In epidermal keratinocytes, Amla showed significant growth-promoting effects, and Seitamirobaran and Acupuncture showed significant growth-promoting effects. As for dermal fibroblasts, centella and berlin showed significant growth-promoting effects. Anemone and Takasaburo, for which hair growth effects have been proposed, did not show significant effects at the cellular level. Tests were conducted on the herb combinations, but the growth effect was generally additive. Therefore, it was difficult to investigate the synergistic effect of the herb combination using cell tests. These results are natural because hair formation involves multiple cells and many factors. However, the cell test was effective in selecting the herbal material used in the following animal tests.
2.マウス発毛試験
実験動物として毛周期の休止期にある6週齢のBDF−1オスマウスを購入した。マウスは6週目で毛周期の退行期に入り、7週目から休止期に入る。一週間の順化後、バリカンおよびシェーバーでマウスの背部を剃毛し、試料を1日1回、1~2mLずつ背部に塗布した。各試料につきマウス5匹を使用した。個体差の影響を避けるために、比較する二つの試料を同じマウスの背部の左右に塗布した。観察と撮影を1週間毎に行い、6~7週間後に発毛効果を下記の評価基準にしたがって判定した。

<マウス試験の結果>
マウス発毛試験の結果は表4に、具体的データは図1に示した。ハーブエキス単独では、アムラが有意な発毛効果を示したが、クマノギクとタカサブロウのエキスは両者とも発毛効果は弱かった。しかし、これら3種のハーブを組合わせると、明確な発毛効果が認められた。6種のハーブを組合わせると、発毛効果がさらに増強され、16種類のハーブ組合わせると、さらに効果が増強された。ハーブ16種のオイル抽出物は同エキス試料と同程度の効果を示した。16種類のハーブに補助成分を添加した試料(表6のレシピBは)は最も顕著な発毛効果を示した。

3.ヒトモニター試験
A.ヘアーオイルのモニター試験
表3に記載したハーブ16種オイル試料について、発毛効果を検討した。アーユルヴェーダは育毛にヘアーオイルを好んで用いるからである。被験者は脱毛症状が進行した比較的若い12名のバングラデシュ人男性(25歳~42歳)を採用した。1日に朝晩2回、1~2mLを6ヶ月頭部に塗布した。下記の評価基準に従って発毛促進効果を評価し、結果を表6に示した。

 B.ヘアートニックのモニター試験
表3に示す2種類のヘアートニック試料(有効成分の濃度が異なるレシピAおよびレシピB)を作成した。試料の作成には、脂溶性成分の可溶化、細胞毒性の軽減、栄養成分等の添加を施した。17名の日本人男性(27歳~60歳)と11名の日本人女性を2群に分け、それぞれの群にレシピAまたはレシピBを1日に朝晩2回、1~2mLを6ヶ月頭部に塗布した。下記の評価基準に従って、脱毛防止、増毛効果、発毛効果を評価し、得られた結果を表6に示した。

<ヒトモニター試験の結果>
 表4&5から明らかなように、ヒトモニター試験において、本発明を特徴づける植物抽出物を含むヘアーオイルおよびヘアートニックが、被験者のほとんどにおいて顕著な脱毛防止効果、増毛効果、および発毛効果を示した。いずれの試験においても、皮膚刺激やアレルギー反応等の皮膚障害を訴えた被験者は皆無であった。これらの成績は、育毛あるいは発毛を目的とする既知の化粧品、医薬部外品、医薬品等のどれよりも優れていると判定される。したがって、本発明を特徴づける植物抽出物を含む頭部用組成物は、新規性、進歩性および有用性に富むと判断した。
