背景技术
某些金属的抗微生物特性在相当长的一段时间内是已知的。这种独特的性质已经为包括农业和医疗保健在内的各个行业所投资,以试图控制感染和污染。
医疗保健环境中常用的一种金属是银。银的抗微生物作用取决于生物活性银离子,从而导致对病原体的细胞膜内的关键的酶系统产生不可逆的破坏,而使细胞死亡。银用作微生物试剂的最有效的条件是那些具有较高温度和过度潮湿的条件。这些条件有助于释放银离子所需的离子交换反应。但是,这些特殊的条件很少在日常医疗机构中再现,因此限制了银在控制感染率方面的功效。相比之下,已表明铜在多种环境条件下具有令人印象深刻的抗微生物功效。

对于在伤口敷料中使用抗微生物材料的兴趣尤其突出。伤口可以是以下六种类型之一:擦伤、切口、撕裂伤、刺伤、撕脱或截肢。一个特别的挑战是治疗例如糖尿病和压力性溃疡等慢性伤口,该伤口会导致皮肤的“屏障”功能的长期破坏,从而增加感染的可能性。

发明内容
在第一方面,本发明提供了一种包括基底和金属组分的抗微生物材料,其中金属组分包括化学键合的铜和锌。
在第二方面,本发明提供了一种制造包括基底和金属组分的抗微生物材料的方法,其中金属组分包括化学键合的铜和锌,该方法包括以下步骤:
a)将铜和锌结合以制备所述金属组分;
b)将金属组分加热到熔融状态;
c)用高速气体破坏所述熔融状态,和;
d)将金属组分与基底结合。

以下说明的提供是为了使本领域的任何技术人员能够制造和使用本发明。对所公开的实施例的各种修改对于本领域技术人员而言都将是显而易见的。
在第一方面,本发明提供了一种包括基底和金属组分的抗微生物材料,其中金属组分包括化学键合的铜和锌。
术语“抗微生物材料”是指具有抗微生物性质例如杀生物或生物抑制(biostatic)性质的材料。在本发明的上下文中,术语“杀生物”应理解为意指可以破坏、阻止、使其无害或对病原生物施加控制作用的物质,而术语“生物抑制”是指可以抑制生物体例如微生物的生长或繁殖。设想本发明将可用于对抗任何微生物,例如任何细菌、病毒和/或真菌。特别地,设想葡萄球菌属(Staphylococcus)和克雷伯菌属(Klebsiella)中的细菌、念珠菌属(Candida)中的真菌以及冠状病毒(Coronaviridae)科的成员将对本发明记载的材料敏感。
本发明提供了具有令人惊讶的高的抗微生物活性的材料。包含本发明材料的产品,例如伤口敷料,将有助于更快的伤口愈合并且降低败血病和感染的发生率。本发明在提供有助于治疗糖尿病性溃疡和其他愈合或闭合缓慢的伤口的产品中特别有用。
使用“基底”,我们意指可以将金属组分掺入到其中的任何合适的结构材料,从而提供可以将金属组分布局于其上或其中的物理介质。在优选的实施方案中,基底是泡沫。使用“泡沫”,我们意指使用一种由基底材料形成的结构,该结构在基底材料内具有捕获气体的口袋。泡沫可以是固体泡沫。固体泡沫的主要组分可以是基于聚合物的材料。泡沫也可以是液体泡沫。

与纯的金属或相关化合物的形式相比,与使用纯铜,使用合金产生了许多有利的性质。具体地,铜锌合金得益于锌的超强抗微生物性质,优异的延展性/可铸性和高的强度。
可预期金属组分的颗粒尺寸为10-80μm,优选尺寸为15-30μm内的任意值。与粗的粉末相比,高度研磨的粉末会释放更多的离子,这些被释放的离子可具有抗微生物作用。
设想金属组分将包含至少60%的铜。该制剂具有增强的抗微生物性质。优选地,金属组分包含75-80%的铜和相应量的20-25%的锌。如上所述,金属组分可以额外地包含其他元素、其化合物以及盐。这些添加物可以赋予所要求保护的材料有益的性质。例如,添加的组分可以进一步增强抗微生物作用或允许所要求保护的产品的寿命延长。
在本发明的一个实施例中,金属组分可以散布在整个基底中。使用“散布”(interspersion),我们意指金属散落在基质材料的颗粒/分子之间。可以将这种结构描述为“浸渍”。金属成分可以均匀地或不均匀地分散(dispersion)在整个基板材料中。本领域技术人员会理解,散布、分散和/或浸渍的程度可以取决于在制造基底材料中使用的聚合物类型和/或将金属组分施加到基底的方法。在本发明的另一个实施方案中,金属组分可以作为涂层存在于基底表面上。在存在涂层的情况下,预期涂层将被布置为使得在使用中它与可能被污染的表面/伤口接触以发挥其抗微生物作用。涂层可以是任何厚度。另外,涂层应理解为存在于基底的至少一个表面上,也可以存在于所有基底表面上。涂层可以部分覆盖或完全覆盖基底的特定的表面。涂层的覆盖程度将取决于所要求保护的产品的预期用途。
设想基底可以是基于聚合物的基底。聚合物是由较小的重复亚单元组成的大分子。优选地,用于本发明的基底可以包括聚氨酯、聚丙烯和/或基于胶原的聚合物。根据本公开,该基材可以包括基于聚合物的水凝胶或基于聚合物的水胶体。热固性和热塑性聚氨酯均可适用于本发明。但是,可以设想,适合于稳定地保持金属组分的任何材料都可以单独使用或组合使用以作为本发明的基底。例如,如果合适,如羊毛、棉、皮革、亚麻、苎麻、丝绸、大麻、竹子、黄麻、人造丝、氯丁橡胶、弹性纤维、橡胶、聚酯这些材料都可以适合用作基底。在某些情况下,应当理解,基底可以是不同类型的聚合物的组合。这样的组合可以在基底上赋予其他的有利的性质以实现所期望的目的或便于制造和存储。具体地,设想可以将藻酸盐和纤维素掺入基底中以增强吸收性、柔韧性和舒适性。技术人员会认识到,基于聚合物的水凝胶由于存在亲水性官能团而特别有利地用于伤口敷料中。该功能可以控制特定表面的水分。
优选地,制造后的基底可以包含以下成分(以最终干燥产品的重量计算的百分比):
a)羧甲基纤维素钠7%
b)表面活性剂3%
c)甘油18%
d)柠檬酸3.5%
e)选定的聚合物68.5%
合适的表面活性剂的实例包括硬脂酸钠、磺基琥珀酸二辛酯钠盐和全氟辛烷磺酸盐。合适的表面活性剂可以属于以下任何组:阴离子、阳离子、非离子或两性离子表面活性剂。如果需要,柠檬酸部分可以被其他弱酸例如乙酸、乳酸和磷酸代替。以上列表的部分e)可以被上述任何聚合物替代。优选地,所选择的聚合物被单独使用;然而,如果认为最终的抗微生物试剂是更有效的并且仍占基底组合物的68.5%,则可以组合使用不同的聚合物。
优选地,3-15重量%的基底由金属组分组成。设想如果需要,在材料中包含其他添加物以改善抗微生物性质。这些添加物可以包括螯合剂、硫酸镁和/或铜肽。这些添加物可以以0.1至1重量%,例如约0.5重量%的量掺入到基底中。术语“螯合剂”用于描述可以与单个金属离子形成多个键从而形成更稳定的络合物的物质。技术人员会认识到此类物质的作用可以增强抗微生物性质。
当它与任何被污染的表面接触时,本发明可为有效的。在优选的实施方案中,本发明可以掺入到适合于在人或动物的不同结构位置的皮肤的表面上施用的伤口辅料中。抗微生物材料优选是透气的。使用“透气”,意指使空气流保持流向伤口或待治疗的其他表面。在应用于伤口敷料时,设想允许伤口干燥或至少不闷热的能力以进一步加强愈合过程。优选地,该材料遍布了间隔开的孔,以促进透气性。技术人员将理解如何根据材料的尺寸和用途来布置这些孔。
本发明提供了高水平的抗微生物活性,因此具有多种应用。本发明包括含有本发明的抗微生物材料的感染控制产品。这样的产品在医疗保健领域中可具有实用性,经常作为医学材料。使用“感染控制产品”,我们意指代任何可治疗、预防或减轻感染的发展和/或蔓延的产品。此类产品的实例包括伤口敷料、绷带、医疗设备、药物容器和用于感染防护的个人防护服。
可以设想,本发明的一种用途可以是将金属组分添加到水胶体材料中以治疗腐烂的伤口。腐烂的伤口是坏死组织自身正在从伤口部位分离的伤口。已知这些类型的伤口是从所述伤口渗出液体,因此使用水胶体敷料将这种液体转化成凝胶形式会是特别有益的。
本发明还提供了包含本公开的抗微生物产物的服装或家用产品。使用“家用产品”,我们意指通常在家中使用的任何产品,例如清洁产品和/或衣服。使用“服装”,我们意指可以穿着于身体的任何结构位置上的任何物品。可以应用本发明的服装的实例包括通常会与穿着者的皮肤紧密接触的内衣(包括袜子、背心、长袜、裤子)。可以利用掺入本发明的材料而提供的其他服装,例如鞋子、围巾、裤子、手套、帽子、围裙、运动或物理疗法的关节支撑物(例如护膝袖、颈部支撑物、支撑内裤(supportive brief)等)。此外,还包括家用清洁产品,例如海绵、抹布(一次性或可重复使用)和毛巾。
本发明还提供包含本发明的抗微生物材料的卫生产品,例如卫生巾。使用“卫生产品”,我们意指主要为个人使用并且旨在与使用者的皮肤或身体孔口(bodily orifice)紧密接触的产品。将本发明的抗微生物材料应用于此类产品将有助于防止任何特定微生物的任何有害堆积,并且降低败血症的可能性。可以掺入本发明的此类卫生产品的实例包括窗帘、床上用品、清洁产品、卫生巾、棉塞和相关的卫生产品、一次性尿布、失禁裤子和垫子、衣服、鞋子以及用于运输所述产品的装置。
在第二方面,本发明提供了一种制造包括基底和金属组分的抗微生物材料的方法,其中所述金属组分包括化学键合的铜和锌。该方法包括以下步骤:a)将铜和锌结合以制备所述金属组分;b)将金属组分加热到熔融状态;c)用高速气体破坏所述熔融状态,和d)将已被破坏的金属组分与基底结合。
因此,一种生产本发明的金属组分的方法可以包括等离子体或气体雾化过程。可以设想,金属的粉末形式可以用在本发明的方法中,按照本领域技术人员所理解的其他的形式也可能是恰当的。
可以设想,等离子体或气体雾化过程会产生粉末形式的金属组分,其可以在适当时与基底结合,如本领域技术人员所理解的那样。
在优选的实施方案中,在等离子或气体雾化过程开始之前,可任选地通过使用机械磨损过程来减小金属组分的尺寸。使用“机械磨损”,我们意指采用能够将金属组分逐渐分成更小的个体的任何过程。该过程可以通过使用许多的磨损设备来实现,这些设备包括但不限于:磨碎机、卧式磨、一维振动磨、3D振动磨和行星式磨。所有上述装置都会使尺寸减小,由于在研磨介质的撞击期间赋予了样品的能量。因此,在本发明的方法中,可以将金属形式的铜和锌研磨成我们所需要的合适形式。
一旦铜和锌结合在一起,就可以进行雾化过程。如本领域技术人员将会理解的,结合铜和锌的方式可以根据要使用的雾化过程而不同。
等离子体雾化要求金属组分为线状以用作原料。如本领域技术人员将会理解的,这通常是金属组分的合金线。与常规的气体雾化相反,等离子体雾化使用等离子炬在单个步骤中瞬间熔化和雾化线状物。然后使用冷却塔将形成的液滴转化为球形粉末。
或者,可以使用常规的气体雾化。这可以包括将铜-锌金属组分加热到约2000℃,以产生所述组分的熔融状态。使用“熔融态”,我们意指所述金属组分在暴露于高温时的液体形式。如本领域技术人员将会理解的,高速气流可流过膨胀喷嘴,从输入室虹吸熔融金属组分。可用于此过程的气体的实例包括氮气、氩气、氦气或空气。技术人员将会认识到,可以在该过程中使用一种以上的气体,并且优选的气体或气体混合物将是惰性的/不反应的。所用气体的选择将取决于所需的最终破坏的金属(粉末)的特性。为了提供合适的金属组分用于本发明的材料,可能需要高速的惰性气体。技术人员将会认识到,所需速度将根据所用的气体而有所不同,但是可能在100-2000m/s的范围内。该过程破坏了熔融金属的液流并且结果是产生高度分散的颗粒,最终形成所需的金属组分的粉末形式。通过上述方法获得的粉末形式具有许多优点;高度球形颗粒的制备,低的氧含量以及对于生产铜和锌的适应性。技术人员还将会认识到,可以存在能够用于实现相同效果的生产金属粉末的替代性方法。
为了生产最终的抗微生物材料,将金属组分添加到基底中。具体地,金属粉末以少量的方式添加,直到将整个产物全部转移到基底。将所得的组合物在室温(20-22℃)下以350rpm混合2小时,随后使其固化。
为了产生水胶体产品,在可以添加粘合剂中的金属组分之前,将水胶体材料加热到240℃。粘合剂组分为凝胶形式,并且可以包含80%的羧甲基纤维素和20%的粘合剂。将水胶体材料和粘合剂组分充分混合,以确保在整个所得的材料中的均匀分布。
本发明还提供了一种治疗伤口感染的方法,该方法包括将本发明的抗微生物材料施用于伤口上。
为了可以更清楚地理解本发明,现在将通过实施例的方式来描述本发明的实施方案。
实施例1
在两种不同的细菌菌株(金黄色葡萄球菌(Staphylococcus aureus)和肺炎克雷伯菌(Klebsiella pneumoniae))上进行抗微生物材料的测试
将每种测试生物体制备成约1x105菌落形成单位(CFU)/mL于0.85%NaCl中。对于每个样品,使用每种测试生物体进行5次平行接种。使用胰蛋白胨大豆琼脂(Tryptone SoyaAgar(TSA))的倾注平板法,在接种的点对接种物进行计数。将已经被接种的样品在24℃±1℃和>95%的湿度下保持24小时。在暴露时间之后,将已经被接种的试样在无菌下移至9ml稀释剂中。剧烈摇晃以确保任何残留的测试生物体完全地再悬浮。将得到的悬浮液铺在TSALT(补充有0.3%大豆卵磷脂和3%Tween 80的TSA)中。将平板在31℃±1℃下培养至少5天。

对于样品“3%CuZn泡沫”和“15%CuZn泡沫”,可以看出在经历24小时的接触时间后这两种细菌菌株的数量均减少了>4log。将其与样品“0%CuZn泡沫”进行比较,后者对于这两种测试生物体均未显示出明显的抗微生物活性。
实施例2
在真菌白色念珠菌(Candida albicans)上进行抗微生物材料的测试
将每种测试生物体制备成约1x106CFU/mL于0.85%NaCl中。对于每个样品,使用适当体积的测试生物体(表2)对于试样进行5次平行接种。使用胰蛋白胨大豆琼脂(TryptoneSoya Agar(TSA))的倾注平板法,在接种的点对接种物进行计数。然后将已经被接种的样品放置于24℃±1℃、>95%的湿度下的培养箱,时间持续1、8或24小时。在所需的暴露时间之后,将已经被接种的试样在无菌下移至9ml稀释剂中。剧烈摇晃以确保任何残留的测试生物体完全地再悬浮。将得到的悬浮液铺在SDALT(补充有0.3%大豆卵磷脂和3%Tween 80的SDA)中。将平板在24℃±1℃下培养至少5天。对于阴性对照,样品用适当体积(表2)的无菌的0.85%NaCl接种,并以与试样相同的方式进行培育和分析。

对于样品“0%CuZn泡沫”,在24℃下1、8或24小时的接触时间之后,观察到白色念珠菌的数量没有明显减少。对于样品“2%CuZn泡沫”,在24℃下1小时的接触时间之后,观察到白色念珠菌的数量的减少大于3log;在24℃下8小时或24小时的接触时间之后,观察到白色念珠菌的数量的减少大于4log。对于样品“3%CuZn泡沫”,在24℃下1小时的接触时间之后,观察到白色念珠菌的数量没有明显减少;在24℃下8小时之后,观察到白色念珠菌的数量的减少大于3log;在24℃下24小时之后,观察到白色念珠菌的数量的减少大于5log。
实施例3:在牛冠状(bovine corona)病毒(BCV)毒株L9上进行抗微生物材料的测试
为了制备该材料,在无菌条件下切成1x1cm的小块,并在折叠步骤后转移到Eppendorf杯中。为了制备测试病毒溶液,培养U373细胞。为了产生病毒,将BCV毒株L9加入已制备的单层中。在24-48小时的培育期后,通过快速的冷冻/融化循环来裂解细胞。去除细胞碎片,并将上清液直接用作测试病毒悬浮液。使用微量滴定法通过终点稀释滴定来测定感染性。通过计算滴定度的降低,与未经处理材料的杀病毒活性进行比较,以评估经过处理的材料的杀病毒活性。

在60分钟的接触时间后,在使用新的绿色/白色尼龙铜浸渍织物下,检测到仅一例材料中有残留病毒。相比之下,检测未经处理的材料,残留病毒可以在所有情况下被检测到。得出以下平均值:≤1.55±0.04(新的绿色/白色尼龙铜浸渍织物)和2.98±0.12(参考)。可见,在60分钟暴露时间后,基于10倍测定,两种材料的差值为log10的值1.43。
Claims (24)
1.一种抗微生物材料,其包括基底和金属组分,其中金属组分包括化学键合的铜和锌。
2.根据权利要求1的抗微生物材料,其中铜和锌形成合金。
3.根据权利要求1或2的抗微生物材料,其中金属组分还包括选自以下的任何金属:锆、银、金、钯、铂、铱、铝、镍、钨、钼、钽、钛、碘和/或其任何合金。
4.根据权利要求1、2或3的抗微生物材料,其中金属组分还包括锆、铜、锌、银、金、钯、铂、铱、铝、镍、钨、钼、钽、钛和/或碘中任意的一种或多种的盐。
5.根据权利要求1-4中任一项的抗微生物材料,其中金属组分包括尺寸为3-50μm,优选15-30μm的颗粒。
6.根据权利要求1-5中任一项的抗微生物材料,其中金属组分包含至少60%的铜。
7.根据权利要求1-6中任一项的抗微生物材料,其中金属组分包含75-80%的铜。
8.根据权利要求1-7中任一项的抗微生物材料,其中金属组分包含20-25%的锌。
9.根据权利要求1-8中任一项所述的抗微生物材料,其中金属组分散布在在整个基底中,和/或其中金属组分在所述基底的表面上形成涂层。
10.根据权利要求9所述的抗微生物材料,其中涂层具有抗微生物性质,并且被布置以便在使用中表面与可能被污染的表面接触。
11.根据权利要求1-10中任一项的抗微生物材料,其中基底包括基于聚合物的基底,优选其中聚合物为聚氨酯、聚丙烯和/或胶原;更优选其中聚合物包含在水凝胶或水胶体中。
12.根据权利要求1-11中任一项的抗微生物材料,其中所述基底是泡沫。
13.根据权利要求1-12中任一项的抗微生物材料,其中所述材料的3-15重量%由金属组分组成。
14.根据权利要求1-13任一项的抗微生物材料,其中该材料还包括螯合化合物、硫酸镁和/或铜肽。
15.根据权利要求1-14中任一项的抗微生物材料,其中所述抗微生物材料是透气的。
16.一种感染控制产品,优选伤口敷料或绷带,其包含权利要求1-15中任一项的抗微生物材料。
17.一种服装或家用产品,其包含权利要求1-15中任一项的抗微生物材料。
18.一种卫生产品,优选卫生巾,其包含权利要求1-15中任一项的抗微生物材料。
19.一种包括基底和金属组分的抗微生物材料的制造方法,其中金属组分包括化学键合的铜和锌,
该方法包括以下步骤:
a)将铜和锌结合以制备所述金属组分;
b)将金属组分加热到熔融状态;
c)用高速气体破坏所述熔融状态,和;
d)将被破坏的金属组分与基底结合。
20.根据权利要求19的方法,其中在步骤(b)之前使用机械磨损方法来减小金属组分的尺寸。
21.根据权利要求19或20的方法,其中步骤(c)产生所述金属组分的粉末形式。
22.根据权利要求19、20或21中任一项的方法,其中在步骤(b)中将所述金属组分加热至2000摄氏度。
23.根据权利要求1-18中任一项的抗微生物材料、感染控制产品、服装、家用产品或卫生产品,其可通过权利要求19-22中任一项的方法获得。
24.一种治疗伤口感染的方法,其包括将权利要求1至15中任一项的抗微生物材料施用于伤口。
