として、本発明は、果物又は野菜又は植物からのみ由来する水抽出性の親油性及び非親油性の生物活性成分から成る水分散性の一次組成物の調製プロセスを提供するものであり、前記プロセスは、
a)選択された果物又は野菜又は植物原料をpH約8〜11及び約20〜25と50℃の間に含まれる温度でホモジナイゼーションに供するステップと、
b)前記ホモジナイズしたものから液体エキスを分離した後、前記液体エキスを中和するステップと、
c)中和した前記液体エキスを最終的に濃縮又は乾燥するステップと
を含む。
以下の説明中において、生物活性化合物という用語が、経口で摂取したとき又は化粧品の状態で付けたときに生物活性又は健康上の影響を示す分子又は成分を意味することは理解されよう。
水道水160g及び炭酸水素ナトリウム2gを、0.5リットルのガラス製反応容器に入れた。0.29%のパルミチン酸ゼアキサンチン(zeaxanthin palmitate)を含有するクコの乾燥果実40gを前記水溶液に加え、混合装置(Polytron(登録商標)、26000rpm)を用いて前記混合物を15分間ホモジナイズした。氷浴で冷却することにより温度を30℃未満に維持した。その結果得られる混合物のpHは8.3であった。次に、前記混合物を2000Gで10分間遠心分離した。残留固形物を除去した(50g)。この液相(150g)を、50%クエン酸水溶液0.8gを加えることにより、pH7に中和した。この赤橙色の液体を凍結乾燥し、赤橙色の粉末33.2gを得た。
2.1.水溶性化合物(主に糖類)の除去
0.29%のパルミチン酸ゼアキサンチンを含有するクコの乾燥果実250gを3.0リットルのガラス製反応容器に入れた。水道水1.5リットルを加え、この混合物を室温で1時間、静かに混合した。次に、この液相を分離し、同条件の下、1リットルの水で前記果物の固形物の2度目の洗浄を実施した。この2つの液相を合わせ、凍結乾燥して、0.02%未満のパルミチン酸ゼアキサンチンを含有する粉末157gを得た。
水洗した前記クコの果実を、pH電極を備えた2リットルの反応容器に入れた。さらに水道水250gを加え、10%の水酸化ナトリウム水溶液を加えることにより、前記混合物のpHを10.5まで上げた。前記混合物を、混合装置(Polytron(登録商標)、26000rpm)で15分間ホモジナイズした。ホモジナイゼーション中に10%の水酸化ナトリウム水溶液を加えることにより、前記混合物のpHを10.5に維持した。氷浴で冷却することにより温度を30℃未満に保持した。次に、前記混合物を2000Gで10分間、遠心分離した。残留固形物を除去した。この液相を85℃で2分間低温殺菌した後、50%のクエン酸水溶液6.8gを加えることによりpH7.0に中和した。この結果得られる液体を凍結乾燥し、赤色の粉末68.8gを得た。
実施例1で調製した前記一次組成物を、ヨーグルト様の発酵乳製品の製造に使用する。このために、2.8%の脂肪を含有し2%の脱脂粉乳及び6%のショ糖を加えた乳製品1Lを調製し低温殺菌した後、温度を42℃に下げた。濃縮していないストレプトコッカスサーモフィラス(Streptococcus thermophilus)の菌株及び非粘性のラクトバチルスブルガリクス(Lactobacillus bulgaricus)の菌株の前培養物を、10%還元乳の粉末及び市販の0.1%酵母エキスを含有する無菌のMSK培地内で再賦活化した。次に、再賦活化したこれら前培養物の各々1%を低温殺菌済の前記乳製品に播種した後、この乳製品をpHが4.5の値に到達するまで32℃で発酵させた。ヨーグルト様の発酵済の前記乳製品に、実施例1にあるような前記一次組成物(1%)を加え、4℃で保存した。
トウモロコシ、トウモロコシグルテン、鶏肉及び魚、塩、ビタミン及びミネラルで飼料混合物を作製した。次に、前処理機(pre−conditioner)から出てくる含湿飼料を押出調理機(extruder−cooker)に投入し、ゼラチン化した。前記押出機から出てくるゼラチン化した前記生地を金型に通して押し出した。前記金型のヘッドから出てくる押出物を、イヌへの給餌に適するように細かく裁断し、約110℃で約20分間乾燥させた後、冷却してペレットを形成した。前記ペレットの結果的な水分活性は約0.6であった。前記ペレットを、獣脂及び実施例1で調製した前記一次組成物を含むコーティング物質の噴霧によりコーティングした。
ハードカプセル剤形中の組成物は、以下の処方を有する。
Claims (13)
- 果物又は野菜又は植物からのみ由来する水抽出性の親油性及び非親油性の生物活性成分から成る水分散性の一次組成物の調製プロセスであって、
a)自然のままの又は洗浄した、選択された果物又は野菜又は植物原料を、pH8〜11及び20〜25℃と30℃未満の間に含まれる温度でホモジナイゼーションに供するステップと、
b)前記ホモジナイズしたものから液体エキスを分離した後、前記液体エキスを中和するステップと、
c)中和した前記液体エキスを最終的に濃縮又は乾燥するステップと
を含む、プロセス。 - 選択された前記果物又は野菜又は植物原料をホモジナイゼーションの前に室温で水洗する、請求項1に記載のプロセス。
- 前記抽出性の親油性生物活性成分が、脂質、カロテノイド、及び親油性ビタミンから成る群から選択される、請求項1及び2のいずれかに記載のプロセス。
- 前記カロテノイドが、リコペン、カロテン、フィトフルエン、フィトエン、カンタキサンチン、アスタキサンチン、β−クリプトキサンチン、カプサンチン、ルテイン、ゼアキサンチンなどのカロテン及びキサントフィル、又は脂肪酸エステル型のカロテン及びキサントフィルである、請求項3に記載のプロセス。
- 前記抽出性の非親油性生物活性成分が、アルカロイド、炭水化物、フラボノイドなどのポリフェノール、水溶性ビタミン、及びミネラルから成る群から選択される、請求項1及び2のいずれかに記載のプロセス。
- 前記フラボノイドが、アピゲニン、ルテオリン若しくはジオスメチンなどのフラボン、ケルセチン、ミリセチン、ケンフェロールなどのフラボノール、ナリンゲニン及びヘスペリジンなどのフラバノン、エピカテキン及びガロカテキンなどのカテキン、ペラルゴニジン、マルビジン、シアニジンなどのアントシアニジン、又はゲニステイン、ダイゼインなどのイソフラボンである、請求項5に記載のプロセス。
- 前記果物、野菜又は植物原料が、クコ、ブルーベリー、クランベリー、シロフサスグリ、アカフサスグリ、クロフサスグリ、クワ、ブラックベリー、スグリ、ラズベリー、サジー、イチゴ、イチゴの木の実、ブドウなどのベリー、或いはリンゴ、メロン、キウイ、サクランボ、ナツメ、プルーン、モモ、カキと、マンダリン、オレンジ、タンジェリン、グレープフルーツなどの柑橘類と、カモミール、キク、ダイダイ、スイカズラ、ジャスミン及びベニバナと、トマト、ホウレンソウ、セロリ、ニンジン、エンドウマメ、ケール、パセリ、クレソン、キャベツ、ブロッコリー、レタス、メキャベツ、カラードグリーン、カブラナ、フェンネル、タマネギ、茶、トウモロコシ、カカオ、コーヒー、タイム若しくはスィートレッドペッパーなどの、フラボノイド、ポリフェノール又はカロテノイドに富むその他任意の果物、野菜、種子、花、又は植物原料である、請求項1〜6のいずれかに記載のプロセス。
- ステップc)に従う乾燥を凍結乾燥により実施する、請求項1〜7のいずれかに記載のプロセス。
- 果物又は野菜又は植物からのみ由来する水抽出性の親油性及び非親油性の生物活性成分から成る水分散性の一次組成物であって、請求項1〜8のいずれかに記載のプロセスにより得られる一次組成物。
- 水分散性の粉末組成物である、請求項9に記載の一次組成物。
- 経口投与用の食品、栄養補助食品、ペットフード製品、ペット用栄養補助食品又は化粧品製剤を調製するための、請求項9及び10のいずれかに記載の一次組成物の使用。
- 皮膚の健康向上、特に皮膚の光防護、又は加齢に対する皮膚組織の防護を意図した経口用又は化粧用組成物を調製するための、請求項9及び10のいずれかに記載の一次組成物の使用。
- 請求項9及び10のいずれかに記載の一次組成物を含む、経口投与用の食品、栄養補助食品、ペットフード製品、ペット用栄養補助食品又は化粧品製剤。
