・ VVのK1Lコード配列を含むK1Lマーカー遺伝子および転写ユニット、好ましくは、その真正プロモーターの転写制御配列を含む、
・ 相同組換えによって組換えMVAから前記マーカーを後に除去するための、K1Lマーカー遺伝子をフランキングする2つのDNA配列、好ましくは、2つの同一挿入DNA断片(例えば、E.coli LacZ由来)、
・ クローニングサイト、好ましくは、任意に異種遺伝子が挿入されるマルチプルクローニングサイト、
・ ワクシニアウィルスに特異的な転写のためのプロモーター、好ましくはワクシニアウィルス誘導プロモーター、またはワクシニアウィルスに特異的な転写を可能にする異種ポックスウィルスプロモーター、または、ワクシニアウィルスに特異的な転写を可能とする合成プロモーター、
という機能性関連組成(functionally linked components)を含み、
前記組成が、上述の本発明の変異MVAゲノム内の非必須部位、MVAゲノム内の欠失III部位、または、MVAゲノム内のK1Lを遺伝子工学で欠失させた部位への、外来遺伝子の標的挿入に必須である2つのMVA-DNA配列によりフランキングされている。

制限エンドヌクレアーゼHindIIIに対するMVAゲノムの概略地図を上に示す。HCVコード配列は、ワクシニアウィルス早期/後期プロモーターP7.5の転写制御下に置かれており、MVAゲノム内の欠失III部位で相同組換えにより挿入されている。flank1およびflank2は、欠失III部位に隣接するMVA-DNA配列であり、MVAゲノムへの組換えをターゲットとするために働く。Rec2は、flank1の末端に対応し、相同組換えにより最終的な組換えウィルスのゲノムからK1L選択性マーカーを除去するための、283bp反復MVA-DNA配列の部分を示す。組換えMVA-HCV-NS3のゲノム構造の説明は以下に示す。

トランジェントK1L選択技術を改良するため、発明者らは、K1Lを有さないMVAの構築(例:MVA(IInew)、図1B)および新規DNAベクター構築物(トランスファープラスミド)(例:pIIIΔHR、図1C)のデザインという2つの手段を採用した。
1.1 MVAおよびMVA変異ウィルスの生育
MVAウィルスは、トリ胚線維芽(CEF)培養において原株であるCVA株の連続継代により生産される、非常に弱毒化されたワクチンウィルスである。ワクシニアのMVA株の生産、特性および使用に関する沿革の一般的なレビューとして、Mayrら(Mayr et al. in Infection 3, 614 [1975].)により開示された要約を参照することができる。CEFへの適合のため、他の細胞システムにおけるMVAウィルスの生育は、非常に制限される。容易にセルラインを維持できるという特徴を示す乳児ハムスター腎臓細胞(BHK−21)は、高効率CEFのように、MVA生育、組換え遺伝子の優れた発現をサポートし、発現ベクターおよび生菌組換えワクチンの発展において、標準化されたMVAの生育のために推奨される(Drexler et al. 1998, J. Gen. Virol., 79, 347-352)。
出来るだけ精製され、宿主細胞に特異的な成分を含まないウィルス調製物を得るために行われる精製工程は、MVAおよびWRウィルスで同一とした(Joklik, Virology 18, 918 [1962], Zwartouw et al., J. gen. Microbiol. 29, 523529 [1962])。感染させて−30℃で保存した細胞培養を解凍し、残存細胞を振り落とすか、プラスチック基板でかき取り、細胞およびウィルスを遠心分離によって培地から除去した(Sorvall centrigue、GSAローター、1時間、500rpm、10℃)。ウィルスおよび細胞粒子からなる沈殿物を、1回PBS(沈殿量の10〜20倍)に懸濁し、懸濁液を上述のように遠心分離した。新たな沈殿物を10倍量のRSB緩衝液(10mM TrisHCl(pH8.0)、10mM KCl、1mM MgCl2)に懸濁し、残存している無傷細胞を分裂させ、細胞膜からウィルス粒子を遊離させるために、懸濁液を超音波(4mm直径チップを備えたLabsonic 1510、Bender and Hobein(Zurich, Switzerland);2×10秒、60ワット、室温)で簡単に処理した。細胞核および大きな細胞デブリスを、後の簡単な懸濁液の遠心分離において除去した(Sorvall GSAローター、DuPont Co., D6353 Bad Nauheim, FRG;3分、3000rpm、10℃)。沈殿物は、上述のように、再度RSB緩衝液に懸濁し、超音波処理、遠心分離処理を行った。遊離ウィルス粒子を含む回収した上清をあわせて、35%スクロース含有10mM Tris−HCl(pH8.0)10mlからなるパッドに層状に広げ、そして、Kontron TST 28.38/17 ローターで遠心分離した(Kontron Instrumente, Zurich, Switzerland; Beckman SW 27 rotorに相当;90分、14,000rpm、10℃)。上清をデカンタし、ウィルス粒子を含む沈殿を10mM Tris−HCl(pH8.0)10mlで回収し、超音波(2×10秒、室温、上述の機器)による簡単な処理でホモジネートし、さらなる精製のためにステップグラジェントに供した。このグラジェントステップは、それぞれ、スクロースの10mM Tris−HCl(pH8.0)溶液5ml(スクロース濃度ステップ:20%、25%、30%、35%および40%)の組成とした。グラジェントは、Kontron TST 28.38/17ローターを用いて、14,000rpm、10℃、35分で遠心した。この遠心後、ウィルス粒子を含むいくつかの分離ゾーンが、30%〜40%スクロースの間におけるグラジェント領域でみられた。この領域をグラジェント(10ml)から吸い上げ、スクロース溶液をPBS(20ml)で希釈し、ウィルス粒子を遠心分離によってそこから沈殿させた(Kontron TST 28.38/17 ローター、90分、14,000rpm、10℃)。純粋なウィルス粒子でほぼ構成されている沈殿を、ウィルス濃度が平均1〜5×109pfu/mlに相当するようにPBSで回収した。精製されたウィルス原液は、後続の実験のため、直接またはPBSで希釈して使用した。
2.1 K1L欠失プラスミド
MVAゲノムから、MVA ORF 022L(Antoine, G., F. Scheiflinger, F. Dorner, and F. G. Falkner. 1998. Virology 244:365-396に開示されているように、全欠失配列は、MVAゲノムのnt20719〜21169から構成される)のプロモーター配列とともに、残りの263bpのK1L配列を除去するために、発明者らは、欠失プラスミドpIInewLZ-gpt-del(pUCII−LZのflank 1)(14)を構築した。このプラスミドは、MVAゲノム内の欠失II部位(the site of deletion II)への相同組換えに予め使用されるものであり、K1L遺伝子を含まず、以下のPCRによって単離された新たなflank(K2L)により置換されている。前記PCRは、MVAゲノム遺伝子から、プライマーIIf1newAおよびIIf1newBを用いて行われる。

さらに、N2Lオープンリーディングフレーム(ORF)の5’領域315bp反復(以下のプライマーdelIIAおよびdelIIBを用いたPCRにより単離)は、LacZ-gpt-カセットの下流に挿入され、これによってpIInewLZ-gpt-delが作製され、N2L遺伝子配列の相同組換えによりマーカーカセットを欠失できる。

MVA(IInew)は、MVA(クローン分離株F6(clonal isolate F6))を感染させた初代トリ線維芽(CEF)へのpIInewLZ-gpt-del DNAのトランスフェクションによって生産され、続いて、上述のような(7)ミコフェノール酸存在下でβ−ガラクトシダーゼを生産するウィルスの選択が行われた。組換えMVAの選択後、選択プレッシャーを除去し(selective pressure was removed)、マーカーフリーのウィルスを単離した(図1B)。
3.1 ベクタープラスミドの構築
さらに、MVAトランスファープラスミドにおけるMVAゲノム配列とMVA反復配列との間での相同組換えの可能性を排除するために、我々は、lacZ遺伝子由来の2つの短い216bp反復配列により挟まれた、K1Lマーカーカセットを含む新たなプラスミドpIIIΔHRをデザインした。
感染/トランスフェクション実験における組換え効率についてのこれらの変化の効果を測定した。CEF細胞を、MVA(F6)またはMVA(IInew)で感染し、pIIIdHR-gfpまたはpIIIΔHR-gfpでトランスフェクトした。サンプルは、48時間で回収され、一部分がRK13モノレイヤーの感染に使用された。3日後、感染させたモノレイヤーを完全に回収し、RK13細胞への第2継代を行った。3日のポスト感染で、多数の可視/gfp蛍光細胞凝集体が、光/UV光顕微鏡観察により測定された。それから、発明者らは、例えば、K1Lマーカーカセットやgfp遺伝子のMVAへの挿入のような、正しい組換え事象のパーセンテージを算出するためにこのデータを使用した(表1)。発明者により作成された通常の選択システム(MVA(F6)およびpIIIdHR-gfp)を使用したところ、第2RK13継代後におけるK1Lおよびgfpに対するて全てのフォーカス組換えの30.5%であることがわかった。MVAゲノムからの残りのK1L配列の除去は、適切な組換え事象をわずかに促進した(45%、MVA(IInew)およびpIIIdHR-gfp)。これに対して、MVAトランスファープラスミドからのflank1反復配列の除去は、残りのK1L配列を持つMVAを使用した場合でさえ、所望の組換え事象(71%)という明らかな増加結果となった。発明者らは、新規MVAおよびgfpトランスファープラスミド(MVA(IInew)およびpIIIΔHR-gfp)を使用した場合に、事実上100%効率を得た。RK13細胞におけるはじめのブラインド継代は、希釈液のプレーティングおよびフォーカス量の測定前に行われた。これは、第1RK13継代におけるGFPポジティブ細胞凝集体の測定をする際、結果が適切な組換え事象の高い数値を示唆し、さらに、次の継代に入る時、約半分のウィルス単離体が、GFPポジティブウィルスフォーカスを誘導できないことが観察されたためである。この観察は、おそらく不安定な単一組換え事象および/またはキャリーオーバープラスミドDNA由来のとランジェントgfp発現に基づく。
HCV-J株(遺伝子型1b)のnonstructural 3(NS3)オープンリーディングフレーム(ORF)を発現する組換えMVA(rMVA)の構築は、内因性K1L配列を欠くMVA親ウィルス(MVA-IInew)を基礎としてrMVAを生産し、異種、非MVA反復要素(LacZ)を有するプラスミドを使用する、我々の新規技術の有益な使用の実施例として供給できる。はじめから、我々は、Staib C.,ら(Staib C., et al. 2000, Biotechniques 28:1137-1148)に開示されているような我々の通常の宿主範囲選択プロトコールを使用する、HCV-J株(遺伝型1b)のHCV NS3遺伝子(MVA-HCV/NS3)の発現のためのrMVAの生産を試みていた。

- DNAベクター構築物であって、
ワクシニアウィルス(VV)K1L遺伝子をコードする配列または機能的に同等な遺伝子をコードする配列、および、外来タンパク質またはその機能性部分をコードするDNA配列を含み、
VVK1Lおよび外来タンパク質コード領域が、それぞれ、MVAゲノム内の非必須部位をフランキングするDNA配列によってフランキングされ、
非必須部位が、MVAゲノムにおいてK1L遺伝子配列およびそのプロモーター配列またはこれらの配列の機能性部分が、機能を有するK1L遺伝子との間の相同組換えを妨げるのに十分な程度に欠失または変異した部位である、DNAベクター構築物。 - 外来タンパク質が、治療ポリペプチドおよび病原性物質ならびにそれらの機能性部分からなる群由来の異種タンパク質である、請求項1記載のDNAベクター構築物。

- 組換えMVAの生産方法であって、MVAゲノムにおけるK1L遺伝子配列およびそのプロモーター配列またはこれらの配列の機能性部分が機能を有するK1L遺伝子との間の相同組換えを妨げるのに十分な程度に欠失または変異しているMVA変異体により、MVA宿主細胞を感染させる工程、請求項1から12のいずれか一項に記載のDNAベクター構築物で、前記宿主細胞をトランスフェクトする工程、および、ウサギ腎臓RK-13細胞、または、MVAもしくは前記変異MVAの増殖生育を可能とするK1L遺伝子機能を本質的に必要とする他の細胞の生育により、回復したMVAを選択する工程を含む組換えMVAの生産方法。
