Description
1. センサの応答時間が短くなるよう、生物分解性材料が、速やかな透過を許さなければならない。これは、生物分解性材料がグルコースに対して高い透過率を有している場合に、達成可能である。
2. グルコースがアッセイ成分に接触すべく生物分解性材料を通って拡散できるが、アッセイ成分が生物分解性材料を通って拡散して体内を自由に動き回ることがないよう、生物分解性材料が、分子量遮断特性を有していなければならない。
・その読み取りが当該センサが体内に埋め込まれたときに外部の光学的手段によって経皮的に照会できる検出可能な光信号であるグルコースのためのアッセイ成分、および
・分析対象の前記アッセイ成分への接触を許容しつつ、前記アッセイ成分を包んでいる生物分解性材料の殻
を有しており、
前記生物分解性材料が、疎水性ユニットと親水性ユニットとを有するコポリマーを含んでいるセンサを提供する。

Claims (23)
- グルコースを生体内で検出するためのセンサであって、
・その読み取りが当該センサが体内に埋め込まれたときに外部の光学的手段によって経皮的に照会できる検出可能な光信号であるグルコースのためのアッセイ成分、および
・分析対象の前記アッセイ成分への接触を許容しつつ、前記アッセイ成分を包んでいる生物分解性材料の殻
を有しており、
前記生物分解性材料が、疎水性ユニットと親水性ユニットとを有するコポリマーを含んでおり、該親水性ユニットのそれぞれが、ポリエチレングリコールと二価の酸とのエステルを含んでいるセンサ。 - 前記コポリマーが、ランダム・コポリマーである請求項1に記載のセンサ。
- 前記コポリマーが、25000Daを超えない分子量遮断限界を有している請求項1または2に記載のセンサ。
- 前記コポリマーが、10000Daを超えない分子量遮断限界を有している請求項3に記載のセンサ。
- 前記コポリマーが、少なくとも5.0×10−10cm2/sの透過率を有している請求項1〜4のいずれか1項に記載のセンサ。
- 前記疎水性ユニットの重量割合が、前記コポリマーの10〜90%である請求項1〜5のいずれか1項に記載のセンサ。
- 前記親水性ユニットが、二価の酸としてテレフタル酸および/またはコハク酸を含んでいる請求項1〜6のいずれか1項に記載のセンサ。
- 前記親水性ユニットが、二価の酸としてテレフタル酸のみを含んでいる請求項7に記載のセンサ。
- 前記親水性ユニットにおいて、コハク酸に対するテレフタル酸の比が、1:2〜2:1である請求項7に記載のセンサ。
- それぞれの親水性ユニットの分子量が、400〜4000である請求項1〜9のいずれか1項に記載のセンサ。
- 前記コポリマーの前記疎水性ユニットが、ブタン−1,4−ジオールと二価の酸とのエステルを含んでいる請求項1〜10のいずれか1項に記載のセンサ。
- 前記疎水性ユニットが、二価の酸としてテレフタル酸および/またはコハク酸を含んでいる請求項11に記載のセンサ。
- 前記疎水性ユニットが、二価の酸としてテレフタル酸のみを含んでいる請求項12に記載のセンサ。
- コハク酸に対するテレフタル酸の比が、1:2〜2:1である請求項12に記載のセンサ。
- 前記アッセイが、結合アッセイである請求項1〜14のいずれか1項に記載のセンサ。
- 前記結合アッセイが、競合結合アッセイであり、その成分が、グルコース結合剤およびグルコース類似物質を含んでいる請求項15に記載のセンサ。
- 前記グルコース類似物質が、第1の発色団でラベル付けされ、前記グルコース結合剤が、第2の発色団でラベル付けされ、第1の発色団または第2の発色団の放射スペクトルが、それぞれ第2の発色団または第1の発色団の吸収スペクトルと重なり合っている請求項16に記載のセンサ。
- 前記結合剤が、抗体、Fabフラグメント、レクチン、ホルモン受容体、薬物受容体、アプタマー、または分子インプリント・ポリマーである請求項16または17に記載のセンサ。
- 前記検出または測定可能な光信号が、蛍光共鳴エネルギー移動、蛍光偏光、蛍光消光、燐光、発光強調、発光消光、回折、またはプラズモン共鳴によって生成される請求項1〜18のいずれか1項に記載のセンサ。
- 請求項1〜19のいずれか1項に記載のセンサの製作方法。
- コアセルベーション、溶媒の気化および/または抽出、噴霧乾燥、噴霧コーティング、噴霧冷却、回転ディスク・アトマイゼーション、流動床コーティング、共押し出し、ならびに/あるいはパン・コーティングを含んでいる請求項20に記載の方法。
- 請求項1〜19のいずれか1項に記載のセンサを使用してグルコースを検出する方法であって、該センサを、ヒトを除く哺乳類の皮膚へと埋め込むこと、外部の光学的手段を使用して経皮的にグルコースを検出または測定すること、および前記生物分解性材料を分解することを含んでいる方法。
