体液中のグルコースの検出および定量の方法ならびに用具発明の背景
グルコース濃度の測定法は、例えばグルコース恒常性が維持されない患者(例え
ば糖尿病または低血糖症)のグルコースレベルを毎日監督するためのような臨床
的設!、ならびに生物医学的研究に応用を有する。
グルコース濃度決定法の最近の方法は種々の発色反応を含み、解糖カスケードに
おけるいかなる数の生成物の性質の分光光度的変化をも測定し、または電気分解
自記法的グルコースセンサーを使用してグルコースの酸化を測定する。例えば米
国特許第4、401、122ではグル:−スのin vivo測定法を開示し、
これは酵素(例えばグルコースオキ/ダーゼ)を皮膚の中または下において酵素
反応生成物(例えば酸素)を電気分解自記法または酵素電極を使用して直接皮膚
を通して検出する。検出された酵素反応生成物の量は基質量の計量法である。

このlieadowおよびSchultzの光学センサーは多《の問題を伴う、

なろう。これはまた他の多くの研究および臨床的応用も有するであろう。
発明の要約
本発明は非一放射性蛍光共鳴エネルギー転移(FRET)を使用する体液中のグ
ルコースの検出および定量方法、ならびに本発明のグルコース検出および測定法
を実行するために使用する用具に関する。
本発明の方法において、体液は特別な結合対、標識リガントおよび該リガンドに
グルコースと拮抗して特異的に結合する標識炭水化物一含有レセプター(標識糖
複合体という)の二つの部材、と接触させられる。結合対のそれぞれは光一吸収
物質で標識される。一般的に、重複する励起状態エネルギーレベルを有するふた
つの光一吸収物賃が使用され、ひとつはりガンドにおよびひとつは糖複合体に固
定される。
特に、本発明において、二つの蛍光的〜標識基買、リガンド(例えばレクチン、
モノクローナル抗体)およびグルコースと拮抗してリガンに特異的に結合する炭
水化物一含有レセプターまたは糖複合体は、体内で、または常法によって得た試

FRETは供与体と受容体とが互いに十分に近接している時に起こる(一般的に
100オングストローム以内)。本発明の方法において、F R. E Tはり
ガンドと試料中の糖複合体が結合するときに起こる。しかし、グルコースが存在
すると、これらの分子は競合し(即ち、グルコースは糖複合体とリガンドの結合
を競合する)。故に試料中のグルコースの存在と濃度は、工不ルギー転移の効率

て供与体分子の励起状態エネルギーが受容体分子に転移し、供与体の蛍光を消滅
させ受容体の増感された発光が起こるという事実を利用する。
このような物質は、それは特異的結合対の部材中に接着または組み込まれ(即ち
、リガンドおよび糖複合体)本発明の方法に使用されるが、例えばフルオロセイ
ンおよびローダミンのような蛍光性物質で有り得る。択一的には、ひとつ(供与

与体消光の割合により検出される。
グルコースとの連絡において、反応物質の配買の多様な様式は、本発明のin
viv9態様の原理に従って採用できる。加えて、グルコースとの連絡において
反応物質のいかなる配置もインスリンボンブのフィードバックを提供するために
変更できる。
広範な観点から本発明のin vitroの態様は、グルコースを含有する体液
試料(例えば、血液、尿、細胞外体液)との連絡における反応物質の配置に関し
、例えば該液を反応物質が上に固定されているディスクチップと接触させること
などである。
グルコースはグルコースを含有する体液との関連で反応物質を配置させることに
より蛍光光度測定計中で検出および定量される。in viν0態様として、グ
ルコースに連絡する反応物質の種々の配置様式が採用できる。
本発明の方法および用具は、広範なグルコース濃度(例えば0.5からlbg/
ml)の検出に使用できる。加えて、この方法は信頼できる、なぜならば供与体
および/受容体の発光最大波長で測定された蛍光強度の割合として、またはグル
コースの関数として供与体の発光最大での消光として測定された反応物質は凝集
および生成物の大きさにより測定を妨害しないからである。
また反応物質は消費されないので、用具は長期間再使用可能である。最後に、虱

本発明は体液中のグルコースを検出し定量する方法および本発明のグルコース検
出および定量を行うために有用な用具に関する。本方法はグルコースによって競
合的に減少させられた特異結合対の部材間の結合の発生および量を測定するため
の非−放射性蛍光共鳴工率ルギー転移(FRET)の工程に基づく。結合対の部
材はリガンド(例えばレクチン、モノクローナル抗体)および炭水化物含有レセ
プター(糖複合体という)であり、これらはグルコースと競合してリガンドに特
異的に結合する。リガンドおよびW複合体の双方とも蛍光的に1!!識されてお

FRETの
FRETは一般的に二つの発蛍光団間のエネルギーの非−放射性転移、ひとつは
エネルギー供与体(D)およびもう一方はエネルギー受容体(A)が関与する。
供与体の発光が受容体の励起スペクトルと重複し、かつ双方の部材がひとつの波
長で光エネルギーを吸収し、異なる波長の光を発光するならば任意の適当に選択
された供与体−受容体対が利用できる。
以下に説明する方法は特定の供与体−受容体対としてフルオロセインとローダミ
ンに関する。ここで使用される、フルオロセインという用語は種々の関連する化
合物およびそれらの誘導体を含む種類をいう。同様にここで使用される、ロー長
で光エネルギーを吸収し、異なる波長の光を発光するならば任意の適当に選択さ
れた供与体−受容体対が利用できる。
以下に説明する方法は特定の供与体−受容体対としてフルオロセインとローダミ
ンに関する。ここで使用される、フルオロセインという用語は種々の関連する化
合物およびそれらの誘導体を含む種類をいう。同様にここで使用される、ローダ
ミンは種々に関連した化合物対およびそれらの誘導体をいう。その他の供与体−
受容体対の例はローダミンに対するNBD N (7−ニドロベンズー2−オキ
ソ−1,3−ジアゾール−4−イル)、エオシンまたはエリトロノス(eryt
hrosjs)に対するNBD、または蛍光、ローダミンに対するダンンル、ロ
ーダミンに対するアクリノンオレンジなどである。
択一的にひとつの分子が(供与体)が蛍光性でもう一方が(受容体)非蛍光性で
あることができる。また、(蛍光性)供与体を励起するために使用する波長では
通常は励起しない受容体との供与体−受容体対を使用することも可能である。
しかし非−放射性F R,E Tは受容体励起を引き起こす。
供与体および受容体はここで”対”と呼ばれるが、二つの”部材“の対は実際間
−基貫でありうる。一般的には二つの部材は異なるであろう(例えばフルオロセ
インとローダミン)。一つの分子(例えばフルオロセイン、ローダミン)を供与
体および受容体の両方に使用することは可能である:この場合、エネルギー転移
は蛍光の偏光の偏りの度合いの減少によって測定される。
FRETは図1に表される。供与体の吸収および発光は、それぞれA (D)
。
E (D)と呼ばれ、受容体の吸収および発光は、それぞれA (A)、E (
A)と呼ばれ3図IAに図式的に示される。供与体発光と受容体吸収スペクトル
の重複した面積が(それは重複積分)重要である。もし励起が波長Iで起こるな
らば、供与体による光は波長■で発光するであろう、しかし受容体は光を波長I
では吸収しないので受容体による波長■ではない。
生じる非−放射性転移の過程は図IBに表される。0分子は光子を吸収し、その
電場ベクターをEにより表す。Dの励起状態は一方の側に正電荷を、もう一方の
側に負電荷を有する双極子として表される。もし受容体分子(A)が十分にDに
近づけば(例えば典型的には100オングストローム)、反対に荷電した双極子
はその上に誘導される(励起状態に上昇する)。この双極子−誘導双極子の相互
反応は、供与体−受容体分子間の第6カとして逆に減少する。
古典的には、部分的エネルギー転移が起こり得る。しかし、これはFRETで生
じるものとは異なり、100かOかの量子力学的出来事である。これは供与体は
そのエネルギーの一部を受容体に与えることができない。すべてのエネルギーは
転移されなければならず、エネルギー転移は、エネルギーレベルが(即ちスペク
トル)重複するときにのみ起こる。Aが励起状態から脱する時、放出光は入射光
に関して回転させられるかまたは入射光偏光の偏りの度合いを減少させられる。
結果として、F R,E Tは■でそれ自体の蛍光強度の減少(!I]ち供与体
発光の減少)、■での蛍光強度の出現(即ち増感した発光の増加)および入射光
に関連した蛍光の偏光の偏りの度合いの現象として明らかになる。
最終的なPRETの顕在化は、励起状態寿命中にある。蛍光は平衡化過程として
見ることができ、その中で分子が励起状態に留まる期間の長さは、入射光により
分子がこの状態に入った時の速度とこの状態から脱した時の速度の和との間の競
合の結果である(蛍光および非−放射過程)。さらに非放射過程、FRET、が
加えられれば(すべてを非電荷とし)、崩壊が好ましく、これは■での供与体寿
命が短縮される。
二つの発蛍光団の励起状態および発光スペクトル重複が十分に接近している時、
供与体分子の励起状態エネルギーは共鳴双極子−誘導双極子相互反応により隣に
ある受容体発蛍光団へ転移される。FRETにおいて、試料または混合物は、供
与体を励起するが受容体分子を直接励起しない波長で照らされる。試料は次に2
種類の波長で監督される。それは供与体発光および受容体発光である。もし供与
体と受容体とが十分に接近していなければPRETは起こらず供与体波長でのみ
発光がおこる。もし供与体と受容体とが十分に接近していればFRETが起こる
。
この相互反応の結果、供与体の寿命の減少、供与体蛍光の消光、受容体蛍光強度
の増強および蛍光強度の偏光の偏りの度合いの減少がおこる。エネルギー転移の
効率、R4、は供与体および受容体分子間距離、Rの上昇に伴って急激に低下す
る。単離された供与体受容体対に関して、エネルギー転移の効率はE t=1/

グルコース濃度測定に関してFRETを使用する概念溶液中のグルコース濃度を
測定するためにFERTを使用する概念は、図2に表される。ひとつの巨大分子
(Mという)は多くの共有的−結合発蛍光団およびグルコース残基を含み、なら
びに糖複合体と呼ばれる。第二巨大分子(Lという)はグルコースに対し高度な
特異性を有するリガンド(例えばコンカナバリン^)および通常は第−巨大分子
上の発蛍光団と同一でない発蛍光団を含む。これらの発蛍光団のひとつは前述の
ように供与体、他の一つは受容体となるべぐ選択される。
この説明の目的のために、供与体分子は糖複合体上に配置され、受容体はリガン
ド上に配置される。その会合は次ぎのように図式される。
DMG+AL−−−−−−DMG−LADMGは供与体−巨大分子−グルコース
を、ALは受容体−リガントを表象し、およびDMG−LAは第一複合体中に存
在するグルコースと第二複合体に存在するりガントとの間の会合を表す。会合す
るとふたつの巨大分子は全供与体と受容体との間で十分に接近しエネルギー転移
が起こる。
遊離グルコースの存在は、遊離グルコースが複合体グルコースとりガントを競合
するために式に競合阻害を導入する。このようにグルコース濃度の増加は糖複合
体に結合するりガント量の減少を産する。比較的グルコース濃度が低い場合は少
量の巨大分子会合が起こるので転移効率は高く留まる。高濃度グルコースでは、
転移効率は低く、これはグルコースがうまく相補的巨大分子のりガントを落とす
。
以下の童で説明するように、グルコース濃度が正常個体およびグルコース恒常性
が変化した個体中(例えば糖尿病および低血糖症)に典型的に見いだされるグル
コース濃度を含有する試料のグルコースの、信頼性のある反復可能な測定を得る
ことが可能であることが示された。さらに使用された反応物(即ち蛍光的−標識
リガントおよび糖複合体)は安定で再使用できる。
ハンクス緩衝化塩溶液(HBSS)中の種々のグルコース1度に関してFRET
が測定される競合実験を行った。これらの実験は実施例3に詳細に説明されてい
る。スペクトルは472nmでの励起蛍光で集められ、発光を500−650n
rr+からスキャンニングした。典型的には、蛍光強度はフルオロセイン(約5
20nm)およびローダミン(約596nm)の発光最大で監督された。これら
の研究におけるエネルギー転移の測定は、グルコース濃度の関数として520n
mおよび596nmでの比(即ちFI520/FI596)がまたは520nm
でのフルオロセインの消光であった。
これらの試みの間多くの観察がなされ、それは本発明の方法が広範囲(即ち正常
、低血糖症及び糖尿病患者に見いだされるグルコース濃度)にわたって信頼性の
あるグルコース検出手段を提供することを示している。最終的に化合物は2−4
週間にわたって室温で安定であった。図3では化合物がフルオロセインの同一の
性質を二週間の期間にわたってグルコース濃度に対応し示した。グラフ中のデー
タは520nmにおけるフルオロセイン強度の変化をOmMからのグルコース濃
度に反応して反映する。
第二にFRETで測定されたグルコース濃度は他のメーターを使用しての測定と
一致する。二重盲検において、FRETは50−300mg/dLの範囲テDi
rect30/30グルコメーターを使用して測定したグルコース濃度の±10
%内のグルコース濃度を予言できた。
加えて、図4はFRET法はグルコース濃度を正確に31mMグルコース(〜6
00mg/dL)まで予言することができ、所望する範囲内およびそれ以上であ
る。正常グルコース濃度は通常80から120mg/dLの間で、ならびに糖尿
病レベルでは500mg/dLを越えることができる。Oがら31mMのグルコ
ース範囲では、FRETは(r2)が0.983の測定係数である直線に近い。
それゆえにグルコース濃度の感知するセンサーは全生理範囲の個体わたって信頼
性がある(即ち正常及び低血糖症(糖尿病))。
本発明の構成要素は再使用できることが分かった。FBGおよびRCは8−11
kDカツトオフ透析チユーブに置かれ、溶液研究に従って正確に測定されたこれ
ら、血清およびFRETの異なるグルコース濃度に対して透析された。HBSS
中のFBGおよびRCの初期容量は2mlであり、50rr+]容量の種々のグ
ルコース濃度に対して透析された。一般的に、F l520/F I、。。比に
おいてプラトゥレベルに達するまでに約30分を要した。図5に示されるように
、透析チューブを種々のグルコース濃度に変えた時反応は可逆的である。ウマ血
清において反応は透析構成要素により影響されなかったが、光分散のためベース
ラインにソフトしなかった。
本発明の方法は患者から適切に得られた大きさの試料中のグルコースを感知また
は定量するために使用できる(例えば10−100μm)。正常ヒト血液のこれ
ら微量試料を使用して行う評価では、グルコース濃度は本質的に同時に行われた
従来法のグルコースメーターでの測定と同じであることが分かった。
本方法の低血糖症のグルコース濃度を測定するための能力(則ち糖尿病患者で観
察されたグルコースレベルを作るためにグルコースを増された正常血液試料)も
評価された。図6はこの実験の生データ−スキャンを表す。第一(図6A)はグ
ルコースを含有しない、472nmで励起された試料の500nmからの発光ス
キャンである。これは二つの発光ピークの比による証拠からかなりのエネルギー
転移を表す。ローダミン(第二)ピークは実際フルオロセイン(第一)ピークよ
りも高い。
図6Bに描かれた第ニスキャンでは、グルコースの低血糖レベル、ここでは31
7mg/dL、の血液試料に対する透析効果を表す。エネルギー転移はフルオロ
セイン発光ピークの増加および同時に起こる増感ローダミン発光の減少の双方が
見られる時に減少する。
本方法は血液中のグルコースレベルの評価に効果的であり、以下のような低血糖
範囲で感度がよい Rh−CanAおよびFBGの混合物]50μmを1500
μlの低血糖試料に対して15分間微透析した。図7は、初期のフルオルゼイン
/ローダミン蛍光比は16よりも少し上であり、このフルオルゼイン/ローダミ
ン蛍光比はグルコース濃度の関数として増加し、約40mMグルコースで飽和し
た。
本発明の方法に基づき、in vivoまたはin vitroで血液中のグル
コース濃度を検出する多くの用具を構成できる。これらの用具は、交換されるま
で長期間にわたり(例えば1力月以上)活性を保つ。
本発明のin viv□の態様では反応物(即ち蛍光的に標識されたりガントお
よび第二発蛍光団て標識された糖複合体)をグルコースと連絡させて配置しく例
えば接触)皮膚でのグルコースの測定に関する。反応物は皮膚の中、上または下
に置くことができる。別の方法として反応物を器官および血管(例えば静脈また
は動脈)に置くことができ、そこでそれらはグルコースと連絡して、次に本方法
により測定できる。皮膚の中、上または下に位置する態様では、グルコースは皮
膚を供与体励起波長で照らし、二つの蛍光物質について波長を監督すことにより
検出される。例えば、蛍光物質がフルオルセインおよびローダミンであれば、蛍
光強度は520nmおよび596nm(即ちそれぞれの発光最大波長)で監督さ
れる。
蛍光光度計により検出されたエネルギー転移の測定は、次に二つの発光最大波長
(例えばFI520/FI596)での蛍光強度の比、またはグルコース濃度の
関数としての発光最大での供与体(即ちフルオロセイン)蛍光の消光である。
グルコースと連絡して反応物を置く種々の態様は本発明の胡」1四態様の原理に
従って採用できる。反応物は反応物を所望する場所に保持する任意の型の支持お
よび包囲物質中で体内に導入され、グルコースが測定できる(即ち本発明で測定
できるその濃度)ように接触または連絡させることができる。例えば、反応物は
微透析管、または内径的1mmおよび壁厚5O−1008mで末端密閉された球
にカプセル化できる。カプセル化されたグルコースセンサーは体内の任意の皮肉
に取り付けられる。もうひとつの態様としては。反応物はシリコーンまたはフル
オロカーボン油と混合して皮下に注入できる。反応物はまた皮膚上に刻み込まれ
たり(tattooed)または経皮肉経路に含有されつる。択一的に、反応物
は皮下料またはすでに体から分取るされた体液(例えば尿、細胞外液)と連絡さ
せて反応物を設置することに関する。グルコースは蛍光光度計中に反応物をグル
コース含有体液と連絡させて置くことにより検出および定量される。
反応物をグルコースと連絡させて配置する種々の様式は、本発明のil vit
ro態様に従って採用できる。例えば、反応物は固体基質に付く (例えば棒)
か、または容器(例えば微透析管)中に含有され得る。反応物を、適当量の反応
物を血液またはグルコースを含有する他の体液基質中で消耗させるペンカートリ
ッジ内にも含有できる。
合本発明は以下の実施例で説明されるが、いずれにしても限定とはされない。
伊I FBGおよびRCの
シグマ(Sigma)から購入したフルオロセインーBS^−グルコース(FB
G)およびモレキュラープローブ(Ilolecular Probes)から
購入したローダミン−CanA(RC)をギブコ(Gibco)からのハンクス
緩衝化塩溶液(HBSS)に最終濃度2■g/腸lで溶解した。溶液を次に10
. OQOgで30分遠心し大粒子を除去した。上澄みを次に回収し10゜00
0M夏アミコン限界濾過容器に置き、限界濾過が膜を通過するまで2.000g
で遠心した。残った物質を2璽1sのHBSSに再溶解し、この手順を限界濾過
物中に遊離のフルオロセインまたはローダミンが検出されなくなるまで繰り返し
た。
最終的に残った物質を回収し2厘10HBSSに再溶解し、10.000gで3
0分間もう一度遠心した。上澄みを回収し、エネルギー転移実験に使用するまで
4℃で保存した。

実施例2 フルオロセインーBS^−グルコースおよびローダミン−ConAの
至適filJFの決定
マンノースおよびグルコースで競合実験を行う前に、フルオロセインーBSへ−
グルコース(FBG)およびローダミン−ConA(RC)の至適濃度を決定し
た。
エネルギー転移に使用する任意の溶液中において、FRETが単に分子同士が溶
液中で相互に十分に接近することによって起こることは避けなければならない。
このように、FRET溶液実験は理想的には1mM以下の濃度で行われるべきで
ある、なぜならその濃度では分子は120八単位離れているからである(特異的
に反応するに十分能れている)。 逆に、これらの実験で、蛍光の濃度は検出さ
れ得るに十分高くなければならず、ならびにふたつの種(FBGおよびRC)は
それらの結合定数(K、S)より上でなければならない。
FBGおよびRCの初期濃度は約2 mg/mlであった。最大転移はFBGの
最終濃度が約2μg/mlおよびRCの最終濃度が約150 ug/x1の時起
こった。FRETの至適化は蛍光フルオロセインの消光により測定され、これは
30 pg/mlで50%最これらの実験に使用した分光蛍光計は、温度台を装
備したパーキン−エルマー41PF−2(Perkin−Elmer MPF−
2)で、アップルIl[Eコンピュータに接続した。
コンカナバリンA、 (ConA)はグルコースに特異的に結合するレクチンで
ある。グルコースの連続的希釈は約2μg/IllのFBGおよび約150μg
/111のRCを使用して、HBSS中で0から5iグルコースである。グルコ
ースの濃縮物である種々のグルコース濃度が増加すると、520nmでの蛍光強
度は増加し、グルコースに対する1ノ′2最大反応は約1.87mg/ロ1であ
った。

- 48.個体中のグルコース濃度を非−侵襲的に監督するセンサーであって、第一 光−吸収物質で標識されたグルコースー結合リガンドである第一部材および第二 光−吸収物質で標識された糖複合体である第二部材から成る特異的結合対から成 り、 第一光−吸収物質の励起状態エネルギーレベルは第二光−吸収物質の励起状態エ ネルギーレベルと重複し、 上記リガンドおよび上記糖複合体は、個体体液中に見い出されるグルコースの生 理的濃度範囲で互いに可逆的に結合し、個体の体液中に存在するグルコースが該 糖複合体と置換して可逆的に該リガンドと結合できるようにし、該センサーの構 成要素は、いったん個体中に設置されると、該センサーは個体の皮膚には存在せ ずに個体の体液中のグルコース濃度を非−侵襲的に監督できるよう構成されてい る、センサー。
- 49.上記特異的結合対の第一部材は蛍光的に標識されたレクチンであり、およ び上記特異的結合対の第二部材は蛍光的に標識された糖複合体である請求項48 記載のセンサー。
- 50.上記特異的結合対の第一部材はローダミン−標識コンカナバリンAであり 、および上記特異的結合対の第二部材はグルコースー結合リガンドに結合できる 蛍光−標識グリコシル化血清アルブミンである請求項48記載のセンサー。
