本発明は自動分析装置、特に試料を試薬と反応させ、それによって生成された反応液を測定することによって試料中の分析項目の分析を行う自動分析装置に関する。

【従来の技術】
典型的な自動分析装置によれば、試料は試薬と反応し、それによって生成された反応液は光吸収測定され、その結果として試料中の目的とする分析項目の吸光度が測定される。この測定された吸光度は、その測定に先立って作成された検量線を用いて濃度に変換される。この場合、製造ロットが異なる試薬や標準液を用いて測定を行った場合でも、分析項目の吸光度から濃度への変換に当たっては、試薬製造ロットや標準液製造ロット等が変わる前の検量線が用いられるのが普通である。

【発明が解決しようとする課題】
しかし、用いられる試薬製造ロットや標準液製造ロット等が異なると、一般に検量線が異なることが多い。これは、試薬製造ロットや標準液製造ロット等が検量線を変える検量線パラメ−タを構成する重要な分析パラメ−タであることを意味する。このように、分析パラメ−タが異なると検量線が異なるにもかかわらず、濃度変換が分析パラメ−タ変更前の検量線を用いて行われるならば、分析結果である、変換された濃度値は誤差を含むことになる。

本発明の一つの目的は分析パラメ−タが異なっても、目的とする分析項目の、それによる誤差を含まない分析結果を簡単に得るのに適した自動分析装置を提供することにある。

【課題を解決するための手段】
本発明に基づく自動分析装置は、一つの観点によれば、試料中の予め定められた一つの分析項目について異なる検量線パラメ−タの下で作成された複数の検量線を予め記憶する記憶装置と、前記異なる検量線パラメ−タのうちの特定の検量線パラメ−タを分析パラメ−タとして、前記試料を試薬と反応させ、それによって生成された反応液を測定する測定装置と、その測定結果に基づいて、前記特定の検量線パラメ−タの下で作成された、前記記憶されている検量線を用いて前記予め定められた一つの分析項目の分析結果を生成する分析結果生成装置とを備えていることを特徴とする。

本発明に基づく自動分析装置は、別の観点によれば、複数の分析ユニットと、一つの分析項目について前記分析ユニット毎に作成された検量線を記憶する記憶装置とを備え、前記それぞれの分析ユニットにおいて試料を試薬と反応させて、それにより生成された反応液をそれぞれ測定し、その測定結果から、その測定結果が得られた対応する分析ユニットについて記憶されている検量線を用いて前記一つの分析項目の分析結果をそれぞれ生成するように構成したことを特徴とする。

【発明の実施の形態】
試料は試薬と反応し、それによって生成された反応液は光吸収測定され、その結果として試料中の分析項目の吸光度測定が行われる。測定された吸光度は分析に先立って作成された検量線を用いて濃度に変換される。なお、吸光度測定に代えて濁度測定が用いられてもよい。

検量線は分析されるべき分析項目を含む標準液とその分析項目用の試薬とを反応させ、それによって生成された反応液を測定することによって作成される。しかし、実際の試料と該試料中の分析項目に応じた試薬との反応特性は試薬の製造ロット、処方及びキットによって異なり、それによって検量線が変わることが多い。標準液についても、その製造ロット、処方、キット及び由来によって試薬との反応性が異なり、これによって検量線が変わる。すなわち、試薬の製造ロット、処方及びキット並びに標準液の製造ロット、処方、キット及び由来は検量線を変える検量線パラメ−タである。

検量線を変える要因となるもの、つまり検量線を変える検量線パラメ−タとしては他に、試料が血清であるか尿であるかによって変更する必要のある試料と試薬との混合比、測定感度に影響がある試料と試薬との反応液の反応時間、同様に感度に影響がある光の測定波長がある。測定装置として複数の分析ユニットが用いられる場合は、分注の正確さ、光度計の特性、反応液の温度制御特性等の違いにより分析ユニット毎に検量線が異なる。つまり、分析ユニットもまた検量線を変える検量線パラメ−タである。

そこで、それぞれの分析項目毎にいろいろな検量線パラメ−タの下で検量線が作成され、その作成されたそれぞれの検量線は記憶装置に予め記憶される。これらの検量線の検量線パラメ−タは実際の測定では分析パラメ−タとなるものである。

測定に当たっては、分析パラメ−タが設定され、その設定された分析パラメ−タの下で測定が開始され、それによって目的とする分析項目の吸光度が測定される。もちろん、その吸光度は濁度であってもよい。測定された分析項目の吸光度は検量線を用いて濃度に変換されるが、この検量線としては、設定された分析パラメ−タと同じ検量線パラメ−タの下で作成された、記憶装置から読み出された検量線が用いられる。

したがって、分析結果である変換された濃度は、分析パラメ−タが変わっても、それによる誤差を含まないことになる。すなわち、精度の高い分析結果が得られることになる。もちろん、検量線は記憶装置に予め記憶されている検量線の中から設定された必要な分析パラメ−タをキ−として選択して読み出すだけで簡単に得られる。すなわち、分析パラメ−タが変わっても、目的とする分析項目の、それによる誤差を含まない分析結果を簡単に得ることができる。

複数の分析ユニットを設け、一つの分析項目をそれぞれの分析ユニットを用いて分析するようにしてもよい。この場合は、一つの分析項目についてそれぞれの分析ユニット毎に作成された検量線が記憶装置に記憶される。それぞれの分析ユニットにおいて試料は試薬と反応し、それによって生成された反応液は光吸収測定され、一つの分析項目についての吸光度がそれぞれ測定される。その吸光度は記憶装置に記憶されている対応する検量線を読み出し、これを用いて濃度に変換される。

したがって、分析結果である変換された濃度は、分析ユニットが違っていても、それによる誤差を含まないことになる。すなわち、精度の高い分析結果が得られることになる。もちろん、その分析結果は記憶装置に記憶されている検量線を用いることによって簡単に得られる。

本発明の実施例を図1〜図4を参照して説明する。図1は血清,血漿及び尿の試料を分析することが可能な自動分析装置の概略構成図である。図1の自動分析装置は図2に示すようなディスペンサ方式で試薬を供給する分析ユニットと、図3に示すようなピペッタ方式で試薬を供給する分析ユニットとを混在して含む。図1の分析ユニット3A,3F及び3Gは固定された分析チャンネルを有し、複数の試薬吐出ノズルのそれぞれが試薬毎に専用化されているディスペンサ方式の分析ユニットである。分析ユニット3B,3C,3D及び3Eは分析チャンネルが固定されずにランダムアクセスされ、1本の試薬ピペッティングノズルで次々と分析項目に応じた試薬を分注するピペッタ方式の分析ユニットである。

図1において、分析ユニット3A,3B及び3Cは、種類が血清である試料を分析処理するように分析条件が設定され、分析ユニット3D及び3Eは血漿試料を分析処理するように、また、分析ユニット3F及び3Gは尿試料を分析処理するようにそれぞれ分析条件が設定される。分析ユニット3A〜3Gは主搬送ライン20から取り込まれた試料ラック1をサンプリング位置に位置づけた後に主搬送ライン20に戻す機能を有する搬送路であるサンプリングライン4A〜4Gと、各サンプリングラインに対応して設けられており、試料ラック1の識別情報又はその試料ラック上の各試料容器の識別情報を読取るための識別情報読取装置51〜57と、試料と試薬との分析項目に応じた反応を反応容器内で進め、反応した液を光学的に測定する反応部5A〜5Gと、試薬供給部とをそれぞれ備えている。それぞれの分析ユニットの試薬供給部の内、26,27,28,29がピペッタ方式のものであり、32,33,34がディスペンサ方式のものである。

ラック送出部17は多数の試料ラック1をセットできるエリアを有し、主搬送ライン20の方へ試料ラック1を1個ずつ送り出す送出機構を有する。ラック回収部18はそれぞれの分析ユニットで分析処理された試料を収容している試料ラック1を回収するエリアを有し、試料ラックを整然と並べて配列する整列機構を有する。一時格納部22は分析ユニットによって試料採取された試料ラック1を測定結果が出力されるまで一時的に格納し、再検査が必要な場合は再検査ラック搬送ライン25を介して検体ラックを再び主搬送ライン20により搬送するように送り出し、再検査が不要な場合は試料ラックをラック回収部18へ送り出す。

制御装置は、全体制御用コンピュータ40と、それぞれの分析ユニットに対応して設けられて、それぞれの分析ユニット内の必要な処理及び制御を行う分析ユニット側コンピュータ6A〜6Gと、フロッピーディスクメモリ41を有する。それぞれの分析ユニットの光度計(濁度計であってもよい)からの出力信号の処理は分析ユニット側コンピュータ6A〜6Gによって分担され、それらに接続された全体制御用コンピュータ40は各分析部の動作,ラック搬送系の動作及びシステム内の必要部分の動作制御をすると共に、各種情報処理に必要な演算及び制御を実行する。コンピュータ間の役割分担はこのようなものに限られず、構成上の必要性に応じて種々の態様に変更でき、あるいは全体制御用コンピュータ40のみを用いて分析ユニット側コンピュータを不要にすることも可能である。全体制御用コンピュータ40には、記憶部45が具備され、データ入力用の操作部42,情報を画面表示するためのCRT43及び測定結果を出力できるプリンタ44が接続されている。

試料ラック1は、例えば図2の例に示すように、試料を収容した試料容器2が複数本、例えば5本ずつ装填される箱状の容器保持体からなるが、この形状以外に種々のものを使用してもよい。試料ラック1の外壁にはラック識別情報を示す識別情報媒体が設けられ、試料容器2の外壁には試料識別情報を示す識別情報媒体が設けられる。これらの識別情報媒体としては、バーコードラベルや磁気記録媒体等が用いられる。試料ラック1に設けられたバーコードは、ラック番号及び試料種別の情報を有する。試料容器2に設けられたバーコードは、それぞれの試料に関する情報、例えば、受付番号,受付年月日,患者氏名,患者番号,試料種別,試料依頼分析項目などの情報を有する。

図1における識別情報読取装置50は主搬送ライン20によって搬送される前の試料ラック1又は試料容器2の識別情報(バーコード)を読取った結果をコンピュータ40に入力する。また、一時格納部22に設けられた識別情報読取装置58は試料ラック1が一時格納部22に入るとき及び出るときに試料ラック又は試料容器のバーコードを読取り全体用コンピュータ40に伝達する。

それぞれの分析ユニット3A〜3Gの試薬供給部に収納される各種分析項目用の試薬ボトル12,12A,12Bには、その外壁に試薬識別情報がバーコードなどで表示されている。試薬識別情報としては、試薬(製造)ロット番号,試薬ボトルのサイズ,使用可能な試薬液量,有効期限,ボトル毎に異なっているシーケンス番号,分析項目コードなどが含まれる。このような試薬識別情報はバーコード読取装置によって読取られ、それぞれの分析ユニット3A〜3Gに対応づけられ、試薬供給部における試薬ボトルのセット位置,使用可能な液量と1回の分注量から計算される試薬の分析可能回数,分析項目の種類,その試薬が収納された分析装置番号などが記憶部45に登録される。

主搬送ライン20は試料ラック1を載せる搬送ベルトとベルト駆動用モータを具備し、検体ラックを所望位置まで連続移送するよう制御部により制御される。各サンプリングライン4A〜4Gはラック引込位置,分注位置及びラック送出位置にラックを停止するように搬送ベルトを間欠的に移動することができる。主搬送ライン20によって搬送される試料ラック1は分析ユニットの列に沿って移動され、制御装置によって指定された分析ユニットの前で停止され、直ちにラック移載機構(後述)によりその指定された分析ユニットのサンプリングラインのラック引込位置に移される。分注位置にて試料分注操作の終了した試料ラック1はサンプリングラインのラック送出位置から主搬送ライン20上へラック移載機構によって引き渡される。

ディスペンサ方式の分析装置の構成例を、図2を参照して説明する。分析装置3Aの反応部5Aは透光性の反応容器46aを有する反応容器列を同心円状に2列備え、各反応容器列毎に光源14aから反応容器46Aを透過した光を分光して複数波長を受光する多波長光度計15a(濁度計であってもよい)を備える。各反応容器列に作用するように、反応部5Aの近傍には、検体用ピペッタポンプ47aに接続されたピペットノズルを有する試料分注器48aと、試薬用ディスペンサポンプ60に接続された第1試薬ノズル群保持部64及び第2試薬ノズル群保持部66と、第1撹拌機構65及び第2撹拌機構67と、反応容器洗浄機構19aとが配置されている。試薬保冷庫62内には、複数の分析項目のための第1試薬及び第2試薬(必要な分析項目用のみ)の試薬ボトル12が配列されており、所定温度に冷却される。それぞれの試薬ボトル12内の試薬液はチューブを介して試薬用ディスペンサポンプ60により反応容器列上の対応する試薬吐出ノズルに供給される。この場合、図1に示す分析装置3Aのディスペンサ方式試薬供給部32は図2の試薬用ディスペンサポンプ60,多数の試薬ボトル12を備えた試薬保冷庫62,第1試薬ノズル群保持部64,第2試薬ノズル群保持部66などを含む。

ラック送出部17から供給される個々の試料ラック1は主搬送ライン20によって搬送され、分析ユニット3Aによる分析処理が必要な場合には、分析ユニット3Aのサンプリングライン4Aに移載される。分注位置にきた試料ラック1上の試料は試料分注器48aのピペットノズルによって反応容器46aに所定量ピペッティング分注される。この反応容器には、反応容器列上の所定の位置で分析項目に対応する試薬が吐出され、反応が進行される。所定時間後、反応容器46a内の反応した液は多波長光度計15aによって光学的特性が測定される。多波長光度計15aから出力された信号は分析ユニット側コンピュータ6Aによる制御下で対数変換器30a及びアナログ・ディジタル変換器31aの処理を受け、全体制御用コンピュータ40に送信される。ディスペンサ方式の分析ユニット3F及び3Gも分析ユニット3Aと同様の構成である。

次に、ピペッタ方式の分析ユニットの構成例を、図3を参照して説明する。分析ユニット3Bの反応部5Bに配列された反応容器46b内では、所定の分析項目に関する検体と試薬の反応が進められる。主搬送ライン20からサンプリングライン4B(図1)に移された検体ラック1は、分注位置に位置づけられ、検体分注器48bのピペットノズルにより指示されている検体が採取され、反応容器46bへ検体の所定量が吐出される。検体分注器48bは検体用ピペッタポンプ47bを有する。反応部5Bは恒温槽10から供給される恒温液によって一定温度(例えば37℃)に保たれる。

図3の分析ユニットのピペッタ方式試薬供給部26は第1試薬用と第2試薬用の二つの試薬ディスク26A及び26Bを具備する。多数の分析項目のために準備された各種の試薬を含む試薬ボトル12A及び12Bには、それらの外壁面に試薬識別情報がバーコードによって表示されており、試薬ボトル12A及び12Bが試薬ディスク26A及び26Bに載置された後、各試薬ボトルの試薬識別情報がバーコード読取装置23A及び23Bによって読取られ、その情報が該試薬ボトルの試薬ディスク上のセット位置,対応分析項目,該試薬ボトルがセットされた分析ユニット番号などと共に記憶部45に登録される。試薬分注器8A及び8Bは旋回及び上下動可能な各ピペットノズルに接続された試薬用ピペットポンプ11を具備する。

試料を分注された反応容器46bの列は回転移動され、分析項目に応じて試薬吸入位置に位置づけられた試薬ボトル12Aから試薬分注器8Aにより所定量の試薬液が吸入され、その第1試薬が試薬添加位置にある反応容器46bに吐出される。撹拌位置にて撹拌機構13Aにより内容物が撹拌された後、反応容器列は複数回移送され、反応容器46bが第2試薬添加位置に達すると、試薬分注器8Bは分析項目に応じて試薬吸入位置に位置づけられた試薬ボトル12Bから試薬液を吸入し、該反応容器にその試薬を吐出する。次いで、反応容器の内容物は撹拌機構13Bによって撹拌される。その後、反応容器列の回転移送に伴って反応容器46bは光源14bからの光束を通過し、反応容器46bの反応液を透過した光が多波長光度計15bによって検出される。分析項目に対応する波長の信号は分析装置側コンピュータ6Bによって制御される対数変換器30b及びアナログ・ディジタル変換器31bによって処理され、ディジタル信号が全体制御用コンピュータ40へ送信される。測定済の反応容器46bは洗浄機構19bによって洗浄され、再使用される。分析ユニット3C,3D及び3Eは、分析ユニット3Bと同様の構成を有する。

試料ラック1がラック送出部17にセットされる前に、各試料に対して依頼元から検査指示依頼された分析項目が各試料番号と共に予め操作部42から全体制御用コンピュータ40に登録される。それぞれの分析項目の分析条件情報は、フロッピーディスクメモリ41に記憶されている。その分析条件の内、分析項目コードは5桁の数字からなる。同種の分析項目のために複数の分析ユニットで共通に使用されるべき分析条件パラメータは光度計における測定波長,検体採取量,検量線校正方法,標準液濃度,標準液の本数,分析値異常のチェック限界値などである。分析条件パラメータのうち、各試薬ボトルに対応して記憶されているパラメータは、第1試薬から第4試薬までの必要な試薬数,5桁の数字からなる試薬ボトルのコード,試薬の分注量,1つの試薬ボトル当りの分析可能なテスト数などである。分析ユニット3A,3B及び3Cは血清試料を、分析ユニット3D及び3Eは血漿試料を、分析ユニット3F及び3Gは尿試料をそれぞれ受け入れ可能に各ユニット条件が設定されており、全体制御用コンピュータに分析ユニット番号と共に受け入れ可能検体種別が登録されている。

各分析ユニット3A〜3Gの試薬供給部に試薬ボトルが収納されるのに伴って各試薬ボトルの試薬識別情報が分析ユニット番号と対応づけられて全体制御用コンピュータ40に登録される。この場合、同じ種類の試料を扱う同じグループの複数の分析ユニットに同一種の分析項目用の試薬が収納される。例えば、血清試料の場合には3A,3B,3Cの分析ユニットが同じグループとして扱われるが、その内、分析ユニット3Aの試薬供給部32には、例えば試料依頼数の多い肝機能検査項目であるGOT,GPT及び緊急検査項目であるカルシウム,UA,BUNのための試薬ボトルが収納され、分析ユニット3Bの試薬供給部26には、例えば肝機能検査項目であるGOT,GPT及び検査依頼数の少ない他の分析項目のための試薬ボトルが収納され、分析ユニット3Cの試薬供給部27には、例えば緊急検査項目であるカルシウム,UA,BUN及び検査依頼数の少ない他の分析装置のための試薬ボトルが収納される。従って、肝機能検査項目は2台の分析装置3A及び3Bによって分析処理可能になり、緊急検査項目は2台の分析装置3A及び3Cによって分析処理可能になる。何台の分析装置にどのような分析項目用の試薬を重複させて収納するかは、それぞれの施設の検査室の実情に応じて操作者により決定される。

各試薬ボトル12,12A,12Bが各試薬供給部に収納されるのに伴って、試薬ボトルに設けられた試薬識別情報が読取られ、試薬ボトルコードをキーとして、分析条件パラメータとして既に登録されている情報が検索され、その試薬ボトルに対応する分析項目,ボトルの大きさ,分析可能なテスト回数,試薬ボトルのセット位置などがそれぞれ関係づけられて全体制御用コンピュータ40に登録される。同時に、同種の分析項目の分析処理が可能な複数の分析ユニットにおける同種の分析項目用の試薬ボトル全数に基づく最大分析可能回数も登録され、必要に応じてCRT43に表示される。

各分析ユニットにとって必要な分析項目用の対応試薬が収納された後、検体の分析処理に先立って各分析ユニット毎に、その分析ユニットによって分析処理可能な全分析項目のための検量線校正操作がそれぞれ実行される。各分析ユニットにセットされた試薬ボトルの違いによって検量線の校正値が相違するので、分析項目毎に個々の分析ユニットで得た検量線校正結果を全体制御コンピュータ40の記憶部45に記憶せしめる。これらの校正結果は、各分析ユニットにおいて該当する分析項目が分析処理されたときの濃度演算に使用される。なお、検量線の校正(作成)に関しては更に後述する。

ラック送出部17上に置かれた試料ラック1の内の1つが主搬送ライン20の方へ押し出されれると、それに伴って、その試料ラック1の識別情報又は試料容器2の識別情報が識別情報読取装置50によって読取られる。読取られた情報に基づいて該検体ラック1上の試料種別が全体制御用コンピュータ40によって判定され、その検体種のために予め条件設定されている分析ユニットグループが選定され、その後の判断結果によりその分析ユニットグループの内の1つが試料搬送先として決定される。ここでは、例えば血清試料が判定され、その試料ラックが搬送されるべき分析ユニット3A,3B,3Cのグループが選定されるものとする。

さらに、試料識別情報の読取りに伴って試料番号及び分析項目の登録状況が照合され、試料ラック1上の各試料のために測定指示されている分析項目が判定され、各試料の各分析項目が分析ユニット3A,3B及び3Cの内のいずれかによって分析処理すべきかが、全体制御用コンピュータ40によって判断される。この場合、全体制御用コンピュータ40は、各分析装置に対して既に分析処理が指示されている分析項目数及びそれらの試料の分注終了までにどの程度の時間を要するかを監視している。特に、複数の分析ユニットによる分析処理が可能な特定の分析項目に関しては、いずれの分析ユニットによって該分析項目を分析処理させるのが効率的であるかが判断される。例えば、特定分析項目であるGOT及びGPTに関し、その時点で処理待ち試料数が最も少ない分析ユニットは3Aと3Bのいずれであるかが判断され、待ち時間の少ない方が指定分析ユニットとされる。このような複数の分析ユニット同士の多忙度の程度に応じて特定分析項目を分析処理すべき分析ユニットを自動的に指定する方法以外に、操作者が予め操作部42から各分析項目の処理に使用すべき分析ユニットの優先順位を入力しておく指定方法も可能である。
