Description
【発明の属する技術分野】
本発明は、敗血症及び敗血症様全身性感染において、プロカルシトニン又はプロカルシトニンの部分ペプチド(partial peptides)の発生に関係する、新規な、実験的に確認された発見から導かれる、新規な診断及び治療の可能性に関する。

【従来の技術】
特許DE 42 27 454及びEP 0 656 121 B1及びUS 5,639,617は、敗血症の危険を有する患者及び敗血症の典型的な症候が見られる患者の血清又は血漿中のプロホルモンのプロカルシトニン及びそこから得られる部分ペプチドの測定が、早期検出にとって、すなわち敗血症に至らしめるかもしれない感染の検出及び非感染性の病因との鑑別、重大性の検出、及び、敗血症及び敗血症様全身性感染の治療の成果の評価にとって、有益な診断手段であること開示している。前記測定は、全身性微生物感染に起因する症状を非感染性の病因の他の症状から区別するための診断に特に有益であることが証明されており、該非感染性病因の他の症状とは、敗血症を示唆するかもしれないが、実際には全身性微生物感染に起因しないものであって、それらの臨床像によるが、例えば、個々の器官の非感染性炎症から、術後の拒絶反応又はガンまでの症状である。さらに、全身性炎症は局所性炎症とは区別され得る。

【発明が解決しようとする課題及び課題を解決するための手段】
プロカルシトニンは、カルシトニンのプレホルモンとして既知となっており、その完全なアミノ酸配列は古くから知られている(FEBS 167 (1984), 93-97頁)。プロカルシトニンは、甲状腺のC細胞において、通常の状態で産生されており、それから、特異的開裂によってホルモンのカルシトニンになり、さらに、部分ペプチドのカタカルシン及び57のアミノ酸を含むN−末端残基(「アミノプロカルシトニン」)となる。

敗血症の間のプロカルシトニンの生成に重要である器官もしくは細胞又は組織に関する実験資料によってサポートされているものもいくつか存在する前記技術文献中には異なる意見もあるが、敗血症のケースでは、甲状腺を完全に除去した患者からでさえも、著しく高いプロカルシトニンレベルが観察されることもあるので、敗血症患者の血中に検出可能なプロカルシトニンが、甲状腺の外で形成されると結論付けることが必要であった。

敗血症において「プロカルシトニン」として測定されるペプチドの性質に関して、実際、その特定のペプチドが、甲状腺においてカルシトニン前駆体として形成される完全な長さの既知のプロカルシトニンと完全に同一である必要はないことは、上述の患者におけるアウトセットから明らかにされている。しかしながら、敗血症のケースで形成されるプロカルシトニンが甲状腺で形成されるプロカルシトニンと異なるのかどうかという疑問は、現在まで答えが得られていない。あり得る違いは、既知のプロカルシトニンの、糖化(グリコシレーション)、リン酸化あるいは一次構造の修飾等の翻訳後の修飾、さらに、変性した、短くされたあるいは長くされたアミノ酸配列であった。今日まで分析アッセイ方法は、カルシトニン前駆体として既知のプロカルシトニンと、敗血症の場合に形成されるプロカルシトニンとの間の違いを明らかにしなかったので、敗血症のケースで形成されるプロカルシトニンは、カルシトニン前駆体と同一であり、ゆえに、既知の116アミノ酸のプロカルシトニン配列を有するペプチド(プロカルシトニン1−116)であると暫定的、一般的に見なされていた。

しかしながら、出願人の研究室における測定によって明らかにされ、本出願の実験部分により詳細に説明されているように、敗血症のケースで形成されるプロカルシトニンは、甲状腺で形成される完全なプロカルシトニン1−116とは、わずかだが重大な違いがある。見出された違いは、それから、新規な診断及び治療方法、そこで使用可能な物質、及び、遂行され得る科学的アプローチにおいて実施可能な多数の科学的結論を導き出した。

本出願において開示される発明の開始点は、敗血症及び敗血症様全身性感染のケースにおいて患者血清中に比較的高濃度で検出可能なプロカルシトニンが、116のアミノ酸を含む完全なプロカルシトニン1−116ではなく、そのアミノ末端がジペプチド分短くなっているが他は同一であり、114のアミノ酸のみのアミノ酸配列を有するプロカルシトニン(プロカルシトニン3−116)であるという驚くべき発見である。

完全なプロカルシトニンと比べて欠落している前記ジペプチドは、Ala-Pro構造を有している。完全なプロカルシトニン配列のアミノ末端の二番目のアミノ酸としてのプロリン残基を含むジペプチドの欠落は、特定のペプチダーゼ、すなわち、いわゆるジペプチジル−(アミノ)−ペプチダーゼIV(DP IV又はDAP IV又はCD 26)が、敗血症のケースで検出されるプロカルシトニン3−116の生成に役割を果たすかもしれないという推測を導く。

全身性感染又は敗血症と関連してジペプチジル−(アミノ)−ペプチダーゼIVの可能な役割を決定するために、発明者は実験により、前記生理的DAP IV濃度と敗血症の検出と相関関係があるのかどうかについての試験を行っている。得られた結果は相関関係を示した。

より正確に得られた結果はさらに、敗血症及び全身性感染のケースの高濃度のプロカルシトニンの発生は孤立した現象ではなく、同様に、他のプロホルモンもまた高濃度で測定可能であり、その結果、そのようなプロホルモンの測定が、プロカルシトニン測定に代えて可能であるか又は個々のケースのプロカルシトニン測定を補足するか、もしくは、さらにそれを診断上の意義のある方法で確認するのに適している、という仮説の展開をもたらした。

敗血症のケースの患者血清中に見出されるのが、完全なプロカルシトニン1−116ではなく、短くなったプロカルシトニン3−116であるとの発見も、敗血症治療にとって潜在的な興味ある問題である。Eric S. Nylenらの論文Crit Care Med 1998, Vol.26, No. 6, 1001-1006頁は、プロカルシトニンが炎症反応を維持し且つ増強しうるという事実に基づいて、敗血症のケースで生じているプロカルシトニンが、例えば代謝排泄物として形成される、診断上重要なマーカーであるというだけでなく、感染によって引き起こされる炎症過程におけるメディエーターとしての活性な役割を果たしているらしいことを示す、実験による発見を記載している。プロカルシトニンのこの役割は現在、論争の課題であり、その開示された試験結果は同意できる説明を与えていない。

アミノ末端でアミノ酸2つ分短くされたプロカルシトニンが敗血症のケースで生じているという上述の発見は、敗血症及び他の炎症性全身性感染のケースで活性な役割を果たすプロカルシトニンがおそらくこの短くされたプロカルシトニン3−116であり、また、完全な長さのプロカルシトニンペプチドを用いて行われた研究が、とりわけこの理由で、異なった又は矛盾した結果を与えたことを示唆している。多数の生理活性ペプチドが、開裂、例えば短いペプチド残基の最初の脱離、によってそれらの実際の活性形態に変換されることは周知である。既知の例はアンギオテンシンであり、著しく異なった生理活性を有するペプチドが、最初にテトラペプチド、次にジペプチド、そして最後に単独のアミノ酸の、連続的な脱離によって、14のアミノ酸を有する不活性なアンギオテンシノーゲンから形成される。その生理活性ペプチドのN−末端の比較的わずかな変更が、免疫学的過程で役割を果たし、対応するペプチドの活性にかなりの変化をもたらすことができるという事実は、ごく最近の数多くの出版物によって確認されているが、敗血症の病理的プロセスに対する言及はなされていない(例えば、J. 【発明の実施の形態】
プロカルシトニン3−116が実際に炎症過程と関連し、特異的分子レセプター又は類似した特異的バインダーがこの短くされたプロカルシトニンに対して存在するのであれば、プロカルシトニン3−116の使用、又は、プロカルシトニン3−116のレセプターと相互作用するアゴニスト及びアンタゴニストの使用によって、敗血症の進行に影響を与え、それがきっかけとなって起こる生理反応に、そして炎症過程にも影響を与えることができる新規な治療の可能性が開かれる。プロカルシトニン3−116の特異的バインダー、例えば、選択的抗体、の使用は、ここで伝えられる発見によって明らかにされる治療アプローチでもある。

結局、敗血症及び全身性感染のケースでジペプチジル−アミノペプチダーゼIVがプロカルシトニン3−116の生成に役割を果たすかもしれないということは、更なる仮説、すなわち、例えば適当な選択的バインダー、抗体又は類似のレセプター分子によってそれをブロックすることによりジペプチジル−アミノペプチダーゼIVの活性に影響を与え、それによって、敗血症及び敗血症様炎症過程に治療上の影響を与えることも可能かもしれないという仮説を導く。

【実施例】
実験セクション
A. 敗血症患者の血清からの内因性プロカルシトニンペプチドの単離と確認
重度の敗血症に苦しむ複数の患者からの血清サンプルを混合することによって、総容量68mlの混合血清が調製された。得られたプール血清中のプロカルシトニン濃度は、市販のプロカルシトニンアッセイ(LUMItest PCT, B.R.A.H.M.S. Diagnostica)を用いて測定したところ、280ng/ml(総量19μg)であった。前記プール血清は、同量のバッファー(68ml;10mM EDTA、1mg/mlマウス−IgG、2mg/ml ヒツジ−IgG、2mg/mlウシ−IgG、0.1mmolロイペプチン、PBS中50μMアマスタチン)と混合し、そのサンプル中に含まれるプロカルシトニンを、アフィニティークロマトグラフィーによって単離及び精製した。

このために、全てのプールサンプルを、0.5ml/minの流速で4回、連続して、アフィニティーカラム(0.5×1cm、抗カルシトニン抗体、PierceからのCarbolinkに結合、プロカルシトニン結合容量約20μg)に送った。それからそのカラムを30mlのPBSで洗浄し、結合したペプチドを、50mM酢酸(pH約2.0)を用いて溶離させた。カラム流出物を280nmでの吸収を用いて持続的にモニターし、酢酸によって溶離したタンパク質フラクションを集めた(最終容量2.0ml)。

この方法で集められた物質は、rpC18 カラムμBondapak 0.4×30mm(Watersより)での逆相HPLCによって精製した。その流速は1ml/minであり、移動相及び溶離条件は以下の表1に示す。

そのカラム流出物は、214nmでの吸収によって持続的に測定され、0.25mlのフラクションが集められた。市販のプロカルシトニンアッセイ(LUMItest PCT, B.R.A.H.M.S. Diagnostica)を用いて、PCT免疫反応性を検出できたフラクションを決定した。主要な免疫反応性を有する部分が、シャープなバンドとして51番目のフラクションに溶離したことが見出された。加えて、不均一な組成及び低いPCT免疫反応性を有するタンパク質フラクションが、39から49のフラクションにて得られた。

図1は、前記rp HPLCの、集められた各フラクションについて決定され、溶離したフラクションの吸光度(OD)を示す曲線に重ね合わせたPCT免疫反応性(ng PCT/mlで表される)を示す。

ポジティブなプロカルシトニン免疫反応性を有する全てのフラクションを、窒素ガス処理によって乾燥させた。その後、それらのサンプルをマススペクトロメトリーで分析し、N−末端塩基配列決定を行った。

前記マススペクトロメトリー分析(MSLDI-TOF法)では、図2に示されるプロファイルが、フラクション50−52に対して得られ、そのプロファイルから、12640±15のモル質量という結果となった。マススペクトロメトリーで調べられた他のフラクション(36−49、53−59)はすべて、12640未満のモル質量で不均一な質量分布を示した。それらの個々の質量は、フラクション50−52の質量の強度と比べて2%未満の強度を与えた。このように、敗血症患者血清中のプロカルシトニン免疫反応性が、12640±15の質量と関連があることが示された。

既知のプロカルシトニン1−116のアミノ酸3−116の配列と完全に対応した配列が得られた。その配列の理論上の質量は12627であったが、これはマススペクトロメトリーにより測定された12640±15の質量と一致する。

したがって、114のアミノ酸を含み且つプロカルシトニン3−116としてデザインされたプロカルシトニンペプチドが敗血症患者の血液中を循環することが示された。そのペプチドは、リン酸化又はグリコシル化等の翻訳後の修飾によって変化しない。

前記プロカルシトニン3−116は、可能性のある内因性プロカルシトニン部分ペプチドとしては、現在に至るまで科学論文で論じられておらず、それゆえに、当業者にとって、今日までに、具体的に、このペプチドを調製し、その性質についてそれを調べる理由もない。しかしながら、上記発見は、今や、遺伝子工学技術によって前記プロカルシトニン3−116の具体的な調製の理由をもたらしている。その調製は以下に記載される。

B.組み換えプロカルシトニン3−116のクローニング、発現及び精製
1.クローニング
プロカルシトニン3−116(以下、PCT 114と略す)をコードするDNAフラグメントを、適当なオリゴヌクレオチドプライマーを用いたPCR増幅によって、ヒト甲状腺cDNAプールから単離した。望ましいフラグメントが通常の手法(Ausubel FM, Brent R, Kingston RE, Moore DD, Seidman JG, Smith JA及びStruhl K (1991), Current Protocols in Molecular Biology, John Wiley & Sons, New York)によってクローニングされ、その正確なヌクレオチド配列がDNA塩基配列決定と、プロカルシトニンをコードする既知のDNA配列との比較とによって確かめられた。

PCT 114をコードするcDNAの発現にはベクターが用いられた。これは、T7プロモーターと、いわゆるpelBタンパク質のシグナルペプチドをコードする領域を含んでいる。このpelBシグナルペプチドは、クローニング及び発現後に形成される融合タンパク質が、発現に用いられたホスト細胞の細胞質を通ってそのペリプラズム部分に輸送されることを保証する。この輸送過程では、そのN−末端シグナルペプチドが、膜上に位置するシグナルペプチダーゼによって、同時に分離される(Stader JA及びSilhavy TJ (1990), Engineering Escherichia coli to secrete heterologous gene products, Methods Enzymol. 185, 166-187)。この手順は、見出された発現生成物が正確に望ましい配列を有することを保証する。この手順では、他の発現方法では必要とされるN−末端メチオニンは存在しない。

前記タイプのベクターへのPCT 114用のcDNAのクローニング、及びこのベクターによるE. coliの形質転換の後、プロカルシトニン3−116が発現した。発現したプロカルシトニン3−116を有するペリプラズムフラクションが、それ自身は既知の方法で単離した(Neu HC及びHeppel LA (1965), The release of enzymes from Escherichia coli by osmotic shock and during the formation of spheroblasts, J. Biol. Chem. 240, 3685-3692)。遠心(100,000g、30分、4℃)、及び、その上澄み液(0.2μm)のろ過の後、得られたろ液をアニオン交換クロマトグラフィーによって分離した。プロカルシトニン免疫反応性を有するフラクションは、敗血症血清からのPCT3−116の単離に関して記載されたように、合わされて、逆相HPLCによって精製された。

プロカルシトニン免疫反応性を有するすべてのフラクションは、合わされて、凍結乾燥された。SDS-PAGEを用いてその物質をチェックすることによって示されるように、つまり、得られる物質は少なくとも純度95%である。

プロカルシトニン3−116として発現し、精製されたペプチドの同一性は、マススペクトロメトリーと配列分析によって確認された。

得られた組み換えプロカルシトニン3−116は、新規な組み換えペプチドであり、この形態で、免疫試薬の調製に用いられることができ、また、治療上の適性、あるいは、上述の出版物(Eric S Nylen, Loc. cit.)との関係で、予防上及び治療上の活性を有するその能力について調べられることができる。

PCTアッセイ用の標準物質の調製のために、上述した、遺伝子工学によるプロカルシトニン3−116の調製方法が、また、完全なプロカルシトニン1−116及びプロカルシトニン2−116の調製にも本質的に同一の形態で用いられた。

C. 通常のヒト血清及び重度の敗血症患者からの血清中におけるジペプチジル−アミノペプチダーゼIV(DAP IV)活性の測定
健常者から、及び、敗血症患者からの、それぞれ20の血清サンプルを、それらのジペプチジル−アミノペプチダーゼIV特異的酵素活性について調べた。このDAP IV酵素活性は、Lys−Pro−4−メトキシ−ベータ−ナフチルアミドを用いるそれ自身は既知の方法で、フルオロメトリーによって測定した。このために、各ケース、2μlの血清が3mlの基質(50μg/ml Lys−Pro−4−メトキシ−ベータ−ナフチルアミド、50mM Tris/HCl、pH7.5)を用いてテストされ、340nmの波長を有する光による励起で生じた蛍光は、410nmの放射波長で持続的に測定された。その蛍光シグナルは、4−メトキシナフチルアミン溶液を用いて校正された。この方法で決定された酵素活性は、nmol/minで記載される。

敗血症血清中のDAP IV酵素活性が実質的に、健常者血清(血液提供者の血清)中の活性よりも低いことは明確である。つまり、血漿又は血清中のDAP IV酵素活性の決定は、患者血清中の敗血症を検出するために用いることもできる。

敗血症のケースでの、実質的に減少したDAP IVの血漿濃度は、一方では、該DAP IVが敗血症のプロセスに関与している証拠として考えることができる。他方では、それらの結果は、プロカルシトニン3−116の形成に責任があるのが血漿中のDAP IV濃度ではないことを示している。どちらかといえば、得られた結果は、プロカルシトニン3−116が、おそらく細胞内で、組織又は細胞結合DAP IVによって形成され、プロカルシトニン生成細胞又はプロカルシトニン貯蔵細胞から遊離されるという結論を示唆している。

DAP IVが活性化T細胞から発現するという効果についての文献(cf. Hegen及びOravecz, Protein Reviews on the WEB; Fleischer, loc. cit.)に含まれる情報は、DAP IVの発現と免疫系の活性状態との間の密接な関係を示しており、これは、敗血性全身性感染のケースでは過剰なストレス下にあり、それゆえに、とりわけプロカルシトニン3−116の形成が非常に増加するという、それら自身を表す典型的な反応を示す。

上述の発見から生じる可能性とは別に、敗血症診断の過程でのDAP IVの測定にとって、上述の結果はまた、敗血症においてカスケード様の様式で生じるプロセスがDAP IVインヒビターによって治療上の影響を受け、その結果、敗血症下でのプロカルシトニン3−116及び他のホルモンあるいは変換されたプロホルモンの遊離を予防又は減少させることが可能であろうということを示し、このプレホルモンの遊離に対して、減少又は予防されるべきという病理学的結論を可能にしているのかもしれない。

D. 敗血症のケースにおけるプロカルシトニン以外のプロホルモン濃度の測定
敗血症のケースで放出されるのが完全なプロホルモンのプロカルシトニンではなく、Xaa-Proジペプチドの分短くなった変性プロホルモンであるという事実は、敗血症のケースで放出されるのはプロカルシトニン3−116だけではなく、プロカルシトニン3−116は単に、例えば、免疫調節能を有するものであり、敗血症のケースではおそらく変換形態で高度に遊離されるプロホルモン又は同様のペプチド全体の中の代表的な1つにすぎないという仮説を導く。

既知のプロホルモン及びこれらのプロホルモンに関する文献に述べられるアミノ酸配列のチェックによって、事実上最も良く知られたプロホルモンが、そのアミノ末端に、Xaa-Proと定義されることができ且つそれゆえにプロカルシトニンの場合に見られたのと同様に除去され得るジペプチドを有することが示された。特に、前記タイプのジペプチドは数多くのプロホルモン又は免疫調節体(immunomodulators)のアミノ末端に存在する。下記の、表の形態の文献データのリストは、いくつかの選択されたプロホルモン又は免疫調節体、それらのアミノ末端に見出されるジペプチド、及び、それらのアミノ酸総数の概要を示す。

表2に示されるプロホルモンは、敗血症のケースで濃度が上昇するかもしれないプロホルモンの例であるが、そのリストは網羅的なものとみなされるべきではない。免疫調節体の場合、ジペプチドの脱離はその活性に影響するであろう。

今日までの実験による発見は、敗血症等の全身性感染のケースでは、一般的に、インターロイキン等のプロホルモン及びペプチド免疫調節体が、おそらく、アミノ末端でのジペプチドの脱離による変性を伴って遊離されること、並びに、これらがおそらく、関連する特定のレセプター又はその他のバインダーとの相互作用によって、免疫応答のカスケード内の更に次のステップを開始させること示す。

通常の血清と敗血症患者からの血清におけるプロカルシトニン決定と平行して、他のプロホルモンの決定も行った。これらは公平に、ランダムに選択された。これらは、(i)プロ−ガストリン放出ペプチド(proGRP)、(ii)プロ−心房性ナトリウム排泄増加性ペプチド(pro-ANP又はpro-ANF)、(iii)プロ−アドレノメデュリン(pro-adrenomedullin, pro-ADM)、(iv)プロ−エンドセリン(pro-END)及び(v)プロ−脳ナトリウム排泄増加性ペプチド(pro-BNP)であった。

D.1. 敗血症患者からの血清及び健常者からの血清中のproGRPの決定
アッセイは、proGRP決定用のものが市販されている。近年の出版物に、proGRPが気管支の小細胞癌の腫瘍マーカーとして記載されている(Petra Stieber et al., J Lab Med 1997, 21 (6): 336-344)。proGRPの決定用アッセイは、トーネンコーポレーションのProGRP ELISA KITTMの名称のものが市販されている。

このキットを用い、この市販のキットの使用のための情報にて指定された手順に従って、図4に示す測定結果が得られた。

プロカルシトニン及びproGRPについて得られた値の比較は、正常な血清と敗血症の血清との間の差違が、プロカルシトニンの場合よりも明確であること、しかし該proGRP濃度がプロカルシトニンの濃度と同様に上昇することを示している。

D.2. 敗血症患者からの血清及び健常者からの血清中のpro-ANF、pro-ADM、pro-END及びpro-BNPの決定
プレホルモンであるpro-ANF、pro-ADM、pro-END及びpro-BNPの決定については、アッセイは、DRG(DRG Instruments GmbH, D-35018 Marburg, ドイツ)からキットの形態で市販されており、製造者の指示に従って次の測定に用いられた。

pro-ANFの決定には、Prepro-ANF 26-55(ヒト)RIAキット;pro-ADMの決定には、Pro-Adrenomedullin 45-92 (ヒト)RIAキット;pro-ENDの決定には、Prepro-Endothelin 18-50(ヒト)RIAキット;及び、pro-BNPの決定には、Prepro-BNP 22-46(ヒト)RIAキットであり、これらは上述のDRG社のものである。

20人の正常者A-Tの群の血清及び敗血症患者20人の血清において、上述のプロホルモンと、それらと同時に、プロカルシトニンとを決定した。その結果は以下の表3に要約されている。表3のデータは、図5から9にグラフでも示している。

示された結果は、正常者に比べると、敗血症患者のケースでは、測定されたプロホルモン全ての値でおおよそ明確な増加を示すが、正常者と敗血症患者の間の差違はプロカルシトニンの決定において最も際立っている。

補足的な文献検索によって、さらに、敗血症性ショックにおけるプロ−オピオメラノコルチン(POMC)関連ペプチドの確認に関する出版物J. Endocr. (1988) 119, 159-165頁が明らかになった。該出版物は、敗血症性ショックにおける内因性オピオイド活性の増加に関する疑問、及び、ステロイドの投与による選択性の影響を検討している。感染プロセスの直接の影響又は抗生物質による測定値の影響は論じられていない。前記出版物における問題に基づいては、他のプロホルモンペプチドを含む報告結果の一般化された議論について、論理的な実現可能性はない。ここに開示される本発明の教示に照らすと、前記出版物は、敗血症のケースにおける一般的なプロホルモンの増加をさらに示したものとして回顧的に解釈することができる。

さらなるプロホルモンをテストすることは、ここに開示された結果を見ると決まり切った測定であると見なされるべきであり、もしそのようなテストがポジティブな結果を導き、それによってさらなるプロホルモン決定が敗血症の診断に用いられるのであれば、本出願の教示の使用によるものであるであろう。

Claims (2)
- 敗血症及び敗血症様全身性感染の、早期検出のため及び検出のため、重症度の評価のため、及びその治療処置の達成度の評価のための方法であって、プロ−ガストリン放出ペプチド(proGRP)、プロ−エンドセリン−1(pro-END)、プロ−脳ナトリウム排泄増加性ペプチド(pro-BNP)、プロ−心房性ナトリウム排泄増加性ペプチド(pro-ANP又はpro-ANF)、プロ−レプチン、プロ−ニューロペプチド−Y、プロ−ソマトスタチン、プロ−ニューロペプチド−YY及びプロ−アドレノメデュリン(pro-ADM)からなる群から選択される少なくとも1つのペプチドプロホルモンの含量、及び/又は、それから導かれる部分ペプチドであって、前記ペプチドプロホルモンから得ることのできる成熟ホルモンではない部分ペプチドの含量が、患者の生理的流体サンプル中で測定され、敗血症又は敗血症様全身性感染の存在、その重症度、及び/又は、治療処置の達成度が、測定された前記ペプチドプロホルモンの存在及び/又は検出量から決定することを特徴とする方法。
- 前記ペプチドプロホルモンの前記測定がイムノアッセイ又は免疫沈降アッセイとして行われ、且つ、敗血症又は敗血症様全身性感染の存在の検出が、測定された該ペプチドプロホルモン濃度が、健常者に見られる同じプロホルモンの値よりも有意に高い場合に行われることを特徴とする、請求項1に記載の方法。
