実施例1 FBGおよびRCの調製 シグマ(Sigma)から購入したフルオロセイン−BSA−
グルコース(FBG)およびモレキュラープローブ(Molec
ular Probes)から購入したローダミン−ConA(RC)を
ギブコ(Gibco)からのハンクス緩衝化塩溶液(HBSS)
に最終濃度2mg/mlで溶解した。溶液を次に10,000gで30
分遠心し大粒子を除去した。上澄みを次に回収し10,000
MWアミコン限界濾過容器に置き、限界濾過が膜を通過す
るまで2,000gで遠心した。残った物質を2mlsのHBSSに再
溶解し、この手順を限界濾過物中に遊離のフルオロセイ
ンまたはローダミンが検出されなくなるまで繰り返し
た。

実施例2 フルオロセイン−BSA−グルコースおよびロ
ーダミン−ConAの至適濃度の決定 マンノースおよびグルコースで競合実験を行う前に、

移に使用する任意の溶液中において、FRETが単に分子同
士が溶液中で相互に十分に接近することによって起こる

験は理想的には1mM以下の濃度で行われるべきである、
なぜならその濃度では分子は120A単位離れているからで
ある(特異的に反応するに十分離れている)。逆に、こ

Claims (2)
- 【請求項1】体液試料中のグルコースを定量する方法で
あって: a)体液試料を蛍光的に標識したグルコース−結合レク
チンおよび蛍光的に標識した相補的糖複合体と接触さ
せ、 該レクチンおよび該糖複合体は体液中に見い出される生
理的濃度のグルコースの存在中で、該体液中のグルコー
スが該糖複合体と置換し、可逆的に該レクチンと結合で
きるように互いに可逆的に結合し; b)蛍光的標識レクチンと蛍光的標識糖複合体の間に起
こる非−放射性蛍光共鳴エネルギー転移の程度を測定
し; c)工程(b)でなされた測定と、非−放射性エネルギ
ー転移の程度とカリブレーション工程で決定された体液
中のグルコース濃度との関係とを比較する、 工程から成る方法。 - 【請求項2】体液試料中のグルコースを定量する方法で
あって: a)体液試料を第一光吸収物質で標識されたグルコース
−結合リガンドである第一部材および第二光吸収物質で
標識された糖複合体である第二部材から成る特異的結合
対と接触させ、一方の光吸収物質の発光スペクトルが他
方の光吸収物質の吸収スペクトルと重複し、 該リガンドおよび該糖複合体は体液中に見い出される生
理的濃度のグルコースの存在中で、該体液中のグルコー
スが該糖複合体と置換し、可逆的に該リガンドと結合で
きるように互いに可逆的に結合するように選択され; b)第一光−吸収と第二光−吸収物質との間に起こる非
−放射性蛍光共鳴エネルギー転移の程度を、第一および
第二光吸収分子に帰する発光の比を測定することにより
決定し; c)工程(b)の結果と、第一光−吸収物質と第二光−
吸収物質との間の非−放射性エネルギー転移の程度とカ
リブレーション工程で測定された体液中のグルコース濃
度との関係とを比較する、 工程から成る方法。
