Description
試験装置(図1)は、サンプル導入ゾーン(1)、赤血球分離ゾーン(2)、検出ゾーン(3)、及び吸引ゾーン(4)が備え付けられた支持部材(5)から構成される。赤血球分離ゾーン(7)と部分的に重複するサンプル導入マトリックス(6)は、サンプル導入ゾーン(1)に配置されている。次に、赤血球分離マトリックス(7)は、線状に固定化物質が塗布されている(9)検出マトリックス(8)(検出ゾーン)と若干重複している。吸引マトリックス(10)は検出マトリックス(8)と若干重複している。検出しようとする分析対象物との複合体を形成するために必要なすべての試薬は、サンプル導入マトリックス(6)に供給する。この場合、サンプル導入ゾーンは、重なり合う2つのフリースからなり、第1フリース(金フリース)はNT−proBNP(MAB18.4.34(27−31))に対する金標識抗体を含浸させたものであり、第2フリース(ビオチンフリース)はNT−proBNP(PAB(39−50))に対するビオチニル化抗体を含有する。ストレプトアビジンで作製された線(9)は検出ゾーン(3)内に設ける。
「ビオチンフリース」:100mM Hepes pH7.4、0.1% Tween(登録商標)、20μg/mlビオチニル化PAB(39−50)
「金フリース」:100mM Hepes pH7.4、OD4 MAB18.4.34(27−31)金結合体
MAB17.3.1(13−16)/PAB(39−50)とMAB18.4.34(27−31)/PAB(39−50)のMAB/PAB組み合わせとElecsys(登録商標)試験キット(PAB(1−21)/PAB(30−50))との相関を、Elecsys(登録商標)2010(Roche Diagnostics)を用いて電気化学発光イムノアッセイにおいて調べた。このため、PAB(39−50)をビオチニル化捕捉試薬として使用し、MABのルテニウム付加F(ab’)2フラグメントを検出試薬として使用した。20μlのサンプル又は標準物質をそれぞれ、75μlの2つの抗体試薬と共に、37℃にて9分間インキュベートした。その後、35μlのストレプトアビジン被覆磁性ポリスチレン粒子を添加し、さらに室温にて9分間インキュベートした。インキュベーション溶液のアリコートの電気発光シグナルを慣用法に従ってElecsys(登録商標)2010で測定し、標準曲線を用いて濃度シグナルに変換した。
NT−proBNP試験ストリップを実施例1と同様にして準備した。試薬フリースのために以下の含浸製剤を使用した:
「ビオチンフリース」:100mM Hepes pH7.4、0.1%Tween(登録商標)、
20μg/mlビオチニル化PAB(39−50)
「金フリース」:100mM Hepes pH7.4、OD4 MAB18.4.34(27−31)又はMAB17.3.1(13−16)金結合体
心不全患者からのNT−proBNP含有血清を健常ドナー由来のヘパリン添加血液サンプルに添加し、等分した。150μlのこれらの「混合(spiked)」血液サンプルを試験ストリップにピペットで導入し、CARDIAC Reader(登録商標)(Roche Diagnostics)で標準的な方法に従って測定した。同じサンプルに由来する血漿を遠心分離し、Elecsys(登録商標)NT−proBNPキットを用いてElecsys(登録商標)1010分析システム(Roche Diagnostics)で測定した。60サンプルをこのようにして調製し、両方のシステムで測定した。図5は、両方のシステムで測定した値を示す。相関はr=0.95で非常に良好であった。

Claims (3)
- NT−proBNPを測定するための免疫クロマトグラフィー試験ストリップの形態をした免疫学的サンドイッチ試験装置であって、
NT−proBNPに対する少なくとも2つの抗体が使用されており、ここで、これらの抗体の1つは、NT−proBNPのアミノ酸13−16またはアミノ酸27−31を含むエピトープの部分に対するモノクローナル抗体(MAB)であり、そして
これらの抗体の1つは、NT−proBNPのアミノ酸39−50を含むエピトープの部分に対するポリクローナル抗体(PAB)である、
ことを特徴とする、上記免疫学的試験装置。 - 以下の抗体の組み合わせ:
DSM ACC2592として寄託された細胞系により産生されるMAB 18.4.34(27−31)とPAB(39−50)との組み合わせ;あるいは
DSM ACC2591として寄託された細胞系により産生されるMAB 17.3.1(13−16)とPAB(39−50)との組み合わせ;
を使用することを特徴とする、請求項1に記載の免疫学的試験装置。 - 請求項1または2に記載の免疫学的試験装置を用いることを特徴とする、NT−proBNPの検出方法。
