Description
本発明の分野は、骨の修復および交換である。より詳細には、本発明は、天然の骨と比較し得る機械的特性を有する、機械加工可能な合成骨代用材料インプラントに関する。
骨の空隙、欠損、および損傷のための処置は、構造的な完全性を提供しかつ新規の骨の形成を誘導しなくてはならない。特に、脊椎固定は、骨融合物の塊の形成において、隣接する椎骨間の架橋を作製することによって脊柱を固定するように設計される。初期の脊椎固定方法は、作用を受ける椎骨にわたる金属プレートまたはロッドを用いる脊柱を固定すること、および埋込まれた金属部品の周りに骨の融合が起こることを可能にすることを含んだ。種々の他の型の金属インプラントもまた、脊椎固定手順において使用されてきた。しかし、金属インプラントの強度は、取り囲む骨を遮蔽する圧力を引き起こし、これは、固定をもたらす天然の骨の成長を遅くする。さらに、金属インプラントは、インビボで天然の骨に再構築され得る永久的な外来性の物体である。さらに、金属製デバイスを移植するための多くの外科的手順は、長くかつ複雑である。
本発明は、天然の骨と比較し得る機械的特性を有し、かつインビボで骨に再構築し得る機械加工可能な骨代用材料インプラントを提供する。このインプラントは、脊椎固定における使用のために十分な強度を提供する。このインプラントは、生理学的条件下で反応して低結晶性ハイドロキシアパタイトを形成し、最終的には骨に改質(例えば、再構築)される、十分に混合された前駆体物質を含む。
本発明は、高い圧縮強度および均一な多孔度を有する骨代用材料インプラントを提供する。このインプラントは、生理学的条件下で反応して低結晶性ハイドロキシアパタイトを形成し、最終的にインビボで骨に再構築される、十分に混合された固体前駆体物質を含む。このインプラントは、密度および強度の増大のために生体適合性ポリマーを含み得る。このインプラントはまた、埋込み部位において有用である治療剤(例えば、抗生物質または骨成長刺激タンパク質)を含み得る。
アモルファスリン酸カルシウムの合成
2167mL蒸留水中、150gリン酸水素二ナトリウム七水和物(Na2HPO4・7H2O)の溶液を調製し攪拌した。83.3g NaOH、50g NaHCO3、および3.3g ピロリン酸ナトリウム十水和物(Na4P2O7・10H2O)を連続してこの溶液に加え、溶液1を生成した。
リン酸二カルシウム二水和物の調製
1L蒸留水中、20gリン酸水素二アンモニウム((NH4)2・HPO4)を溶解して、0.300モル/Lの濃度を有する溶液3を調製した。溶液3のpHが7.0から9.0の間であることを確認した。
骨代用材料だぼ
重量で等量部のアモルファスリン酸カルシウム(ACP)およびリン酸二カルシウム二水和物(DCPD)の混合物の100gのロット(約250mL)を、3440mL アルミナセラミックの広口瓶中で、ボールミリングによって均一な粉末にまで粉砕した。ミリングを、750mLの媒体(Y2O3によって安定化された高純度のジルコニウムオキサイドの10mm直径のボール)を使用して、100rpm、3時間実行した。得られる粉末を、>125μmのサイズの粒子を除去するためにふるいにかけた。直径17mmおよび23mm長の固体円筒形だぼが、一軸性圧縮を使用して、ふるいをかけた粉末から形成した。一軸性成形が実行され、型の中で0.25トンの力を粉末に適用することによって円筒形状を作製した。次いで、等方圧プレスを、30,000psiから44,000psiの間で、室温での低温(CIP)、または約66℃で暖温(WIP)のいずれかで実行した。ミリング後に加えたポリマー粉末、またはポリマー繊維を、いくつかのだぼにおいて含めた。いくつかのだぼは、等方圧プレス後に、1100℃で、アルゴン大気中で、焼結させた。
コポリマー粉末を伴う骨代用材料だぼ

Claims (15)
- i)アモルファスリン酸カルシウムと第2のリン酸カルシウムの粒子の間の固体-固体混合と反応形態への相転換を生じる固体反応を促進する力の下での、該粒子の高エネルギー衝撃ミリングであって、該粒子が1.2と1.68との間のカルシウム対リン酸の原子の比を有し、該第2のリン酸カルシウムが、リン酸二カルシウム二水和物、メタリン酸カルシウム、リン酸七カルシウム、リン酸三カルシウム、ピロリン酸カルシウム二水和物、ピロリン酸カルシウム、およびリン酸八カルシウムからなる群より選択され、かつ、該ミリングが、該粒子がナノメータースケールで混合され、その界面で予め反応させられている粉末を形成する、高エネルギー衝撃ミリング、ならびに
ii)乾燥圧縮粉末骨インプラントを形成するための該粉末の等方圧プレス
によって形成される乾燥圧縮粉末骨インプラントであって、
該インプラントは、組成物的に均一であり、少なくとも60MPaの圧縮強度を有し、水和により低結晶性ハイドロキシアパタイトを形成することができ、該低結晶性ハイドロキシアパタイトは、ナノメータースケールの結晶構造を有し、かつ広範な、十分に規定されていないX線回折パターンにより特徴付けられる、乾燥圧縮粉末骨インプラント。 - 機械加工物である、請求項1記載の骨インプラント。
- 第2のリン酸カルシウムはアモルファスリン酸カルシウムとは異なる結晶性を有する、請求項1記載の骨インプラント。
- 第2のリン酸カルシウムがリン酸二カルシウム二水和物である、請求項1記載の骨インプラント。
- 非毒性であり、かつ患者において所望でない生理学的な応答を引き起こさないポリマー粉末をさらに含む、請求項1記載の骨インプラント。
- 少なくとも120MPaの圧縮強度を有する、請求項5記載の骨インプラント。
