本発明は、質量分析用の検体試料の調製に使用される試料調製プレートに関する。特に、本発明は、表面での小さな液滴の操作が容易であり、試料の損失が最少化されていながら、1種または複数の検体を含む試料を濃縮し、また、汚染物質を除去することにより試料を精製するために使用される試料プレーに関する。
3pmoleのウシ血清アルブミン(BSA)のin situトリプシン消化と、それに続く消化ペプチドのナノスプレ質量分析の例が図5に示されている。BSAのトリプシン分解ペプチドが矢印で示されている。
Claims (16)
- プラットホームを備え、
前記プラットホームは基板と1つまたは複数のウェルをもち、
各ウェルが、ひとつ以上の周囲の疎水性壁面領域、および、親水性中央基底領域、を有する容器であり、
各ウェルの容積が0.5μLから100μLであって、水性試料を前記親水性中央領域上で濃縮するように構成される
ことを特徴とする、質量分析用試料プレート。 - プラットホームを備え、
前記プラットホームは基板と1つまたは複数のウェルをもち、
各ウェルが、ひとつ以上の周囲の疎水性壁面領域、および、親水性中央基底領域、を有する容器であり、
各ウェルの容積が100μLであって、水性試料を前記親水性中央基底領域上で濃縮するように構成される
ことを特徴とする、質量分析用試料プレート。 - 前記周囲の疎水性壁面領域が、Teflon(登録商標)、ポリ(ジメチルシロキサン)、および類似物からなる群から選択される材料からなる請求項1もしくは2に記載の試料プレート。
- 前記親水性中央基底領域が、ポリスチレン、および類似物からなる群から選択される材料からなる請求項1もしくは2に記載の試料プレート。
- 前記基板が、プラスチック、PTFE、Teflon、ポリプロピレン、プラスチック、ステンレス鋼、アルミニウム、ガラス、シリコン、および類似物から選択される材料からなる請求項1もしくは2に記載の試料プレート。
- (a)プラットホームを備え、前記プラットホームは基板と1つまたは複数のウェルをもち、各ウェルはひとつ以上の周囲の疎水性壁面領域と親水性中央基底領域とをもつ容器であり、各ウェルの容積が0.5μLから100μLであって、水性試料を前記親水性中央基底領域上で濃縮するように構成される、質量分析用試料プレートを供用すること;
(b)前記試料プレート内のウェルに液体試料物質を付けること;および
(c)前記ウェル内で試料物質が濃縮されて残されるように前記試料を乾燥させること;
を含む質量分析用試料調製方法。 - (a)プラットホームを備え、前記プラットホームは基板と1つまたは複数のウェルをもち、各ウェルはひとつ以上の周囲の疎水性壁面領域と親水性中央基底領域とをもつ容器であり、各ウェルの容積が100μLであって、水性試料を前記親水性中央基底領域上で濃縮するように構成される、質量分析用試料プレートを供用すること;
(b)前記試料プレート内のウェルに液体試料物質を付けること;および
(c)前記ウェル内で試料物質が濃縮されて残されるように前記試料を乾燥させること;
を含む質量分析用試料調製方法。 - 前記周囲の疎水性壁面領域が、Teflon(登録商標)、ポリ(ジメチルシロキサン)、および類似物からなる群から選択される材料からなる請求項6もしくは7に記載の方法。
- 前記親水性中央基底領域が、ポリスチレン、および類似物からなる群から選択される材料からなる請求項6もしくは7に記載の方法。
- 前記基板が、プラスチック、ポリプロピレン、Teflon、ステンレス鋼、アルミニウム、ガラス、シリコンなどから選択される材料からなる請求項6もしくは7に記載の方法。
- (d)前記試料に溶剤の液滴を付けること;および
(e)(d)の前記試料を、スタティックナノスプレMS分析のために、導電性被覆された、引き伸ばされた先端部をもつホウ珪酸ガラスキャピラリ、または、微細加工されたノズルへ移すこと;
をさらに含む請求項6もしくは7に記載の方法。 - 前記溶剤が有機成分と酸成分を含み、前記有機成分が前記溶剤の約40%以上を占め、前記酸が前記溶剤の約0.1%から10%の間であり、残りの成分が水である請求項11に記載の方法。
- (d)前記試料に溶剤の液滴を付けること;
(e)(d)の前記試料をMALDIターゲットプレート上に移すこと;および
(f)MALDI MS分析のために、MALDIマトリックスと(e)の混合物を作ること;
をさらに含む請求項6もしくは7に記載の方法。 - 前記溶剤が有機成分と酸成分を含み、前記有機成分が前記溶剤の約40%以上を占め、前記酸が前記溶剤の約0.1%から10%の間であり、残りの成分が水である請求項13に記載の方法。
- (d)前記試料に溶剤の液滴を付けること;および
(e)前記液滴を試料バイアルに移すこと;
