本出願は、米国仮特許出願、2001年2月23日出願の第60/270,817号と2002年1月11日出願の第60/347,443号の優先権を主張するものである。
カロテノイドは、植物の光合成に関わる重要な天然物である。カロテノイドは、動物および人間の栄養補給剤として、又、食品着色剤及び化粧品にも使用される。カロテノイドは、健康にとって重要ではあるが、動物や人間はそれらを作り出すことが出来ず、従って、これらの化合物は、食餌を通じて、果実と野菜から摂取されなければならない。
本発明は、従来技術の問題点を解決するものである。
本発明は、少なくとも1つの酸誘導部位と少なくとも1つのアルコール誘導部位とを有するエステルに関し、前記少なくとも1つのアルコール誘導部位が、モノヒドロキシカロテノイド、ジヒドロキシカロテノイド、及びポリヒドロキシカロテノイドから成るグループから選択されるヒドロキシカロテノイドを含み、前記酸誘導部位が、モノカルボン酸カロテノイド、ジカルボン酸カロテノイド、及びポリカルボン酸カロテノイドから成るグループから選択されるカルボン酸カロテノイドを含む。好適なヒドロキシカロテノイドには、ルテイン、ゼアキサンチン、クリプトキサンチン、ビオラキサンチン、カロテンジオール、ヒドロキシカロテン、ヒドロキシリコピン(hydroxylycopene)、アロキサンチン、デヒドロクリプトキサンチン(dehydrocryptoxanthin)が含まれる。好適なカルボン酸カロテノイドには、ビキシン、ノルビキシン、β−アポ−8−カロテン酸、クロセチン、ジアポカロテン酸、カルボキシルカロテン、アザフリンが含まれる。
本発明は、酸誘導部位とアルコール誘導部位との両方がカロテノイドである新規なクラスのエステルに関する。この開示の目的のために、ここで、「アルコール誘導部位」と「アルコール部位」という用語は、共に、エステル分子の、アルコールから誘導されるフラグメントを指すものと理解することができる。同様に、「酸誘導部位」と「酸部位」という用語は、共に、エステル分子の、カルボン酸から誘導されるフラグメントを指すものと理解することができる。
図1に示すルテインモノビキシンエステルを、以下の手順によって合成した。ビキシン(500mg,1.27mmol)を、乾燥ジクロロメタン(50mL)中で、塩化オキサリル(500μL,5.25mmol)とN,N−ジメチルホルムアミド(1滴)との添加によって、ビキシノイル塩化物に変換した。前記混合物を、室温で1時間攪拌し、その後、減圧下で乾燥状態にまで蒸発させてビキシノイル塩化物を生成した。
図2に示すルテインジビキシンエステルを、以下の手順によって合成した。ビキシン(500mg,1.39mmol)を、乾燥トルエン(10mL)中で、塩化オキサリル(1mL,10.5mmol)の添加によって、ビキシノイル塩化物に変換し、窒素雰囲気下で1時間還流し、血のように赤い溶液を得た。前記混合物を、僅かに冷却させ、その後、減圧下で乾燥状態にまで蒸発させてビキシノイル塩化物を生成した。
ルテインモノ−β−アポ−8’−カロテンエステルを、以下のように調製した。β−アポ−8’−カロテン酸の酸塩化物は、例2のビキシノイル塩化物と同様に調製された。前記酸塩化物(1.03g,2.5mmol)をベンゼン(10mL)中で溶解させた。これに、ルテイン(3.7g,6.55mmol)とピリジン(1.2mL,12.8mmol)とのベンゼン(10mL)溶液をアルゴン雰囲気下で添加した。反応を、室温で3日間攪拌した。
Claims (3)
- 少なくとも1つの酸誘導部位と少なくとも1つのアルコール誘導部位とを有するエステルであって、前記少なくとも1つのアルコール誘導部位が、モノヒドロキシカロテノイド、ジヒドロキシカロテノイド、及びポリヒドロキシカロテノイドから成るグループから選択されるヒドロキシカロテノイドを含み、前記少なくとも1つの酸誘導部位が、モノカルボン酸カロテノイド、ジカルボン酸カロテノイド、及びポリカルボン酸カロテノイドから成るグループから選択されるカルボン酸カロテノイドを含むエステル。
- 請求項1のエステルであって、前記ヒドロキシカロテノイドは、ルテイン、ゼアキサンチン、クリプトキサンチン、ビオラキサンチン、カロテンジオール、ヒドロキシカロテン、ヒドロキシリコピン、アロキサンチン、デヒドロクリプトキサンチンから成るグループから選択されるエステル。
- 請求項1のエステルであって、前記カルボン酸カロテノイドは、ビキシン、ノルビキシン、β−アポ−8’−カロテン酸、クロセチン、ジアポカロテン酸、カルボキシルカロテン、およびアザフリンから成るグループから選択されるエステル。
