Description
核酸を含有するサンプルから、核酸を遊離するための第一試薬を混合し、核酸の遊離を促す第一工程と、
遊離した核酸を含む液と第二試薬を混合した後、圧力変化により核酸捕捉チップ内に吸引し、固相と混合液を接触させ吐出する第二a工程と
固相を洗浄するために第二試薬を、圧力変化により核酸捕捉チップ内に吸引し、固相と二試薬を接触させた後、吐出する第二b工程と
固相、および、核酸捕捉チップ内に残存する、第一試薬、および/または、第二試薬を除去するために、第三試薬を圧力変化により核酸捕捉チップ内に吸引し、固相と第三試薬を接触させた後、吐出する第三a工程と
核酸捕捉チップ内から第三試薬の除去を促すために、核酸捕捉チップ内に通気を行う第三b工程と
残存する第三試薬を濯ぐために、第四試薬を圧力変化により核酸捕捉チップ内に吸引し、固相と第四試薬を接触させた後、吐出する第四工程と
固相から核酸を溶離するために、第五試薬を圧力変化により核酸捕捉チップ内に吸引し、固相と第五試薬を接触させた後、吐出する第五工程
を含む事を特徴とする。なお、固相が、粒状物或いは繊維状物からなる場合は、平均孔径は平均間隔のことを意味し、25μm以下であることを特徴とする。

10mmol/L ビシン(pH8.5)、0.1mmol/L EDTAを用いる。
図1は核酸捕捉用チップの構成を示す図である。核酸捕捉用チップ31は、上端が圧力変化を発生させる器具、あるいは、自動装置の接続ノズルの先端に気密に勘合される内径を有しており、下端に向けて徐々に内径が細くなるように形成される。核酸捕捉用チップ31は透明もしくは半透明な合成樹脂からなる。チップ31の先端側には、固相が流出することを防止するため、および、固相の平均間隔を規定するための円板状の阻止部材101aをチップ31の上端より圧入し、固相102を挟む形で、阻止部材101bを固相102が所定の平均間隔を維持できるように設ける。これらの阻止部材101a,101b は、液体及び気体が容易に通過し得る多数の孔を有するが、その孔は固相102の流出を阻止できる大きさである。
固相として石英ウール(東芝ケミカル製、グレードB、ウールの直径 6〜12μm)5mgを用いる。
なお、ここで第三試薬には、25mmol/L酢酸カリウム(和光純薬工業(株)製、試薬特級)を含む50%エタノール(和光純薬工業(株)製、試薬特級)を使用した。
なお、ここで第四試薬には、50mmol/L酢酸カリウム(和光純薬工業(株)製、試薬特級)を使用した。
Claims (16)
- シリカを含有した繊維状物を液体に接触可能な状態で内蔵した核酸回収器具であって、シリカ含有繊維状物が多数の通水部を有し、その通水部における繊維と繊維の平均間隔が13μm以上25μm以下であり、且つ、該核酸回収器具が圧力を変化させる器具に接続されていることを特徴とする、核酸回収器具。
- 前記圧力を変化させる器具がポンプである、請求項1記載の核酸回収器具。
- 前記繊維状物が石英ウールである、請求項1又は2記載の核酸回収器具。
- 前記石英ウール間の平均間隔が13μmから21μmである、請求項3記載の核酸回収器具。
- 前記石英ウールの直径が6μmから12μmである、請求項3記載の核酸回収器具。
- 前記繊維状物の距離を保持するために、流体流通可能な阻止部材で、繊維状物を挟み込んだ形状とする、請求項1記載の核酸回収器具。
- 前記阻止部材の材質が石英粒子を焼結した物である、請求項6記載の核酸回収器具。
- 核酸回収器具が核酸捕捉チップである、請求項1又は2記載の核酸回収器具。
- シリカを含有した繊維状物を液体に接触可能な状態で内蔵した核酸回収器具を備える自動核酸回収装置であって、シリカ含有繊維状物が多数の通水部を有し、その通水部における繊維と繊維の平均間隔が13μm以上25μm以下であり、且つ、該核酸回収器具は圧力を変化させる器具に接続されていることを特徴とする、自動核酸回収装置。
- 前記圧力を変化させる器具がポンプである、請求項9記載の自動核酸回収装置。
- 前記繊維状物が石英ウールである、請求項9又は10記載の自動核酸回収装置。
- 前記石英ウール間の平均間隔が13μmから21μmである、請求項11記載の自動核酸回収装置。
- 前記石英ウールの直径が6μmから12μmである、請求項11記載の自動核酸回収装置。
- 前記繊維状物の距離を保持するために、流体流通可能な阻止部材で、繊維状物を挟み込んだ形状とする、請求項9記載の自動核酸回収装置。
- 前記阻止部材の材質が石英粒子を焼結した物である、請求項14記載の自動核酸回収装置。
- 核酸回収器具が核酸捕捉チップである、請求項9又は10記載の自動核酸回収装置。
