ウルケニア種SAM 2179株をじゃま板を有する300ml容量の三角フラスコ中の培地(50ml)において培養した。
発酵培地:グルコース 150g/L
コーンスティープリカー 3.75g/L
KH2PO4 3g/L
Na2SO4 1g/L
MgCl2・6H2O 1g/L
CaCl2・2H2O 0.3g/L
(NH4)2SO4 5g/L;
50mLフラスコあたりのCaCO3添加:培地1.1 0g/L

培地1.5 10g/L;
pH値をNaOH及びオートクレーブで6.0に設定。
グルコースが完全に消費するまでウルケニア種SAM 2179株をじゃま板を有する300ml容量の三角フラスコ中の培地(50ml)において培養した。
発酵培地:グルコース 150g/L
コーンスティープリカー 3.75g/L
KH2PO4 3g/L
Na2SO4 1g/L
MgCl2・6H2O 1g/L
CaCl2・2H2O 0.3g/L
(NH4)2SO4 5g/L;
50mLフラスコあたりのCaCO3添加:培地1.4 5g/L
培地1.5 10g/L;
pH値をNaOH及びオートクレーブで6.0に設定。
ウルケニア種SAM 2179株をじゃま板を有する300ml容量の三角フラスコ中の培地(50ml)においてグルコースが完全に消費するまで培養した。7.5g/LのCaCO3濃度及び修飾前培養により発酵が最適化された。DH1培地中の静置培養の代わりに前培養を用いる場合、同一培地で振とう培養を用いた(48時間、150rpm及び28℃)。
前培養培地:DH1培地

酵母抽出物(g/L) :12.5[Difco]
トロピックマリン(g/L) :16.65[ドイツ国ワールテンベルクに所在のDr.Biener GmbH];
HClでpH値を6.0に設定;
発酵培地:グルコース 150g/L

KH2PO4 3g/L
Na2SO4 1g/L
MgCl2・6H2O 1g/L
CaCl2・2H2O 0.3g/L
(NH4)2SO4 5g/L;
50mLフラスコあたりのCaCO3添加:培地1.6 7.5g/L;
pH値をNaOH及びオートクレーブで6.0に設定。
ウルケニア種SAM 2179株をグルコースが完全に消費するまで5L容量の発酵装置において培養した。
発酵培地:グルコース 150g/L
コーンスティープリカー 3.75g/L
KH2PO4 3g/L
Na2SO4 1g/L
MgCl2・6H2O 1g/L
CaCl2・2H2O 0.3g/L
(NH4)2SO4 5g/L;
pHコントロールありの発酵の場合:H3PO4及びオートクレーブでpH値を4.0に設定;
pHコントロールなしの発酵の場合:NaOH及びオートクレーブに加えて7.5g/のでCaCO3でpH値を6.0に設定。
シゾキトリウム種SR 21株をじゃま板を有する300ml容量の三角フラスコ中の培地(50ml)においてグルコースが完全に消費するまで培養した。
前培養培地:GY培地

酵母抽出物(g/L) :12.5[Difco]
トロピックマリン(g/L):16.65[ドイツ国ワールテンベルクに所在のDr.Biener GmbH]
HClでpH値を6.0に設定;

Claims (8)
- CaCO3をpH値安定化のための必須手段として含む発酵培地においてヤブレツボカビ(Thraustochytriales)目の微生物を培養する方法であって、
当該発酵培地のCaCO3含量が7.5±0.5 g/Lであり、
発酵開始前に対応する酸または塩基を添加することにより発酵培地のpHを5〜7の範囲に設定し、
当該微生物が乾燥バイオマスあたり10%を超えるDHAを産生し、
培養後、当該微生物および/または培地からPUFAを単離することを含む培養方法。 - 当該微生物が乾燥バイオマスの重量単位あたり25重量%以上の油を産生する請求項1に記載の方法。
- 当該微生物が乾燥バイオマスの重量単位あたり14重量%以上のDHAを産生する請求項1に記載の方法。
- 微生物が乾燥バイオマスあたり1%以上のDPAを産生する請求項1〜3のいずれか1項に記載の方法。
- 当該培地がグルコース、コーンスティープリカー、塩化マグネシウム、塩化カルシウム、炭酸カルシウム、硫酸ナトリウム、硫酸アンモニウム及びリン酸水素カリウムを含む請求項1〜4のいずれか1項に記載の方法。
- 培養を10〜40℃で実施する請求項1〜5のいずれか1項に記載の方法。
- 培養を1〜10日間、実施する請求項1〜6のいずれか1項に記載の方法。
- CaCO3をpH値安定化のための必須手段として含むヤブレツボカビ(Thraustochytriales)目の微生物を培養するための培地であって、
当該培地のCaCO3含量が7.5±0.5 g/Lであり、
発酵開始前に対応する酸または塩基を添加することにより当該培地のpHを5〜7の範囲に設定し、
当該微生物が乾燥バイオマスあたり10%を超えるDHAを産生し、
当該培地の使用は、培養することおよびその後当該微生物および/または培地からPUFAを単離することを含む培地。
