Description
β−カロテンの変換の測定
アジズ(Aziz)に従うβ−カロテン分解の測定
酵素活性は、A.ベン・アジズ(A.Ben Aziz)(1971年),Phytochemistry 10,1445に従って、β−カロテン変換活性として決定することができる。本明細書において、1酵素単位は、1分間あたりに1マイクログラムのβ−カロテンを変換する酵素の量として規定される(さらに、アジズ(Aziz)単位と称される)。
酵素活性はまた、ゾロン(Zorn)ら(2003年),Appl.Microbiol.Biotechnol.62:331−336に従って、β−カロテン変換活性として決定することができる。本明細書において、1酵素単位は、1分間あたりに1マイクロモルのβ−カロテンを変換する酵素の量として規定される(さらに、ゾロン(Zorn)単位と称される)。アッセイは以下の通りの行われる:100μlのβ−カロテンストック溶液(下記を参照のこと)を添加する前に、サンプルを含有する1.5mlの酵素を、キュベット中、27℃で5分間、プレインキュベートした。必要であれば、濃縮された培養上清を、クエン酸/リン酸緩衝液pH5.5(この緩衝液は、43mlの0.1Mクエン酸と56mlの2M Na2PO4溶液とを混合することによって調製した)で希釈した。吸収の減少を、温度制御されたセルホルダー中、分光光度計を使用して、15分間、450nmおよび27℃で、モニターした。曲線を、直線性について確認し、以下の等式に従い、曲線の直線部分で酵素活性を算出した:

β−カロテンストック溶液を、以下のようにして調製した:5mgのβ−カロテンおよび500mgのトゥイーン(Tween)−80を50mLのジクロロメタンに溶解した。ジクロロメタンを、ロータリーエバポレーター中、40℃および800mbarでエバポレートした。ほぼすべてのジクロロメタンがエバポレートされたら、30mlの水を添加し、残留するジクロロメタンを、ロータリーエバポレーターにおいて、そして最終的に窒素流において除去した。得られる溶液をろ過し、メスフラスコ中50mlまで水を満たした。溶液は、冷所(冷蔵庫)に貯蔵しなければならず、数日間のみ安定である。
漂白は、ゲリナス(Gelinas)、Cereal Chem.75,810−184(1998年)によって示される通り、内相または生地からのカロテノイドの抽出後に決定した。カロテノイドは、ゲリナス(Gelinas)(1998年)によって示される通り、パンの内相からの全脂質抽出を介して決定した。
ニオイヒメホウライタケ(Marasmius scorodonius)から得られるβ−カロテン変換酵素の培養およびその活性の決定
ニオイヒメホウライタケ(Marasmius scorodonius)から得られるβ−カロテン変換酵素の培養およびその活性の決定を、ゾルン(Zorn)ら(2003年)によって記載のように行った。ここで、ニオイヒメホウライタケ(Marasmius scorodonius)の培養コレクション(the Centraal Bureau voor Schimmelcultures−Utrecht、蘭国、寄託番号CBS850.87から入手可能)由来の菌糸体を使用して、乳化したβ−カロテンで補充した寒天プレートに播種した。プレートのインキュベーションは、24℃で14日間実施した。100mlの標準栄養溶液(SNL、30g/リットルのグルコースH2O;4.5g/リットルのアスパラギンH2O;1.5g/リットルのKH2PO4;0.5g/リットルのMgSO4;3.0g/リットルの酵母抽出物;5mg/lのCuSO4 *5aq、80mg/lのFeC13 *6aq、90mg/lのZnSO4 *7aq、30mg/lのMnSO4 *1aqおよび40mg/lのEDTAを含有する1ml/リットルの滅菌された微量栄養素溶液を含有する;滅菌前に1N NaOHでpHを6.0に調整した)を含有する300ml振盪フラスコに菌糸体を播種し、24℃で7日間、150rpmでの振盪インキュベーターにおいて、インキュベートした。微生物混入の不在についてプレ培養物を確認し、ウルトラ・タラックス(Ultra Turrax)により均質化し、主培養(500ml三角フラスコ中250mL)への播種に使用した。2日目から連日、2mlのサンプルを抜き出し、遠心分離して菌糸体を取り出し、分光光度アッセイにおいて活性を測定した。培養4日後、β分解活性は、1リットルの無細胞上清あたり約0.3ゾルン(Zorn)単位であった。
パプ製パン試験
標準的なベーキングプロセスでは、200gの小麦粉(160gの小麦粉(Kolibri(登録商標)−Meneba、蘭国)および40グラムの小麦粉(Ibis(登録商標)−Meneba、蘭国)の混合物)、1.4gのFermipan(登録商標)乾燥酵母(DSMベーカリー・イングラディエンツ(DSM Bakery Ingredients)、Delft、蘭国)、4gの塩、50ppmのアスコルビン酸、4ppmの真菌α−アミラーゼBakezyme(登録商標)P500(DSMフード・スペシャルティーズ(DSM Food Specialties)、Delft、蘭国)、60ppmの真菌ヘミセルラーゼBakezyme(登録商標)HS2000(DSMフード・スペシャルティーズ(DSM Food Specialties)、Delft、蘭国)および表1に記載の量のβ−カロテン分解酵素および116mlの水から、ピンミキサー中、6分間および15秒間で、パプ製パン(pup loaves)を調製した。生地の温度は28℃であった。混合直後、生地を各150gの2片に分け、丸めて、発酵キャビネット(proofing cabinet)において45分間、30℃で発酵させ、成形し、パンニングした。30℃で70分間の最終発酵後、生地を20分間、225℃でベイクした。
ミニチーズの調製
ミニチュアチーズを、シャキール−Ur−レーマン(Shakeel−Ur−Rehman)ら((Protocol for the manufacture of miniature cheeses、Lait,78,(1998年),607−620頁)により記載の通りに、生成した。生の牛乳を、30分間、63℃で加熱することによって、低温殺菌した。低温殺菌した乳汁を広口プラスチック製ボトル(ボトルあたり200mL)に移し、31℃まで冷却した。続いて、0.72mlのスターターカルチャーDS 5LT1(DSMヒストB.V.(DSM Gist B.V.)、Delft、蘭国)を、遠心ボトル中の200mlの低温殺菌した乳汁のそれぞれに添加し、乳汁を20分間、成熟させた。次いで、CaCl2(200mLの成熟させた乳汁あたり132μLの1mol.L−1溶液)を添加し、続いて、凝固剤(1mlあたり0.04IMCU)を添加した。実験が漂白酵素IまたはIIの使用に関与する場合、この酵素を凝固剤と共に添加した。

- 前記穀粉をベースとする製品は、小麦粉から作製される、請求項1に記載のプロセス。
- 前記酵素は、微生物から誘導される酵素調製物として添加されるか、前記酵素を産生することが可能な微生物によってインサイチュで産生される、請求項1又は2に記載のプロセス。
