濁するように激しく振盪した。得られた懸濁液を、TSALT(0.3%大豆レシチン及び3%Tween 80を添加したTSA)との混釈平板とした。プレートを31℃±1℃で少なくとも5日間インキュベートした。
真菌カンジダ・アルビカンス(Candida albicans)に対する抗菌材料の試験
試験菌を、0.85%NaCl中、約1×106CFU/mLとなるように調製した。
各試料について、5つの複製した試験片に適切な量の試験菌を接種した(表2)。サブローデキストロース寒天(Sabouraud Dextrose Agar)(SDA)の混釈平板で接種時の接種菌数を測定した。次いで、接種後の試料を24℃±1℃、湿度95%以上のインキュベータに1、8又は24時間静置した。所要の曝露時間が経過した後、接種した試験片を無菌的に取り出して希釈剤9mLに移した。これを、残存している試験菌が確実に充分に再懸濁するように激しく振盪した。得られた懸濁液を、SDALT(0.3%大豆レシチン及び3%Tween 80を添加したSDA)との混釈平板とした。プレートを24℃±1℃で少なくとも5日間培養した。陰性対照として、試料に適量(表2)の無菌0.85%NaClを接種し、試験試料と一緒に培養及び分析した。

ウシコロナウイルス(BCV)L9株に対する抗菌材料の試験
材料を準備するために1×1cmの小片を無菌条件下で切断し、折り畳んでからエッペンドルフカップに移した。試験ウイルス液を調製するために、U373細胞を培養した。ウイルスを産生させるために、調製した単層にBCV L9株を添加した。24~48時間インキュベートした後、細胞を急速な凍結融解サイクルに供して溶解した。細胞残屑を除去し、上清をそのまま試験用ウイルス懸濁液として使用した。感染価を、マイクロタイター法を用いるエンドポイント希釈測定(end point dilution titration)により決定した。処理した材料の殺ウイルス活性を、未処理材料の殺ウイルス活性と比較して力価の低下を算出することにより評価した。
Claims (24)
- 基材及び金属成分を含む抗菌材料であって、前記金属成分は化学結合した銅及び亜鉛を含む、抗菌材料。
- 前記銅及び亜鉛は合金を形成している、請求項1に記載の抗菌材料。
- 前記金属成分は、ジルコニウム、銀、金、パラジウム、白金、イリジウム、アルミニウム、ニッケル、タングステン、モリブデン、タンタル、チタン、ヨウ素及び/又はこれらの合金からなる群から選択される任意の金属を更に含む、請求項1又は2に記載の抗菌材料。
- 前記金属成分は、ジルコニウム、銅、亜鉛、銀、金、パラジウム、白金、イリジウム、アルミニウム、ニッケル、タングステン、モリブデン、タンタル、チタン及び/又はヨウ素の何れかの1種又は複数種の塩を更に含む、請求項1、2又は3に記載の抗菌材料。
