Description
Procon 2000(Procon 2000はタンパク質含有量が75重量%)(Central Soya Company、USA)12重量%溶液を調製した。5%アルカラーゼを加え、この大豆タンパクを60℃で6時間加水分解した。加水分解後、pHを6.8に調節し、HTST低温殺菌器(約3分、110℃)にこの溶液を通すことによって、酵素を不活性化した。続いて、この溶液をハイフロ珪藻土ろ過機(Hyflo diatomaecious earth filter)に通した。
大豆タンパクを、実施例1で述べたように加水分解した。加水分解物溶液を、カチオン交換樹脂Lewatit S2328に充填した(流速=5カラム体積/時間)。続いて、ペプチドを1MのNaClで溶離した。
エンドウタンパク質(Propulse 975、ParrHeim Food、カナダ)15重量%溶液を調製した。5%アルカラーゼを加え、このエンドウタンパクを60℃で3時間加水分解した。次に、pHを7.0に調節し、HTST(約3分、110℃)にこの溶液を通すことによって、酵素を不活性化した。この溶液を、HFK131限外ろ過ユニット(Koch Membrane Systems、USA)に通した。この透過液を、以下のプロセスステップに適用した。
エンドウタンパク質を、実施例2で述べたように加水分解した。この加水分解物溶液を、カチオン交換樹脂Amberlite IRC50に充填した(流速=5カラム体積/時間)。続いて、ペプチドを1MのNaClで溶離した。
Claims (16)
- 少なくとも1種のタンパク源からアルギニン及びリジンの含有量がタンパク質含有量の少なくとも20重量%であるペプチド混合物を調製する方法であって、
a)少なくとも1種のタンパク源のタンパク質をペプチドに切断するステップと、
b)前記ペプチドをカチオン交換樹脂に結合させるステップと、
c)前記カチオン交換樹脂を0.05〜2.0規定のアルカリ洗浄液で洗浄するステップと、
d)結合したペプチドをイオン強度が最大で200mM、pHが5.0〜9.0である水性溶離液で溶離するステップと
を含む方法。 - ステップb)とステップc)の間に前洗浄ステップを含み、前記カチオン交換樹脂が最大で200mM、pHが5.0〜9.0である前洗浄液で洗浄される、請求項1に記載の方法。
- 前記ペプチド混合物が、タンパク質含有量の少なくとも30重量%であるアルギニン及びリジンの含有量を有する、請求項1又は2に記載の方法。
- ステップa)が、酵素加水分解を含む、前記請求項1〜3のいずれかに記載の方法。
- ステップb)の前にろ過ステップがくる、前記請求項1〜4のいずれかに記載の方法。
- 前記ろ過ステップが、カットオフが10kDaである膜で行われる、請求項5に記載の方法。
- 前記少なくとも1種のタンパク源が、少なくとも2種の異なるタンパク質を含む、前記請求項1〜6のいずれかに記載の方法。
- 前記少なくとも1種のタンパク源が、野菜由来である、前記請求項1〜7のいずれかに記載の方法。
- 前記少なくとも1種のタンパク源が、大豆、エンドウ及び綿実からなる群から選択される、前記請求項1〜8のいずれかに記載の方法。
- 前記少なくとも1種のタンパク源が、少なくとも10重量%のアルギニン及びリジンを含む、前記請求項1〜9のいずれかに記載の方法。
- アルギニン及びリジンを少なくとも20重量%含む、アルギニン及びリジンに富んだ分子量2,000Da未満のペプチドの混合物を含み、請求項1〜10のいずれかに記載の方法によって得られる製剤。
- アルギニンを少なくとも10重量%、リジンを少なくとも10重量%含む、請求項11に記載の製剤。
- アルギニン及びリジンを少なくとも30重量%含む、請求項11または12に記載の製剤。
- アルギニンを少なくとも15重量%、リジンを少なくとも15重量%含む、請求項11〜13のいずれかに記載の製剤。
- 医薬品、サプリメント、飲物又は食物中の有効化合物としての請求項11から14までのいずれかに記載の製剤の使用。
- 成長ホルモンレベルを刺激するため、免疫応答を刺激するため、又は血糖値の調節を改善するための請求項15に記載の使用。
