【発明の属する技術分野】
本発明は、特定の生薬抽出物を有効成分として含んでなる頭部用組成物に関する技術分野の発明である。より具体的には、キク科(Compositae)ハマグルマ属(Wedelia)に属する植物であるクマノギクの抽出物を配合することにより、優れた脱毛防止、ふけ・痒み抑制作用効果を有するとともに、毛乳頭細胞を活性化することによって、毛髪伸長の促進をする等の効果を有する頭部用組成物に関し、この組成物は特に養毛用組成物、毛乳頭活性化用組成物、さらにはフケ防止用組成物として有用な組成物である。

【従来の技術】
高齢化社会、ストレス社会といわれる現代社会では、頭部毛髪が様々な原因により脱毛の危機にさらされる機会がますます多くなってきている。
これに対応して、より優れた「養毛料」を提供すべく様々な試みがなされている。
養毛料が毛髪に与える効果として主なものに、発毛誘導効果(発毛促進効果,成長期誘導効果)、毛髪を太くする効果、毛髪成長期延長効果、5α−レダクターゼ阻害効果、血行促進効果、殺菌効果、フケ防止効果、保湿効果、抗酸化効果等の効果が挙げられる。

しかしながら、前記のように種々の試みがなされているにもかかわらず、従来の養毛料では、その脱毛防止,発毛効果等の養毛作用は必ずしも十分なものではなかった。これはおそらく脱毛の原因がさまざまであり、また発毛の機構も非常に複雑であるためと考えられている。従って、このような毛髪の複雑な機構に幅広く対応して、様々な用途に応用可能な頭部用組成物を提供することが望まれている。また、これと同時に毛髪のより具体的な構造や機能に着目した養毛用組成物を提供することも極めて有用であり、そのような薬剤を提供することが望まれている。

【発明が解決しようとする課題】
そこで、本発明の課題は、上記の「幅広い対応」という要素と「具体的な構造と機能に着目した」という要素を併せ持つ頭部用組成物を提供することにある。すなわち、本発明は多様な脱毛の原因に対処すべく、優れた脱毛防止効果,発毛促進効果等の養毛作用やフケ,痒みの抑制作用、並びに毛周期と深い関係を有する毛乳頭細胞を活性化することによって毛髪伸長の促進をする個別的な養毛効果を有する頭部用組成物、そして養毛用組成物、毛乳頭細胞活性化用組成物、さらには、フケ防止用組成物を提供することを課題とする。

【課題を解決するための手段】
本発明者は、有用物質の探索対象を脱毛防止効果及び発毛促進効果等の養毛作用を示すことが示唆されていない植物体抽出物に広げ、上記目的を達成すべく検討してきた。その結果、キク科(Compositae)ハマグルマ属(Wedelia)に属する民間薬または生薬として使用されることのある植物であるクマノギクの抽出物が、優れた脱毛防止効果、発毛促進効果等の養毛作用及びフケ、痒み抑制作用を示すことを見い出した。さらに、この抽出物が毛乳頭に直接働きかけて、毛髪成長期延長効果を有する成分であるか否かについて、本発明者が見出した育毛薬剤検定方法を用いて鋭意検討したところ、所望する毛乳頭活性化能乃至毛髪成長期延長効果が認められることを見出し、本発明を完成するに至った。

すなわち本発明は、クマノギク(学名:Wedelia chinensis MERR 若しくはWedelia calendulacea L.)の抽出物を含んでなる頭部用組成物を提供する発明である。

本発明において「頭部用組成物」は、上記の有効成分の頭髪における多様な効能を発揮することが可能な頭髪用の用途に用いることができる組成物を意味するものであり、以下に述べる「養毛用組成物」、「毛乳頭活性化用組成物」、及び、「フケ防止用組成物」を包含する概念である。

「養毛用組成物」とは、発毛促進、脱毛防止、さらにふけや痒みの抑制作用等、並びに毛乳頭細胞活性化作用を包含した総合的な作用により、主に「発毛促進」「脱毛防止」という現象を通じて、毛髪の状態を維持改善する「養毛作用」を発揮し得る頭部用組成物を意味するものである。
また、「フケ防止用組成物」とは、文字通り、フケの発生を防止し得る組成物を意味するものである。

さらに「毛乳頭活性化用組成物」は、主に後述する育毛検定方法によって毛乳頭を刺激することによって、毛周期における成長期を延長させる作用と同休止期から成長期への移行を促進する作用を有する成分を含んでなる、特に「毛乳頭」という特定の作用点に着目した毛髪関連組成物であり、いわば「個別効能を有する養毛用組成物」としての特徴を有する。

【発明の実施の形態】
以下、本発明の実施の形態について説明する。本発明頭部用組成物は、キク科(Compositae)ハマグルマ属(Wedelia)に属するクマノギクの抽出物を有効成分とする毛髪関連組成物である。

クマノギク(学名: Wedelia chinensis MERR もしくは Wedelia calendulacea L. )は、日本ではクマノギク,ハマグルマ又はシオカゼと呼ばれているが、本明細書においては「クマノギク」としてこれをあらわす。

クマノギクの抽出物に関する報告はこれまでにないばかりか、養毛用途を含めた頭部用組成物としての応用は全くない。

本発明頭部用組成物に配合される抽出物は、この植物の葉、地下茎を含む茎、根、果実、樹皮等、植物全草を抽出溶媒と共に浸漬または加熱還流した後、濾過し、濃縮して得られる。本発明に用いられる抽出溶媒は、通常抽出に用いられる溶媒であれば特に限定されず、例えば熱水やメタノール、エタノール、イソプロパノール、n−ブタノール等の低級アルコール、あるいはプロピレングリコール、1,3−ブチレングリコール等の多価アルコール、あるいはこれらの含水アルコール類、アセトン、酢酸エチルエステル等の有機溶媒を単独又は組み合わせて用いることができる。低級アルコールを使用する場合、得られる抽出液をそのまま用いることもできるが、抽出溶媒を留去し、必要により乾燥した後、本発明頭部用組成物に配合してもよい。

本発明頭部用組成物におけるクマノギクの抽出物の配合量は、組成物の形態,施用方法に応じて変動しうるのである。しかし、後述の実施例に記載の方法に従って得られる抽出物を使用する場合、一般的には組成物全重量中抽出物(乾燥物基準)が0.000001〜5.0重量%、好ましくは同 0.00001〜1.0重量%である。この配合量が組成物全重量中の0.000001重量%未満であると、本発明の所期の効果が十分に発揮されず好ましくなく、同5.0重量%を超えると製剤化が困難になる傾向が強くなり好ましくない。また、組成物全重量中の1.0重量%以上配合しても配合量の増加に見合った効果の向上は認められなくなる。

このように、クマノギクの抽出物を配合することにより、優れた脱毛防止効果、発毛促進効果等の養毛作用及びフケ、痒み抑制作用を示す養毛用組成物、ないし、フケ防止用組成物等としての形態を採り得る頭部用組成物が提供される。

また、後述する実施例において明らかにされるように、クマノギクの抽出物は、特に毛乳頭細胞に直接働きかけて、毛周期における成長期の延長作用を示す成分である、という特徴を可能な限り活用することが可能な態様の毛乳頭活性化用組成物も提供される。

このような本発明頭部用組成物には、上記必須成分以外に、通常化粧品、医薬部外品や医薬品等の養毛料に用いられる成分、例えば、油性成分、保湿剤、増粘剤、金属封鎖剤、その他の活性成分、酸化防止剤、紫外線吸収剤、界面活性剤、アルコール類、粉末成分、色剤、水性成分、水、各種皮膚栄養剤等を必要に応じて適宜配合することができる。

具体的には、油分として、例えば高級脂肪酸、固形パラフィン、流動パラフィン、シリコーン油、スクワラン等;保湿剤として、例えばヒアルロン酸、プロピレングリコール、マルチトール、アテロコラーゲン、乳酸ナトリウム等;増粘剤、マルメロ粘質物、カルボキシビニ−ルポリマー、キサンタンガム等;金属封鎖剤として、例えばエデト酸二ナトリウム、エデト酸三ナトリウム、クエン酸ナトリウム、ポリリン酸ナトリウム、メタリン酸ナトリウム、グルコン酸;その他の活性成分として、例えばカフェイン、タンニン、ベラパミル、トラネキサム酸およびその誘導体、甘草抽出物、グラブリジン、火棘の果実の熱水抽出物、各種生薬、酢酸トコフェロール、グリチルリチン酸およびその誘導体またはその塩等の薬剤、ビタミンC、アスコルビン酸リン酸マグネシウム、アスコルビン酸グルコシド、アルブチン、コウジ酸等の他の美白剤、グルコース、フルクトース、マンノース、ショ糖、トレハロース等の糖類などが挙げられる。

また、補助成分として、これらの製剤を調製する上で上記油性成分、保湿剤、増粘剤に加えて、その他の活性成分として、例えばモノオレイン酸グリセリル等の油分、ニコチン酸アミド、ニコチン酸ベンジル、ビタミンE アセテート、センブリ抽出物、塩化カルプロニウム、センブリエキス、アセチルコリン誘導体等の血管拡張剤、セリン、メチオニン等のアミノ酸類、ビタミンB6、ビタミンE 及びその誘導体、ビオチン等のビタミン類、パントテン酸及びその誘導体グリチルレチン酸及びその誘導体、ニコチン酸、ニコチン酸メチル、ニコチン酸トコフェロールなどのニコチン酸エステル類、セファランチン等の皮膚機能亢進剤、エストラジオ−ル等の女性ホルモン剤等を同時に配合してもよい。

さらに、通常、養毛料に用いられる添加剤、例えばヒノキチオ−ル、ヘキサクロロフェン、ベンザルコニウムクロリド、セチルピリジニウムクロリド、ウンデシレン酸、トリクロロカルバニリドおよびビチオノール等の抗菌剤、メントール等の清涼剤、サリチル酸、亜鉛およびその誘導体、乳酸およびそのアルキルエステルなどの薬剤、クエン酸等の有機酸類、アルギニン等のアミノ酸類等が本発明の効果を損なわない範囲で適宜配合することができる。

本発明頭部組成物の性状は、例えば、軟膏、クリーム、乳液、ローション、パック等、従来養毛料に用いるものであればいずれでもよく、剤型は特に問わない。また、本発明の養毛料としては、例えば、ヘアートニック、頭部用クリーム、頭部用ムース、シャンプー、リンス、頭部用ローション等の剤型をとることもできる。

【実施例】
次に実施例をあげて本発明をさらに具体的に説明するが、本発明はこれらにより限定されるものでない。なお、配合量は特記しない限り重量%である。
まず、A.発毛効果、B.育毛効果、C.培養毛乳頭細胞を用いた細胞増殖に関する試験、D.実使用試験とその結果について順に説明する。

A.発毛試験
1.抽出物の調製(調製例1)
市販のクマノギク(乾燥物) 2000gを、8リットルのメタノールに室温で5日間浸漬した。抽出液から溶媒を留去し、次いで乾燥してクマノギクのメタノールエキス乾燥物 162.4gを得た。

2.試料の調製
上記で得られたメタノールエキス乾燥物 0.1%、0.5%、1%、2%を、それぞれ75%エタノール溶液に溶解して試料1〜3を得た。

3.発毛試験方法及びその結果
対照試料として 0.1%クロトン油の75%エタノール溶液を用い、本発明の液状の試料1〜4(第1表参照のこと)について発毛試験行った。
すなわち、実験動物として毛周期の休止期にあるC3H/HeNCrJマウスを使用し、小川らの方法(ノーマルアンドアブノーマル、エピダーマル、ディファレンシエ−ション[Normal and Abnormal Epidermal Differentiation ]、M .Seiji およびI .A .Bernstein 編集、第159 〜170 ページ、1982年、東大出版)に従って行なった。

すなわち、マウスを1群10匹とし、それぞれ被検試料1〜4と対照試料( 0.1%クロトン油の75%エタノール溶液)用の5群に分け、バリカンおよびシェ−バーでマウスの背部を剃毛し、それぞれの試料を1日1回、 0.1mlずつ塗布した。18日、24日後に毛の再生面積を測定した。結果は再生面積の平均値で表した。その結果を第1表に示す。

第1表より、クマノギク抽出物は、マウスの発毛試験において優れた発毛効果を示した。また、1.0%以上の濃度ではその効果の向上は認められなかった。この結果より、クマノギクの抽出物を配合した本発明頭部用組成物は、優れた発毛効果を有することは明らかになり、この組成物は養毛用途に適した頭部用組成物であることが判明した。

B.育毛試験(トリコグラム試験)
1.抽出物の調製
上記発毛試験例で調製した調製例1の方法で抽出物を調製した。

2.試料の調製
上記方法で得られたメタノールエキス乾燥物 0.05、0.5および1%を、70%
エタノール溶液(90%)、オレイン酸ナトリウム(0.01%)、ドデシルベンゼンスルホン酸(0.49%)、硬化ヒマシ油エチレンオキシド(40モル)付加物( 0.5%)及びイオン交換水(8%)と混合撹拌して溶解させた。さらにイオン交換水(残余)を添加混合して、液状の試料5、6および7を得た。

3.育毛試験方法及びその結果
本発明の養毛料の脱毛防止、発毛効果等の養毛作用を調べるために、ヒトに対して、以下の方法でトリコグラム試験を実施した。
試料の使用前と使用後の抜去毛髪の毛根を顕徴鏡下で観察し、毛根の形態から休止期毛根数を計数し、その割合の増減によって養毛作用を比較した。尚、休止期毛根とは成長の止まった毛の毛根であり、脱毛を訴える人は正常な人よりもこの休止期毛根の割合が多いことが認められている。

被験試料及び対照試料の各試料をそれぞれ男性被験者10名の頭皮に1日2回、1回2mlずつ6ケ月間連続して塗布し、塗布直前および6ケ月間塗布終了直後に被験者1名につき 100本ずつ毛髪を抜去し、それぞれの毛根を調ベ、実使用テストを行った。結果を第2表に示す。

この結果より、クマノギクの抽出物を配合した本発明頭部用組成物は、毛周期の成長期を延長させること基づく優れた発毛効果を有すること明らかになり、上記の試験Aにおける結果と同様に、この組成物は養毛用途に適した頭部用組成物であることが判明した。

C.培養毛乳頭細胞を用いた細胞増殖試験
1.抽出物の調製
上記抽出物の調製は、上記発毛試験例で調製した調製例1の方法で抽出物を調製した。さらに、メタノールエキス乾燥物をDMSOで0.2重量%の溶液に調製して、これを無血清培地(MEM)に希釈してクマノギク抽出物をそれぞれ1.0×10−9〜1.0×10−6%を含む溶液を調製した。

2.毛乳頭細胞の採取
整形外科手術によって摘出された32歳男性の後頭部皮膚(5mm×1.5cm)から、脂肪組織を分離して、そこから毛包を摘出し、毛球部より毛乳頭細胞を単離した。単離した毛乳頭細胞を20%FBSを含むMEMで2週間培養した〔37℃,5%CO2 〕。毛乳頭細胞から細胞のアウトグロースが確認された時点で、培地を10%FBSを含むMEM(MEM+10%FBS)に交換して同様の条件で培養した。以降、1週間に2回の割合で培養液(MEM+10%FBS)を交換して細胞を維持した。
培養開始より4週間後に継代培養を行い、以後細胞が十分増殖した時点で再度継代して、この継代を繰り返した。

3.試験方法
継代数3代目の毛乳頭細胞を用い、MEM+10%FBS培地で,10000及び細胞/mlの細胞密度の細胞懸濁液を調製した。この細胞懸濁液を200μl ずつ、96ウエルのマイクロプレートに分注し(つまり,2000細胞/ウエル)、37℃,5%CO2 で3日間インキュベートを行い細胞を付着させた。培養開始72時間後、対照は培養液を無血清のMEMに交換した。被検体系は対象物質であるクマノギク抽出物を含む無血清培地(MEM)に交換した(各試料8〜11)。
対照および被検体系をいずれもさらに4日間培養した。

培養終了後、それぞれの系にアラマーブルー(alamar blue)(BIOSOURSE社製) を20μl 添加後、さらに8時間培養し、マイクロプレートリーダー(Micro plate reader:Bio RAD社製) で570nmと595nmの吸光度を測定した。添付の使用説明書に従って、吸光度の測定結果によりアラマーブルーの還元率を算出した。この還元率は細胞数と相関することから対照および被検体系での細胞増殖率を比較した。

測定したクマノギク抽出物(試料8〜11)における、上記細胞増殖促進指標を下記第3表に示す。

第3表より明らかなように、本発明のクマノギク抽出物は、培養毛乳頭細胞活性化試験において有意な毛乳頭細胞増殖促進効果を示した。このことより、本発明頭部用組成物は、毛乳頭細胞に直接働きかけて養毛効果を発揮する「毛乳頭活性化用組成物」としての用途に特に適していることが判明した。

上記処方にて製造したヘアートニックについて実使用でフケ、発毛、脱毛等の症状に対する効果を検討した。フケ、発毛、脱毛等の症状を呈する10名の男性(年齢25才〜55才) に1日1〜2回、1〜3mlずつ4ヵ月にわたって投与し、以下に述べる評価基準で、フケ防止効果、発毛促進効果および脱毛防止効果を試験した。その結果を第3表に示す。

第4表から明らかなように、このヘアートニックは、発毛促進、フケ防止及び脱毛防止に対して優れた効果を発揮することが明らかになった。
このことは、上述したように本発明頭部用組成物は、養毛用途に適していることは勿論のこと、フケ防止等の他の目的を有する頭部用組成物としても非常に有用であることが明らかになった。

以下に、種々の剤型の本発明による養毛料の処方例を実施例として説明する。なお、これらの処方の本発明頭部用組成物について、上記の試験を行ったところ、いずれの組成物においても発毛効果、育毛効果、培養毛乳頭細胞増殖効果及び実使用において、非常に優れた結果を示した。
