Abstract
【課題】アンドロゲン受容体特異的オリゴヌクレオチドを細胞内に効率的に伝達する二本鎖オリゴヌクレオチド構造体及びナノ粒子、ならびに前記オリゴヌクレオチド構造体を含む脱毛予防又は発毛用組成物を提供する。【解決手段】二本鎖オリゴヌクレオチドの両末端に親水性物質及び疎水性物質が単純共有結合又はリンカー媒介(linker-mediated)共有結合で接合された形態の構造を有する二本鎖オリゴヌクレオチド構造体とする。また、水溶液で前記二本鎖オリゴヌクレオチド構造体が疎水性相互作用によって自己集合して生成され得るナノ粒子とする。【効果】本発明に係る二本鎖オリゴヌクレオチド構造体及びこれを有効成分として含む脱毛予防又は発毛用組成物は、副作用無しで高い効率でアンドロゲン受容体の発現を抑制させ、脱毛、特に、アンドロゲン性脱毛、円形脱毛、休止期脱毛の予防及び発毛誘導に優れた効果を示すことができる。【選択図】図6
Description
本発明は、アンドロゲン受容体特異的配列を含む二本鎖オリゴヌクレオチド構造体及びこれを含む脱毛予防及び発毛用組成物に関し、より詳しく、アンドロゲン受容体特異的配列のヌクレオチドを細胞内に効率的に伝達するために、二本鎖オリゴヌクレオチドの両末端に親水性物質及び疎水性物質が単純共有結合又はリンカー媒介(linker-mediated)共有結合で接合された形態の構造を有する二本鎖オリゴオリゴヌクレオチド構造体、水溶液で前記二本鎖オリゴオリゴヌクレオチド構造体が疎水性相互作用によって自己集合(self-assembling)して生成され得るナノ粒子、及び前記二本鎖オリゴヌクレオチド構造体を含む脱毛予防及び発毛用組成物に関する。
毛髪は、身体の保護と外見に重要な役割を担い、毛髪を管理する目的は、頭皮を保護し、健康な毛髪状態を維持し、外見を良くすることなどにある。脱毛は、成長周期によって成長を止めた毛髪が自然に抜けることを指し、一般に、深刻な脱毛は主に男性に発生する遺伝的現象として認識されてきた。しかし、最近では、業務的ストレス、環境汚染、有害環境への露出、誤った食習慣によって脱毛が発生するなど、環境的要因の重要性が台頭しており、脱毛症は、毛髪が存在すべき部位に毛髪がない状態を意味する疾患として認識されることになった。脱毛症は、毛嚢が破壊され、線維組織として回復されて永久的な脱毛状態となる瘢痕性脱毛症と、組織が線維化せず、毛嚢もそのまま保存される非瘢痕性脱毛症とに分類され、非瘢痕性脱毛症には、休止期脱毛症、遺伝性アンドロゲン脱毛症、円形脱毛症、生長期脱毛症がある。
毛髪は、経時的に成長期(Growing stage)、衰退期(Degenerating stage)、休止期(Resting stage)、脱落期(Exogen stage)などの周期を有し、これを毛周期という。成長期の寿命は通常2~8年と、全体毛髪の約90%を占め、毛乳頭と接触する毛口の下半部に毛母細胞の分裂が持続して髪の毛が生成される。成長期が過ぎるとしばらくの間成長を止める時期が来るが、これを衰退期という。これは、毛髪の生成と発育が止まる休止期に移る時期であり、毛髪の根にも変化が起き、毛母細胞と色素細胞の活動が止めながらケラチンが生成されず、毛髪の成長が中断される。休止期には毛口部が収縮し、脱落期になってこそ毛髪が抜けるが、この時期にはタンパク質分解酵素が関与すると知られている。毛髪の成長を調節する因子としては、アンドロゲン、エストロゲン、甲状腺ホルモン、ステロイド、プロラクチン及び成長ホルモンなどが関与すると考えられており、中でもアンドロゲンが最も重要な調節因子として知られている。ホルモンが脱毛に関与するという最も一般的な例は、出産後の一時的な脱毛であり、妊娠をするとエストロゲンが増加し、毛周期が生長期から休止期に進行することを抑制するが、出産後にエストロゲンが急激に減少しながら休止期への進行が加速化し、休止期脱毛が発生する。すなわち、このようにホルモン依存の脱毛症があるが、その他にも脱毛の原因は、遺伝的要因、男性ホルモン、老化、血液循環障害、ストレス、スーパオキシドラジカル(superoxide radical)などを選ぶことができ、このような原因に従ってその対応策が変わり得る。男性ホルモンが原因である脱毛に対しては、DHT抑制剤(DHT blockers)を治療剤として使用しており、該抑制剤の基本的な機序は、5-アルファ還元酵素(5-α-reductase)により、テストステロンが活性度の高いジヒドロテストステロン(Dihydrotestosterone;以下、DHT)に転換されることを防ぐ役割を担う。一方、DHTはテストステロンに比べてアンドロゲン受容体(AR)との結合能力が5倍以上高いため、毛嚢のタンパク質合成を遅延させてDHTの過剰生成を防ぎ、アンドロゲン受容体への結合を遮断する物質を治療剤として使用している(Dallob AL et al.,1994.J.Clin.Endocrinol.Metab.79,703-709;Ellsworth,K and Harris G.,1995,Biochem.Biophys.Res.Commun.215,774-780;Kaufman KD.,2002.Mol and Cell Endocrinology.198,85-89)。
1942年にHamiltonが脱毛と男性ホルモンとの関係を明らかにしたが、アンドロゲン性脱毛(Androgenetic alopecia,AGA)は、毛根細胞内に存在するテストステロン(Testosterone)が、強力な代謝体であるDHTに転換され、DHT(Dihydrotestosterone)が毛嚢のアンドロゲン受容体(Androgen receptor,AR)と結合して細胞内代謝を活性化させるアデニルシクラーゼ(Adenyl cyclase)の活性を抑制することによって、細胞内のcAMPの濃度を下げて糖代謝を低下させ、その結果としてエネルギー供給が阻害されてタンパク質合成が遅れることによって毛嚢の成長期が短縮し、このような現象が反復される過程で休止期の毛嚢の比率が増加し、次第に細くて短い毛髪が生えてしまう。すなわち、アンドロゲン性脱毛の発生には、毛根細胞内に存在するテストステロンとアンドロゲン受容体の過剰発現に関連したホルモン成分であるDHT受容体及び5-アルファ還元酵素(5-α-reductase)の活性が重要な因子とされており、テストステロンが5-アルファ還元酵素(5-α-reductase)によってDHT(Dihydrotestosterone)に過剰生産され、このような代謝物が毛髪周期阻害剤の生産を刺激することにより、成長期が短縮し且つ毛嚢の毛髪生成能力を抑制させると知られている(Kaufman KD.,2002.Mol and Cell Endocrinology.198,89-85;Naito et al.,2008.Br.J.Dermatol.159,300~305)。
テストステロンと比較したとき、DHTは、アンドロゲン受容体(AR)との結合力が5倍以上高いものと知られており、アンドロゲンに特異的な細胞及び組織では、DHTがテストステロンに比べてアンドロゲン活性に大きく関与することが知られている。このような代謝過程を担当する5-アルファ還元酵素(5-α-reductase)は、2種類のサブタイプ(subtype)が存在し、組織によってその役割が多少異なるが、Type1の5-アルファ還元酵素(5-α-reductase)は皮脂腺(sebaceous gland)に存在し、Type2の5-アルファ還元酵素(5-α-reductase)は、泌尿生殖管(genitourinary tract)と毛嚢(hair follicles)などに主に存在する。
DHTの過剰生成を抑制するために、5-アルファ還元酵素(5-α-reductase)を標的に使用する薬物としてフィナステリド(Finasteride)とデュタステリド(Dutasteride)があり、フィナステリドはType2の5-アルファ還元酵素(5-α-reductase)にのみ作用し、デュタステリドはType1、2の5-アルファ還元酵素(5-α-reductase)に作用して、前立腺関連疾患への効果が大きいものと知られている。特に、中でも、ハゲ治療剤としてFDA承認を受けた薬物は、フィナステリドを主成分とするプロペシア(Propecia)である。現在まで開発された脱毛治療剤は主に単一化合物が主流であり、血行促進のためのミノキシジル(minoxidil)、男性ホルモン抑制物質としてフィナステリド、デュタステリドがあり、最近では、JAK阻害薬(JAK inhibitor)(ルキソリチニブ(ruxolitinib)、トファシチニブ(tofacitinib))に対する薬物がFDA承認を受けた。しかし、これらの物質よりも優れた効果を有する物質を探すための研究は継続されている。
アンドロゲン受容体(androgen receptor)は、110KDaのステロイド性受容体であり、重要な機能の一つは、アンドロゲンに関連した遺伝子の転写である。アンドロゲン受容体は、前立腺癌、前立腺肥大症、男性型脱毛、筋肉消失及び多毛症のような男性ホルモンに関連した疾患において重要な役割を担い、このような理由から、前立腺癌と男性型ハゲのように男性にのみ現れる疾患の治療に標的として用いられた。アンドロゲンと総称される男性ホルモンの場合、テストステロンは脳下垂体、副腎、睾丸などで生成され、標的臓器の細胞内に入ってテストステロン5-アルファ還元酵素(5-α-reductase)によってジヒドロテストステロン(DHT)に還元されてから受容体と結合し、アンドロゲンとしての作用を示す。したがって、上述したように、テストステロンをDHTに還元する5-アルファ還元酵素(5-α-reductase)の作用を阻害することによってDHTを生成することを抑制する方法と、テストステロンから生成されたDHTが受容体と結合することを阻害することによってアンドロゲンの作用を抑制する方法により当該疾患に対する治療剤開発が摸索されている。
遺伝子の発現を抑制する技術は、疾病治療のための治療剤開発及び標的検証において重要な道具である。この技術のうち、干渉RNA(RNA interference;以下、‘RNAi’という。)は、その役目が発見されて以来、種々の哺乳動物細胞(mammalian cell)において配列特異的mRNAに作用するという事実が明らかにされた(Silence of the transcripts:RNA interference in medicine.J Mol Med(2005)83:764-773)。長い鎖のRNA二本鎖が細胞に伝達されると、伝達されたRNA二本鎖はダイサー(Dicer)というエンドヌクレアーゼ(endonuclease)によって21~23個の二本鎖(base pair,bp)としてプロセシングされた短い干渉RNA(small interfering RNA;以下、‘siRNA’という。)に変換され、siRNAはRISC(RNA-induced silencing complex)に結合し、ガイド(アンチセンス)鎖がターゲットmRNAを認識して分解する過程によって、ターゲット遺伝子の発現を配列特異的に阻害する(NUCLEIC-ACID THERAPEUTICS:BASIC PRINCIPLES AND RECENT APPLICATIONS.Nature Reviews Drug Discovery.2002.1,503-514)。
バートランド(Bertrand)研究陣によれば、同じターゲット遺伝子に対するsiRNAがアンチセンスオリゴヌクレオチド(Antisense oligonucleotide,ASO)に比べて生体内/外(in vitro及びin vivo)でmRNA発現の阻害効果に優れ、当該効果が長く持続する効果を有することが明らかにされた(Comparison of antisense oligonucleotides and siRNAs in cell culture and in vivo.Biochem.Biophys.Res.Commun.2002.296:1000-1004)。また、siRNAの作用機序は、ターゲットmRNAと相補的に結合して配列特異的にターゲット遺伝子の発現を調節するので、既存の抗体ベースの医薬品又は化学物質医薬品(small molecule drug)に比べて、適用できる対象が画期的に拡大可能であるという長所を有する(Progress Towards in Vivo Use of siRNAs.MOLECULAR THERAPY.200613(4):664-670)。
siRNAの優れた効果及び様々な使用範囲にもかかわらず、siRNAが治療剤として開発されるためには、体内におけるsiRNAの安定性(stability)の改善及び細胞伝達効率の改善によってsiRNAがターゲット細胞に効果的に伝達されるようにする必要がある(Harnessing in vivo siRNA delivery for drug discovery and therapeutic development.Drug Discov Today.2006 Jan;11(1-2):67-73)。
上記の問題を解決するべく、体内安定性の改善のためにsiRNAの一部のヌクレオチド又は骨格(backbone)を核酸分解酵素抵抗性を有するように修飾(modification)したり、或いはウイルス性ベクター(viral vector)、リポソーム又はナノ粒子(nanoparticle)などの伝達体の利用などに関して活発に研究されている。
アデノウイルスやレトロウイルスなどのウイルス性ベクターを用いた伝達システムは形質移入効率(transfection efficacy)が高いが、免疫原性(immunogenicity)及び発癌性(oncogenicity)が高い。これに対し、ナノ粒子を含む非ウイルス性(non-viral)伝達システムは、ウイルス性伝達システムに比べて細胞伝達効率は低いが、生体内(in vivo)における安定性(stability)が高く、ターゲット特異的に伝達が可能であり、内包されているRNAiオリゴヌクレオチドを細胞又は組織に吸収(uptake)及び内在化(internalization)させるなどの改善された伝達効果が高いだけでなく、細胞毒性及び免疫誘発(immune stimulation)が殆どないという長所があり、現在は、ウイルス性伝達システムに比べて有力な伝達方法として評価されている(Nonviral delivery of synthetic siRNA s in vivo.J Clin Invest.2007 December 3;117(12):3623-3632)。
前記非ウイルス性伝達システムのうちナノ伝達体(nanocarrier)を利用する方法は、リポソーム、陽イオン高分子複合体などの様々な高分子を使用することによってナノ粒子を形成し、siRNAをこのようなナノ粒子(nanoparticle)、すなわち、ナノ伝達体(nanocarrier)に担持して細胞に伝達する形態を有する。ナノ伝達体を利用する方法の中で主に活用される方法には、高分子ナノ粒子(polymeric nanoparticle)、高分子ミセル(polymer micelle)、リポプレックス(lipoplex)などがあるが、このうち、リポプレックスを用いた方法は、陽イオン性脂質で構成され、細胞のエンドソーム(endosome)の陰イオン性脂質と相互作用してエンドソームの脱安定化効果を誘発して細胞内に伝達する役割を担う(Proc.Natl.Acad.Sci.15;93(21):11493-8,1996)。
siRNAの細胞内伝達効率性を向上させるために、siRNAに生体適合性高分子である親水性物質(例えば、ポリエチレングリコール(polyethylene glycol,PEG))を単純共有結合又はリンカー媒介(linker-mediated)共有結合で接合させたsiRNA接合体を用いて、siRNAの安定性確保及び効率的な細胞膜透過性のための技術が開発された(大韓民国登録特許第883471号)。しかし、siRNAの化学的修飾及びポリエチレングリコール(polyethylene glycol,PEG)を接合させること(PEGylation)だけでは生体内での低い安定性とターゲット臓器への伝達が円滑でないという短所が依然としてある。このような短所を解決するために、オリゴヌクレオチド、特に、siRNAのような二本鎖オリゴヌクレオチドに、親水性及び疎水性物質が結合した二本鎖オリゴヌクレオチド構造体が開発されたが、前記構造体は、疎水性物質の疎水性相互作用によってSAMiRNATM(self assembled micelle inhibitory RNA)と命名された自己集合(self-assembled)ナノ粒子を形成するが(大韓民国登録特許第1224828号)、SAMiRNATM技術は、既存の伝達技術に比べてサイズが非常に小さいながらも均一な(homogenous)ナノ粒子を得ることができるという長所を有する。
SAMiRNATM技術の具体的な例として、親水性物質としてPEG(polyethylene glycol)又はHEG(Hexaethylenglycol)が使用されるが、PEGは合成ポリマー(synthetic polymer)であり、通常、医薬品、特にタンパク質の水溶性(solubility)増加及び薬物動態学(pharmacokinetics)の調節のために用いられる。PEGは多分散系(polydisperse)物質であり、一バッチ(batch)のポリマーは、異なる個数の単量体(monomer)の総和からなって、分子量がガウス曲線形態を示し、多分散指数(polydisperse value,Mw/Mn)で物質の同質性程度を表現する。すなわち、PEGが低い分子量(3~5kDa)のとき、約1.01の多分散指数を示し、高い分子量(20kDa)のとき、約1.2という高い多分散指数を示し、高い分子量であるほど物質の同質性が相対的に低い特徴を示す(F.M.Veronese.Peptide and protein PEGylation:a review of problems and solutions.Biomaterials(2001)22:405-417)。したがって、PEGを医薬品に結合させた場合、接合体にPEGの多分散的特徴が反映され、単一物質の検証が容易ではないという短所から、PEGの合成及び精製過程の改善によって低い多分散指数を有する物質を生産する趨勢であるが、特に、分子量の小さい物質にPEGを結合させた場合、結合が容易になされたかどうか確認し難い不具合があるなど、物質の多分散性特徴による問題点があった。(Francesco M.Veronese and Gianfranco Pasut.PEGylation,successful approach to drug delivery.DRUG DISCOVERY TODAY(2005)10(21):1451-1458)。
そのため、最近では、既存の自己集合ナノ粒子であるSAMiRNATM技術の改良された形態として、SAMiRNATMを構成する二本鎖ヌクレオチド構造体の親水性物質を一定の分子量を有する均一な1~15個の単量体(monomer)と必要によってリンカー(linker)を含む基本単位にブロック(block)化し、これを必要に応じて適切な個数で使用することによって、既存のSAMiRNATMに比べてより小さいサイズを有し、また、多分散性が画期的に改善された新しい形態の伝達体技術が開発された。
一方、脱毛に関連した世界市場規模は2024年には118億ドルに達する見込みであると報告(Grand View Research,Inc)され、米国男性の7人中の4人、中国男性の5人中の1人がハゲ頭であり、その原因の90%以上が男性型脱毛症(androgenetic alopecia)であると知られている。しかし、現在まで開発された大部分の脱毛治療剤がDHTと5-アルファ還元酵素(5-α-reductase)を標的にしており、アンドロゲンと直接の連関性を持つアンドロゲン受容体を標的にした治療剤及び発毛製品に対する開発はないことが確認された。
そこで、本発明者らは、アンドロゲンと直接の連関性を有するアンドロゲン受容体を標的にした発毛関連製品の開発のために鋭意努力した結果、特定アンドロゲン受容体特異的配列がアンドロゲン受容体の発現を効果的に阻害でき、これを含む二本鎖オリゴヌクレオチド構造体及びこれを含む組成物が脱毛予防又は発毛に優れた効果を示すことを確認し、本発明を完成するに至った。
本発明の目的は、アンドロゲン受容体特異的であり、非常に高い効率でその発現を阻害できる新規オリゴヌクレオチド配列及び該配列を毛根細胞に効果的に伝達するための二本鎖オリゴヌクレオチド構造体を提供することにある。
本発明の他の目的は、前記二本鎖オリゴヌクレオチド構造体を含むナノ粒子(nanoparticle)を提供することにある。
本発明のさらに他の目的は、前記新規オリゴヌクレオチド配列又は前記二本鎖オリゴヌクレオチド構造体を有効成分として含む脱毛予防又は発毛用医薬組成物を提供することにある。
本発明のさらに他の目的は、前記新規オリゴヌクレオチド配列又は前記二本鎖オリゴヌクレオチド構造体を有効成分として含む脱毛予防又は発毛用化粧料組成物を提供することにある。
前記目的を達成するために、本発明は、下記構造式(1)の構造を有する二本鎖オリゴヌクレオチド構造体を提供する。

前記構造式(1)で、Aは親水性物質、Bは疎水性物質、X及びYはそれぞれ独立に、単純共有結合又はリンカー媒介の共有結合を意味し、Rは、配列番号6、58、68、99、107、109、260、270、284、298、348、358、359及び434からなる群から選ばれるいずれか一つの配列を含むセンス鎖及びこれと相補的な配列を含むアンチセンス鎖を含むアンドロゲン受容体特異的なオリゴヌクレオチドを意味する。
本発明は、また、前記二本鎖オリゴヌクレオチド構造体を含むナノ粒子(nanoparticle)を提供する。
本発明は、また、前記二本鎖オリゴヌクレオチド構造体又はナノ粒子を有効成分として含む脱毛予防又は発毛用医薬組成物を提供する。
本発明は、また、前記二本鎖オリゴヌクレオチド構造体又はナノ粒子を有効成分として含む脱毛予防又は発毛用化粧料組成物を提供する。
本発明は、また、発毛が必要な客体に、本発明に係る構造体、ナノ粒子又は医薬組成物を投与したり、或いは発毛が必要な部位に、本発明に係る構造体、ナノ粒子又は医薬組成物を塗布する段階を含む脱毛治療方法を提供する。
本発明は、また、脱毛予防或いは発毛が必要な客体又は該当の部位に、本発明に係る構造体、ナノ粒子、又は化粧料組成物を投与又は塗布する段階を含む脱毛予防又は発毛促進方法を提供する。

特に断りのない限り、本明細書で使われる全ての技術的及び科学的用語は、本発明の属する技術の分野における熟練した専門家によって通常理解されるのと同じ意味を有する。一般に、本明細書における命名法及び以下に記述する実験方法は、本技術分野でよく知られており、通常用いられるものである。
本発明では、アンドロゲン受容体を標的にして発現を阻害できるオリゴヌクレオチドを選別するために、アンドロゲン受容体の全体に対して2-塩基スライディングウィンドウアルゴリズム(2-base sliding window algorithm)を適用して候補配列リストを選定し、その中から、他の遺伝子とRNA配列に対して同一性(identity)が15塩基以下である468個の候補配列を最終選別し、公知の先行文献(米国公開特許US2007-0141009)に開示された76個のsiRNA配列を含めて総544個のオリゴヌクレオチド配列を用いてアンドロゲン受容体阻害程度を実験した結果、特に効果の良い14種のオリゴヌクレオチドが選別できた。また、前記オリゴヌクレオチドを二本鎖オリゴヌクレオチド構造体として作製して細胞内伝達効率を増加させ、脱毛防止及び発毛効果を向上させることができた。
したがって、本発明は、一観点において、配列番号6、58、68、99、107、109、260、270、284、298、348、358、359及び434からなる群から選ばれるいずれか一つの配列を含むセンス鎖及びこれと相補的な配列を含むアンチセンス鎖を含むアンドロゲン受容体特異的な二本鎖オリゴヌクレオチドに関する。
本発明における二本鎖オリゴヌクレオチドは、一般的なRNAi(RNA interference)作用を有する全ての物質を含む概念であり、前記アンドロゲン受容体特異的配列にはアンドロゲン受容体特異的shRNA、ASOなども含まれることは、本発明の属する技術の分野における通常の知識を有する者には明らかである。siRNAを標的細胞内に伝達するための従来の方法は、依然として、細胞膜を通過して細胞内に伝達されること、及び細胞内でエンドソームから抜け出て細胞質に移動しながらその活性が減少し、生体内に存在する分解酵素によって容易に分解してしまう問題点が解決されずにいる。さらに他の形態であるアンチセンスオリゴであるDNAと標的mRNA分解のためのsiRNAを接合したDNA-RNAハイブリッド(hybrid)は、生体内で既存の二本鎖オリゴRNAに比べて安定的であり、DNA部分は標的タンパク質と結合できるアプタマー塩基配列を有するので、標的細胞内に効率的に伝達され、RNAがタンパク質として発現することを抑制するsiRNA塩基配列を有するので、標的細胞内の標的mRNAと結合して遺伝子発現を抑制する。このようなDNA-RNA混成粒子は生体物質だけで構成されるので、毒性がなく、体内に存在する核酸分解酵素であるDNaseとRNaseに対して大きい抵抗性を有することから、RNAiの新しい技術であるといえよう。
また、アンドロゲン受容体に対する特異性が維持される限り、前記配列番号1~配列番号468からなる群から選ばれるいずれか一つの配列を含むセンス鎖又はこれに相補的なアンチセンス鎖において、一つ以上の塩基が置換、欠失、又は挿入された配列を含むセンス鎖及びアンチセンス鎖を含むアンドロゲン受容体特異的siRNAも本発明の権利範囲に含まれることは、本発明の属する技術の分野における通常の知識を有する者には明らかである。
前記配列番号1~468は、ヒトアンドロゲン受容体特異的な配列であり、アンドロゲン受容体mRNAの他の部位には15塩基配列以下の相同性を有するRNAセンス鎖配列である(表2参照)。一方、配列番号469~544は、既存の特許(US2007-0141009)で知られたヒトアンドロゲン受容体特異的siRNA配列を表す(表3参照)。
本発明では、既存の特許で開示されたアンドロゲン受容体特異的オリゴヌクレオチド配列と細胞内の活性を比較した結果、優れた効率性を有しながらもヒトの他のmRNAと相同性が少ないRNA配列が発明できた。本発明に係るオリゴヌクレオチドは、好ましくは、配列番号6、58、68、99、107、109、260、270、284、298、348、358、359及び434からなる群から選ばれるいずれか一つの配列をセンス鎖として含むアンドロゲン受容体特異的オリゴヌクレオチドであり、より好ましくは、配列番号68又は109の配列をセンス鎖として含むアンドロゲン受容体特異的オリゴヌクレオチドである。
本発明に係るオリゴヌクレオチドのセンス鎖又はアンチセンス鎖は、19~31個のヌクレオチドからなることが好ましく、前記配列番号1~配列番号468から選ばれたいずれか一つの配列を含むセンス鎖及びこれに相補的なアンチセンス鎖を含む。
本発明で提供されるアンドロゲン受容体特異的オリゴヌクレオチドは、該当の遺伝子を暗号化するmRNAと相補的に結合し得るように設計された塩基配列を有するので、該当の遺伝子の発現を効果的に抑制できることが特徴である。また、前記オリゴヌクレオチドの3’末端に1つ又は2つ以上の非結合(unpaired)されたヌクレオチドを含む構造であるオーバーハング(overhang)を含むことができる。
また、前記オリゴヌクレオチドの生体内安定性向上のために、核酸分解酵素抵抗性の付与及び非特異的免疫反応の減少のための様々な修飾(modification)を含むことができる。前記オリゴヌクレオチドを構成する第1又は第2オリゴヌクレオチドの修飾は、1つ以上のヌクレオチドにおける糖構造の2’炭素位置で-OH基が-CH3(メチル)、-OCH3(メトキシ)、-NH2、-F(フッ素)、-O-2-メトキシエチル-O-プロピル(propyl)、-O-2-メチルチオエチル(methylthioethyl)、-O-3-アミノプロピル、-O-3-ジメチルアミノプロピル、-O-N-メチルアセトアミド又は-O-ジメチルアミドオキシエチルへの置換による修飾;ヌクレオチドにおける糖(sugar)構造内の酸素が硫黄に置換された修飾;又は、ヌクレオチド結合のホスホロチオエート(phosphorothioate)又はボラノホスフェート(boranophosphate)、メチルホスフェート(methyl phosphonate)結合への修飾から選ばれた1つ以上の修飾が組み合わされて用いられてよく、PNA(peptide nucleic acid)、LNA(locked nucleic acid)又はUNA(unlocked nucleic acid)形態への修飾も利用可能である(Ann.Rev.Med.55,61-652004;US5,660,985;US5,958,691;US6,531,584;US5,808,023;US6,326,358;US6,175,001;Bioorg.Med.Chem.Lett.14:1139-1143,2003;RNA,9:1034-1048,2003;Nucleic Acid Res.31:589-595,2003;Nucleic Acids Research,38(17)5761-5773,2010;Nucleic Acids Research,39(5)1823-1832,2011)。
本発明で提供されるアンドロゲン受容体特異的オリゴヌクレオチドは、該当の遺伝子の発現を阻害させる他に、該当のタンパク質の発現も顕著に阻害させる。
本発明の他の様態として、アンドロゲン受容体特異的二本鎖オリゴヌクレオチドの生体内への効率的な伝達及び安定性向上のために二本鎖オリゴヌクレオチドの両末端に親水性物質及び疎水性物質が接合された形態の接合体を提供する。
上記のように、二本鎖オリゴヌクレオチドに親水性物質及び疎水性物質が結合した二本鎖オリゴヌクレオチド接合体の場合、疎水性物質の疎水性相互作用によって自己集合ナノ粒子を形成するが(大韓民国特許登録番号第1224828号)、このようなナノ粒子は、体内への伝達効率及び体内における安定性に極めて優れるだけでなく、粒子サイズの均一性に優れ、品質管理(Quality control)がし易いので、薬物としての製造工程が簡単であるという長所がある。
したがって、本発明は、他の観点において、下記の構造式(1)の構造を有する二本鎖オリゴヌクレオチド構造体に関する。
A-X-R-Y-B 構造式(1)
前記構造式(1)で、Aは親水性物質、Bは疎水性物質、X及びYはそれぞれ独立に、単純共有結合又はリンカー媒介の共有結合を意味し、Rは、配列番号6、58、68、99、107、109、260、270、284、298、348、358、359及び434からなる群から選ばれるいずれか1つの配列を含むセンス鎖及びこれと相補的な配列を含むアンチセンス鎖を含むアンドロゲン受容体特異的なオリゴヌクレオチドを意味する。
より好ましくは、本発明に係るアンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体は、下記構造式(2)の構造を有する。
前記構造式(2)で、A、B、X及びYは、前記構造式(1)における定義と同一であり、Sはアンドロゲン受容体特異的オリゴヌクレオチドのセンス鎖、ASはアンドロゲン受容体特異的オリゴヌクレオチドのアンチセンス鎖を意味する。
より好ましくは、アンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体は、下記構造式(3)又は(4)の構造を有する。
前記構造式(3)及び構造式(4)で、A、B、S、AS、X及びYは、前記構造式(1)における定義と同一であり、5’及び3’は、アンドロゲン受容体特異的オリゴヌクレオチドセンス鎖の5’末端及び3’末端を意味する。
上記の構造式(1)~構造式(4)における前記アンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体は、アンチセンス鎖の5’末端にリン酸基(phosphate group)が1個~3個結合してよく、RNAの代わりにshRNAが使用されてもよいことは、本発明の属する技術の分野における通常の知識を有する者には明らかである。
前記構造式(1)~構造式(4)における親水性物質は、分子量が200~10,000である高分子物質が好ましく、より好ましくは、1,000~2,000である高分子物質である。例えば、親水性高分子物質には、ポリエチレングリコール、ポリビニルピロリドン、ポリオキサゾリンなどの非イオン性親水性高分子化合物を使用することが好ましいが、必ずしもこれに限定されない。
特に、構造式(1)~構造式(4)における親水性物質(A)は、下記構造式(5)又は構造式(6)のような形態の親水性物質ブロック(block)形態で利用可能であるが、このような親水性物質ブロックを必要によって適切な個数(構造式(5)又は構造式(6)におけるn)を使用することによって、一般合成高分子物質などを使用する場合に発生し得る多分散性による問題点を大きく改善することができる。

前記構造式(5)で、A’は親水性物質単量体(monomer)、Jはm個の親水性物質単量体間、又はm個の親水性物質単量体とsiRNAを互いに連結するリンカー、mは1~15の整数、nは1~10の整数を意味し、(A’m-J)又は(J-A’m)で表示される反復単位が、親水性物質ブロックの基本単位に該当する。
前記構造式(5)又は構造式(6)のような親水性物質ブロックを有する場合、本発明に係るアンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体は、下記構造式(7)又は構造式(8)のような構造を有することができる。

前記構造式(7)及び構造式(8)で、X、R、Y及びBは、構造式(1)における定義と同一であり、A’、J、m及びnは、構造式(5)及び構造式(6)における定義と同一である。
前記構造式(5)及び構造式(6)で、親水性物質単量体(A’)は、非イオン性親水性高分子の単量体のうち、本発明の目的に符合するものであればいずれも使用可能であり、好ましくは、表1に記載の化合物(1)~化合物(3)から選ばれた単量体、より好ましくは、化合物(1)の単量体が使用されてよく、化合物(1)において、Gは、好ましくはCH2、O、S及びNHから選ばれてよい。
特に、親水性物質単量体の中でも特に化合物(1)で表示される単量体は、様々な官能基を導入することができ、生体内親和性が良く、免疫反応を少なく誘導するなど、生体適合性(bio-compatibility)に優れる他にも、構造式(7)又は構造式(8)による構造体内に含まれたオリゴヌクレオチドの生体内安定性を増加させ、伝達効率を増加させることができるという長所を有し、本発明に係る構造体の製造に非常に適している。
前記構造式(5)~構造式(8)における親水性物質は、総分子量が1,000~2,000の範囲内であることが特に好ましい。したがって、例えば、構造式(7)及び構造式(8)において化合物(1)によるヘキサエチレングリコール(Hexaethylene glycol)、すなわち、GがOで、mが6である物質が用いられる場合、ヘキサエチレングリコールスぺーサ(spacer)の分子量が344であるから、反復回数(n)は3~5であることが好ましい。特に、本発明は、必要によって前記構造式(5)及び構造式(6)において(A’m-J)又は(J-A’m)で表示される親水性基の反復単位、すなわち、親水性物質ブロック(block)が、nと表示される適切な個数で使用されてよいことを特徴とする。前記各親水性物質ブロック内に含まれる親水性物質単量体であるAとリンカーであるJは、独立に、各親水性物質ブロック間に同一であってもよく、異なってもよい。すなわち、親水性物質ブロックが3個用いられる場合(n=3)、一番目のブロックには化合物(1)による親水性物質単量体が、二番目のブロックには化合物(2)による親水性物質単量体が、三番目ブロックには化合物(3)による親水性物質単量体が用いられるなど、全ての親水性物質ブロック別に異なる親水性物質単量体が用いられてもよく、全ての親水性物質ブロックに化合物(1)~化合物(3)による親水性物質単量体から選ばれたいずれか1つの親水性物質単量体が同一に用いられてもよい。同様に、親水性物質単量体の結合を媒介するリンカーも、各親水性物質ブロック別にいずれも同一のリンカーが用いられてもよく、各親水性物質ブロック別に異なるリンカーが用いられてもよい。また、親水性物質単量体の個数であるmも、各親水性物質ブロック間に同一であってもよく、異なってもよい。すなわち、一番目の親水性物質ブロックでは親水性物質単量体が3個連結(m=3)され、二番目の親水性物質ブロックでは親水性物質単量体が5個(m=5)、三番目の親水性物質ブロックでは親水性物質単量体が4個連結(m=4)されるなど、異なる個数の親水性物質単量体が用いられてもよく、全ての親水性物質ブロックで同一個数の親水性物質単量体が用いられてもよい。
また、本発明において、前記リンカー(J)は、PO3
-、SO3及びCO2からなる群から選ばれることが好ましいが、これに制限されるものではなく、使用される親水性物質の単量体などによって本発明の目的に符合するいかなるリンカーも使用可能であることは、通常の技術者には明らかである。
前記構造式(1)~構造式(4)、構造式(7)及び構造式(8)における疎水性物質(B)は、疎水性相互作用によって構造式(1)~構造式(4)、構造式(7)及び構造式(8)によるオリゴヌクレオチド構造体で構成されたナノ粒子を形成する役割を担う。前記疎水性物質は、分子量が250~1,000であることが好ましく、ステロイド(steroid)誘導体、グリセリド(glyceride)誘導体、グリセロールエーテル(glycerol ether)、ポリプロピレングリコール(polypropylene glycol)、C12~C50の不飽和又は飽和炭化水素(hydrocarbon)、ジアシルホスファチジルコリン(diacylphosphatidylcholine)、脂肪酸(fatty acid)、リン脂質(phospholipid)、リポポリアミン(lipopolyamine)などが用いられてよいが、これに制限されるものではなく、本発明の目的に符合するいかなる疎水性物質も使用可能であるという点は、本発明の属する技術の分野における通常の知識を有する者には明らかである。
前記ステロイド(steroid)誘導体は、コレステロール、コレスタノール、コール酸、ギ酸コレステロール、ギ酸コレスタニル及びコレステリルアミンからなる群であってよく、前記グリセリド誘導体は、モノ-、ジ-及びトリ-グリセリドなどから選ばれてよいが、このとき、グリセリドの脂肪酸は、C12~C50の不飽和又は飽和脂肪酸が好ましい。
特に、前記疎水性物質の中でも飽和又は不飽和炭化水素又はコレステロールが、本発明に係るオリゴヌクレオチド構造体の合成段階で容易に結合させることができるという長所から好ましく、C24炭化水素、特にジスルフィド結合(disulfide bond)を含む形態が最も好ましい。
前記疎水性物質は、親水性物質の反対側末端(distal end)に結合し、オリゴヌクレオチドのセンス鎖又はアンチセンス鎖のいずれの位置に結合しても構わない。
本発明に係る構造式(1)~構造式(4)、構造式(7)及び構造式(8)における親水性物質又は疎水性物質とアンドロゲン受容体特異的オリゴヌクレオチドは、単純共有結合又はリンカー媒介の共有結合(X又はY)によって結合する。前記共有結合を媒介するリンカーは、親水性物質又は疎水性物質とアンドロゲン受容体特異的オリゴヌクレオチドの末端で共有結合し、必要によって特定環境で分解可能な結合を提供する限り、特に限定されるものではない。したがって、前記リンカーは、本発明に係る二本鎖オリゴヌクレオチド構造体の製造過程中にアンドロゲン受容体特異的オリゴヌクレオチド及び/又は親水性物質(又は、疎水性物質)を活性化するために結合させるいかなる化合物も使用可能である。前記共有結合は、非分解性結合又は分解性結合のいずれであっても構わない。このとき、非分解性結合にはアミド結合又はリン酸化結合があり、分解性結合にはジスルフィド結合、酸分解性結合、エステル結合、アンヒドリド結合、生分解性結合又は酵素分解性結合などがあるが、これに限定されるものではない。
また、前記構造式(1)~構造式(4)、構造式(7)及び構造式(8)におけるR(又は、S及びAS)で表示されるアンドロゲン受容体特異的オリゴヌクレオチドは、アンドロゲン受容体のmRNAに特異的に結合できる特性を有する配列であればいずれも使用可能であり、好ましくは、本発明では、配列番号6、58、68、99、107、109、260、270、284、298、348、358、359及び434からなる群から選ばれるいずれか1つの配列を含むセンス鎖とそれに相補的な配列を含むアンチセンス鎖で構成される。
特に、本発明に係る前記構造式(1)~構造式(4)、構造式(7)及び構造式(8)に含まれるsiRNAは、好ましくは、配列番号6、58、68、99、107、109、260、270、284、298、348、358、359及び434からなる群から選ばれるいずれか1つの配列を含むセンス鎖とこれに相補的な配列を含むアンチセンス鎖からなるアンドロゲン受容体特異的オリゴヌクレオチドであってもよい。
本発明は、また、本発明に係るアンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体において、前記構造体内の親水性物質のオリゴヌクレオチドと結合した反対側末端部位にアミン基又はポリヒスチジン(polyhistidine)基がさらに導入されてよい。
これは、本発明に係るアンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体の伝達体の細胞内導入とエンドソーム脱出を容易にするためのものであり、既に量子ドット(Quantum dot)、デンドリマー(Dendrimer)、リポソーム(liposome)などの伝達体の細胞内導入とエンドソーム脱出を容易にするためにアミン基の導入とポリヒスチジン基が利用できるという点及びその効果が報告されたことがある。
具体的に、伝達体の末端或いは外側に修飾された1級アミン基は、生体内pHで陽性子化しながら負電荷を帯びる遺伝子と静電気的相互作用によって結合体を形成し、細胞内流入後にエンドソームの低いpHで緩衝効果を有する内部3級アミンによってエンドソームの脱出が容易になることによって、リソソーム分解から伝達体を保護できると知られており(高分子ベースのハイブリッド物質を用いた遺伝子伝達及び発現抑制。Polymer Sci.Technol.,Vol.23,No.3,pp254-259)、非必須アミノ酸の一つであるヒスチジンは、残基(-R)にイミダゾリン(pKa3 6.04)を有するので、エンドソームとリソソームにおいて緩衝能力(buffering capacity)を増加させる効果があり、リポソームをはじめとする非ウイルス性遺伝子伝達体(non-viral gene carrier)においてエンドソーム脱出効率を高めるためにヒスチジン修飾が利用可能であるという点が知られている(Novel histidine-conjugated galactosylated cationic liposomes for efficient hepatocyte selective gene transfer in human hepatoma HepG2 cells.J.Controlled Release 118,pp262-270)。
前記アミン基又はポリヒスチジン(polyhistidine)基は、1つ以上のリンカーを介して親水性物質又は親水性物質ブロックと連結されてよい。
本発明の構造式(1)による二本鎖オリゴヌクレオチド構造体の親水性物質にアミン基又はポリヒスチジン(polyhistidine)基が導入される場合には、構造式(7)のような構造を有することができる。

Pは、アミン基又はポリヒスチジン基を意味し、J1とJ2はリンカーであって、J1は及びJ2は、独立に、単純共有結合、PO3 -、SO3、CO2、C2-12アルキル、アルケニル、アルキニルから選ばれてよいが、これに限定されるものではなく、使用される親水性物質によって本発明の目的に符合するJ1とJ2はいかなるリンカーも使用可能であることは、通常の技術者には明らかである。
Pは、アミン基又はポリヒスチジン基を意味し、J1とJ2はリンカーであって、J1は及びJ2は、独立に、単純共有結合、PO3 -、SO3、CO2、C2-12アルキル、アルケニル、アルキニルから選ばれてよいが、これに限定されるものではなく、使用される親水性物質によって本発明の目的に符合するJ1とJ2はいかなるリンカーも使用可能であることは、通常の技術者には明らかである。
好ましくは、アミン基が導入された場合には、J2は単純共有結合又はPO3
-、J1はC6アルキルであることが好ましいが、これに限定されない。
また、ポリヒスチジン(polyhistidine)基が導入された場合には、構造式(9)では、J2は単純共有結合又はPO3
-、J1は化合物(4)が好ましいが、これに限定されない。
また、構造式(9)による二本鎖オリゴヌクレオチド構造体の親水性物質が、構造式(5)又は構造式(6)による親水性物質ブロックであり、これにアミン基又はポリヒスチジン(polyhistidine)基が導入される場合には、構造式(10)又は構造式(11)のような構造を有することができる。

前記構造式(10)及び構造式(11)で、X、R、Y、B、A’、J、m及びnは、構造式(5)又は構造式(6)における定義と同一であり、P、J1及びJ2は、構造式(9)における定義と同一である。
特に、前記構造式(10)及び構造式(11)において、親水性物質は、アンドロゲン受容体特異的オリゴヌクレオチドセンス鎖の3’末端に結合した形態であることが好ましく、この場合、前記構造式(9)~構造式(11)は、次の構造式(12)~構造式(14)の形態を有することができる。
前記構造式(12)~構造式(14)で、X、R、Y、B、A、A’、J、m、n、P、J1及びJ2は、前記構造式(9)~構造式(11)における定義と同一であり、5’及び3’は、アンドロゲン受容体特異的オリゴヌクレオチドセンス鎖の5’末端及び3’末端を意味する。
本発明で導入可能なアミン基としては、1級~3級アミン基が用いられてよく、1級アミン基が用いられることが特に好ましい。前記導入されたアミン基は、アミン塩として存在してもよいが、例えば、1級アミン基の塩は、NH3
+の形態で存在してよい。
また、本発明で導入可能なポリヒスチジン基は、3~10個のヒスチジンを含むことが好ましく、特に、好ましくは5~8個、最も好ましくは6個のヒスチジンを含むことができる。さらに、ヒスチジンの他に1つ以上のシステインも含まれてよい。
一方、本発明に係るアンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体及びこれから形成されたナノ粒子にターゲッティングモイエティが具備されると、効率的にターゲット細胞への伝達を促進するので、比較的低い濃度の投与量でもターゲット細胞に伝達され、高いターゲット遺伝子発現調節機能を示すことができ、他の臓器及び細胞への非特異的なアンドロゲン受容体特異的オリゴヌクレオチドの伝達を防止することができる。
これによって、本発明は、前記構造式(1)~構造式(4)、構造式(7)及び構造式(8)による構造体に、リガンド(L)、特に受容体媒介エンドサイトーシス(receptor-mediated endocytosis,RME)によりターゲット細胞内在化(internalization)を増進させる受容体と特異的に結合する特性を有するリガンド(ligand)がさらに結合した二本鎖オリゴヌクレオチド構造体を提供し、例えば、構造式(1)による二本鎖オリゴヌクレオチド構造体にリガンドが結合した形態は、下記構造式(15)のような構造を有する。

前記構造式(15)で、A、B、X及びYは、前記構造式(1)における定義と同一であり、Lは、受容体媒介エンドサイトーシス(receptor-mediated endocytosis,RME)によりターゲット細胞内在化(internalization)を増進させる受容体と特異的に結合する特性を有するリガンドを意味し、iは1~5の整数、好ましくは1~3の整数である。
前記構造式(15)におけるリガンドは、好ましくはターゲット細胞特異的に細胞内在化(internalization)を増進させるRME特性を有するターゲット受容体特異的抗体、アプタマー、ペプチド;又は、葉酸(Folate、一般に、folateとfolic acidが交差使用されており、本発明における葉酸は、自然状態又は人体で活性化状態であるfolateを意味する。)、N-アセチルガラクトサミン(N-acetyl Galactosamine,NAG)などのヘキソアミン(hexoamine)、ブドウ糖(glucose)、マンノース(mannose)をはじめとする糖又は炭水化物(carbohydrate)などの化学物質などから選ばれてよいが、これに限定されるものではない。
また、前記構造式(15)における親水性物質Aは、構造式(5)及び構造式(6)による親水性物質ブロックの形態で用いられてよい。
本発明のさらに他の様態として、アンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体を含むナノ粒子を提供する。
上述したように、アンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体は、疎水性及び親水性物質の両方を含んでいる両親媒性であり、親水性部分は、体内に存在する水分子と水素結合などの相互作用によって親和力を有しているので外側に向かうようになり、疎水性物質は、その同士間の疎水性相互作用(hydrophobic interaction)によって内側に向かうようになるので、熱力学的に安定したナノ粒子を形成する。すなわち、ナノ粒子の中心に疎水性物質が位置し、アンドロゲン受容体特異的オリゴヌクレオチドの外側の方向に親水性物質が位置することにより、アンドロゲン受容体特異的オリゴヌクレオチドを保護する形態のナノ粒子を形成する。このように形成されたナノ粒子は、アンドロゲン受容体特異的オリゴヌクレオチドの細胞内伝達及びオリゴヌクレオチド効能を向上させる。
本発明に係るナノ粒子は、同一の配列を有するオリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体だけで構成されてもよく、異なる配列を有するオリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体で構成されてもよいことを特徴とするが、本発明における異なる配列を有するオリゴヌクレオチドは、異なるターゲット遺伝子、例えば、アンドロゲン受容体特異的なオリゴヌクレオチドであってもよく、同一のターゲット遺伝子特異性を有しながらその配列が異なる場合も含まれると解釈される。
また、アンドロゲン受容体特異的なオリゴヌクレオチド以外に他の脱毛関連遺伝子特異的なsiRNAを含む二本鎖オリゴヌクレオチド構造体が本発明に係るナノ粒子に含まれてもよい。
本発明は、さらに他の観点として、アンドロゲン受容体特異的二本鎖オリゴヌクレオチド、これを含む二本鎖オリゴヌクレオチド構造体及び/又は前記二本鎖オリゴヌクレオチド構造体を含むナノ粒子を有効成分として含有する脱毛、特に、アンドロゲン性脱毛の予防又は発毛用医薬組成物に関する。
前記医薬組成物は、軟膏、ペースト、ゲル、ゼリー、セラム(serum)、エアゾールスプレー、非エアゾールスプレー、フォーム、クリーム、ローション、溶液又は懸濁液の剤形から選ばれる剤形に用いられることを特徴とし得るが、これに限定されない。
本発明に係る組成物は、テストステロンの代謝産物であるDHTがアンドロゲン受容体に結合する自体を抑制して脱毛予防又は発毛誘導に効果を示す。
本発明では、本発明に係る二本鎖オリゴヌクレオチド又はその構造体以外の、アンドロゲン受容体以外の他の脱毛疾患関連遺伝子に特異的な二本鎖オリゴヌクレオチド、又はこれを含む二本鎖オリゴヌクレオチド構造体が、本発明に係る組成物にさらに含まれてもよい。
本発明に係る組成物は、アンドロゲン受容体の上位又は下位信号伝達に関与する遺伝子に関連した脱毛、特に、アンドロゲン性脱毛に適用されてよいが、これに限定されるものではない。
本発明の組成物には、前記の有効成分の他に、さらに薬剤学的に許容可能な担体を1種以上含んで製造できる。薬剤学的に許容可能な担体は、本発明の有効成分と両立可能である必要があり、食塩水、滅菌水、リンゲル液、緩衝食塩水、デキストロース溶液、マルトデキストリン溶液、グリセロール、エタノール及びこれら成分のうち1成分又は2以上の成分を混合して使用することができ、必要によって、抗酸化剤、緩衝液、静菌剤などの他の通常の添加剤を添加することができる。また、希釈剤、分散剤、界面活性剤、結合剤及び潤滑剤をさらに添加し、水溶液、懸濁液、乳濁液などのような注射用剤形に製剤化できる。特に、凍結乾燥(lyophilized)した形態の剤形に製剤化して提供することが好ましい。凍結乾燥剤形の製造のために、本発明の属する技術の分野に一般に知られている方法が用いられてよく、凍結乾燥のための安定化剤が追加されてもよい。なお、当分野の適正な方法で又はレミングトン薬学科学(Remington’s pharmaceutical Science,Mack Publishing company,Easton PA)に開示されている方法を用いて、各疾病によって又は成分によって好ましく製剤化されてもよい。
本発明の組成物に含まれる有効成分などの含有量及び投与方法は、通常の個人の症候と脱毛の深刻度に基づき、本技術分野における通常の専門家が決定できる。また、散剤、錠剤、注射剤、軟膏剤などの様々な形態で製剤化でき、単位投与量又は多回投与量容器、例えば、密封したアンプル及び瓶などで提供されてよい。
本発明は、さらに他の観点において、アンドロゲン受容体特異的二本鎖オリゴヌクレオチド、これを含む二本鎖オリゴヌクレオチド構造体及び/又は前記二本鎖オリゴヌクレオチド構造体を含むナノ粒子を有効成分として含有する脱毛、特に、アンドロゲン性脱毛の予防又は発毛用化粧料組成物に関する。
前記組成物は、これに限定されないが、ヘアトニック、ヘアコンディショナー、ヘアエッセンス、ヘアローション、ヘア栄養ローション、ヘアシャンプー、ヘアリンス、ヘアトリートメント、ヘアクリーム、ヘア栄養クリーム、ヘアモイスチャークリーム、ヘアマッサージクリーム、ヘアワックス、ヘアエアゾール、ヘアパック、ヘア栄養パック、ヘア石鹸、ヘアクレンジングフォーム、ヘアオイル、毛髪乾燥剤、毛髪保存処理剤、毛髪染色剤、毛髪用ウェーブ剤、毛髪脱色剤、ヘアゲル、ヘアグレーズ、ヘアドレッシング、ヘアラッカー、ヘアモイスチャーライザー、ヘアムース又はヘアスプレーの剤形から選ばれる剤形に用いられることを特徴とし得る。
本発明は、さらに他の観点において、発毛が必要な客体に本発明に係る構造体、ナノ粒子又は医薬組成物を投与したり、或いは発毛が必要な部位に本発明に係る構造体、ナノ粒子又は医薬組成物を塗布する段階を含む脱毛治療方法に関する。
本発明は、また、脱毛予防或いは発毛が必要な客体又は該当の部位に、本発明に係る構造体、ナノ粒子又は化粧料組成物を投与又は塗布する段階を含む脱毛予防又は発毛促進方法に関する。
本発明は、また、脱毛予防又は発毛を促進するための前記二本鎖オリゴヌクレオチド構造体の用途に関する。
本発明は、また、脱毛予防又は発毛を促進するための治療剤(medicine)又は化粧料製造のための前記二本鎖オリゴヌクレオチド構造体の用途に関する。
本発明において、脱毛は、アンドロゲン性脱毛、円形脱毛、休止期脱毛をいずれも含む。
以下、実施例を挙げて本発明をより詳細に説明する。これらの実施例は単に本発明をより具体的に説明するためのものであり、本発明の範囲がこれらの実施例によって制限されないということは、当該技術の分野における通常の知識を有する者にとって明らかである。
実施例1.アンドロゲン受容体をターゲットとするオリゴヌクレオチドスクリーニングのためのアルゴリズムと候補配列選定
siRNA基礎治療剤高速大量スクリーニング(siRNA based drug high-throughput screening)は、mRNA全体に対して1-塩基(1-base)又は2-塩基(2-base)スライディングウィンドウアルゴリズム(sliding window algorithm)を適用して可能な全ての候補配列を生成し、相同性フィルタリング(homology filtering)を行って不要な候補配列を除去し、最終的に選別された全てのオリゴヌクレオチドに対して該当の遺伝子発現阻害程度を確認する方法である。
siRNA基礎治療剤高速大量スクリーニング(siRNA based drug high-throughput screening)は、mRNA全体に対して1-塩基(1-base)又は2-塩基(2-base)スライディングウィンドウアルゴリズム(sliding window algorithm)を適用して可能な全ての候補配列を生成し、相同性フィルタリング(homology filtering)を行って不要な候補配列を除去し、最終的に選別された全てのオリゴヌクレオチドに対して該当の遺伝子発現阻害程度を確認する方法である。
まず、アンドロゲン受容体に対するオリゴヌクレオチド候補配列に対するデザイン過程は、ヒトアンドロゲン受容体mRNAであるNM_000044.3(isoform 1、10,661bp)、NM_001011645.2(isoform 2、8112bp)のエクソン地図(exon map)を確認して同型タンパク質共通部位(isoform common region)を抽出し、抽出した同型タンパク質共通部位(isoform common region)に対して2-塩基スライディングウィンドウアルゴリズム(2-base sliding window algorithm)を適用して、19個の塩基で構成された3,956個の候補配列を選別した。
選別されたオリゴヌクレオチド候補配列リストをヒト総参考配列RNA(human total reference seq RNA)に対するBLAST e-value 100以下で行い、他の遺伝子とRNA配列に対して同一性(identity)が15塩基以下である468個の候補配列を最終選別した。このとき、既に公知された先行文献(米国公開特許US2007-0141009)で言及された76個のsiRNA配列を含めて総544個のオリゴヌクレオチド配列を用いてアンドロゲン受容体阻害程度実験を行った。
実施例2.二本鎖オリゴヌクレオチド構造体の合成
本発明で製造された二本鎖オリゴヌクレオチド構造体(SAMiRNA)は、下記構造式のような構造を有する。
本発明で製造された二本鎖オリゴヌクレオチド構造体(SAMiRNA)は、下記構造式のような構造を有する。
合成過程は、ヌクレオシド(nucleoside)が結合した固形支持体(CPG)上から始めて遮断除去(deblocking)、結合(coupling)、キャッピング(capping)及び酸化(oxidation)からなるサイクルを反復することによって、所望の配列のRNA一本鎖を得る。当該一連の二本鎖オリゴRNAの合成過程には、RNA合成器(384 synthesizer,BIONEER、韓国)を用いた。
二本鎖オリゴヌクレオチド構造体のセンス鎖は、ポリエチレングリコール(polyethylene glycol,PEG)-CPGを支持体にしてβ-シアノエチルホスホロアミダイトを用いてDNA骨格構造をなすホスホジエステル結合を連結していく方法で行って、3’末端部位にポリエチレングリコールが結合したセンス鎖の二本鎖オリゴヌクレオチド-親水性物質構造体を合成した後、ジスルフィド結合が含まれているC24を5’末端に結合させた。センス鎖とアニーリングを行うアンチセンス鎖の場合も、β-シアノエチルホスホロアミダイトを用いてRNA骨格構造をなすホスホジエステル結合を連結していく方法を用いて、センス鎖と相補的な配列のアンチセンス鎖を製造した後、5’末端にリン酸基を結合させるために化学的リン酸化試薬(Chemical Phosphorylation reagent,CPR)を用いて、5’末端にリン酸基が結合したアンチセンス鎖を製造した。
合成が完了すると、60℃の恒温水槽(water bath)で28%(v/v)アンモニア(ammonia)を処理して合成されたオリゴヌクレオチド一本鎖及びオリゴヌクレオチド-高分子構造体をCPGから切り離した後、脱保護(deprotection)反応を用いて保護残基を除去した。保護残基が除去されたオリゴヌクレオチド一本鎖及びオリゴヌクレオチド-高分子構造体は、70℃のオーブンでN-メチルピロリドン(N-methylpyrolidon)、トリエチルアミン(triethylamine)及びトリエチルアミントリハイドロフルオリド(triethylaminetrihydrofluoride)を体積比10:3:4の比率で処理して2’を除去した。前記反応物から、高速液体クロマトグラフィー(High Performance Liquid Chromatography,HPLC)により、オリゴヌクレオチド一本鎖、オリゴヌクレオチド-高分子構造体及びリガンドが結合したオリゴヌクレオチド-高分子構造体を分離し、これをMALDI-TOF質量分析器(MALDI TOF-MS,SHIMADZU、日本)で分子量を測定し、合成しようとする塩基配列及びオリゴヌクレオチド-高分子構造体と符合するかどうか確認した。その後、それぞれの二本鎖オリゴヌクレオチド構造体を製造するために、センス鎖とアンチセンス鎖を同量混合して1Xアニーリングバッファ(30mM HEPES、100mM酢酸カリウム(Potassium acetate)、2mM酢酸マグネシウム(Magnesium acetate)、pH7.0以上に入れ、90℃恒温水槽で3分反応させた後、さらに37℃で反応させ、目的するSAMiRNA、monoSAMiRNA(n=1)、monoSAMiRNA(n=2)、monoSAMiRNA(n=3)及びmonoSAMiRNA(n=4)をそれぞれ製造した。製造された二本鎖オリゴヌクレオチド構造体は、電気泳動を用いてアニーリングを確認した。
実施例3.アンドロゲン受容体をターゲットにしてRNAiを誘導するSAMiRNAナノ粒子スクリーニング
3.1 SAMiRNAナノ粒子の製造及び粒度分析
実施例2で合成されたアンドロゲン受容体配列をターゲットとする544種のSAMiRNAの粒度分析のためにZetasizer Nano ZS(Malvern,UK)を用いてSAMiRNAのサイズ及び多分散指数(polydispersity index)を測定した結果、無作為に選別されたSAMiRNAに対するナノ粒子のサイズ及び多分散指数は下記表4の通りであり、代表的なグラフは、図3のように測定された。
3.1 SAMiRNAナノ粒子の製造及び粒度分析
実施例2で合成されたアンドロゲン受容体配列をターゲットとする544種のSAMiRNAの粒度分析のためにZetasizer Nano ZS(Malvern,UK)を用いてSAMiRNAのサイズ及び多分散指数(polydispersity index)を測定した結果、無作為に選別されたSAMiRNAに対するナノ粒子のサイズ及び多分散指数は下記表4の通りであり、代表的なグラフは、図3のように測定された。
3.2 SAMiRNAナノ粒子の細胞内処理方法
アンドロゲン受容体の発現を抑制するSAMiRNA発掘のために、ヒト由来前立腺癌細胞株であるLNCaPを利用し、LNCaP細胞株は、10%ウシ胎児血清(Hyclone,US)及び1%ペニシリン-ストレプトマイシン(Hyclone,US)が含まれたRPMI培地(Hyclone,US)を用いて、37℃、5%CO2条件で培養した。上と同じ培地を用いてLNCaP細胞株を12ウェルプレート(Costar,US)に4×104cells/wellの条件で分注し、翌日、SAMiRNAを1X DPBSで希釈して細胞に50nMとなるように処理した。SAMiRNAの処理は、12時間ごとに1回ずつ処理する条件で総4回処理し、37℃、5%CO2条件で培養した。

3.3 アンドロゲン受容体mRNA発現抑制効能分析を用いたSAMiRNAスクリーニング
実施例3.2と同じ方法でSAMiRNAが処理された細胞から抽出されたRNAを、AccuPower(登録商標) RocketScriptTM Cycle RT Premix with oligo(dT)20を用いてcDNAとして合成した後、Taqmanプローブ方式のマルチプレクスqPCR方法を用いて、SAMiRNA対照試料(control sample)に対比してアンドロゲン受容体遺伝子の相対的発現率を分析した。
実施例3.2と同じ方法でSAMiRNAが処理された細胞から抽出されたRNAを、AccuPower(登録商標) RocketScriptTM Cycle RT Premix with oligo(dT)20を用いてcDNAとして合成した後、Taqmanプローブ方式のマルチプレクスqPCR方法を用いて、SAMiRNA対照試料(control sample)に対比してアンドロゲン受容体遺伝子の相対的発現率を分析した。
その結果、図4に示すように、アンドロゲン受容体を標的とする544種のSAMiRNAから60%以上のアンドロゲン受容体mRNA抑制能を示す配列のうち、既存特許(US2007-0141009A)で言及された配列9種と表2の配列14種が選別され(図5)、14種の配列に対するアンドロゲン受容体mRNA抑制能再現評価は、図6に示した。アンドロゲン受容体遺伝子発現を最も効果的に抑制する2種のSAMiRNAを最終選別した。当該SAMiRNAの配列情報は、下記表5の通りである。
3.4 選別されたSAMiRNAのアンドロゲン受容体タンパク質発現抑制効能評価
実施例3.3で選別された68、109番の配列を含めて、共に選別された14種のSAMiRNAが効果的にアンドロゲン受容体タンパク質の発現を抑制するかどうか確認するために、ウェスタンブロット(Western blot,WB)分析を行った。6ウェルプレート(Costar,US)にLNCaP細胞株を1.2×105cell/wellずつ分注した後、37℃、5%CO2条件で培養した。翌日、リポフェクタミン(invitrogen,USA)を用いて50nM濃度で形質移入(transfection)を行った。48時間培養後、培地を除去し、プロテアーゼ阻害剤カクテル(protease inhibitor cocktail)(Sigma Aldrich,USA)が含まれた細胞溶解バッファ(cell lysis buffer)(Cell signaling technology,USA)を用いてタンパク質を分離した。BCAアッセイキット(Thermo,USA)を用いてタンパク質の量を定量した後、20ugのタンパク質を5倍レムリサンプルバッファー(Laemmli’s 5x sample buffer)と共に95℃で10分間沸かした。変性されたタンパク質をSDS-ポリアクリルアミドゲルに電気泳動した後、PVDFメンブレインに移した。メンブレインをブロッキング溶液(5%脱脂粉乳含有TBS及び0.05% Tween 20)に浸して室温で1時間処理した後、1次抗体であるAR抗体(1:2000,Santa cruz,USA)及びGAPDH抗体(1:5000,Cell Signaling Technology,USA)と共に4℃冷蔵庫に一晩反応させた。TBSTで3回洗浄した後、ホースラディッシュペルオキシダーゼ(horse-radish peroxidase)標識2次抗体(Cell signaling technology)と常温で1時間反応した後、化学発光試薬であるSuperSignal(登録商標)ピコ化学発光基質(Pico Chemiluminescent Substrate)(Thermo,USA)を用いてタンパク質バンドを検出した。

14種のSAMiRNAに対するアンドロゲン受容体タンパク質発現抑制能は、図7のように確認され、タンパク質発現においても配列番号68と109の抑制能が最も優れていた。
3.5 ヒト由来毛根細胞である毛乳頭真皮細胞(Hair Follicle Dermal Papilla Cell,HFDPC)においてアンドロゲン受容体タンパク質発現抑制効能評価
実施例3.4から最終的に選別された配列番号68及び109が実際にヒトの毛根細胞でアンドロゲン受容体タンパク質発現を抑制させるかどうか確認するために、ヒト由来毛根細胞である毛乳頭真皮細胞(HFDPC)を対象にしてタンパク質発現抑制の有無を確認した(図8)。両配列ともアンドロゲン受容体タンパク質の発現を抑制できることが確認された。
実施例3.4から最終的に選別された配列番号68及び109が実際にヒトの毛根細胞でアンドロゲン受容体タンパク質発現を抑制させるかどうか確認するために、ヒト由来毛根細胞である毛乳頭真皮細胞(HFDPC)を対象にしてタンパク質発現抑制の有無を確認した(図8)。両配列ともアンドロゲン受容体タンパク質の発現を抑制できることが確認された。
実施例4. SAMiRNAナノ粒子の皮膚内伝達効果確認
最終的に選別された配列番号68及び109で作製したSAMiRNA-AR#68、SAMiRNA-AR#109が実際にヒトの毛根に伝達されるかを確認するために、ヒトの毛髪を対象にして遺伝子伝達効果を確認した。

毛髪は、実験当日に髪の毛の先端部分を取って抜いて採取し、毛根から1cm程度に切った後、96ウェルプレートに200ul M199培地(10%FBS+1%ペニシリン)に1時間インキュベーションした。その後、遺伝子伝達の有無を観察するために、蛍光物質(FAM dye)が標識されたSAMiRNA 2uM、10uMが含まれているM199培地200ulに24時間インキュベーションした。物質処理24時間後、DPBSを用いて3回の洗浄作業を行い、最終的に3.7%ホルムアルデヒド、2%FBSがあるPBSで20分間毛根固定作業を行った。
固定の完了した毛根を、OCT化合物(OCT compound)が入っているベースモールド(base mold)に植え、あらかじめ凍らせておいたステンレスプレート上に乗せてOCT化合物を完全に凍らせた。凍らせた組織は-70℃に保管し、組織切片器で切る前に、組織切片がし易くなるように-20℃に30分程度置いた。切片組織は、10um厚でスライドに乗せて1時間乾燥させ、乾燥が終わると、マウント過程を経るが、このとき、DAPI含有のマウンティングメディアムを用いた。共焦点レーザースキャン顕微鏡(Confocal Laser Scan Microscope(LSM5 LIVE CONFIGURATION VARIOTWO VRGB))で蛍光を観察した結果、SAMiRNAが髪の毛組織の毛根細胞に伝達されることが確認された(図9)。
以上、本発明内容の特定の部分を詳細に記述したところ、当業界における通常の知識を有する者にとって、このような具体的技術は単に好ましい実施様態に過ぎず、これによって本発明の範囲が制限されない点は明らかであろう。したがって、本発明の実質的な範囲は、添付する請求項及びそれらの等価物によって定義されるといえよう。
本発明に係るアンドロゲン受容体特異的オリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体及びこれを有効成分として含む脱毛予防又は発毛用組成物は、副作用無しで高い効率でアンドロゲン受容体の発現を抑制させ、脱毛、特に、アンドロゲン性脱毛、円形脱毛、休止期脱毛の予防及び発毛に優れた効果を示すことができる。

前記構造式(2)で、Sは、請求項1に記載のオリゴヌクレオチドのセンス鎖、ASはアンチセンス鎖を意味し、A、B、X及びYは、請求項1における定義と同一である。 - 下記構造式(3)又は構造式(4)の構造を有する、請求項2に記載の二本鎖オリゴヌクレオチド構造体。
前記構造式(3)及び構造式(4)で、A、B、X、Y、S及びASは、請求項2における定義と同一であり、5’及び3’は、オリゴヌクレオチドセンス鎖の5’末端及び3’末端を意味する。 - 親水性物質の分子量は、200~10,000であることを特徴とする、請求項1に記載の二本鎖オリゴヌクレオチド構造体。
- 親水性物質は、ポリエチレングリコール(PEG)、ポリビニルピロリドン及びポリオキサゾリンからなる群から選ばれるいずれか1つであることを特徴とする、請求項4に記載の二本鎖オリゴヌクレオチド構造体。
- 親水性物質は、下記構造式(5)又は構造式(6)の構造を有することを特徴とする、請求項1に記載の二本鎖オリゴヌクレオチド構造体。

前記構造式(5)及び構造式(6)で、A’は親水性物質単量体(monomer)を、Jは、m個の親水性物質単量体間、又はm個の親水性物質単量体とオリゴヌクレオチドとを互いに連結するリンカーを、mは1~15の整数、nは1~10の整数を意味し、
親水性物質単量体A’は、化合物(1)~化合物(3)から選ばれたいずれか1つの化合物であり、リンカー(J)は、PO3 -、SO3及びCO2からなる群から選ばれる。
- 前記疎水性物質の分子量は、250~1,000であることを特徴とする、請求項1に記載の二本鎖オリゴヌクレオチド構造体。
- 疎水性物質は、ステロイド(steroid)誘導体、グリセリド(glyceride)誘導体、グリセロールエーテル(glycerol ether)、ポリプロピレングリコール(polypropylene glycol)、C12~C50の不飽和又は飽和炭化水素(hydrocarbon)、ジアシルホスファチジルコリン(diacylphosphatidylcholine)、脂肪酸(fatty acid)、リン脂質(phospholipid)及びリポポリアミン(lipopolyamine)からなる群から選ばれることを特徴とする、請求項7に記載の二本鎖オリゴヌクレオチド構造体。
- 前記ステロイド(steroid)誘導体は、コレステロール、コレスタノール、コール酸、ギ酸コレステロール、ギ酸コレスタニル及びコレステリルアミンからなる群から選ばれることを特徴とする、請求項8に記載の二本鎖オリゴヌクレオチド構造体。
- 前記グリセリド誘導体は、モノ-、ジ-及びトリ-グリセリドから選ばれることを特徴とする、請求項8に記載の二本鎖オリゴヌクレオチド構造体。
- 前記X及びYで表示される共有結合は、非分解性結合又は分解性結合であることを特徴とする、請求項1に記載の二本鎖オリゴヌクレオチド構造体。
- 前記非分解性結合は、アミド結合又はリン酸結合であることを特徴とする、請求項11に記載の二本鎖オリゴヌクレオチド構造体。
- 前記分解性結合は、ジスルフィド結合、酸分解性結合、エステル結合、アンヒドリド結合、生分解性結合又は酵素分解性結合であることを特徴とする、請求項11に記載の二本鎖オリゴヌクレオチド構造体。
- 親水性物質に受容体媒介エンドサイトーシス(receptor-mediated endocytosis,RME)によってターゲット細胞内在化(internalization)を増進させる受容体と特異的に結合する特性を有するリガンド(ligand)がさらに結合したことを特徴とする、請求項1に記載の二本鎖オリゴヌクレオチド構造体。
- 前記リガンドは、ターゲット受容体特異的抗体、アプタマー、ペプチド、葉酸(folate)、N-アセチルガラクトサミン(N-acetyl Galactosamine,NAG)、ブドウ糖(glucose)及びマンノース(mannose)からなる群から選ばれることを特徴とする、請求項14に記載の二本鎖オリゴヌクレオチド構造体。
- 前記親水性物質のオリゴヌクレオチドと結合した反対側末端部位に、アミン基又はポリヒスチジン(polyhistidine)基がさらに導入されたことを特徴とする、請求項1に記載の二本鎖オリゴヌクレオチド構造体。
- 前記アミン基又はポリヒスチジン(polyhistidine)基は、1つ以上のリンカーを介して親水性物質又は親水性ブロックと連結されたことを特徴とする、請求項16に記載の二本鎖オリゴヌクレオチド構造体。
- 前記アミン基は1級~3級アミン基から選ばれたいずれか1つであることを特徴とする、請求項16に記載の二本鎖オリゴヌクレオチド構造体。
- 前記ポリヒスチジン(polyhistidine)基は、3~10個のヒスチジンを含むことを特徴とする、請求項16に記載の二本鎖オリゴヌクレオチド構造体。
- 請求項1~19のいずれか一項に記載の二本鎖オリゴヌクレオチド構造体を含むナノ粒子(nanoparticle)。
- 異なる配列を有するオリゴヌクレオチドを含む二本鎖オリゴヌクレオチド構造体が混合されていることを特徴とする、請求項20に記載のナノ粒子。
- 請求項1~19のいずれか一項に記載の二本鎖オリゴヌクレオチド構造体を有効成分として含む、脱毛予防又は発毛用医薬組成物。
- 請求項20に記載のナノ粒子を有効成分として含む、脱毛予防又は発毛用医薬組成物。
- 前記医薬組成物は、軟膏、ペースト、ゲル、ゼリー、セラム(serum)、エアゾールスプレー、非エアゾールスプレー、フォーム、クリーム、ローション、溶液又は懸濁液の剤形から選ばれる剤形に用いられることを特徴とする、請求項22に記載の医薬組成物。
- 前記医薬組成物は、軟膏、ペースト、ゲル、ゼリー、セラム(serum)、エアゾールスプレー、非エアゾールスプレー、フォーム、クリーム、ローション、溶液又は懸濁液の剤形から選ばれる剤形に用いられることを特徴とする、請求項23に記載の医薬組成物。
- 請求項1~19のいずれか一項に記載の二本鎖オリゴヌクレオチド構造体を有効成分として含む、脱毛予防又は発毛用化粧料組成物。
- 請求項20に記載のナノ粒子を有効成分として含む、脱毛予防又は発毛用化粧料組成物。
- 前記組成物は、ヘアトニック、ヘアコンディショナー、ヘアエッセンス、ヘアローション、ヘア栄養ローション、ヘアシャンプー、ヘアリンス、ヘアトリートメント、ヘアクリーム、ヘア栄養クリーム、ヘアモイスチャークリーム、ヘアマッサージクリーム、ヘアワックス、ヘアエアゾール、ヘアパック、ヘア栄養パック、ヘア石鹸、ヘアクレンジングフォーム、ヘアオイル、毛髪乾燥剤、毛髪保存処理剤、毛髪染色剤、毛髪用ウェーブ剤、毛髪脱色剤、ヘアゲル、ヘアグレーズ、ヘアドレッシング、ヘアラッカー、ヘアモイスチャーライザー、ヘアムース又はヘアスプレーの剤形から選ばれる剤形に用いられることを特徴とする、請求項26に記載の化粧料組成物。
