Claims (21)
- 目的タンパク質;
エンカプスリンタンパク質;及び
RID(RNA interacting domain)タンパク質;をコーディングするポリヌクレオチドを含むことを特徴とする、ワクチン生産用組換え発現ベクター。 - 前記エンカプスリンタンパク質は、配列番号1のアミノ酸配列又はその一部配列を含むことを特徴とする、請求項1に記載のワクチン生産用組換え発現ベクター。
- 前記RIDタンパク質は、前記ワクチンの発現量又は水溶性を増加させるための融合パートナーであることを特徴とする、請求項1に記載のワクチン生産用組換え発現ベクター。
- 前記RIDタンパク質は、配列番号12、配列番号13又は配列番号14のアミノ酸配列又はその一部配列を含むことを特徴とする、請求項1に記載のワクチン生産用組換え発現ベクター。
- 前記目的タンパク質、エンカプスリンタンパク質及びRID(RNA interacting domain)タンパク質の間には、少なくとも一つのリンカータンパク質が位置し、
前記リンカータンパク質は、配列番号15、配列番号16、配列番号17、配列番号18、配列番号19又は配列番号20の遺伝子配列を含むことを特徴とする、請求項1に記載のワクチン生産用組換え発現ベクター。 - 前記ポリヌクレオチドは、RIDタンパク質の切断のためにTEVタンパク質を追加でコーディングすることを特徴とする、請求項1に記載のワクチン生産用組換え発現ベクター。
- 前記目的タンパク質は、ウイルス抗原タンパク質、抗体、細胞受容体、酵素、構造タンパク質、血清及び細胞タンパク質からなる群より選択されるいずれか一つのタンパク質であることを特徴とする、請求項1に記載のワクチン生産用組換え発現ベクター。
- 前記ウイルス抗原タンパク質は、ロタウイルス(Rotavirus)抗原タンパク質、子宮頸部癌ウイルス抗原タンパク質、デングウイルス(Dengue virus)抗原タンパク質及びコロナウイルス抗原タンパク質からなる群より選択されたいずれか一つのタンパク質であることを特徴とする、請求項7に記載のワクチン生産用組換え発現ベクター。
- 請求項1~8のうちいずれか一項に記載の発現ベクターに形質転換された宿主細胞。
- 前記宿主細胞は、大腸菌(E. coli)であることを特徴とする、請求項9に記載の形質転換された宿主細胞。
- 目的タンパク質;エンカプスリンタンパク質;及びRID(RNA interacting domain)タンパク質;を含むことを特徴とする、融合タンパク質。
- 前記目的タンパク質は、エンカプスリンタンパク質のC-末端に連結されることを特徴とする、請求項11に記載の融合タンパク質。
- 前記目的タンパク質、エンカプスリンタンパク質及びRID(RNA interacting domain)タンパク質の間には、少なくとも一つのリンカータンパク質が位置し、
前記リンカータンパク質は、配列番号15、配列番号16、配列番号17、配列番号18、配列番号19又は配列番号20の遺伝子配列を含むことを特徴とする、請求項11に記載の融合タンパク質。 - a)目的タンパク質;エンカプスリンタンパク質;及びRIDタンパク質;をコーディングするポリヌクレオチドを含むワクチン生産用組換え発現ベクターを生産する段階;
b)前記発現ベクターを宿主細胞に導入して形質転換体を生産する段階;及び
c)前記形質転換体を培養して組換え融合タンパク質の発現を誘導し、これを収得する段階;を含むことを特徴とする、ワクチン生産方法。 - 前記RIDタンパク質は、前記ワクチンの発現量又は水溶性を増加させるための融合パートナーであることを特徴とする、請求項14に記載のワクチン生産方法。
- 前記RIDタンパク質は、配列番号12、配列番号13又は配列番号14のアミノ酸配列又はその一部配列を含むことを特徴とする、請求項14に記載のワクチン生産方法。
- 前記宿主細胞は、大腸菌(E. coli)であることを特徴とする、請求項14に記載のワクチン生産方法。
- 前記エンカプスリンタンパク質は、配列番号1のアミノ酸配列又はその一部配列を含むことを特徴とする、請求項14に記載のワクチン生産方法。
- 前記目的タンパク質は、ウイルス抗原タンパク質、抗体、細胞受容体、酵素、構造タンパク質、血清及び細胞タンパク質からなる群より選択されたいずれか一つのタンパク質であることを特徴とする、請求項14に記載のワクチン生産方法。
- 前記ウイルス抗原タンパク質は、ロタウイルス(Rotavirus)抗原タンパク質、子宮頸部癌ウイルス抗原タンパク質、デングウイルス(Dengue virus)抗原タンパク質及びコロナウイルス抗原タンパク質からなる群より選択されたいずれか一つのタンパク質であることを特徴とする、請求項19に記載のワクチン生産方法。
