Abstract
Description
一態様では、本発明は、細胞内のPD1遺伝子に対して遺伝子編集を行う方法を提供し、前記方法は、ヌクレアーゼ及びsgRNAを用いてPD1遺伝子に対して遺伝子編集を行うステップを含み、前記sgRNAはヌクレアーゼを誘導してPPD1遺伝子を切断して切断部位を形成する。
(1)NCBIによってヒトPD1ゲノム配列を照会し、エクソンNo.1、エクソンNo.2又は重要なタンパク質コード領域を選択してsgRNA設計を行い、設計したsgRNAの標的配列は次のとおりである。

sgRNA3: gtctgggcggtgctacaact(SEQ ID No.2)

2-1 T7E1酵素切断:
(1)Cas9、sgRNAを細胞に形質導入し、ゲノムDNAを抽出する。
(1)Cas9、sgRNAを細胞に形質導入し、2~3日後に細胞を収集する。
(1)Cas9、sgRNA及び外因性DNAを細胞に形質導入し、7日後に細胞を収集する。
以下、sgRNA9を例としてCRISPR/Cas9技術を組み合わせ、PD1がノックアウトされた強化CD19-CART細胞をワンステップで構築する。

100μl仕様のエレクトロポレーションカップに適用:
(1)82μl Solution+18μl supplement/各エレクトロポレーションカップで、エレクトロポレーショントータルに応じて、エレクトロポレーション液mixを調製し、均一に混合し、室温で放置。
上記「donor」外因性ドナーDNAを外因性DNA配列とし、2つの異なるドナー(Donor-1及びDonor-2)のT細胞において、PD1-sgRNA9部位のいずれでも高い部位特異的組み込み率を有することを証明し、図5に示すように、部位特異的組み込み効率は20%~30%に達する。
Claims (13)
- 細胞内のPD1遺伝子に対して遺伝子編集を行う方法であって、
ヌクレアーゼ及びsgRNAを細胞に導入することにより、PD1遺伝子に対して遺伝子編集を行うステップを含み、前記sgRNAはヌクレアーゼによるPPD1遺伝子の切断を誘導して切断部位を形成し、
PD1を標的とする前記sgRNAの標的配列は、SEQ ID No.1~6のいずれかに示す配列を含み、好ましくは、PD1を標的とする前記sgRNAの標的配列は、SEQ ID No.4及び/又はSEQ ID No.5に示す配列を含む、ことを特徴とする方法。 - 前記ヌクレアーゼは、Cas9、Cas3、Cas8a、Cas8b、Cas10d、Cse1、Csy1、Csn2、Cas4、Cas10、Csm2、Cmr5、Fok1、Cpf1から選択される1つ又は任意の複数であり、好ましくは、前記ヌクレアーゼは、Cas9であり、さらに好ましくは、前記Cas9は、肺炎レンサ球菌、化膿レンサ球菌又はストレプトコッカス・サーモフィルスに由来するCas9から選択される、ことを特徴とする請求項1に記載の方法。
- 前記sgRNAは塩基の化学修飾をさらに含み、好ましくは、前記化学修飾は、メチル化修飾、メトキシ修飾、フッ素化修飾又はチオ修飾のうちの1つ又は任意の複数である、ことを特徴とする請求項1又は2に記載の方法。

PD1を標的とする前記sgRNAの標的配列は、SEQ ID No.1~6のいずれかに示す配列を含み、好ましくは、PD1を標的とする前記sgRNAの標的配列は、SEQ ID No.4及び/又はSEQ ID No.5に示す配列を含む、ことを特徴とするsgRNA。 - 前記sgRNAは、塩基の化学修飾をさらに含み、好ましくは、前記化学修飾は、メチル化修飾、メトキシ修飾、フッ素化修飾又はチオ修飾のうちの1つ又は任意の複数である、ことを特徴とする請求項11に記載のsgRNA。
- 請求項11又は12に記載のsgRNAの細胞内のPD1遺伝子に対する遺伝子編集における使用。
