本発明の合成HIVコード配列(発現カセット)は、発現のレベルを評価するため、およびGag含有構築物の場合には、VLPの産生を評価するために、多数の異なる発現ベクターにクローニングされ得る。HIVポリペプチドに対する合成DNAフラグメントは、真核生物発現ベクター(一過性発現ベクター、CMVプロモーターに基づく哺乳動物ベクター、およびバキュロウイルス発現系において使用するためのシャトルベクターが挙げられる)にクローニングされ得る。対応する野生型配列もまた、同じベクターにクローニングされ得る。

(合成コード配列についての発現アッセイ)
野生型配列は、合成HIV由来の配列がクローニングされたベクターと同じ特徴を有する発現ベクターにクローニングされる。
(発現のウエスタンブロット分析)
本明細書中に記載されるHIV発現カセットをトランスフェクトした細胞のウエスタンブロット分析を、共有に係るWO 00/39302に記載されるように本質的に実施する。簡単に言えば、ヒト293細胞を、実施例2に記載されるように、ネイティブまたは合成のHIV発現カセットを含む、pCMV6aのベースのベクターでトランスフェクトする。細胞を、トランスフェクション後、60時間培養する。上清を記載したように調製する。細胞溶解物を、以下のように調製する。細胞を、リン酸緩衝化生理食塩水で1回洗浄し、界面活性剤[0.1M Tris−HCl(pH7.5)中の1% NP40(Sigma Chemical Co.St.Louis,MO)]で溶解し、そして溶解物を新しいチューブに移す。SDS−ポリアクリルアミドゲル(前もって調製した(pre−cast)8〜16%;Novex、San Diego、CA)に、20μlの上清または12.5μlの細胞溶解液を充填する。タンパク質スタンダードもまた、充填する(5μl、広いサイズ範囲のスタンダード;BioRad Laboratories,Hercules,CA)。電気泳動を行い、BioRad Transfer Chamber(BioRad Laboratories,Hercules,CA)を用いて、製造業者(Millipore)によって推奨される転写緩衝液を用いて、100ボルトで90分間、転写を実施し、タンパク質をImmobilon P膜(Millipore Corp.,Bedford,MA)に転写させる。この膜を、HIV−1−ポジティブヒト患者血清にさらし、そしてo−フェニレンジアミンジヒドロクロリド(OPD;Sigma)を用いて免疫染色する。
合成HIV発現カセットのインビボ免疫原性
A.免疫
合成HIV発現カセットの免疫原性を評価するために、マウス研究を実施し得る。プラスミドDNA(例えば、本発明の合成配列を含む発現カセットを有するpCMVKM2)を、100μlの総注射容量において以下の最終濃度まで希釈する:20μg、2μg、0.2μgおよび0.02μg。希釈DNAの起こり得るネガティブな希釈効果を克服するために、各サンプル中の総DNA濃度を、ベクター(pCMVKM2)単独を使用して、20μgまで上昇させる。コントロールとして、ネイティブな対応するポリペプチドをコードする発現カセットを含むプラスミドDNAを、同様に扱う。4匹のBalb/cマウス(Charles River,Boston,MA)の12群に、種々の投薬量を使用して筋肉内免疫する(50μl/肢、前脛骨筋中に筋肉内注射)。
体液性免疫応答を、マウス血清の適切な抗HIV抗体ELISA(酵素結合免疫吸着アッセイ)を用いて、免疫後0および4週目(群5〜12)、そしてさらにそれぞれ免疫後6および8週目、2回目の免疫後2および4週目(群1〜4)に検査する。
特異的細胞傷害性Tリンパ球(CTL)の頻度を、ペプチドをパルスしたBalb/cマウスCD4細胞の標準的なクロム放出アッセイにより評価する。HIVタンパク質発現ワクシニアウイルスに感染したCD−8細胞を、ポジティブコントロールとして使用する(vv−タンパク質)。簡潔には、脾臓細胞(エフェクター細胞、E)を、(上記のように免疫した)BALB/cマウスから得る。細胞を培養し、再刺激し、そして例えば(Doe,B.およびWalker,C.M.,AIDS 10(7):793−794,1996に)記載されたGagペプチドでパルスした標的細胞に対するCTL活性についてアッセイする。細胞傷害活性を、標準的な51Cr放出アッセイにおいて測定する。標的(T)細胞をエフェクター(E)細胞とともに種々のE:T比で4時間培養し、そして二連のウェルからの平均cpmを使用して特異的51Cr放出(%)を算出する。
(合成HIV発現カセットのインビボ免疫原性)
(A.一般的免疫化方法)
合成HIV発現カセットの免疫原性を評価するために、モルモット、ウサギ、マウス、アカゲザル(rhesus macaque)およびヒヒを用いる研究が実施される。これらの研究は、代表的には以下のように構成される:DNA免疫化単独(単回または複数回);タンパク質免疫化(追加免疫)をともなうDNA免疫化;シンドビス粒子免疫化をともなうDNA免疫化;シンドビス粒子単独による免疫化。
実験は、モルモットにおいて以下のように実施され得る。各々が6匹のモルモットを含む群は、0、4、および12週に、本明細書に記載される1つ以上の配列を含む発現カセットをコードするプラスミドDNAで、筋肉内または粘膜により免疫化する。次いで、動物は、約18週に、プラスミドブーストの配列(単数または複数)によりコードされるHIVタンパク質および/またはその他のHIVタンパク質の単回投与量(筋肉内、皮内または粘膜による)で追加免疫される。抗体力価(幾何平均力価)を、第3のDNA免疫化の2週間後および上記タンパク質追加免疫2週間後に測定する。これらの結果を用いて、免疫応答を生成する合成構築物の有用性、およびDNA免疫化後の免疫応答を増大するタンパク質追加免疫を提供する利点を証明する。
実験は、ウサギにおいて以下のように実施され得る。ウサギを、本明細書に記載のHIVタンパク質をコードするプラスミドDNAで、筋肉内、粘膜により、または皮内で免疫化する(Biojectニードルレスシリンジを用いる)。この核酸免疫化は、初期免疫化の後、タンパク質追加免疫をともなう。代表的には、本発明の合成HIVポリペプチドをコードするポリヌクレオチドを含む構築物は、高度に免疫原性であり、そしてウサギにおいてわずか2回の免疫化後に実質的な抗原結合抗体応答を生成する。
任意の免疫化動物モデルにおいて、体液性免疫応答は、免疫化後の種々の時点で、抗HIV抗体ELISA(酵素結合イムノソルベント検定法)で、免疫化動物からの血清標本中でチェックされる。血清の抗体力価は、上記のように抗HIV抗体ELISAにより決定される。要約すれば、免疫化動物からの血清は、動物を免疫化するために用いられた、上記DNAによりコードされたHIVポリペプチド/タンパク質(単数または複数)、および/またはポリペプチドに対して惹起された抗体についてスクリーニングされる。ELISAマイクロタイタープレートのウェルは、選択されたHIVポリペプチド/タンパク質で一晩コートされ、そして4回洗浄される;次いで、ブロッキングが、PBS−0.2%Tween(Sigma)で2時間行われる。ブロッキング溶液を除去した後、100μlの希釈されたマウス血清を添加する。その後、血清を、1/25希釈で、かつ系列3倍希釈によって試験する。マイクロタイタープレートを、4回洗浄し、そして二次の、ペルオキシダーゼ連結抗マウスIgG抗体(Pierce、Rockford、IL)とインキューベートする。ELISAプレートを洗浄し、そしてウェルあたり100μlの3、3’、5、5’−テトラメチルベンジジン(TMB;Pierce)を添加した。各ウェルの吸光度を15分後に測定する。力価は、代表的には、最大の半分の吸光度(O.D.)を与えた血清の希釈の逆数として報告される。
(本発明の合成HIVポリヌクレオチドを含む発現カセットを用いる、ヒヒおよびアカゲザルのDNA免疫化)
(A.ヒヒ)
4匹のヒヒを、3回(0、4および8週)、本明細書に記載の遺伝子送達ヒビクルを用い、両側で大腿四頭筋肉中に筋肉内に、または粘膜により免疫化する。各免疫化の後2週間で動物から採血し、そしてHIV抗体ELISAを、単離された血漿を用いて実施する。このELISAは、二次抗体複合体が、1:500の希釈で用いた、抗ヒトIgG、g鎖特異的ペルオキシダーゼ複合体(Sigma Chemical Co.,St.Louis、MD63178)であることを除いて、本質的に上記のように実施する。50μg/mlの酵母抽出物を、血漿サンプルと抗体複合体の希釈物に添加しても良く、ヒヒ中にある既存の酵母抗体に起因する非特異的バックグラウンドを低減する。リンパ球増殖応答は、第4の免疫化後2週(14週目)にヒヒで観察され、そしてHIVポリペプチドでの追加免疫(44週および76週)後に実質的に高められる。このような増殖結果は、Tヘルパー細胞機能の誘導の指標である。
本発明のポリヌクレオチドをコードする、合成のコドン改変HIVポリペプチドの改良された能力は、発現プラスミド中に構築されるとき、アカゲザルで確認され得る。代表的には、アカゲザルは、改変HIVポリヌクレオチドの2または3の1mg投与量の後、検出可能なHIV特異的CTLを有する。要するに、これらの結果は、合成HIV DNAは、非ヒト霊長類で免疫原性であることを証明する。中和抗体もまた検出され得る。

(HIV−1ワクチンのアクセサリー遺伝子成分:変異したTat、RevおよびNef型C抗原の機能分析)

上記のように、本明細書に記載の任意のポリヌクレオチドまたは発現カセットの免疫原性は、容易に評価される。以下の表(表D)は、ウサギにおけるエレクトロポレーションを用いる、個々のサブタイプのワクチンおよび混合されたサブタイプのワクチンの両方の、サブタイプBおよびCエンベローププラスミドの免疫原性の比較を含む例示の手順である。このような方法は、任意の他のHIVポリペプチドに等しく適用可能であることが明らかである。
(GagおよびPol構築物を用いるマウス免疫化研究)
細胞性免疫応答および体液性免疫応答を、以下の構築物について(本質的に実施例4に記載のように)評価した:Gag、GagProtease(+FS)(GP1、最適化されたプロテアーゼコドンおよびINSの不活性化;GP2、プロテアーゼINSの不活性化のみ)、フレームシフトをもつGagPolΔインテグラーゼ(gagFSpol)、およびGagPolΔインテグラーゼインフレーム(GagPol)(図91を参照のこと)。GagPolΔインテグラーゼインフレームの複数のバージョンもまた、弱力化(GagPolAtt)または非機能的プロテアーゼ(GagPolIna)とともに設計した。
(C型誘導エンベロープポリペプチドを使用しての中和抗体のタンパク質発現、免疫原性および産生)
サブタイプCの主要な単離物TV1、(これは南アフリカの個体から回収された)由来のエンベロープ(Env)ワクチンを、以下のようにウサギで試験した。遺伝子カセットを設計し、gp120(表面抗原)、gp140(表面抗原および膜貫通タンパク質gp41の外側ドメイン)、および可変ループ領域(V2およびV1V2)の欠失を有するか、有さないHIV−1エンベロープポリタンパク質の全長(gp120およびgp41)のgp160形態を発現する。全ての遺伝子を、Rev非依存様式で、コード化糖タンパク質の発現を増強するように配列改変し、次いで、これらを、上記のようにDNAワクチンおよびタンパク質生産適用のためのpCMVベースのプラスミドへとクローン化した。これらの配列を本明細書中に記載されるようにコドン最適化した。簡潔に述べると、全ての改変されたエンベロープ遺伝子を、カイロンpCMV連結ベクター、好ましくはEcoRI/XhoI部位へとクローン化した。
TV1 Env抗原の全長(gp160)、短縮型gp140(Env外側ドメインのみ)およびgp120のネイティブバージョンを、本明細書中に記載の発現カセットから生産した。gp140コード配列を、293T細胞へと一過性にトランスフェクトした。遺伝子産物の発現レベルを、組織内抗原捕獲ELISAによって評価した。TV001c8.2、TV001c8.5およびTV002c12.1のネイティブ配列から構築されたエンベロープ遺伝子は、インビトロで正しいタンパク質を発現し、gpl40TV001c8.2は、最も高い発現レベルを示した。さらに、TV1由来クローン8.2から発現したEnvタンパク質は、CD4レセプタータンパク質を結合することを見出した(これは、発現タンパク質のこの特性が、機能的コンホメーションにおいて維持されることを示す)。発現したTV1 gp160タンパク質結果のレセプター結合特性/機能をまた、細胞融合アッセイによって確認した。
一過性発現実験は、TV100ウイルス単離物およびTV002ウイルス単離物由来のエンベロープ遺伝子が、所望のタンパク質産物を発現することを示した。18人の感染した南アフリカ個体(サブタイプBおよびC)由来の血清を使用して、これらの2つのウイルス株の相対的中和感受性は、以下の通りであった。TV2株は、1:10の血清希釈で、18/18の血清によって中和され、1:50希釈では16/18;1:250希釈では15/18で中和された。比較すると、TV1単離物は、1:10希釈で15/18:1:50希釈で6/18のみ中和され;そして1:250希釈で検体のひとつも中和されなかった。さらに、TV001患者の血清は、試験された全ての希釈物でTV002単離物に対して中和活性を示した。対照的に、TV002は、1:10の血清希釈でのみTV001の中和を示した。これらの結果は、TV001単離物が、TV002よりもその感染した宿主において、より広範でかつ強力は中和抗体応答を誘導し得ることを示唆する。
TV1 Env DNA(合成発現カセットを含む)およびタンパク質ワクチンを、以下の表Hに示されるように投与した。
(Rhesus Macaquesにおける免疫学的応答)
表Iに示すように構成された免疫研究において、細胞性免疫応答および体液性免疫応答を、3群のrhesus macaques(各群は、4匹の動物で構成された)において評価した。免疫組成物についての投与経路は、いずれの場合においてもエレクトロポレーションであった。抗体価を、2回目の免疫の2週間後について表Iに示す。

Claims (53)
- 2つ以上の免疫原性HIV−1ポリペプチドをコードする合成ポリヌクレオチドであって、該ポリペプチドの少なくとも2つは、異なるHIV−1サブタイプ由来であり、ここで、該合成ポリヌクレオチドは、配列番号9〜配列番号93のうちの一つに記載のポリヌクレオチド配列を含む、合成ポリヌクレオチド。
- 前記HIV−1サブタイプがサブタイプBおよびサブタイプCである、請求項1に記載の合成ポリヌクレオチド。
- 前記HIVポリペプチドが、Gag、Env、Pol、Tat、Rev、Nef、Vpr、Vpu、Vifおよびこれらの組み合わせからなる群から選択される、請求項1に記載の合成ポリヌクレオチド。
- 前記ポリヌクレオチドがTat、RevおよびNefをコードする、請求項1に記載の合成ポリヌクレオチド。
- 前記ポリヌクレオチドがVif、VprおよびVpuをコードする、請求項1に記載の合成ポリヌクレオチド。
- 前記HIV−1ポリペプチドの1つ以上が1つ以上の突然変異を含む、請求項1に記載の合成ポリヌクレオチド。
- 前記HIV−1ポリペプチドがvif、vprまたはvpuを含む、請求項6に記載の合成ポリヌクレオチド。
- さらなる抗原性ポリペプチドをコードする配列をさらに含む、請求項1に記載の合成ポリヌクレオチド。
- 請求項1〜8のいずれかの合成ポリヌクレオチドを含む、発現カセット。
- 選択された宿主細胞における使用のための組換え発現系であって、該組換え発現系は、請求項9に記載の発現カセットを含み、ここで前記ポリヌクレオチド配列は、該選択された宿主細胞における発現に適合する制御エレメントに作動可能に連結される、組換え発現系。
- 請求項10に記載の組換え発現系であって、前記制御エレメントが、転写プロモーター、転写エンハンサーエレメント、転写終結シグナル、ポリアデニル化配列、翻訳開始の最適化のための配列、および翻訳終結配列からなる群から選択される、組換え発現系。
- 前記転写プロモーターが、CMV、CMV+イントロンA、SV40、RSV、HIV−Ltr、MMLV−ltr、およびメタロチオネインからなる群から選択される、請求項11に記載の組換え発現系。
- 前記ポリヌクレオチド配列が、前記選択された細胞における発現に適合する制御エレメントと作動可能に連結される、請求項9に記載の発現カセットを含む細胞。
- 前記細胞が哺乳動物細胞である、請求項13に記載の細胞。
