(c)炭水化物、脂肪及び窒素源から成る群より選択される少なくとも1つのエネルギー源、及び場合により
(d)ビタミンD
を含有する栄養組成物、例えば動物栄養補助飼料を提供する。
(i)ダイズの形態又は何らかの適切なダイズ抽出物又は濃縮物の形態、例えばダイズ粉、乾燥モヤシ、豆乳の形態、又はダイズからの乾燥水抽出物として使用し得る、ダイズ由来タンパク質;又は
(ii)乳タンパク質、例えば、乳清粉末、カゼインカルシウムなどのカゼイン塩及び/又は全脂粉乳又は好ましくは脱脂粉乳の形態で使用し得る、乳清由来タンパク質又はカゼイン塩、及び/又は

総エネルギー供給量の5から45%の、ダイズ由来タンパク質、乳タンパク質、必須アミノ酸の混合物及びアルギニンから成る群より選択される1又はそれ以上の窒素源;
総エネルギー供給量の0.1から15%の、ω−3及びω−6多不飽和脂肪酸を含む1又はそれ以上の脂肪源
を含む。
総エネルギー供給量の20から35%の、ダイズ由来タンパク質、脱脂粉乳及びカゼイン塩から成る群より選択される1又はそれ以上の窒素源;
総エネルギー供給量の3から12%の、ω−3及びω−6多不飽和脂肪酸を含む1又はそれ以上の脂肪源を含む。
(a)本発明の有効成分、例えばγ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシド、
(b)医薬として許容される担体、例えば動物によって生理学的に良好に耐容される担体
を含有する個別単位用量形態の医薬組成物、例えば動物薬又は獣医用組成物に関する。
1.1 親水性タマネギ画分、画分Aを得るために、吸着カラムクロマトグラフィーを使用する。85%エタノール水溶液を使用してカラムにスラリー充填する。充填後、固定相(Amberlite XAD−4)をエタノール400ml、85%エタノール水溶液500ml、水2500ml及び85%エタノール水溶液500mlで段階的に洗浄する。洗浄後、固定相を、分離のために使用する第一溶媒、すなわち15%エタノール水溶液(v/v)で予備処理する。
サッカリドを含む画分を「画分A2」と称し、サッカリドを含まない画分を「画分A1」と称する。
用量をmg/mlで示し、結果を吸収孔/酒石酸抵抗性酸性ホスファターゼ陽性(TRAP+)多核細胞±SEMとして示す。
1倍比例量(12mg/ml)で試験した画分A1は、破骨細胞活性を有意に阻害しない。しかし、2倍用量(24mg/ml)は、破骨細胞活性を0.144の吸収孔/細胞比に有意に低下させ(12mg/mlに比して−40%)、30mg/mlではより一層強力な破骨活性の阻害、すなわち0.015及び0.020吸収孔/細胞(12mg/mlに比して−90%)が測定できる。画分A1は破骨細胞活性を阻害する化合物を含むと結論される。

用量をmg/mlで示し、結果を吸収孔/酒石酸抵抗性酸性ホスファターゼ陽性(TRAP+)多核細胞±SEMとして示す。分画の間の損失を相殺するため、画分A1−2を除き、全ての画分を、画分Aと比較してそれぞれの収量の1倍、2倍及び3倍比例量で試験する。
画分A1−1、A1−2及びA1−3において阻害は測定されない。これら全ての画分の吸収孔/細胞比は、陰性対照のSEMの95%信頼区間内に位置する。全く糖類を含まない画分A1−4は、3倍用量で有意の破骨細胞活性阻害を示す。1倍及び2倍用量での細胞活性の見かけ上の刺激は、細胞数の低下によるものである。画分A1−4をさらなる分画のために選択する。
0.00625%ギ酸で緩衝した無勾配水/アセトニトリル系を溶媒として使用して、画分A1−4をSR−RP−HPLCで4つの画分、A1−4A、A1−4B、A1−4C及びA1−4Dにさらに分画する。画分A1−4Bは画分A1−4の2つの少量の化合物を含み、画分A1−4Cは画分A1−4の最も主要な化合物から成り、画分A1−4A及びA1−4Dは前留出と後留出(prerun and afterrun)である。画分A1−4AとA1−4Dをさらなる試験のために一緒にプールする。全体として、各ランの実施時に0.125mgを適用し、各画分をプールする、65回の単一HPLCの実施により、画分A1−4 8.125mgを分離する。分画は、HPLC装置の出口でタップを切り替えることによって手動で実施する。
画分A1に相当する3倍比例量の画分A1−4及び画分A1−4Cは骨吸収を有意に(p<0.5)阻害する。画分A1−4CはA1−4と同様に破骨活性を阻害し、A1−4Cが画分A1−4の唯一の活性化合物であることを示す。構造解明実験に十分な量を回収するためにA1−4Cの単離を反復する。
2.1 A1−4CのRP−HPLC−ESI−MS2
A1−4Cの構造に関する第一構造情報を得るためにHPLC−ESI−MS装置を用いて質量分析を実施する。前記MSは四重極イオントラップを備える。フラグメンテーションは、35%の衝突エネルギーを使用して、正に荷電したイオン化分子をヘリウムガスと衝突させることによって達成される。
ESI−MSによって得た結果は、画分A1−4Cがg−GPeCSOであるという推定をさらに裏づけ、先のHPLC−ESI−MS実験結果と矛盾しない。
トリメチルシリル−プロパンスルホン酸を外部標準として使用して、D2O中で測定を実施する。
3.1 抽出。タマネギの乾燥薄片(Allium cepa L,Liliaceae)を粉砕し、400gを、60℃で1時間、エタノール水溶液(85%v/v)2000mLで2回抽出し、ろ過した。エタノールの蒸発後、水性残留物を凍結乾燥し、−20℃で保存した。
Claims (11)
- 骨吸収上昇を特徴とする疾患又は状態の治療、検査又は予防のための、ヒト又は動物用薬剤の製造におけるγ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドの使用であって、前記薬剤が少なくとも2mMのγ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドを含む、前記使用。
- 骨吸収上昇を特徴とする疾患又は状態が、パジェット病、腫瘍誘導性骨疾患又は骨粗しょう症又はこれらの任意の組合せである、請求項1に記載の使用。
- 少なくとも2mMのγ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドを含む、骨吸収上昇を特徴とする疾患又は状態の治療、検査又は予防のための医薬組成物。
- 骨吸収上昇を特徴とする疾患又は状態が、パジェット病、腫瘍誘導性骨疾患又は骨粗しょう症又はこれらの任意の組合せである、請求項3に記載の医薬組成物。
- 少なくとも2mMのγ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドを含有する、骨吸収を阻害するための医薬組成物。
- 少なくとも2mMのγ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドを含有する、骨吸収上昇の食事療法に使用するための医薬組成物。
- γ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドと医薬として許容される担体を含有する、個別単位用量形態の請求項3〜6のいずれか一項に記載の医薬組成物。
- 動物にとって生理学的に許容される1又はそれ以上の担体と少なくとも2mMのγ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドの混合物を均質化することを含む、骨吸収上昇を特徴とする動物における疾患又は状態の治療又は予防のため又は動物における骨吸収上昇の管理のための獣医用組成物を製造するための方法。
- 骨吸収上昇を特徴とする疾患又は状態が、パジェット病、腫瘍誘導性骨疾患又は骨粗しょう症又はこれらの任意の組合せである、請求項8に記載の方法。
- γ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドが破骨細胞の吸収活性を用量依存的に阻害する、請求項1または2に記載の使用。
- γ−L−グルタミル−トランス−S−1−プロペニル−L−システインスルホキシドが破骨細胞の吸収活性を用量依存的に阻害する、請求項3〜7のいずれか一項に記載の医薬組成物。
