Abstract
Description
本出願は、2020年1月27日に出願された米国特許仮出願第62/966335号の優先権の利益を主張し、その全体が参照により本明細書に組み込まれる。
リパーゼ原液を、連続撹拌下で水を含有する容器に添加し、続いて2-メチルイミダゾール(2M)及び酢酸亜鉛無水物(dehydrate)(0.4M)を添加した。溶液は、0.25及び0.125mg/mLのリパーゼ、25~800mMの2-メチルイミダゾール、及び0.625~30mMの酢酸亜鉛二水和物の最終濃度を有した。室温でさらに24時間撹拌を続けた。溶液を6700×gで5分間遠心分離し、続いて上清を除去した。上清をBCAアッセイに使用した。再分散した(redispersed)ペレットを水で3回洗浄した後、リパーゼ特異的アッセイを用いてペレット及び上清について酵素活性を測定した。封入リパーゼの活性は49%であった。
リパーゼ原液を、連続撹拌下で水を含有する容器に添加し、続いて2,5-ジヒドロキシテレフタル酸(0.3M)及び酢酸亜鉛無水物(0.4M)を添加した。溶液は、0.25及び0.125mg/mLのリパーゼ、3~25mMの2,5-ジヒドロキシテレフタル酸、及び7~58.6mMの酢酸亜鉛無水物の最終濃度を有した。室温でさらに24時間撹拌を続けた。溶液を6700×gで5分間遠心分離し、続いて上清を除去した。上清をBCAアッセイに使用した。再分散したペレットを水で3回洗浄した後、リパーゼ特異的アッセイを用いてペレット及び上清について酵素活性を測定した。封入リパーゼの活性は33%であった。
リパーゼ原液を、連続撹拌下で水を含有する容器に添加し、続いてテレフタル酸(0.1M)及び硝酸アルミニウム九水和物(1.3M)を添加した。溶液は、0.25及び0.125mg/mLリパーゼ、9~44mMテレフタル酸、及び17~67mM硝酸アルミニウム九水和物の最終濃度を有した。室温でさらに24時間撹拌を続けた。溶液を6700×gで5分間遠心分離し、続いて上清を除去した。上清をBCAアッセイに使用した。再分散したペレットを水で3回洗浄した後、リパーゼ特異的アッセイを用いてペレット及び上清について酵素活性を測定した。封入リパーゼの活性は75%であった。
試験1:界面活性剤あり
プロテアーゼ及び(A)リパーゼ(封入されていない/純粋なリパーゼ)、(B)実施例1、(C)実施例2、及び(D)実施例3を含有する水性試験溶液を調製した。各試験溶液を半分に分けた。試験溶液の半分に界面活性剤混合物を加えた。リパーゼ活性の測定は、界面活性剤混合物の添加時に開始した。さらに、4時間、1日、2日、7日、14日、21日、及び28日目に活性を測定した。すべての溶液は0.05mg/mLリパーゼ、0.9mg/mLプロテアーゼ、及び20%界面活性剤(存在する場合)の最終濃度を有し、22℃又は37℃の温度に維持された。
プロテアーゼ及び(A)リパーゼ(封入されていない/純粋なリパーゼ)、(B)実施例1、(C)実施例2、及び(D)実施例3を含有する水性溶液を調製した。すべての溶液は0.05mg/mLのリパーゼ及び0.9mg/mLのプロテアーゼの濃度を有し、22℃の温度に維持された。リパーゼ活性の測定は、リパーゼ溶液へのプロテアーゼの添加時に開始した。さらに、4時間、1日、2日、7日、14日、21日、及び28日目に活性を測定した。
Claims (38)
- 内部容積を画定する構造体を含む金属-有機構造体(MOF)と、
内部容積内に配置された酵素と、
界面活性剤と
を含む組成物。 - 前記酵素がヒドロラーゼである、請求項1に記載の組成物。
- 前記酵素が、ペプチダーゼ、リパーゼ、グルコシダーゼ、又はそれらの2つ以上の組み合わせを含む、請求項1又は請求項2に記載の組成物。
- 前記界面活性剤が、非イオン性界面活性剤、アニオン性界面活性剤、カチオン性界面活性剤、双性イオン性界面活性剤、又はそれらの2つ以上の組み合わせを含む、請求項1~3のいずれか一項に記載の組成物。
- 前記非イオン性界面活性剤が、アルキルアルコキシル化アルコール、アルコールエトキシレート、アルコールプロポキシレート、プロポキシル化脂肪アルコール、アルコキシル化脂肪酸アルキルエステル、アルキルフェノールエトキシレート、ノニルフェノールエトキシレート、アルキルポリグリコシド、アルコキシル化アミン、脂肪酸モノエタノールアミド、脂肪酸ジエタノールアミド、エトキシル化脂肪酸モノエタノールアミド、プロポキシル化脂肪酸モノエタノールアミド、ポリヒドロキシアルキル脂肪酸アミド、グルコサミンのN-アシルN-アルキル誘導体、商品名SPAN及びTWEENで入手可能な製品、又はそれらの2つ以上の組み合わせを含む、請求項4に記載の組成物。
- 前記アニオン性界面活性剤が、サルフェート、スルホネート、又はそれらの組み合わせを含む、請求項4又は請求項5に記載の組成物。
- 前記スルホネートが、アルカン-2,3-ジイルビス(サルフェート)、アルキルサルフェート、又はそれらの組み合わせを含む、請求項6に記載の組成物。
- 前記サルフェートがアルコールエーテルサルフェートを含む、請求項7に記載の組成物。
- 前記スルホネートが、直鎖アルキルベンゼンスルホネート、アルキルベンゼンスルホネートの異性体、分岐アルキルベンゼンスルホネート、フェニルアルカンスルホネート、α-オレフィンスルホネート、オレフィンスルホネート、アルケンスルホネート、ヒドロキシアルカンスルホネート及びジスルホネート、第二級アルカンスルホネート、パラフィンスルホネート、エステルスルホネート、スルホン化脂肪酸グリセロールエステル、α-スルホ脂肪酸メチルエステル、アルキル-若しくはアルケニルコハク酸、ドデセニル/テトラデセニルコハク酸、アミノ酸の脂肪酸誘導体、スルホコハク酸若しくは石鹸のジエステル及びモノエステル、又はそれらの2つ以上の組み合わせを含む、請求項6~8のいずれか一項に記載の組成物。
- 前記スルホネートがアルキルベンゼンスルホネート(LAS)を含む、請求項9に記載の組成物。
- 前記カチオン性界面活性剤が、アルキルピリジニウム化合物、アルキル第四級アンモニウム化合物、アルキル第四級ホスホニウム化合物、アルキル第三級スルホニウム化合物、又はそれらの2つ以上の組み合わせを含む、請求項4~10のいずれか一項に記載の組成物。
- 前記双性イオン性界面活性剤が、アミンオキシド、ベタイン、又はそれらの2つ以上の組み合わせを含む、請求項4~11のいずれか一項に記載の組成物。
- 前記組成物が、約10重量%~約40重量%の前記界面活性剤を含む、請求項1~12のいずれか一項に記載の組成物。
- 前記組成物が、約20重量%~約30重量%の前記界面活性剤を含む、請求項1~13のいずれか一項に記載の組成物。
- 前記組成物が、0ppm超~約1500ppmの酵素を含む、請求項1~14のいずれか一項に記載の組成物。
- 前記組成物が、約40ppm~約900ppmの酵素を含む、請求項1~15のいずれか一項に記載の組成物。
- 前記組成物が、中性からアルカリ性のpHを有する、請求項1~16のいずれか一項に記載の組成物。
- 前記組成物が、約7~約9のpHを有する、請求項1~17のいずれか一項に記載の組成物。
- 前記酵素及び前記MOFが共有結合していない、請求項1~18のいずれか一項に記載の組成物。
- 前記構造体が孔隙を画定する、請求項1~19のいずれか一項に記載の組成物。
- 前記孔隙が、前記酵素を前記内部容積から逃がさないようにするサイズを有する、請求項20に記載の組成物。
- 前記孔隙が、前記酵素よりも低い分子量を有する化合物が前記内部容積に入ることを可能にするサイズを有する、請求項20又は21に記載の組成物。
- 前記化合物が、前記酵素よりも少なくとも約25%低い分子量を有する、請求項22に記載の組成物。
- 前記化合物が、前記酵素よりも少なくとも約50%低い分子量を有する、請求項22又は請求項23に記載の組成物。
- 前記化合物が、前記酵素よりも少なくとも約75%低い分子量を有する、請求項22~24のいずれか一項に記載の組成物。
- 前記MOFが、反応したMOF前駆体を含む、請求項1~25のいずれか一項に記載の組成物。
- 前記MOF前駆体が有機化合物及び金属塩を含む、請求項26に記載の組成物。
- 前記有機化合物が、イミダゾール基、トリアゾール基、又は少なくとも2つのカルボン酸基を有する化合物を含む、請求項27に記載の組成物。
- 前記有機化合物が、2-メチルイミダゾール、イミダゾレート-2-カルボキシアルデヒド、3-メチル-1,2,4-トリアゾール、1つ又は複数のヒドロキシル基又は複数のアミノ基で置換されていてもよいテレフタル酸、又はそれらの組み合わせを含む、請求項27又は請求項28に記載の組成物。
- 前記金属塩が、Zn、Al、Co、Fe、Sc、Ca、Mg、Mn、Li、Ni、Cu、Zr、又はそれらの2つ以上の組み合わせを含む、請求項27~29のいずれか一項に記載の組成物。
- 前記金属塩が、Zn(NO3)2、Zn(OAc)2、Al(NO)3、又はそれらの2つ以上の組合せを含む、請求項27~30のいずれか一項に記載の組成物。
- 前記MOFが、ZIF-8、MOF-74、MIL-53、MAF-7、ZIF-90、又はそれらの2つ以上の組み合わせを含む、請求項1~31のいずれか一項に記載の組成物。

- 前記MOF前駆体及び前記酵素を、水を含む容器に添加することを含む、請求項27~35のいずれか一項に記載の組成物を製造する方法。
- 前記MOF前駆体及び前記酵素を共沈させて、内部容積を画定する構造体を含むMOFを形成し、前記酵素を前記内部容積内に配置させる、請求項36に記載の方法。
- 前記界面活性剤を添加することをさらに含む、請求項37に記載の方法。
