蛍光光度計を皮膚の近くに保持し、センサに位置合わせすることによって測定値を取得する。センサ内で生成された蛍光信号を経皮的に測定するとき、皮膚が信号を吸収することを考慮に入れることが必要である。実験により、人間の皮膚の吸収率は、400nm〜900nmの範囲で最も低いことがわかっている。得られる最終出力値は、2つの蛍光色素からの蛍光強度の正規化した比であり、以下の関係式(式1)で定義される。
ただし、Cはセンサ内の化学種のモル数をセンサ容積で割った商を表す。この濃度の尺度を用いると、固定された化学種および溶液中の化学種は同じように扱われる。

Claims (10)
- ほ乳動物の体内に植え込まれると、マクロファージによって摂取され、かつ植込み部位から離れるように輸送され得る適切なサイズの複数の粒子を含む、分析物のin vivo測定用センサであって、各粒子が、外部光学的手段によって経皮的に検出可能または測定可能な光信号の読出し値を与えるアッセイ成分を含み、各粒子を生分解性ゲル中に閉じ込めてマクロファージによる摂取を妨げるところのセンサ。
- 前記粒子の寸法が、最大でも5ミクロン未満である、請求項1に記載のセンサ。
- 前記アッセイが結合アッセイである、請求項1または2に記載のセンサ。
- 前記結合アッセイが競合結合アッセイであり、その成分が、分析物結合剤および分析物類似体を含む、請求項3に記載のセンサ。
- 前記分析物類似体を第1発色団で標識し、前記分析物結合剤を第2発色団で標識し、前記第1発色団または第2発色団の発光スペクトルが、前記第2発色団または第1発色団の吸収スペクトルにそれぞれ重なり合う、請求項4に記載のセンサ。
- 前記結合剤が、抗体、Fabフラグメント、レクチン、ホルモン受容体、薬物受容体、アプタマーまたは分子鋳型ポリマーである、請求項4または5に記載のセンサ。
- 前記検出可能または測定可能な光信号が、蛍光共鳴エネルギー移動、蛍光偏光、蛍光消光、リン光、発光増強、発光消光、回折またはプラズモン共鳴によって生成される、請求項1から6のいずれか1つに記載のセンサ。
- 前記粒子が生分解性でない、請求項1から7のいずれか1つに記載のセンサ。
- 前記分析物がブドウ糖である、請求項1から8のいずれか1つに記載のセンサ。
- ヒトを除くほ乳動物の皮膚内にセンサを植え込むことと、外部光学的手段を用いて分析物を経皮的に検出または測定することと、生分解性材料を分解させることと、マクロファージによって粒子を摂取させることと、マクロファージによって前記粒子を植込み部位から移動させることとを含む、請求項1から9のいずれか1つに記載のセンサを用いて分析物を検出する方法。
