Description
(1−1)連続流式PCR装置の製作
図2aに示したように、本発明による連続流式PCR装置において、3つの加熱ブロック21、22、23は銅を用いて製作し、断熱ブロック12はベークライトを用いて製作した。
図3b乃至図3eに示したように、多重連続流式PCR装置を製作した。前記装置において、加熱ブロック及び断熱ブロックの材質としては前記実施例(1−1)と同様にそれぞれ銅及びベークライトを用いた。
前記実施例(1−1)で製造した装置を用いてキャピラリーチューブ内でのPCR混合液の連続流相でPCRを遂行した。
PCRが終了した試料のDNA増幅の有無を確認するために、ゲル電気泳動を施した。前記実施例2で収得した10μLのPCR産物をTBE緩衝溶液上で2%アガロースゲル電気泳動で分析した。DNA増幅水準を確認するために、既存の市販の機器(MBS 0.2G,Hybaid, U.K.)を用いて増幅した陽性対照区用試料及びサイズマーカーを一緒にロードした。市販の機器におけるPCR条件は95℃で2分、そして95℃で30秒、60℃で1分、72℃で2分であった。このようなサイクルを33回繰り返し、72℃にて5分間放置した。前記PCR産物を4℃に冷却してPCR反応を終了した。
本発明者らは、本発明による連続流式PCR装置が互いに異なる組成を有するPCR混合液を順次に注入した場合でも各々の鋳型DNAに対する増幅反応を行うことができるか否かを調べた。

Claims (21)
- 複数の加熱ブロック−断熱ブロック組立体が温度調節可能な複数の加熱ブロックと組み立てられて2つ以上の独立的な反応の遂行を可能にし、それぞれの組立体上に流体注入口である第1末端と流体排出口である第2末端を有し、第1末端から第2末端への連続的な流体のフローを許容するキャピラリーチューブが巻きつけられていることを特徴とする高性能多重連続流式反応装置。
- ポリメラーゼ連鎖反応(PCR)を遂行することを目的とする請求項1記載の高性能多重連続流式反応装置。
- 前記加熱ブロックが、ヒーター及び温度センサーを用いて互いに異なる温度に調節されることを特徴とする請求項1記載の高性能多重連続流式反応装置。

- 前記断熱ブロックがベークライト又はアクリルポリマー樹脂を用いて製造されたことを特徴とする請求項1記載の高性能多重連続流式反応装置。
- 前記キャピラリーチューブが、ガラス、溶融シリカ、ポリテトラフルオロエチレン(PTFE)及びポリエチレンからなる群から選択された材質で製造されたことを特徴とする請求項1記載の高性能多重連続流式反応装置。
- 前記キャピラリーチューブの外壁が、ポリイミド又はポリテトラフルオロエチレンでコーティングされたことを特徴とする請求項1記載の高性能多重連続流式反応装置。
- 前記キャピラリーチューブの内壁が、トリメチルクロロシラン、ジメチルジクロロシラン、メチルトリクロロシラン、トリメチルメトキシシラン、ジメチルジメトキシシラン、及びメチルトリメトキシシランからなる群から選択された少なくとも1つ以上の物質でコーティングされたことを特徴とする請求項1記載の高性能多重連続流式反応装置。
- 前記キャピラリーチューブが、前記加熱ブロック−断熱ブロック組立体の外面に形成された螺旋溝に沿って固定されたことを特徴とする請求項1記載の高性能多重連続流式反応装置。
- 前記キャピラリーチューブが、前記加熱ブロック−断熱ブロック組立体の外面周囲に10回乃至50回巻き付けられていることを特徴とする請求項1記載の高性能多重連続流式反応装置。
- 蛍光を誘発する光源、蛍光を検出する検出器、及び光線をキャピラリーチューブに照射した後、発生する蛍光を検出器に集束させる光学装置を含む蛍光誘導装置と、
前記蛍光誘導装置及びキャピラリーチューブの相対的な位置を変化させることができる走査装置とを更に含み、実時間反応進行度を探知できることを特徴とする請求項1記載の高性能多重連続流式反応装置。 - 前記反応が実時間ポリメラーゼ連鎖反応であることを特徴とする請求項11記載の高性能多重連続流式反応装置。
- (a)請求項1に記載の装置におけるキャピラリーチューブの第1末端に1つ以上のPCR混合液を注入する段階と、
(b)適切な速度でPCR混合液の流速を制御しながら第2末端から排出されるPCR産物を回収する段階を含むことを特徴とする高性能多重連続流式核酸増幅方法。 - 請求項1に記載の装置におけるキャピラリーチューブを95〜100℃、45〜65℃及び65〜72℃に設定された加熱ブロックに順次に又は繰り返して接触させることを特徴とする請求項13記載の高性能多重連続流式核酸増幅方法。
- 前記キャピラリーチューブが、前記加熱ブロック−断熱ブロック組立体に10回乃至50回繰り返して接触することを特徴とする請求項13記載の高性能多重連続流式核酸増幅方法。
- 前記PCR混合液が、MgCl2、dNTP(dATP、dCTP、dGTP、及びdTTP)混合液、1つ以上のプライマー、1つ以上の耐熱性DNAポリメラーゼ、耐熱性DNAポリメラーゼバッファー、及び1つ以上の鋳型DNAを含むことを特徴とする請求項13記載の高性能多重連続流式核酸増幅方法。
- 前記プライマーが分子指標(molecular beacon)であることを特徴とする請求項16記載の高性能多重連続流式核酸増幅方法。
- 前記PCR混合液が、二本鎖DNAに特異的に結合して蛍光を放出することができる1つ以上のインターカレーティング色素(intercalating dye)を更に含むことを特徴とする請求項16記載の高性能多重連続流式核酸増幅方法。
- 前記PCR混合液が、ポンプによりキャピラリーチューブの第1末端から第2末端まで移動することを特徴とする請求項13記載の高性能多重連続流式核酸増幅方法。
- 前記段階(a)におけるPCR混合液が、連続的又は断続的に注入されることを特徴とする請求項13記載の高性能多重連続流式核酸増幅方法。
- 断続的に注入されるPCR混合液が互いに異なる組成を有する場合、1つの組成を有する組成物を注入した後、有機溶媒又は空気層を注入する段階を更に含むことを特徴とする請求項20記載の高性能多重連続流式核酸増幅方法。
